Characterization of Hempseed Protein in Cheungsam from Korea (한국산 청삼 헴프단백질의 특성 분석)

  • The neutrichemical analysis of hempseed from Cheungsam was performed in order to apply hempseed to food industry. Cheungsam hempseed was mainly composed of crude fat (30.52%) and protein (30.02%). The hempseed fat contained about 88% of polyunsaturated fatty acids, and the omega-6 to omega-3 ratio was determined to be approximately 3:1. Hempseed from Cheungsam was rich in essential amino acids. Moreover, hempseed protein from Cheungsam, extracted by acid-base precipitation method, showed free radical scavenging activity. The results suggest that Cheungsam hempseed protein could be utilized as a superior source of protein nutrition.

Chemical Compositions of Corni Fructus and Separating Properties of Its Flesh by drying (산수유 열매의 화학성분과 건조에 따른 과육분리특성)

  • An attempt was made to investigate the some constituents and separation of flesh by drying of Corni Fructus for utilization as processing foods of Cornus officianalis S. et Z. Moisture and reducing sugar contents of Corni Fructus flesh were 40.4% and 18.6%, respectively. The content of anthocyanin was 183.0 mg/100g(dry base). The free sugars of Corni Fructus were consisted of glucose and fructose and their contents were 7.9% and 9.0%, respectively. The compositions of organic acids were malic and succinic acids which contained 1.9% and 0.5%, respectively. The titrable acidity as malic acid and pH were 3.6% and 3.19. The decrease ratio of weight and the drying temperature as the optimum conditions for the separation of flesh and seed of Corni Fructus were $30{\sim}35%$ and $60^{\circ}C$.

Variation of the Electrokinetic Potential and Surface Energy Profile of a Binary Mixture Dispersion with Mixing Ratio (이종혼합부유물질의 양에 따른 electrokinetic potential 및 surface energy profile의 변화 양상)

  • Different colloidal particles generally co-exist in the water and wastewater. Thus, there needs to identify practical electrokinetic characteristics of the particles, comparing with the case when each colloidal material is independently distributed. In this study, changes of overall zeta potential was examined through mixed dispersions of $TiO_{2}$ and $MnO_{2}$. The mixing ratios were classified into 3-type in order to distinguish the effects of the proportions of each particle from those of total concentration in colloidal suspensions. The types are single colloidal dispersions of $TiO_{2}$ and $MnO_{2}$ (1:0, 0:1), mixed dispersions at different ratios (0.75:0.25, 0.5:0.5, 0.25:0.75), and a mixed dispersion with doubled concentration (1:1), respectively. It showed that the overall variation of zeta potential as a function of pH was intensified in a colloidal dispersion with the ratio of 1:1. It was concerned that the double action of ion would contribute to this result. On the one hand, the zeta potentials of each colloidal dispersion commonly decreased at the state of strong acid and base under the influence of compression of the electric double layer. The changing patterns were also considered through calculating total interaction energy between colloidal particles based on DLVO theory and measuring turbidity of the colloidal dispersions.

A Study for Comparison of Dietary Quality and Vitamin K Intake of Vegetarians with Carnivores (채식선호자와 육식선호자의 식사의 질 및 비타민 K 섭취 비교 연구)

  • The prevalence of chronic diseases have been rising in the developing countries because of their increased animal foods consumption and Western lifestyle. Lately, vegetarian diet that exclude animal products get public attention. The purpose of this study was to evaluate the nutritional status and dietary quality of vegetarians, and their consumption of vitamin K and was also assessed. Vegetarians including strict vegan and lacto-ovo-vegetarian consumed their diet at least over 6 months. Carnivores were gender and age matched with vegetarians and they consumed over 50% of protein and fat from animal sources. Current nutrient intakes and dietary quality were assessed using 3-day food records and intake of vitamin K was calculated from the data base of 'Provisional Table on the vitamin K contents of foods, USA'. Blood sample were collected and biochemical parameters and plasma phylloquinone concentrations were analyzed. Anthropometric data from vegetarian and carnivore were not significantly different. The intake of calories, protein, vitamin $B_2$, Ca and Zn of the vegetarians were remarkably lower than RDA for each nutrient. Moreover, index of nutritional quality and nutrient adequacy ratio of vegetarians were lower than those of carnivore. Vegetarian consumed less fat and the ratio of n-6/n-3 fatty acid was lower in vegetarian. The intake of essential amino acids in vegetarian was significantly lower than that of carnivore. The vitamin K consumption and plasma phylloquinone concentration of vegetarian were significantly higher than those of carnivore (p<0.05). The dietary vitamin K consumption was positively correlated with plasma phylloquinone levels in vegetarian (p<0.01).

