Role of Selenium in Alteration of Erythrocyte Parameters in Bovine Fluorosis

  • Signs of dental discolouration, difficulty in mastication, bony exostosis and debility were observed in cattle from Qingtongxia Ningxia, China where fluoride concentration in drinking water, soil, fodder, serum, bone, teeth, haircoat and urine were significantly higher than the corresponding health site. The problem of fluorosis in beef cattle is attributable to water containing toxic levels of fluoride. The objective of this paper was therefore to evaluate the influence of fluoride on erythrocyte parameters in cattle under high fluoride and low selenium conditions, as well as the protective efficacy of selenium exposure in feedstuff for bovine endemic fluorosis. Sixteen 6 to 7 year-old high fluoride beef cattle were randomly allotted into two groups each with eight cows: high fluoride control group, and supplemented with 0.25 mg/kg selenium per day for 83 days respectively. In addition, eight 6 to 7 year-old normal control beef cattle were selected from a non-high fluoride site. Blood samples were collected on day 0, 30 and 83 for erythrocyte parameters analysis and scanning electronic microscopy. The results indicated that erythrocytes, hemoglobin, packed cell volume values and $Na^+-K^+$ ATPase activity from affected cattle on the high fluoride site were significantly reduced during the period as compared with the corresponding samples of normal control cattle, a great number of echinocytes were present in peripheral blood, and subsequent anaemia. However, affected cattle exposed to selenium revealed increasable erythrocyte parameters, the extent of elevation in these values being dependent on the duration of supplementation with selenium. These findings suggest that fluoride exposure can cause erythrocyte damage, whereas selenium supplementation can antagonize fluoride-induced generation of free radicals and cumulative effects of lipid peroxidation in erythrocytes. Selenium supplementation may help to alleviate the possible hazards associated with bovine endemic fluorosis.

감마선조사 쇠고기의 유전독성 및 급성독성학적 안전성평가 (Genotoxicological and Acute Toxicological Safeties of Gamma Irradiated Beef)

  • 우육의 위생화를 위한 방사선 조사기술의 이용 가능성을 검토할 목적으로 방사선 조사 쇠고기(5 kGy)를 대상으로 유전독성 및 급성독성 시험을 통한 안전성 평가를 실시하였다. 감마선 조사 및 비조사 쇠고기 현탁액의 S. typhimurium TA98, TA100, TA1535 및 TA1537에 대한 복귀변이 집락수를 조사한 결과, 대사활성계 도입 및 부재시 모두, 모든 시험균주에서 시험 적용 농도인 $0.1{\sim}8.3{\;}mg/plate$의 범위에서 복귀변이 집락수의 농도 의존적인 증가 혹은 감소를 보이지 않아 감마선 조사 쇠고기(5 kGy)는 돌연변이원성이 없는 것으로 판단되었다. 또한, 설치류 망상적혈구를 이용하여 감마선 조사된 쇠고기의 염색체 이상 시험을 수행한 결과, 감마선 조사 쇠고기는 시험적용 용량인 $1250{\sim}2500{\;}mg/plate$1의 범위에서 소핵을 가진 망상적혈구의 출현율이 음성대조군과 유의한 차이를 나타내지 않아 소핵을 유발하지 않음을 확인하였다. 조사 우육의 급성독성시험 시험결과, 전 기간을 통하여 마우스 암수 모두에서 조사우육(5 kGy)의 경구투여시 폐사동물은 관찰되지 않았으며, 체중변화도 대조군과 유의적인 차이를 보이지 않았다. 또한, 마우스와 랫드 암수 모두에서 본 시험물질에 의한다고 생각되는 어떠한 임상증상이나 유의할 만한 병변도 관찰되지 않았다.

