매자나무중 Berberine함량(含量)의 계절적(季節的) 변화(變化)에 관한 연구(硏究) (Studies on the Seasonal Variation of Berberine Contents in Berberis koreana)

  • The determination of berberine in Berberis koreana was examined by the high performance liquid chromatography using ${\mu}-Bondapak$ $C_{18}$ column and $CH_3CN/phosphate$ buffer (pH 5.2) (60/40) as a mobile phase. The seasonal variations of berberine contents in Berberis koreana were as follows; 1) In roots, the average berberine content was 0.94% with the highest level of 1.32% in October. 2) In stems, their average berberine content was about 0.1% and in March, April, October and November, the contents were relatively high. 3) In leaves, however, the content was as low as 0.005%. According to the experimental results obtained, we found that Berberis koreana roots can serve as the useful natural plant resources for the berberine and October is the optimal season for the collection.

매자나무성분이 마우스 수면 및 체온에 미치는 영향 (Effects of Berberis koreana Palibin on Sleep Duration and Rectal Temperature in Mouse)

  • Berberis koreana Palibin belonging to Berberidaceae family, a common herb in Korea, has been contained some quantity of Berberine analogue and other ingredients. Authors therefore paid attention to its pharmacological actions and examined the effects on sleep duration and rectal temperature in mouse with crystal (A) from Berberis koreana Palibin in Korean native plans. The experiment searching for the effect on sleep duration was performed with pretreatment of Berberis Koreana Palibin crystal (A) 30 min before the administration of 25 % ethanol, and its crystal were also administered intraperitoneally with the intention to examine the effect on rectal temperature in mouse. The results of the experiment were as follows; 1. Crystal (A) from Berberis koreana Palibin was made by extraction with ethanol and HCI. 2. Crystal (A) enhanced the hypnotic activity of alcohol in concentratins of 0.1 mg/10g or 0.15 mg/10g. 3. Rectal temperatures in mice were significantly reduced with administration of crystal (A) in concentrations of 0.1 mg/10g or 0.15 mg/10g. 4. The maximal reduction of rectal temperature and potentiation of the hypnotic activity were observed at 30 min after its administration. From the above results, it is clear that crystal (A) from Berberis koreana Palibin exerts the potentiation of hypnotic action of alcohol and reduction of rectal temperature in normal mouse. Its pharmacological effects are probably derived from the action upon the central nervous system.

매자나무뿌리 알콜 추출물의 항진균 작용 (Antifungal Activity of Alcohol Extract and Crystal(A) of Berberis Koreana Palibin)

  • The fungistatic effects of extract and crystal (A) of Berberis Koreans Palibin, a common shrub in Korea, were obeserved and compared with undecylenic acid and vegetable oil. In vitro studies, the spores of fungi were inoculated on Sabouraud's glucose agar media which contained compounds of various concentration, and the growth of the fungi was observed for 3 weeks. The species of the fungi used in these experiments were Epidermophyton floccosum, Microsporum gypseum, Microsporum audouini, Microsporum canis, Microsporum nanum, Microsporum cookei, Trichophyton rubrum, Trichophyton mentagrophytes, Trichophyton tonsurans and Trichophyton verrucosum. The results of the studies were as follows: 1. The growth of E. floccosum, M. audouni, M. canis, M. nanum, M. cookei, was inhibited in Sabouraud's glucose agar media containing extract and crystal (A) of Berberis Koreana Palibin 1 mg/ml.

매자나무 Ethyl acetate 분획물의 예쁜 꼬마선충 내의 항산화 효과 (Antioxidant Activity of Ethyl acetate Fraction of Berberis koreana Palibin in Caenorhabditis elegans)

  • Ethyl acetate (EA) soluble fraction of the Berberis amurensis (Berberidaceae) methanol extract showed the potent DPPH radical scavenging activity through Caenorhabditis elegans model system. The EA fraction was measured for the activity of superoxide dismutase (SOD), catalase, and oxidative stress tolerance by using C. elegans along with reactive oxygen species (ROS) level. In addition, SOD-3 expression was conducted using a transgenic strain (CF1553) to confirm that the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the EA fraction. As a result, the EA soluble fraction of B. amurensis increased SOD and catalase activity, and decreased ROS accumulation in a dose-dependent manner. Furthermore, the EA fraction-treated CF1553 worm showed higher SOD-3::GFP intensity than the control worm.

매자나무 세포독성성분 분석 (Analysis of Cytotoxic Constituent of Berberis koreana Palibin)

  • Methanol extracts of five Berberidaceae species were examined against tissue factor inhibitory and tumour cell growth inhibitory activity. Methanol extracts of Berberis koreana Palibin showed a strong cytotoxicity activity against SK-MEL-2 (Melanoma) tumour cell lines with more than 90% in $25{\mu}g/m\ell$ and against A549 (Lung carcinoma), SK-OV-3 (Ovarian cancer), XF498 (CNS cancer) and HCTl5 (Colon cancer), other Berberidaceae species except B. koreana species have no effect on the tumour cells. Biologically active compound, therefore, was isolated through the activity guided fractionation and purification. The structure was confirmed by NMR. FT-IR and MS to 2-(3,4-dihydroxybenzyl)-ethyl alcohol. It showed cytotoxicity activity against SNU-C4 tumour cell lines with 50.7% in $50{\mu}g/m\ell$. Methanol extracts of 5 Berberidacae species have no effect on the tissue factor inhibitory activity.

