Cytochrome P4501B1(CYP1B1) is known to be inducible by xenobiotic compounds such as policyclic aromatic hydrocarbon(PAH) and dioxins such as 2,3,7,8-tetrachloro-dibenzo-p-dioxin(TCDD). And these induction of CYP1B1 is also regulated by many categories of chemicals. In order to investigate the effects of several chemicals on CYP1B1 gene expression in Hepa-I and MCF-7 cells, 5' flanking DNA of human CYP1B1 was cloned into pGL3 basic vector containing luciferase gene, and then transfected into these cells. After treatment of chemicals, the luciferase activity was measured. CYP1B1 enzyme metabolize PAHs and estradiol. CYP1B1 metabolize estradiol to 4-hydrozyestradiol that is considered as carcinogenic metabolite. Luciferase activity was induced about 20 folds over that control by 1 nM TCDD (2,3,7,8-tetrachloto-p-dioxin). Recent industrialized society, human has been widely been exposed to widespread environmental contaminants such as PAHs(polycyclic aromatic hydrocarbon) that are originated from the imcomplete combustion of hydrocarbons. PAHs are known to be ligands of the AhR(aryl hydrocarbon receptor). Induction of cytochrome P4501B1(CYP1B1) in cell culture is widely used as a biomarker for PAHs. Therefore we have studied the effect of PAHs in the human breast cancer cells MCF-7 to evaluate bioactivity of PAHs. We have used the United State of America EPA selected 13 different PAHs, PAHs mixtures and extracts from environmental samples to evaluate the bioassay system. We examined effects of PAHs on the CYP1B1-luciferase reporter gene and CYP1B1 mRNA level. Benzo(k)fluoranthene and dibenzo(a, h)anthracene showed strong response to CYP1B1 promoter activity stimulation, and also CYP1B1 mRNAs increase in MCF-7 cells in a concentration-dependent manner. Acenaphthene, anthracene, benzo(b)fluoranthene, fluorene, fluoranthene, anphthanlene, pyrene, phenanthrene and carbazole were weak responders in MCF-7 cells. RT-PCR analysis indicated that PAHs significantly up-regulate the level of CYP1B1 mRNA.
This study was performed to determine the effects of antimutagenicity of Porturaca oleracea L. in Korea. In Ames test, the ethanol extract of Poturaca oleracea L. inhibited mutagenic activity of N-methyl-N'-nitro-N-nitrosoguanidine(MNNG), , 4-nitroquinoline-1-oxide(4NQO), benzo($\alpha$)pyrene (B($\alpha$)P) and 3-amino-1,4-dimethyl-5H-pyrido-(4,3-b)indole (Trp-P-1) in Salmonella typhimurium TA98 and TA100. But hot-water extract Poturaca oleracea L. only Inhibited mutagenic activity of MNNG in Salmonella typhimurium TA100, On 4NQO, the ethanol extract 100-1,600$\mu\textrm{g}$/plate of Porturaca oleracea L. showed a slight inhibitory effect of 13-48%, 4-47% in TA98 and TA100, respectively, but on MNNG, it showed higher inhibitory effect of 6-86% in TA100, And the treatment of 1,600$\mu\textrm{g}$/plate of ethanol extract of Porturacea L. had strong antimutagenicity with 74-87% inhibition against TA98 and TA100 induced by B(a)P and with 85-93% inhibition against TA98 and TA100 induced by Trp-P-1. The ethanol extract was fractionated with ether. chloroform, ethylacetate, butanol and water. Among them, most of the fraction except water fraction showed strong antimutagenicity effects against mutation induced by 4-NQO, MNNG, B(a)P and Trp-P-1. Chloroform fraction had strong antimutagenicity with 91% inhibition against TA100 induced by MNNG, diethyl etherfraction had strong antimutagenicity with 92%, 98% inhibition against TA98 and TA100 induced by 4NQO, Chloroform fraction had strong antimutagenicity with 97% inhibition against TA100 induced by B (a)P and diethyl etherfraction had strong antimutagenicity with 98% inhibition against both strain Induced by Trp-P-1, respectively.

Pine has been known as a traditional medicinal plant and as showing a physically beneficial function to a human being. Therefore, this study was performed to investigate the physiological activities of main pine neddles. Ethanol extracts from pinus needles did net exhibit any mutagenicity. On the contrary, inhibitory effects of ethanol extract were observed on mutagenicity induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG), 4-nitroquinoline-1-oxide (4NQO), 3-amino-1,4-dimethyl-5H-pyrido-(4,3-b)indol (Trp-P-1) and benzo(a)pyrene $(B({\alpha})P)$ using Salmonella typhimurium reversion assay. On direct-acting mutagen (MNNG, 4NQO) and indirect-acting mutagen (Trp-P-1, $(B({\alpha})P)$, we observed higher inhibitory effect. Stepwise fractionation of the ethanol extract was done by using ether, chloroform, ethyl acetate, butanol and water to obtain effective fraction. Among them, water fractions $(100\;{\mu}g/plate)$ of Pinus thunbergii, Pinus rigida, Pinus densiflora and Pinus koraiensis showed high inhibition of 91.65%, 94.7%, 84.22% and 79.02%, respectively, on the mutagenicity of MNNG in Salmonella typhimurium TA100.

