Identification of a PAS Domain-containing Protein in a Mammalian Hibernator, Murina leucogaster

  • Mammalian hibernation is a type of natural adaptation that allows organisms to avoid harsh environment and to increase the possibility of survival. To investigate the molecular link between circadian and hibernating rhythms in the greater tube-nosed bats, Murina leucogaster, we set out to identify circadian genes that are expressed in bats, with specific focus on the PAS domain by using PCR-based screens. We could isolate a eDNA clone, designated as LPAS1, that encodes a protein of 521 amino acid residues. LPAS1 is closely related with CLOCK family with the highest homology to human CLOCK. Based on RT-PCR analyses, LPAS1 transcripts are ubiquitously present in tissues from both summer active and winter dormant periods. Given that LPAS1 is a member of the bHLH-PAS protein superfamily but lacks polyglutamine transactivation domains, it is likely to function as a repressor for endogenous CLOCK to hinder its roles in promoting transcription. Our result will open a new avenue to further examine the functional interconnection between the circadian clock and the circannual clock such as mammalian hibernation.

복식에 표현된 한.중 박쥐문양의 형태적 비교 (A Comparative Study of Bat Patterns in Clothes and Personal Ornaments of China and Korea)

  • We, in this thesis, examined the similarities and differences of bat patterns in China and Korea, along with a consideration of how two peoples understood bats and how their understanding of bats developed and also influenced the bat patterns. As for research period, the Ming(明) dynasty and the Qing(淸) dynasty in China and the Chosun dynasty in Korea were considered. The subjects of this study included clothes, personal ornaments, and embroidery. This research was done by comparing and analyzing the similarities and differences of two countries' bat patterns. The results of comparing and analyzing Chinese and Korean bat patterns were as follows: In terms of similarity, people of two countries understood bat patterns as the symbol of happiness. Therefore, they were used in clothes, personal ornaments, and daily necessities. In terms of difference, bat patterns of two countries had different shapes. In China, colorful and realistic single bat patterns were shown as intensive composition, whereas in Korea plain and simple bat patterns were used. These differences resulted from different way of understanding of bat patterns. It can be concluded that the bat patterns can be a great cultural commodity with high commercial values, if the traditional pattern is used as a unique element of the Oriental designs and altered and developed to attract consumers' attention.

Characteristics of Auditory Stereocilia in the Apical Turn of the Echolocating Bats by Scanning Electron Microscopy

  • The auditory system of the Korean greater horseshoe bat (Rhinolophus ferrumequinum korai, RFK) is adapted to its own echolocation signal, which consist of constant frequency (CF) element and frequency modulated (FM) element. In contrast, the Japanese long-fingered bat (Miniopterus schreibersii fuliginosus, MSF) emits FM signals. In the present study, the characteristics of stereocilia in RFK (a CF/FM bat) and MSF (a FM bat) were studied in the apical turn of the cochlea where the lower frequencies are transduced. Stereocilia lengths and numbers were quantitatively measured in RFK by scanning electron microscopy and compared with those of MSF. Each inner hair cells (IHCs) of RFK possessed three rows of stereocilia, whereas MSF possessed five rows of stereocilia. Gradients in stereocilia lengths and numbers of stereocilia of the IHCs of RFK were found to be less pronounced and fewer, respectively, than those of MSF. Each outer hair cells (OHCs) possessed three rows of stereocilia in both species. OHCs stereocilia in RFK that distinguished it from MSF were a shorter length and a greater number of stereocilia. These features suggest that the apical cochleas of RFK are adapted for the processing of higher frequency echolocation calls rather than that of MSF.

Elemental analysis of the liver, kidney, and intestine tissues from a Hodgson's bat (Myotis formosus tsuensis)

  • Hodgson's bats are critically endangered in South Korea. This study analyzed the concentrations of elements in liver, kidney, and intestine tissues from a Hodgson's bat found dead in the wild. The concentrations of essential elements followed the order Fe > Zn > Cu > Mn > Se in the three tissues. Hg was detected at the highest concentrations among the non-essential elements analyzed in the liver and kidney tissues, while As was the most highly concentrated non-essential element in the intestine. To the best of our knowledge, this is the first study of tissue element concentrations in Hodgson's bats.