초고압 저온처리에 의한 매자나무의 면역 활성 (Effect of Immune Activity on Berberis koreana Palibin by Ultra High Pressure Low Temperature Process)

  • This study was performed to investigate a methodology of improving immune activities of Berberis koreana Palibin by ultra high pressure on low temperature extraction process. Extraction yield was enhanced up to more than 35% by above process, compare to the control and extraction average. The cytotoxicity on human kidney cell (HEK293) was showed below 20.4%, 21.6% in adding 1.0 mg/ml of the highest concentration. Generally, the extracts by ultra high pressure extraction process showed less toxicity about 5% than the other processes. It could tell that toxic materials that existing in the plant could be reduced or broken by ultra high pressure process due to can be broken bond such the hydrogen bond, the electrostatic bond, the van der Waals bond, and the hydrophobic bond, can be broken by high pressure. The result could be employed to develop a new type of functional food from B. koreana Palibin by low temperature high pressure process.

구상나무림(林)의 군락생태학적 연구 (Ecological Studies on the Vegetational Characteristics of the Abies koreana Forest)

  • 우리나라 특산 수종인 구상나무림(林)의 보존과 조림 및 임업경영에 필요한 정보를 체계화 시킬 목적으로 ZM방식을 통한 군락분류를 하였던 바 다음과 같이 요약할 수 있었다. 구상나무림(林)은 구상나무-신갈나무군락과 구상나무 제주조릿대군락으로 구분되었으며 구상나무 신갈나무군락은 미역줄나무군, 금마타리군 및 전형군으로 구분되었고, 미역줄나무군은 전형소군, 참나물소군 미 원추리 소군으로 구분되었다. 구상나무-제주조릿대군락은 매발톱나무군, 마삭줄군 및 전형군으로 구분되었으며 매발톱나무군은 전형소군과 노루귀소군으로, 마삭줄근은 노루귀소군과 전형소군으로 구분되는 등 총 10개의 식생단위로 구분되었다. 일치법으로 검토한 바 구상나무-신갈나무군락은 소백산맥에 위치하는 지리산, 덕유산, 가야산에서 출현하였고, 구상나무-제주조릿대군락은 한라산에서 출현하는 등 지리적으로 구분되었고, 군락단위간에는 해발과 지형이 복합적으로 영향을 미치는 경향이었다. 식생단위와 구상나무의 흉고직경 및 수고간에는 구상나무-신갈나무군락의 평균흉고직경과 평균수고가 각각 28.4cm, 10.6m로서 구상나무-제주조릿대군락의 평균흉고직경과 수고가 23.6cm, 6.3m인데 비하여 큰 편이었고 구상나무-신갈나무군락 중에서는 금마티리군이 평균흉고직경과 수고가 20cm, 5m로서 가장 작었으며, 구상나무-제주조릿대군락 중에서는 전형군의 평균흉고직경과 수고가 4cm와 3m로서 가장 작았다.

유산균 발효된 매자나무 추출물이 마우스 혈청중의 Melatonin 및 Serotonin의 함량에 미치는 영향 (The Effect of Extracts of Berberis koreana Bark by Lactobacillus Fermentation on the Concentration of Serotonin and Melatonin in the Serum of Treated ICR Mice)

  • This study was to investigate the effect of fermentation extracts on the concentration of serotonin and melatonin in the serum of the ICR mice. The ICR mice were divided into water control group, lactobacillus fermentation solution including (Lactobacillus paracasei and Bifidobacterium longum B6) control group, positive control group (milk and doxylamine succinate), negative control group (caffein) and the groups treated with the extracts of Berberis koreana bark (WE: water extracts, FE-L.P: fermentation extracts of Lactobacillus paracasei, FE-B.L: fermentation extracts of Bifidobacterium longum B6). After ten-day feeding treatment, the mean concentration of serotonin for water control, WE, FE-L.P and FEB. L group was 134.72, 183.01, 232.09 and $223.78 ng/m{\ell}$, respectively. The mean concentration for FE-L.P and FE-B.L group were approximately 66% larger than that for water control group. The mean concentration of melatonin for water control, WE, FE-L.P and FE-B.L group was 76.92, 106.66, 157.56 and $141.81pg/m{\ell}$, respectively. The mean concentration of melatonin for FE-L.P and FE-B.L group were also larger than that for water control group. Our results indicated that the fermentation extracts of Berberis koreana bark have relatively greater potential to induce secretion of serotonin and melatonin. Therefore, the fermentation extracts have antidepressant effect.

초고압 및 초음파 추출공정을 이용한 매자나무 수피의 향장활성 증진 (Enhancement of Cosmeceutical Activities of Berberis koreana Bark by High Pressure and Ultrasonification Extraction Processes)

  • This study was performed to investigate the enhancement of cosmeceutical activities of Berberis koreana bark by different extraction processes. The extracts are WE (water extract at $100^{\circ}C$, control), USE (ultrasonification for 1 hours at $60^{\circ}C$ with water), HPE (high pressure for 5 minutes at $60^{\circ}C$ with water) and USE + HPE (ultrasonification process for 1 hours after high pressure for 5 minutes at $60^{\circ}C$ with water), respectively. The cytotoxicity of the extracts was in the range of 24.02~26.94% at 1.0 mg/ml concentration. The USE + HPE showed the lowest cytotoxicity. Compared to the WE, total phenolic and flavonoid contents in the USE + HPE increased to 121.5% and 154.2%. The USE + HPE showed the highest activity at 1.0 mg/ml concentration in 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging, inhibition activity of xanthine oxidase and superoxide dismutase (SOD)-like test, respectively. Tyrosinase inhibition of WE, USE, HPE and USE + HPE at 1.0 mg/ml concentration was measured as 17.72, 19.62, 22.83 and 24.16%, respectively. Hyaluronidase inhibition activities of the USE + HPE were higher than 20.8%~29.5% of the WE. Our results suggested that the extracts from ultrasonification process after high pressure extraction has relatively high cosmeceutical activities, and that the bark of Berberis koreana could be considered as a candidate of new functional cosmetic agents.