구치용 도재소부금관과 전부도재관에 파절을 일으키는 한국음식에 관한 연구 (A comparative study on the correlation between Korean foods and the fractures of PFG and all ceramic crowns for posterior applications)

  • 연구목적: 4종 구치용도재관(Full-porcelain-occlusal-surfaced PFG, Half-porcelain-occlusal-surfaced PFG, Empress 2, Ice Zirkon)과 선별된 한국음식의 fracture load와 dynamic cyclic load를 측정하여 구치용 도재관에 파절을 일으킬 가능성이 있는 한국음식을 선별하는데 있다. 연구재료 및 방법: 4종의 각 porcelain 보철물 system 당 15개의 축대칭을 이루는 crown을 제작했다. 이때 occlusal reduction은 1.5-2.0 mm로 했다(중심부 1.5 mm, 교두부 2.0 mm). 각 15개의 시편의 교합면 중앙부에 직경 5 mm의 stainless steel ball을 위치시킨 후 Instron 4465 universal testing machine(Instron, Norwood, MA USA)을 이용하여 5 mm/min의 crosshead speed로 수직 부하를 주어 파절을 일으키는 최대 부하(N)를 기록했다. 이후, 한국음식 중 삶은 게, 닭(뼈포함), 소갈비(뼈포함), 마른 오징어, 건멸치, 사탕, 호두껍질을 표본으로 설정하고 이들을 파절시키는 최대 부하(N)를 universal testing machine(Instron 4465) 에서 측정하여 기록했다. 각 항목당 15번을 측정했다. 음식물을 파절시킬 때 필요한 최대부하와 각 보철물의 파절저항을 비교하여 한국의 식습관과 도재를 이용한 보철물 파절의 상관관계를 조사하였다. fracture loads는 analysis of variance 와 Post Hoc tests를 이용해서 분석하였다($\alpha$=0.05). 차후에 위에서 얻은 결과를 바탕으로 Hydraulic Dynamic Fatigue Testing Machine(858 Bionix II, MTS systems, Eden Prairie, MN USA)를 이용하여 4종의 각 porcelain 보철물 system당 5개의 crown에 14Hz Cyclic Load를 가하여 crown에 파절을 일으키는 masticatory cycle수를 알아 보았다. Load 수치는 41.0 N(건멸치 파절강도), 169.0 N(마른오징어 파절강도), 382.9 N(닭뼈 파절강도), 2224.8 N(사탕 파절강도)로 설정하였다. 결과: 95% confidence intervals for mean fracture load는 2599.3-2809.1 N(완전도재교합면 PFG), 3689.4-3819.9 N(반도재교합면 PFG), 1501.2-1867.9 N(Ice Zirkon), 803.2-1188.5 N(Empress 2)로 나왔고 95% confidence intervals for dynamic cyclic load on fracture는 instron 상에서 도재보철물에 파절을 일으키지 않은 load인 2224.8 N(사탕 파절강도)와 382.9 N(닭뼈 파절강도)로 실험했을 때, 2224.8 N에서 4796.8-9321.2 cycles(완전도재교합면 PFG), 2224.8 N에서 881705.1-1143565.7 cycles(반도재교합면 PFG), 382.9 N에서 979993.0-1145773.4 cycles(Ice Zirkon), 382.9 N에서 564.1-954.7 cycles(Empress 2)로 나왔다. 결론: 통계학적으로 유의할 만한 차이가 그룹들 간 fracture load에서 나타났다. 한국음식물 중 소갈비(뼈포함)와 사탕(자두맛캔디)은 구치용 도재보철물을 파절시킬 가능성이 있는 음식물로 밝혀졌다. 단일수직부하에서는 파절이 생기지 않는 경우라 할지라도 dynamic cyclic load를 줄 경우 일정 주기 후에 파절이 생기는 결과를 얻을 수 있었다.

Effect of Ranging Methods on Sarcomere Length and Palatability Varies between and within Muscles

  • Current study was conducted to investigate the effect of different hanging techniques (achilles tendon, aitch bone and pelvic ligament) on variations in sarcomere length and palatability within and between muscles using thirty-four Angus steer sides. Results showed that tenderstretch(by either suspension method) resulted in longer sarcomeres for most positions within the major leg muscle than did conventional hanging method, but in some minor muscles(eg., mm. gluteus profundus, gastronemius, and gracilis) tenderstetch allowed the muscles fibres to shorten. Some tenderstretched muscles(e.g., m. gluteus profundus, 1.5 ${\mu}m$) appeared not to toughen even at very low sarcomere lengths, while others toughened at higher sarcomere lengths. The current data demonstrated that the effect of tenderstretch on the length of sarcomeres and its influence on palatability varied between muscles, Overall the difference between the two tenderstretch methods was for the aitch method to produce meat that was 3.2 units more palatable than the ligament hanging method.

