Effect of electron donor for reductive dechlorination of PCE using biobarrier (Biobarrier를 이용한 PCE의 환원적 탈염소화시 전자공여체의 영향)

  • The applicability of in situ microbial filter or biobarrier technology for the remediation of soil and groundwater contaminated with chlorinated solvents was investigated. The efficiency and rates of reductive dechlorination of chlorinated solvents are known to be highly dependent on hydrogen concentration. In this study, the effect of electron donors on the reductive dechlorination of PCE was investigated using vermicompost (worm casting) and peat as permeable reactive barrier medium The effect of organic acids (lactate, butyrate and benzoate), yeast extract and vitamin $B_{l2}$ on the reductive dechlorination was investigated. Compared to the control (no electron donor added), addition of electron donors stimulated the dechlorinated rate. Among the electron donor treatments, lactate/benzoate amendment exhibited the highest dechlorination rate. Since vermicompost and peat are inexpensive and biodegradable and have high sorption capacity, they could be successfully used as biobarrier media, especially when electron donors (for example, lactate/benzoate) are added.d.

  • From 120 soil and activated sludge, the strains able to grow on benzoate and m-Toluate have been isolated after selective enrichment which were later identified as Psudomonas sp. according to its morphological and biochemical characteristics. Ben-2 strain contained two plasmid DNA having about 120 Kb and below 2.0 Kb by agarose gel electrophoresis. Form the comparative investigation of catechol 1,2-oxygenase and catechol 2,3-oxygenase activities in Ben-2 strain and its cured strain, Ben-2 strain has both of these two enzymes while cured strain has catechol 1,2-oxygenase only.

  • Catabolic pathways for the degradation of various aromatics by Sphingomonas yanoikuyae Bl are intertwined, joining at the level of substituted benzoates, which are further degraded vita ring cleavage reactions. The mutant strain EK497, which was constructed by deleting a large DNA region containing most of the genes for biphenyl, naphthalene, m-xylene, and m-toluate degradation, was unable to grow on all of the aromatics tested except for benzoate as the sole source of carbon and energy.S. yanoikuyae EK497 was found to possess only catechol ortho-ring cleavage activity due to deletion of the genes for the meta-cleavage pathway. Wild-type S. yanoikuyae Bl grown on benzoate has both catechol orthoand meta-cleavage activity. However, m-xylene and m-toluate, which are metabolized through methylbenzoate, and biphenyl, which is metabolized through benzoate, induce only the meta-cleavage pathway, suggesting the presence of a substrate-dependent induction mechanism.

  • A strain able to grow up m-toluate minimal medium has been isolated after selective enrichment and given the name T81X, which was later identified as pseudomonase putida according to its morphological and biochemical characteristics. After treatment with plasmied specific curing agent, mitomycin C, followed by replica plating on m-toluate and xylene minimal agar plate, T81Xstrain has been shown to harbour a curable plasmid relating to the m-toluate and xylene metabolism. Spontaneous curing frquency of this plasmid was also greatly enhanced by growing on benzoate minimal medium. After then, it was also xylene metabogrowing on benzoate minimal medium. After then, it was found to be conjugally nontransmissible. From the comparative investigation of catechol 1,2-oxygenase and catechol 2,3-oxygenase activities in wild type and cured strain on various growth substrate, it appeared that T81X strain has both of these two enzymes while cured strain has catechol 1,2-oxygenase only. Growing on m-toluate minimal medium T81X strain should carry the genetic information necessary for coding the catechol 1,2-oxygense induced by m-toluate or benzoate, on that curable plasmid.

  • We synthesized chiral choleseric liquid crystalline monomer p-phenylene bis-(p-3-acryloxy-2(S)-methylpropoxy)benzoate (1) and achiral nematic liquid crystalline monomer, p-phenylene p-acryloxy-6-hexyloxy-p-octyloxybenzoate (2) for the construction of cholesteric polymer network with a pitch gradient. In this research we attempted to synthesize cholesteric polymer film which have a wide reflection bandwidth in the region of visible wavelength with the synthesized chiral cholesteric monomer 1 and nematic monomer 2 under various photopoly merization conditions. We succeeded to construct cholesteric polymer film network with a pitch gradient covering more than 200 nm bandwidth in the visible wavelength by diffusion controlled photostructuring method.