동굴성 박쥐 7종의 온도선호도와 동면처 선택 (The Thermal Preference and the Selection of Hibernacula in Seven Cave-dwelling Bats)

  • 동굴을 잠자리로 이용하는 동굴성 박쥐의 동면생태에 관한 연구를 위하여 2005년부터 2011년까지 6회의 동면기간 동안 수행하였다. 조사기간 동안 박쥐가 출현된 140개의 동면장소에서 관박쥐, 문둥이박쥐, 검은집박쥐, 긴가락박쥐, 관코박쥐, 대륙쇠큰수염박쥐, 흰배윗수염박쥐, 붉은박쥐, 큰발윗수염박쥐, 물윗수염박쥐, 토끼박쥐등 총 13,288개체를 확인하였다. 동굴성 박쥐 7종의 온도선호도는 종 간 차이를 나타냈다. 동굴성 박쥐 7종 가운데 붉은박쥐의 평균 체온은 $13.64{\pm}0.76^{\circ}C$로 가장 높았고, 다음으로 관박쥐와 긴가락박쥐의 평균 체온은 $9.19{\pm}2.35^{\circ}C$$7.83{\pm}1.94^{\circ}C$였다. 반면 저온을 선택한 관코박쥐의 체온은 $2.64{\pm}0.98^{\circ}C$, 토끼박쥐는 $2.78{\pm}0.98^{\circ}C$, 검은집박쥐는 $2.76{\pm}1.68^{\circ}C$, 물윗수염박쥐는 $4.4{\pm}1.1^{\circ}C$로 다른 3종(붉은박쥐, 긴가락박쥐, 관박쥐)의 온도선호도와 차이를 나타냈다. 종 간 차이를 나타낸 7종의 체온은 대기온도($T_a$)에 비해 암벽온도($T_r$)와 밀접하게 상관되었다. 동굴성 박쥐 7종의 동면장소 별 출현된 박쥐의 종 수는 동면장소의 온도특성에 의해 차이를 나타냈다. 동면장소의 온도가 고온특성을 나타내고 대기온도 범위가 좁은 동면장소(HTR, 동면저의 최저온도가 $7^{\circ}C$ 이상)에 출현된 박쥐 종수는 $1.44{\pm}0.53$로 가장 낮았지만, 동면장소의 온도가 저온특성이고 대기온도 범위가 좁은 동면장소(LTR, 최고온도가 $7^{\circ}C$ 이하인 곳)에서 출현된 박쥐의 종수는 $2.77{\pm}1.72$였다. 동면장소의 온도가 저온과 고온특성을 모두 포함하는 대기온도 범위가 넓은 동면장소(WR)에서 $3.02{\pm}1.36$종으로 가장 높게 나타났다.

교외지역에 서식하는 Myotis aurascens의 주간휴식지 선택 및 행동권 크기 (Diurnal Roosts Selection and Home Range Size in the Myotis Aurascens (Chiroptera: Vespertilionidae) Inhabiting a Rural Area)

  • Between July and October 2011, radio-tracking was used to analyze the characteristics of home ranges and day roosts of Myotis aurascens by using 3 individuals (male: 2, female: 1). Bat capturing was conducted at a bridge and a nearby forest in Ulju-gun, Ulsan-si. We attached radio transmitters (0.32 g) to the bats and monitored them by using a radio receiver with a Yagi antenna. Home-range analysis of M. aurascens by using 100% minimum convex polygon (MCP) and 95% MCP showed an average of 106.5 ha and 89.3 ha, respectively, and 50% kernel home range (KHR) showed an average of 8.4 ha. Home range overlap of the 3 bats was observed at the bridge and at nearby water bodies as the core areas, and the size of the home range overlap was 7.3 ha by 100% MCP, 5.9 ha by 95% MCP, and 1.6 ha by 50% KHR. The home range for each bat consisted of the main foraging sites, and the types of foraging sites were similar. M. aurascens-01(M-01) used the bridge and nearby water bodies as the nightly main core areas, M. aurascens-02(M-02) used rice fields and water bodies adjacent to the forest as core areas, and M. aurascens-03(M-03) used water bodies and resident areas as core areas. Although rice fields and resident sites represented the core areas of the home ranges of M-02 and M-03, habitat use was the highest near water bodies as the core area for all the 3 bats. The types of day roosts in this study were a wooden house, canopies of a broad-leaved woodland, and banks of rice fields. The roosts in the wooden house and canopies of the broad-leaved woodland were located within the forest, and the roost in the banks of rice fields was also adjacent to the forest. Our results revealed that the main home range and foraging sites of M. aurascens were located near water bodies as the core area, and forests and places adjacent to the forests were used as day roosts.