Antimutagenic effect of Doenjang (Korean soy paste) on various carcinogens in Salmonella typhimurium strains of TA98 and TA100 were studied. By the addition of methanol extract of Doenjang to aflatoxin B₁(AFB₁)in the experimental system, the mutagenicity of AFB₁ on the strains of TA98 and TA100 was com-pletely inhibited. The methenol extract of the Doenjang also inhibited the mutagencities induced by direct mutagens such as N-methy1-N'-nitro-N-nitroguanidine(MNNG)and 4-nitroquinoline-1-oxide(4-NQO), and another indirect mutagens of benzo(a) pyrene(BaP) and dimethylnitrosamine(DMN). From the solvents and thin layer chromatographic(TLC) fractionations, free fatty acid(s), especially linoleic acid in Doenjang seemed to be one of the active antimutagenic compounds.
Tea is one of the most frequently consumed drinks due to its favourite taste and the health benefit. Tea is produced by several processes and drying is very important step to develop the flavour and destroys the enzymes in tea. However, during drying tea, polycyclic aromatic hydrocarbons some of which are carcinogen and genotoxin are naturally produced. The risk of PAHs by drinking tea was characterized by determining contents of 4 PAHs in tea. 4 PAHs including Benz(a)anthracene (BaA), Chrysene (CHR), Benzo(b)fluoranthene (BbF) and Benzo(a)pyrene (BaP) were investigated by GC-MS in total 468 tea products, which were contaminated up to $4.63ng\;g^{-1}$. Mate tea was the most highly contaminated by BaA, CHR, BbF and BaP and followed by Solomon's seal and Chrysanthemum. The Margin of Exposures calculated by the concentration of BaA, CHR, BbF and BaP and consumption amount of tea were higher than 10,000, and the risk of PAHs in tea were low concern to public health.

The balance between metabolic activation of xenobiotics and detoxification of their active metabolites may playa vital role in controlling mutagenic and carcinogenic processes. To assess the possible role of P. ginseng C.A. Meyer in detoxification of xenobiotics, we studied the effects of ginseng on several parameters of the monooxygenasd system, including benzo(a) pyrene monooxygenase(AHH) and benzo(a) pyrene epoxide hydratase(EH) as well as effects of ginseng on the conjugation system. Test animals receiving ginseng saponin-fraction induced epoxide hydratase activity to over $150\%$ (20mg/kg b.w.) of the control and increased glutathione transferase activity (GSH-T) up to $140\%$ (20mg/kg b.w.) of the control, whereas no significant changes were observed in the benzopyrene monooxygenase activity (AHH). Such a selective induction of the inactivation enzyme epoxide hydratase, combined with a marked elevation of the detoxifying enzyme glutathione transferase, without a concurrent induction of benzopyrene monooxygenase which is responsible for the formation of carcinogenic intermediates, demonstrates that ginseng has the potential to alter the metabolic course of carcinogenic polycyclic aromatic hydrocarbons, and thereby enhance detoxification. Thus, ginseng may play an important role in the prevention of tumors caused by carcinogens.

The analysis of the change in the herbal tea composition according to the difference in processing conditions result. This study was. investigated the antigenotoxic effects of Aster scaber Thunb root extract on the mutagenesis induced by benzo($\alpha$)pyrene(B($\alpha$)P) The treatment with B($\alpha$ )P at 150 mg/kg significantly Increased the incidence of MNPCE(p<0.05) The amount of 50, 100, 150 and 200 mg/kg of ethanol extract from Aster scaber were administered to animals immediately after injection of B($\alpha$)P. Significant reductions(p<0.05) with 24.5, 22.6, 59.8 and 79.4%. respectively, ware observed in the frequencies of MNPCE compared to positive control. When the fractions of hexane. chloroform, ethyl acetate. butanol and water from ethanol extract were treated with concentration of 10 mg/kg, the suppression rates of the MNPCE were 3.9, 35.3, 40.2 11.8 and 49.0%, respectively. And also, the strong suppression rate of the MNPCE treated with above five fractions in the concentration of 80 mg/kg showed 78.4, 65.7, 75.5, 68.6 and 77.5%, respectively compared to positive control. These results indicate that the five fractions in the concentration of 80 mg/kg from Aster scaber ethanol extract have a strong modulatory effect on B($\alpha$ )P induced the MNPCE.
The mutagenicities and antimutagenicities of butanol (PL I) and water (PL II) extracts from the filtrate of the cultured broth of Phellinus linteus were examined using the Ames/Salmonella test. No mutagenic activity of PL I and PL II was found in Salmonella typhimurium strains TA98 and TA100, either with or without S9 activation. In contrast, PL I and PL II showed inhibitory effects on the mutagenic activities induced by the directly-acting mutagens, 4-nitro-o-phenylenediamine (NPD) using the tester strain TA98 and sodium azide (NaN3) using the tester strain TA 100 in the absence of S9 mix. PLI and PL II also showed inhibitory effects on the mutagenicities of the indirectly-acting mutagens, 2-aminofluorene (2-AF) using the tester strain TA98 and benzo[a]pyrene (B[a]P) using the tester strain TA 100 in the presence of S9. These results suggest that P. linteus has an antimutagenic activity and may play a role in the prevention of cancer.

The effect of Herricium erinaceus on the mutagenicity in salmonella assay and quinone reductase activity in hapalclc7 cells were studied. Antimutagenic as evaluated by Ames test, the extract and fractions of JHerricium erinaceus had no effects on the mutagenicity by themselves. However, methanol extract and fractions from Hericium erinaceus showed strong inhibitory effect on the mutagenesis induced by N-methyl -N'-nitor-N- nitroso-guanidine (MNNG) and benzo(a)pyrene (B(a)P). Among the solvent fractions of methanol extract, the hexane fraction, the chloroform fraction and the ethylacetate fraction exhibited stronger inhibitory activity against MNNG and B(a)P induced mutagenesis than butanol and water fractions. The methanol extract, the extract, the chloroform and the ethylacetate fractions of Hericium erinaceus induced the activity of quinone reductase, an anticarcinogenic marker enzyme, in murine hepalclc7 cells while the others did have little effect on the enzyme activity.