  • The applicability of biobarrier or in situ microbial filter technology for the remediation of groundwater contaminated with chlorinated solvent was investigated through batch microcosm study. The efficiency and rates of reductive dechlorination of tetrachloroethylene (PCE) are known to be highly dependent on hydrogen concentration. In this study, the effect of electron donors on the reductive dechlorination of PCE was investigated using vermicompost (or worm casting) and peat as a biobarrier medium. The effect of organic acids (lactate, butyrate and benzoate), yeast extract and vitamin $B_{12}$ on the reductive dechlorination was investigated. In the absence of biobarrier medium (adsorbent), addition of electron donors stimulated the dechlorination rate of PCE compared to the control experiment (i.e., no electron donor added). Among the treatments, addition of lactate or lactate/benzoate as hydrogen donor exhibited the highest dechlorination rate ($k_1=0.0260{\sim}0.0266\;day^{-1}$). In case of using vermicompost as a biobarrier medium, amendment of lactate/benzoate exhibited the highest dechlorination rate following with a pseudo-first-order degradation rate constant of $k_1=0.0849\;day^{-1}$. In contrast, when Pahokee peat was used as a biobarrier medium, either butyrate or lactate addition exhibited the highest dechlorination rate with $k_1$ values of 0.1092 and $0.1067\;day^{-1}$, respectively. The results of this study showed the potential applicability of in situ biobarrier technology using vermicompost or peat as a barrier material for the remediation of groundwater contaminated with chlorinated solvent.

  • An experiment was performed to investigate the combined effect of preservatives and the synergistic effect of sodium chloride to them on the inhibition of bacterial growth. Escherichia coli and Salmonella typhimurium were cultured with or without shaking in liquid media (pH 6) of tryptone-glucose-yeast extract or tryptic soy broth which contained 0.1% potassium sorbate and/or 0.03% sodium benzoate, equivalent to half of the maximum permissible levels, respectively. The growth of E. coli was more inhibited with one or both of the two preservatives by shaking culture than by non-shaking culture. For S. typhimurium the single treatment of the preservatives did not show inhibitory effect whereas the combined treatment of them showed bacteriostatic effect in shaking culture and a prolongation of lag phase in non-shaking culture. Addition of 2% sodium chloride to either potassium sorbate or potassium sorbate plus sodium benzoate remarkably increased the growth inhibition of E. coli for non-shaking cultivation but no effect observed for shaking cultivation. S. typhimurium was more sensitive to the addition of sodium chloride than E. coli in both shaking and non-shaking culture to show lower viable cell counts than initial numbers.

  • 2-Ethoxy-4-acetamidobenzoate (III) was synthesized by ethylation of N-formyl compound obtained with formylation of methyl 4-aminosalicylate and 2-ethoxy-4-acetamidobenzamide (VIII) was synthesized by ethylation of 4-acetamidosalicylamide (VII) formed by reacting methyl 4-acetamido-2-acetoxy benzoate with concentrated ammonia water under pressure. 2-Ethoxy-4-acetamido benzamide (VIII) was also synthesized by acetylation of 2-ethoxy-4-aminobenzamide (VI) obtained by pressure eraction of methyl 2-ethoxy-4-formylamido-benzoate (I) with concentrated ammonia water.

  • Eighty two bacterial strains have been isolated from five different soil and sewage samples by selective enrichment culture on m-toluate minimal medium. Two of these were identified as Pseudomonas capacia, one as P. putida, one as Yersinia intermedia, and one as Flavobaeterium odoratum. P. cepacia SUB37 appeared to carry plasmid superficially similar to TOL plasmid previously described in p. putida mt-2 and other two plasmids from Flavobacterium odorutum and Y. intermedia larger than that of p. putida mt-2. p. cepacia SUB37 was sensitive to streptomycin but resistant to rifampicin. P. cepacia SUB37 carrying plasmid metabolizes the hydrocarbons to benzoate and toluates via the corresponding alcohols and aldehydes. By the curing experiment, it appears that P. cepacia SUB37 carries TOL plasmid encoding for the enzymes responsible for the catabolism of toluene and xylene via benzoate and the toluates and then by meta pathway in the process of degradation of aromatic hydrocarbons. p. cepacia SUB37 degraded m-toluate rapidly to be very low level when it was fully grown.
