The Effect of Mugwort Extracts on the Benzo(a)pyrene-induced Hepatotoxicity in Rats (Benzo(a)pyrene에 의해 유도된 간기능 장해에 미치는 쑥의 효과)

  • Mugwort has been used as a Korean folk medicine in treating liver diseases acting as an analgesics, sedative, diuresis, choleretics. This study was perfomed to evaluate the effect of mugwort extracts on the changes of enzyme activities, lipid accumulation of the serum and liver, when hepatotoxicity was induced by benzo(a)pyrene. The results are as follows: 1. Mugwort water extract administration prevented the increase of serum and liver AST, ALT, LDH, ${\gamma}$-GTP, liver ALP activities and bilirubin content caused by B(a)P injection. 2. The increase of serum and liver ALT, LDH, ${\gamma}$-GTP, serum AST activities and liver bilirubin contents in B(a)P treated group were decreased by mugwort methanol extract treatment. 3. Serum and liver total cholesterol, phospholipid, triglyceride level and serum HDL-cholesterol level were increased by B(a)P treatment. After combined treatment of mugwort water and methanul extracts, these lipid content were significantly decreased. 4. The hepatotropic effect of mugwort water extract and after-treatment against B(a)P induced hepatotoxicity was superior to that of methanol extract and pretreatment.

  • A modified method was investigated for the determination of benzo(a)pyrene metabolites generated by the rat liver microsomes based on the HPLC technique with radioactive flow detection. By adding $[^3H]-dexamethasone$ to the B(a)P metabolites mixture metabolized by the microsome, the poor yield of solvent extaction of B(a)P metabolites was compensated.

Effect of Benzo[a]pyrene on Genes Related to the Cell Cycle and Cytochrome P450 of Saccharomyces cerevisiae

  • Benzo[a]pyrene (B[a]P) is an environmental pollutant that has been implicated in carcinogenesis. Saccharomyces cerevisiae was treated with B[a]P, and the responses of its cytochrome P450 (CYP) enzyme and DNA-damage checkpoint genes were examined through gene expression profiles using a reverse transcription polymerase chain reaction (RT-PCR). The DNA-damage checkpoint genes tested were the chk1 and pds1 genes, involved in a metaphase arrest, the swi6 gene targeted by G1 arrest, the pol2 gene related to S phase arrest, and the cln2 gene encoding a cyclin protein, all of which are based on rad9 and rad24. Among these genes, no noticeable effect was found when the cells were exposed to various concentrations of B[a]P. However, the transcriptional activity of CYP51 was significantly different when the cells were exposed to B[a]P. Accordingly, the present results indicate that cytochrome P450 plays a more significant role than DNA-damage checkpoint genes in the response of S. cerevisiae to B[a]P.

Benzo(a)pyrene Reduction in Sesame Oil Using Microwaving Method (Microwaving을 이용한 참기름의 Benzo(a)pyrene 저감화)

  • Sesame oil has superior oxidation stability and unique roasting flavor. Accordingly, this has been used for edible oil as well as a seasoning material for a long time in Korea. But sesame oil is a simple pressed oil, unrefined. During manufacturing process of roasting-expression, benzo(a)pyrene[B(a)P] formed as a strong carcinogenic substance causes a social problem. Detection of B(a)P in sesame oil was due to residual content in raw-sesame seeds and formation in roasting-expressing process. Especially, maximal forming process was roasting. Accordingly, in this study applied the traditional roasting method by roaster and microwaving method as a new type. Best roasting time by microwaving was for 5~10 min, B(a)P content in sesame oil was 0.53~0.79 ${\mu}g/kg$. These B(a)P contents showed 1/2 level than direct roasting method by roaster. As a result, B(a)P contents in sesame oil appeared the difference of more than 2 times according to roasting condition of sesame seed. For minimizing of B(a)P content in sesame oil is demanded roasting of sesame by microwaving than direct roasting by roaster.