  • In this study, I analyzed the effect of slagging caused by blending bituminous coal and subbituminous coal while maintaining the generator output, combustion conditions, and ventilation conditions for 870MW thermal power plant designed with bituminous coal. Accordingly I proposed an acceptable method of blending coal method. the blending ratio of sub-bituminous coal was adjusted to 10%, 20%, 40%, 60%, 80%, etc. to confirm ultimate analysis, proximate analysis, ash fusion temperature change, slagging indices, etc. Proper blending coal conditions are blending with sub-bituminous coal at 40% or less, ratio of base component to acid component(B/A) is 0.4 or less or 1 or more, total alkali(TA) is 3.5 or less, fusion slagging index(Rfs) is 1,345℃ or more, and ash content is 13% or less in ultimate analysis, the ash content in proximate analysis is 15% or less, and the initial deformation temperature(IDT) should be at least 1,200℃ or more

Microstructure and Properties of Organic-Inorganic Hybrids(PDMS/SiO$_2$) Through Variations in Sol-Gel Processing (졸-겔공정의 변수조절을 통해 제조된 유기-무기복합체 (PDMS/SiO$_2$)의 미세구조와 특성)

  • SiO$_2$ and PDMS/SiO$_2$ xerogels which are derived PDMS into TEOS have been synthesized by sol-gel process and controlled pore size and distribution through 2 step acid/base catalyzed processes using HCI and NH$_4$OH as a catalyst. In HCl catalyzed SiO$_2$ and PDMS/SiO$_2$ xerogels, pH and gellation time of xerogel were 2.3~2.5 and 12~13 days, respectively, and the shape of xerogel was identified to pellet type and column type. Under acidic condition of final reaction solution, the hydrolysis rate is accelerating, resulting in long gel times. The shape of xerogel is pellet type. In contrast, under less acidic condition, the condensation rate is accelerating, resulting in shorter gel times and the shape of xerogel is column type. The surface area and average Pore size were changed 400$\rightarrow$600($\m^2$/g) and 15$\rightarrow$28$\AA$, respectively, depending to the increase of the mole ratio of HCl/NH$_4$OH, and represented uniform pore size distribution. It is that all the alkoxide groups are hydrolyzed by HCl after the first step and the condensation rate is enhanced by NH$_4$OH. The regular backbone structures of silica are formed at low temperature and the uniform pores are produced by heat treatment.

Effect of ZnCl2 Co-catalyst in the Synthesis of Dimethyl Carbonate from Ethylene Carbonate and Methanol by Using Base Catalysts (염기 촉매를 이용한 디메틸카보네이트 합성에서 ZnCl2 조촉매의 영향)

  • The synthesis of dimethyl carbonate(DMC) is a promising reaction for the use of naturally abundant carbon dioxide. DMC has gained considerable interest owing to its versatile chemical reactivity and unique properties such as high oxygen content, low toxicity, and excellent biodegradability. In this study, the synthesis of DMC through the transesterification of ethylene carbonate(EC) with methanol was investigated by using ionic liquid and metal oxide catalysts. The screening test of different catalysts revealed that choline hydroxide ([Choline][OH]) and 1-n-butyl-3-methyl imidazolium hydroxide([BMIm][OH]) had better catalytic performance than metal salts catalysts such as MgO, ZnO and CaO. The effects of reaction parameters such as reaction temperature, MeOH/EC mole ratio, and carbon dioxide pressure on the reactivity of [Choline][OH] catalyst were discussed. High temperature and high MeOH/EC mole ratio were favorable for high conversion of EC. However, the yield of DMC showed a maximum when carbon dioxide pressure was 1.34 MPa, and then it decreased for higher carbon dioxide pressure. Zinc chloride($ZnCl_2$) was used as co-catalyst with the ionic liquid catalyst. The mixed catalyst showed a synergy effect on the EC conversion and DMC yield probably due to the acid-base properties of the catalysts.

Molecular Cloning of Thermostable $\alpha$-Amylase and Maltogenci Amylase Genes from Bacillus licheniformis and Characterization of their Enzymatic Properties (Bacillus licheniformis의 내열성 $\alpha$-amylase 및 maltogenic amylase 유전자의 분리와 그 효소 특성)

  • The genes encoding the thermostable $\alpha$-amylase and maltogenic amylase from Bacillus lichenciformis were cloned and expressed in E. coli. The recombinant plasmid pTA322 was found to contain a 3.1kb EcoRI genomic DNA fragment of the thermostable $\alpha$-amylase. The cloned $\alpha$-amylase was compared with the B. licheniformis native $\alpha$-amylase. Both $\alpha$-amylase have the same optimal temperature of $70^{\circ}C$ and are stable in the pH range of 6 and 9. The complete nucleotide sequences of the thermostable $\alpha$-amylase gene were determined. It was composed of one open reading rame of 1,536 bp. Start and stop codons are ATG and TAG. From the amino acid sequence deduced from the nucleotide sequence, the cloned thermostable $\alpha$-amylase is composed of 483 amino acid residues and its molecular weight is 55,200 daltons. The content of guanine and cytosine is $47.46mol\%$ and that of third base codon was $53_41mol\%$. The recombinant plasmid, pIJ322 encoding the maltogenic amylase contains a 3.5kb EcoRI-BamHI genomic DNA fragment. The optimal reaction temperature and pH of the maltogenci amylase were $50^{\circ}C$ and 7, respectively. The maltogenic amylase was capable of hydrolysing pullulan, starch and cyclodextrin to produce maltose from starch and panose from pullulan. The maltogenic amylase also showed the transferring activity. The maltogenic amylase gene is composed of one open reading frame of 1,734bp. Start and stop codons are ATG and ATG. At 2bp upstream from start codon, the nucleotide sequence AAAGGGGGAA seems to be the ribosome-binding site(RBS, Shine-Dalgarno sequence). A putative promoter(-35 and-10 regions) was found to be GTTAACA and TGATAAT. From deduced amino acid sequence from the nucleotide srquence, this enzyme was comosed of 578 amino acid residues and its molecular weight was 77,233 daltons. The content of guanine and cytosine was $48.1mol\%$. The new recombinant plasmid, pTMA322 constructed by inserting the thermostable $\alpha$-amylase gene in the EcoRI site of pIJ322 to produce both the thermostable $\alpha$-amylase and the maltogenic amylase were expressed in the E. coli. The two enzymes expressed from E. coli containing pTMA322 was reacted with the $15\%$ starch slurry at $40^{\circ}C$ for 24hours. The distribution of the branched oligosaccharides produced by the single-step process was of the ratio 50 : 50 between small oligosaccharide up DP3 and large oligosaccharide above DP3.

