Pseudolithoderma subextensum (Sphacelariales, Phaeophyceae): a new record of crustose brown alga from Korea

  • Pseudolithoderma subextensum is a crustose brown algal species in the family Lithodermataceae and order Sphacelariales. This species is distributed in several regions across the world including, Europe, Western Atlantic, Middle East and Asia (Hong Kong and Japan). Recent floristic surveys along the Korean coastal shores have revealed new records of encrusting brown algae. In this study, we report P. subextensum as a new record from Korea. Morpho-anatomical and molecular studies on Ralfsia-like specimens from Korea identified some of them as P. subextensum. Pseudolithoderma subextensum is characterized by dark chestnut brown crust with a hypothallial basal layer and erect perithallial filaments, tufts of hairs occasionally arising from the basal layer, several discoid shaped chloroplasts per cell, plurangia arising terminally on erect filaments and without sterile cells, and unangia arising terminally on erect filaments, elongated cylindrical and without paraphyses. Phylogenetic analyses based on COI-5P (545bp) reveal that P. subextensum are nested within Lithodermataceae and forms the same clade with P. roscoffense. The genetic divergences for COI-5P between them is 24.5%.

A STUDY OF THE RADIATION EFFECTS ON THE BASAL CELL OF THE RAT TONGUE EPITHELIUM ACCORDING TO THE EXPOSURE TIMING (방사선 조사시기에 따른 백서 설상피의 기저세포에 미치는 영향에 관한 연구)

  • The purpose of this study was to investigate chronic radiation effects on the basal cell of the rat tongue epithelium according to different irradiation timing. Forty-two female rats were devided into 5 experimental groups according to different irradiation timing and were irradiated single dose of 396cGy by MK cell irradiator using Cs-137. Experimental rats were sacrificed at the 2nd week, 4th week and 6th week after birth. The specimens were examined with light microscope and transmission electron microscope. The following results were obtained. 1. The first changes after irraditation were vacuoles. The vacuoles were chiefly observed in the cytoplasm, perinuclei area, and nuclei. 2. The most severe degenerative changes in the basal cell layer were observed in all experimental groups. ; cellular disarrangement, vacuole formation, widening of intercellular space, enlarged mitochondria & rER, and chromatin clumping were seen. 3. The cellular degenerative changes were most severe at the 4th week after birth in all experimental group, and the basal cell hyperplasia was seen at the 6th week in the most of experimental groups 4. The experimental groups 3 and 4 show more severe and more prolonged cellular degeneration than experimental groups 1 and 2, which were irradiated in pregnancy, and experimental group 5, which was irradiated after tongue maturation.

Ultrastructure of Integument of the Smooth Lumpsucker, Aptocyclus ventricosus (Pallas, 1769) (Teleostei: Cyclopteridae) (뚝지, Aptocyclus ventricosus 피부의 미세구조)

  • This study describes the cell type, ultrastructure and histochemical characteristics as a preliminary study for the research on integument of the smooth lumpsucker, Aptocyclus ventricosus in accordance with the physiological and environmental changes using light and electron microscopes. The SEM revealed the presence of well-developed finger printing structure in the skin surface. The skin surface of the smooth lumpsucker showed an irregular folds in cross section of light microscope. Integument is composed of outer epidermal and inner dermal layer. The epidermal layer is a stratified layer composed of epithelial cells, mucous cells, vacuolar cells, and granular cells. Epithelial cells are classified into superficial, intermediated, and basal cell. The superficial cells were the squamous with well-developed microridges on the free surface, and the microridges were covered with glycocalyx. The mucous cells of unicellular gland were mainly distributed in the apical layer of epidermis and contained mucosal materials of neutral glycoprotein. The vacuolar cells of unicellular gland were mainly distributed in the mid and basal layer of epidermis. The proportion of mucous cells and vacuolar cells were $7.0({\pm}1.07)%$ and $40.6({\pm}3.31)%$ of epidermal area, respectively. The granular cells contained membrane bounded secretory granules with high electron density and developed cell organelles in the cytoplasm. The dermal layer was loose connective tissue layer and composed of mainly collagen fibers. It also contained blood vessels and chromatophores of melanophores and reflecting platelets.