Bioactive Conjugated Linoleic Acid (CLA) in Milk

  • Conjugated linoleic acid (CLA) isomers are found naturally in foods, such as milk, milk products, beef and others, from biohydrogenation of vegetable oils. They are heterogenous group of isomers of linoleic acid in the family of polyunsaturated fatty acids. Among the isomers of linoleic acid cis9, trans11- CLA (c9, t11-CLA) and trans10, cis12- CLA (t10, c12-CLA) are found to be biologically active isomers. These biologically active isomers either individual or combined found to be health beneficial in various diseases, such as cancer, diabetes, obesity, and atherosclerosis, conclusive participation in physiological processes are necessary. This review focused on the current study of CLA in prevention of disease, such as cancer, diabetes and atherosclerosis, and their effective function in body fat reduction, improvement of bone and muscle mass at a cellular, clinical and systematic level.

Bioactivities and Potential Mechanisms of Action for Conjugated Fatty Acids

  • Since conjugated linoleic acid (CLA) was identified as a principal anticancer component from ground beef in the 1980s, CLA research has discovered that CLA has a wide range of biologically beneficial effects. Clinical studies with CLA are on the rise, and it is apparent that CLA may not be as effective in humans as in rodents, in particular its anti-obesity aspect. In addition, research with regard to other conjugated fatty acids as well as CLA metabolites is still in its infancy. Investigation of bioactivities for other conjugated fatty acids and CLA metabolites may help to extend the understanding of CLA and its mechanisms of actions. This may pose an opportunity to use CLA more efficiently and expand the future use of other conjugated fatty acids as pharmacological agents to assist current treatments.

소스(Sauce)의 인식과 기호도 및 품질요인에 대한 평가분석 (An Evaluation of the Recognition, Preferences and Quality Factors on Sauces)

  • This study was performed to investigate and analyze the level of recognition, preferences and quality factors of sauces. A total of 1,340 subjects, 660 males and 680 females in the Daegu and Kyeongbuk areas participated in a questionnaire for this study. The results were summarized as follows: Among the respondents, 41.1% answered that they learned about the sauces from a western style restaurant. 73.4% of the respondents suggested that the taste and the quality of sauces had an influence on their preferences of western dishes. 51.2% of the respondents recognized that sauces are mainly used when baking and broiling; it was widely known that demiglace sauce is used on steaks. The preferred colors of sauces from most popular to least popular were brown, blond, white, yellow and red. The overall favorite stock material of demiglace sauce was beef bone. Foods that were preferred to be served with sauces were found to be beef, pea seafood, fish, shellfish, poultry and cereals from most preferred to least preferred. The favorite sauces were brown stock sauce, tomato sauce, liquor sauce, and cream sauce from most preferred to least preferred. The oil and butter sauces were preferred the least. Using a scale of 1 to 5 with 5 being the highest, the respondents rated the way sauces were cooked as the most important the quality-determining factor, with a mean value of 4.31. The raw ingredient as the most important factor was rated 4.45. Among sensory factors, the respondents agreed taste was that the most important characteristic, with a mean value of 4.65. 36.0% evaluated the overall quality of sauces as good and 47.0% as ordinary when served with western dishes. 44.1% of the respondents stated that the taste is was the topmost characteristic that needed to be improved, and that flavor and color were next in order.

대추농축액을 첨가한 브라운소스의 품질특성 (Quality Characteristics of Brown Sauce Added with Jujube Concentrate)

  • Brown sauce is very frequently used for western meat dishes, and this study aims to develop brown sauce for Asians and Koreans by adding jujube concentrate which has natural sugar and is used in Chinese medicine. The ingredients and sensory characteristics of brown sauce added with jujube concentrate were examined and the results are as follows. First, the moisture content increased with increased amount of jujube concentrate added, while the pH decreased. In the results of the sensory evaluation, the sample added with 5% jujube concentrate had the highest score, and the one added with 9% jujube concentrate had the lowest score in color. In the flavor evaluation, the sample added with 5% jujube concentrate had the highest score, and the one added with 9% jujube concentrate had the lowest score. In overall acceptability, the sample added with 5% jujube concentrate had the highest score, and the scores were high in order of 3%, 7%, and 9% concentrates when the samples were heated for 16 hours. Also, when making brown sauce made with beef leg bones, their ingredients continued to flow out even heated for more than 10 hours. Therefore, it was considered that more delicious and healthier sauce could be made by adding jujubes when making beef leg bone brown sauce. Based on these results, even though brown sauce originated from western foods, it can be used to Koreans' taste by adding Korean ingredients. Moreover, more recipes can be made from the brown sauce added with jujube concentrate for more Asians including Koreans to enjoy.