The analysis of products from base-catalyzed depolymerization of kraft lignin (크라프트 리그닌의 염기 촉매 분해(BCD)에 의한 부산물의 조성 분석)

  • The based-catalyzed depolymerization (BCD) of kraft lignin isolated from black liquor which the chemical pulping of a mixture of various Southeast Asia hardwood chips was carried out in a batch reactor in the presence of different NaOH concentrations with supercritical methanol. The S:G ratio of the kraft lignin determined by pyrolysis-GC/MS analysis turned out roughly 1.4:1 and main products were vanillic acid, syringol and 3-methoxy catechol. The diethyl ether extracts as phenolic monomers from BCD reaction were produced similar yield among different NaOH concentrations. The 21 compounds were identified by GC/MS analysis in all experiments and major products were catechol, 3-methoxycatechol, 4-methylcatechol, syringol and isovanillic acid. However, it had been shown to be different monomer contents depending on the dosage of NaOH. Catechol, 4-methylcatechol and 3-methoxycatechol were shown to be the dominant monomer from BCD reaction using 7.5 and 3.25% of NaOH concentration whereas syringol, isovanillic acid, 3-methoxycatechol and 4-methylcatechol were determined to be the most typical products under the condition of 1.63% NaOH.

Studies on the Semicarbazone Formation of Mono substituted Benzaldehydes (일치환 Bezaldehyde 의 Semicarbazone 생성 반응에 관한 연구)

  • Semicarbazone formation of nine monosubstituted benzaldehydes was studied kinetically in 20% methanol buffer solution at 15, 25, 35, and $45^{circ}C$. The rate of p-nitrobenzaldehyde semicarbazone formation is 2.7 times as fast as that of benzaldehyde, while p-hvdroxybenzaldehyde is 3.6 times as slow as that of benzaldehyde. Activation energies for p-chlorobenzaldehyde, benzaldehyde, p-methylbenzaldehyde, p-methoxybenzaldshyde, p-hydroxybenzaldehyde, and p-dimethylaminobenzaldehyde semicarbazone formation are calculated as 5.80, 6.19, 6.57, 7.06, 8.03, and 6.46 kcal/mol respectively. It is concluded from the effect of ionic strength that the reaction is affected by not ions but neutral molecules involving hydrogen bonding between oxygen atom of carbonyl group and hydrogen atom of acid-catalyst, and concerted attack of the necleophilic reagent, free base on carbonyl compound. Also, the effect of solvent composition is small in 20% and 50% methanol (and ethanol) aqueous solutions. The ${\rho}-{\sigma}$ plots for the rates of semicarbazone formation at pH 7.1 show a linear ${\rho}-{\sigma}$ relationship (${\rho}=0.14l$, in contrast to that at pH 2.75 and pH 5.4 corresponding to ${\rho}-{\sigma}$ correlations reparted by Jencks. The rate of semicarbazone formation at pH 5.4 show a relationship which is convex upward, resulting in a break in the curve but at pH 2.75, slight difference from a linear relationship. As a result of studying citric acid catalysis, second-order rate constants increase linearly with citric acid concentration and show a 2 times increase as the catalyst concentration is varied from 0.025 to 0.1 mol/1 at pH 2,9, but slight increase at pH 5.3. The rate-determining step is addition below pH 5 but is dehydration between pH 5 and 7. Conclusively, the rate-determining step of the reaction changes from dehydration to addition in respect to hydrogen ion activity near pH 5. The ortho: para rate ratio of the hydroxybenzaldehydes for semicarbazone formation is about 17 at $15^{\circ}C$. It is concluded that the results constitute strong evidence in favor of greater stabilization of p- than o-hydroxybenzaldehyde by substituent which donate electrons by resonance and is due to hydrogen bonding between the carbon-bound hydrogen of the-CHO group and the oxygen atom of the substituent.