Effect of Ginseng Extracts on the Binding to DNA of Benzo(a)pyrene Metabolites in uitro in Rats (DNA와 Benzo(a)pyrene 대사물질 결합형성에 미치는 인삼 추출물의 영향)

  • Reactive metabolites generated by benzo(a)pyrene(BP) monooxygenase(AHH) interact with nucleophiles in DNA and cause mutation and carcinogenesis. We studied the effect of Panax ginseng C.A. Meyer, which induce epoxide hydratase(EH) activity without concomitant induction of AHH activity, on the binding of BP metabolites to DNA in uitro in Sprague Dawley rats. DNA-BP metabolite adducts can be resolved into at least five distinct peaks by elution of a Sephadex LH-20 column with a water methanol gradieNt. These peaks are arbitrarily designated A(most polar) through I(least polar). Of the 5 peaks tentatively assigned to 7,8 biol-9,10-oxide(A),7,8·oxide(B),4,5-oxide(C), and further metabolites of 9-OH-BP(D & E), peaks A, C, D, and I were reduced to 70, 85, 80, and 30% of controls, respectively, and there was no significant change in peak B. In connection with this DNA binding study, BP metabolizing enzymes including AHH, EH, demethylase(DM) activity and cyt. P-450 contents were also investigated in order to compare the BP treated control with ginseng and BP treated test groups. The results showed that the EH activity was increased by 139% over the BP control, the Cyt. P-450 content was increased by 180% over the control value, and DM and AHH activities were also increased to some degree for the BP test group, but there was no significant effect of the ginseng treatment.

Antigenotoxic Effects of Synurus deltoides Extract on Benzo[a]pyrene Induced Mutagenesis

  • This study investigated the antigenotoxic effects of Synurus deltoides extract on the mutagenesis induced by benzo[a]pyene(B[a]P). About 80% and 90% antimutagenic effects were observed in the presence of over 200$\mu\textrm{g}$/plate of methanol extract of Synurus deltoides against Salmonella typhimurium TA98 and TA100 induced by B[a]P, respectively. The methanol extract itself did not induce an increased frequency of micronucleated polychromatic erythrocytes(MNPCE) irrespective of the sampling time(up to 72h), while the treatment with benzo[a]pyrene(B[a]P) at 150mg/kg significantly increased (p<0.05) the incidence of MNPCE. The strongest relative frequency of MNPCE was observed at 36h after injection of B[a]P and the most significant reduction (p<0.05) in the frequencies of MNPCE was occurred at the feeding of the methanol extract to mice 12 h before injection of B[a]P. The most significant reduction (p<0.05) with 48% was observed in the frequencies of MNPCE when 50 mg/kg of the methanol extract was given to the mice 12h before injection of B[a]P, while the strongest relative frequency inhibition was 54% at the multiple feeding of 5mg/kg of the methanol extract e time every day for 5 days on th frequencies of MNPCE induced by 150 mg/kg of B[a]P. These results indicate that the methanol extract of Synurus deltoides have a strong modulatory effect on benzo[a]pyrene induced MNPCE.

  • Benzo(a)pyrene(B(a)P), an extensively studied polycyclic aromatic hydrocarbon(PAH), is a common contaminant produced through the burning of fossil fuels, particularly coal, and from the exhaust products of internal combustion engines. It produces a wide range of toxicities, including carcinogenicity in experimental animals. B(a)P has been shown to suppress systemic immunity in experimental animals, which may contribute to the growth of the chemical-induced tumors. Using colorimetric MTT assay natural killer(NK) cell-mediated growth inhibition of tomor cell was measured in normal and B(a)P-exposed C57BL/6 mice. Non-adherent splenocytes of normal or B(a)P-exposed mice were cultured with Yac-1 cells at four different effector/target(E/T) cell ratios ranging from 200/1, 100/1, 50/1, and 25/1 in an assay volume of 0.1 ml. After the optical density of culture wells containing MTT solution was measured at a wavelength of 540 nm, the percentage of dead cells relative to the control target cell number was calculated. The NK activity of B(a)P-exposed mice was markedly lower than that of non-exposed mice group at all E/T ratios. These results indicated that suppression of NK cell activity may play a role in allowing for the growth of tumors.