활동 및 동면동물의 정자 두부 Plasma Membrane의 성분 및 활성에 관한 비교 연구 (Comparative Study on Components and Activities of Sperm Head Plasma Membrane in Active and Hibernating Animals)

  • 북부 온대지방에 서식하는 박쥐는 그 대부분이 동면을하며, 그 생식유형은 특이하여 가을에 교미하여 사정된 정자는 자성생식도내에 긴 동면기간 동안 저장되어 있는 것으로 알려져 있다(예: 한국큰관박쥐). 환언하면 저장되어 있던 정자는 이듬해 봄에 배란되는 난자와 수정하여 초여름에 출산하게 된다. 본 연구는 월동하는 정자 두부 Plasma membrane의 성분과 활성 등 형태적 및 기능적 특성을 관찰 분석함으로써, 인공임신율을 높일 수 있는 보다 효과적인 한냉온도 정자저장방법을 모색하기 위하여 시도되었다. 동면하는 한국큰관박쥐와 햄스타 및 비동면동물(마우스)의 정자 Plasma membrane이, 실온과 한냉온도(박쥐-동면온도)에서, 생존율, 첨체반응을, 단백질분포, 능동수송에 관여되는 효소$(Na^+-K^+-ATPase)$의 활성 및 주사전자현미경적 조직화학상을 분석하였다. 본 연구에서 얻은 실험결과에 의거하면, 동면동물인 한국큰관박쥐와 햄스타 및 비동면동물인 마우스 정자의 생존율과 첨체반응율은, 실온과 한냉(박쥐-동면)온도간에 의의있는 차이를 발견할 수 없었고, 단백질 분포와 $Na^+-K^+-ATPase$ 효소활성 및 주사전자현미경적 조직화학적 소견에 있어서도 두 온도간에 의의있는 차이는 없었다. 이와 같이 실온과 한냉온도 간에 형태적 기능적 차이가 없다고 하는 사실은, 동면동물이든 비동면동물이든 Spermatozoa plasma membrane이 동면중 또는 동면 이후에 안정적인 수정을 보장해 주는 형태적 및 기능적 보호구조임을 시사해 준다.

제주도 큰발윗수염박쥐(Myotis macrodactylus)의 유전적 집단 구조와 계통 유연관계 (Genetic Population Structure and Phylogenetic Relationship of the Large-footed Bat (Myotis macrodactylus) on Jeju Island)

  • 본 연구는 미토콘드리아 DNA (mtDNA) cytochrome B (CYTB)와 NADH dehydrogenase subunit 1 (ND1) 유전자 서열의 다형성을 근거로 제주도 큰발윗수염박쥐 집단의 유전적 집단 구조와 계통 유연관계를 조사하는 데 목적이 있다. 동아시아 박쥐에서 CYTB 유전자 haplotype은 14개의 haplotype들이 발견되었고, ND1은 9개의 haplotype 들이 발견되었다. 집단별 haplotype의 분포는 지역-특이적인 양상을 보였다. ND1 haplotype 분석결과에서 제주도 집단은 4개의 haplotype을 나타내고, 한라산 소집단과 서부지역 소집단은 3개 haplotype을 나타내었으나, 동부지역 소집단에서는 제주도 전체에서 공통으로 발견되는 1개(Nd03)의 haplotype만 출현하였다. NJ tree에서 제주도 집단은 강원도 집단보다 일본 집단과 더 근연으로 확인되었다. 중국과 일본의 모계선조 계보 사이의 분화 시점은 0.789±0.063 MYBP으로 추정되었고, 제주도와 일본의 모계선조는 약 17만 년(0.168±0.013 MYBP) 전에 분리된 것 으로 판단된다. 제주도 집단은 적어도 5만 년 이전에 이주한 것으로 보인다. 또한 ND1 haplotype 분석결과는 제주도 집단이 이주 후에도 지역 내에서의 적어도 2회 이상의 유전적 분화를 겪었다는 것을 보여주고 있다. 본 연구 결과는 동아시아 큰발윗수염박쥐의 계통 유연관계를 이해하는 데 중요한 기초자료가 될 것이며, 향후 한반도의 남부와 중국, 러시아 등에서 시료 확보를 통해 집단 간 진화적 상관관계를 이해하는 데 필요한 설득력 있는 자료가 마련되어야 할 것이다.

애기박쥐과에 속하는 한국산 박쥐류 6종에 대한 핵형학적 연구 (A Karvotvpic Studv on Six Korean Vespertilionid Bats)

  • The karyotypes of 4 Korean Myotis species (M. mystucinus gracilis, M. jormous tsuensis, M. duubentonii ussuriensis and M. macrodoctvlusp and 2 Pipistrelfus species (p. coreensis and p. abramusl belonging to the Vespertilionidae were examined. The 4 Mvotis species had the karyotypes of 2n : 44 with FN : 50 (M. m. gracilis and M. f. tsuensisl or 52 (M. d. ussuriensis and M. mncrodactylus). Furthermore the karyotype of R obrumus (2n : 26, FN : 441 seemed advanced compared with that of U coreensis (2n : 44, FN : 501 which is similar to the original karyotype of Mvotis species (2n : 44, FN : 50).