The difference of castration timing of Korean Hanwoo bulls does not significantly affect the carcass characteristics

  • It is already well known that castration improves marbling quality but exact timing of castration is still highly debated in beef cattle production industry. After castration, blood hormonal changes occur in steer and objective of this study was to investigate the effects of growth hormone (GH) levels on adipocyte differentiation in stromal vascular cells (SVCs) and transdifferentiation into adipocytes in C2C12 myoblasts. Total GH concentrations were measured via enzyme-linked immunosorbent assay (ELISA) in 24 male calves and 4 female calves. Cell proliferation, cellular triglyceride (TG) accumulation, and the cell's lipolytic capability were measured in C2C12 myoblasts and SVCs. Myogenic, adipogenic, and brown adipocyte-specific gene expression was measured via real-time polymerase chain reaction (PCR) using SYBR green. Serum GH levels were the highest in late-castrated calves. Treatment with 5 ng/mL GH resulted in greater TG accumulation as well as increased CCAAT-enhancer-binding protein (C/EBP)α and peroxisome proliferator-activated receptor (PPAR)γ expression compared to that after treatment with 15 ng/mL GH. Treatment with 5 ng/mL GH also resulted in lower myogenin (myo)G and myoD expression compared to that after treatment with 15 ng/mL GH. The expression of bone morphogenetic protein (BMP) 7 after treatment with 5 ng/mL GH was higher than that after treatment with 15 ng/mL GH. But carcass characteristics data showed no significant difference between early and late castrated steers. Therefore, our results indicate that castration timing does not seem to be inevitable determinate of carcass qualities, particularly carcass weight and marbling score in Hanwoo beef cattle.

Effects of Size and Rate of Maturing on Carcass Composition of Pasture- or Feedlot- Developed Steers

  • Steers (n = 335) of known genetic backgrounds from four fundamentally different growth types were subjected to two production systems to study the main effects and possible interactive effects on carcass composition. Growth types were animals with genetic potential for large mature weight (LL), intermediate mature weight-late maturing (IL), intermediate mature weight-early maturing (IE), and small mature weight-early maturing (SE). Each year, in a nine year study, calves of each growth type were weaned and five steers of each growth type were developed on pasture or feedlot and harvested at approximately 20 and 14 mo of age, respectively. Data recorded were chilled carcass weight and percentages of forequarter, foreshank, chuck, rib, plate, brisket, hindquarter, round, rump, shortloin, sirloin, flank, lean, fat, bone, and retail cuts. The growth $type{\times}production$ system interaction was an important source of variation in chilled carcass weight (p = 0.0395) and percentage retail cuts (p = 0.001), lean (p = 0.001), fat (p = 0.001), rump (p = 0.0454), shortloin (p = 0.0487), and flank (p = 0.001). The ranking of the growth $type{\times}production$ system means for percentage lean was LL-pasture>IL-pasture = IE-pasture = SE-pasture>LL-feedlot, IL-feedlot>IE-feedlot = SE-feedlot. The growth $type{\times}production$ system interaction was non-significant (p>0.05) for forequarter, foreshank, chuck, rib, plate, brisket, hindquarter, round and bone. Growth types of IE and SE yielded greater (p<0.05) mean forequarter than did growth types of IL and LL ($51.6{\pm}0.3$ and $51.5{\pm}0.3$ vs. $51.1{\pm}0.3$ and $50.8{\pm}0.3%$). Mean bone was highest (p<0.05) for the LL growth type and lowest (p<0.05) for the SE growth type ($19.5{\pm}0.5$ vs. $16.8{\pm}0.5%$). Mean bone was greater (p<0.05) for the pastured steers than for the feedlot steers ($21.8{\pm}0.8$ vs. $14.5{\pm}0.6%$). These data indicate that growth type responded differently in the two production systems and that these results should be helpful in the match of genetics to production resources.