Effect of Puerariae Radix Methanol Extract on Benzo(a)pyrenc -in - duced Hepatotoxicity in Rats (갈근 메탄올 엑기스가 흰쥐에 있어서 Benzo(a)pyrene에 의해 유도된 간장해에 미치 는 영향)

  • The present study was conducted to evaluate the hepatoprotective effect of puerariae Radix methanol extract on benzo(a) pyrene(B(a)P) - induced liver injuries in rats. In vitro experiment, primary cultured hepatocytes (5X105 cells/$m\ell$) were cultured for 20~24 hours after adding puerariae Radix mehtanol extract(32$\mu\textrm{g}$/$m\ell$) and B(a)P(50 uM). In vivo experiment, Puerariae Radix methanol extract(0.25 g/kg/day, per os) was administered for 7 days and B(a)P(0.1 mg/kg/day, intraperitoneally) was given after the last administration of extract. And then the hepatoprotective effect of Puerariae Radix methanol extract was investigated biochemically through in vitro and in vivo experiments. Namely, activities of enzymes (GOT, GPT and LDH) were measured and 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide(MTT) assay were carried out in vitro cell culture study and GOT, GPT, LDH and ALP activities and HDL-cholesterol, total cholesterol and triglyceride contents were performed in vivo study. In vitro experiment, as a result of enzyme activity measurement(GOT, GPT and LDH) and MTT assay, GOT,GPT and LDH activities changed by B(a)P were recovered to normal levels and hepatocytes impaired by B(a)P were recovered to normal. In vivo experiment, Puerariae Radix methanol extract significantly decreased the enzyme activities(GOT, GPT, ALP and LDH in serum and GPT and ALP in tissue) and lipid contents in comparison to B(a)P-treated group.

Assessment of In Vitro Oocyte Maturation in Two Gobiid Fish Species, Chasmichthys dolichognathus and Tridentiger trigonocephalus after Exposure to Benzo[a]pyrene

  • Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous environmental contaminants derived from incomplete combustion of carbons and crude oil. In this study, we investigated the effects of benzo[a]pyrene (B[a]P), a representative PAHs on in vitro sex steroid hormone production and germinal vesicle breakdown (GVBD) using isolated oocytes of longchin goby (Chasmichthys dolichognathus) and chameleon goby (Tridentiger trigonocephalus). Oocytes in diameters of 0.8-0.9 (end vitellogenic stage) and 0.9-1.0 mm (germinal vesicle migratory stage) from longchin goby and 0.5 mm (fully vitellogenic stage) from chameleon goby were used. In GVBD assay, B[a]P at 10 nM stimulated GVBD in the oocytes of 0.8-0.9 mm from longchin goby. B[a]P at 1 nM stimulated GVBD in the oocytes with diameter 0.5 mm from chameleon goby. In steroid production from oocytes of longchin goby, B[a]P at 100 nM decreased testosterone (T) production, B[a]P at 1,000 nM increased estraiol-17 (J (E2) production and 10 and 100 nM increased $17,20{\beta}$-dihydroxy-4-pregnen-3-one ($17{\alpha}20{\beta}P$) production in the oocytes with diameter 0.8-0.9 mm. B[a]P at 1,000 nM increased E2 production, 100 and 1,000 nM increased $17{\alpha}20{\beta}P$ production in the oocytes with diameter 0.9-1.0 mm. In steroid production of oocytes from chameleon goby, B[a]P at 1,000 nM increased $E_2$ production. B[a]P at 10 nM increased $17{\alpha}20{\beta}P$ production. In the ratio of $E_2$ to T ($E_2$/T), B[a]P at 100 and 1,000 nM increased $E_2$/T in the oocytes of longchin goby. B[a]P at 100 nM also increased $E_2$/T in the oocytes of chameleon goby. Taken together, these results suggest that B[a]P have not only weak estrogenic effects but progestogenic effects on oocyte maturation.

Improving Method of Pre-treatment for Detection Benzo($a$)pyrene Contents in Concentrates (농축물의 Benzo($a$)pyrene 함량 측정을 위한 전처리 방법의 개선)

  • I assessed the B($a$)P content from the Ginseng extract, Red ginseng extract, and Ssangwha extract which have high viscosities. It wasn't easy to extract oil from these samples, consequently measuring of B($a$)P was difficult. In order to know the exact detecting contents, I injected standard material of B($a$)P to the above extractions and pre-treated for measurement but it was also difficult to measure of contents exactly. To improve of detecting method, I removed mucinous materials using a 85% phosphoric acid solution or 10% citric acid solution and then processed continuously with $60^{\circ}C$ hot water. The analysis revealed that extracting the samples contained B($a$)P determined the rate of each 70%, 55%, 67% could increase. As a result the detecting method of B($a$)P contents could be improved.