Effects of Chilsun-Whan on Intestinal Mucosa and Gastrointestinal Transit Time in Rats (칠선환이 흰쥐 장점막과 위장관의 통과속도에 미치는 영향)

  • Constipation is a common clinical problem that comprises of symptoms that include excessive straining, hard feces, feeling of incomplete evacuation and infrequent defecation. Although many conditions, such as metabolic problems, fiber deficiency, anorectal problem, an drug, can cause constipation. This study was examined the effects of Chilsun-Whan on intestinal mucosa and gastrointestinal transit time and plasma lipids in rats. Adult male rats were fed for weeks on diets containing no addition(basal diet group), 5% cellulose(cellulose group) and 2.5% . Chilsun-Whan group(Chilsun-Whan group). The results were as follows; 1. The fecal weght was significantly increased 2 times in Chilsun-Whan administrated group compare to basal diet group. 2. The gastrointestinal transit times was significantly decreased in Chilsun-Whan administrated group compare to basal diet. 3. Carmine red mixed with Chilsun-Whan, as a marker, was administered through a gastric tube for stomach or intracecally by a chronically implanted catheter for colon transit. Small intestinal transit and large intestinal transit time were significantly decreased in Chilsun-Whan administrated group compare to basal diet. 4. The height of jejunal villi was developed in Chilsun-Whan administrated group compare to basal diet The thickness of mucosa and muscle layer of colonic mucosa were significantly developed in Chilsun-Whan administrated group compare to basal diet group. 5. The change of goblet cell in colonic mucosa was increased acid mucin stained alcian blue in Chilsun-Whan administrated group compare to basal diet and cellulose group. 6. HDL-cholesterol of plasma lipid was increased in Chilsun-Whan administrated group compare to basal diet and cellulose groups. Theses results suggests that Chilsun-Whan may be used in prevention and treatment of constipation resulting in increase of fecal weight, decrease of gastrointestinal transit time. development of intestinal villi, intensify of stainability of acid mucin in colon.

Desquamative Diseases of Gingiva (임상가를 위한 특집 3 - 잇몸이 벗겨지는 질환)

  • The gingiva consists of an epithelial layer and an underlying connective tissue layer. The oral epithelium is a keratinized, stratified, squamous epithelium. The epithelium can be divided into the following cell layer: basal layer, prickle cell layer, granular cell layer and keratinized cell layer. The desquamative disease of gingiva means exfoliative diseases of epithelial layer on the gingiva. The chronic desqumative gingivitis is usually related to the dematologic disorders that produce cutaneous and mucous membrane blisters. The cicatricial pemphigoid and lichen planus are representative diseases of the dermatologic cases. Patients may be asymptomatic or symptomatic. When symptomatic, their complaints range from a mild burning sentation to an severe pain. The clinical examination must be considered with a thorough history, and routine histologic and immunofluorescence studies. A systemic approach needs to achieve accurate diagnosis and treatment of the gingival desquamative diseases.

Morphological Study on the Osphradium of Rapana venosa (Gastropoda : Muricidae) (피뿔고동 ( Rapana venosa Valenciennes )의 Osphardium 에 관한 형태학적 연구)

  • The authors observed histochemical and ultrastructural characters on the osphradium of Rapana venosa Valenciennes using light microscope, scanning and transmission electron microscpes. The results were as follows:1)The basic structure of osphradium was bipectinated shape, which consisted of a septum situating in the center of osphradium and numerous osphradial leaflets. On the other hand, Epidermis of ospradial leaflets formed the structure of pseudostratified ciliated columnar epithelium which was composed of an epithelial cell layer, a basal cel layer and a neuropile. 2) Ciliated dpithelial cells:A large number of these cells were observed on the lateral and ventral regions but a small number of them were observed on the dorsal region. These cells had cylindrical microvilli, slender mitochondria and serve fibers.3) Supporting cells: These cells had cylindrical microvilli, spongy layer, electron dense granules, mitochondria and nerve fibers4) Four types secretory epothelial cells: Four distinct types of secretory epithelial cells were recognized and were arbitrily designated as Type I, Type II, Type III and Type IV.cell type I: These cells contained electron denwe granules(diameter, 0.94-1.56${\mu}{\textrm}{m}$), well developed Golgi apparatus and rough endoplasmic reticula, cell type II: These cills contained two types of granules of the different electron density. One was high electron density granules which were 0.4-1.0${\mu}{\textrm}{m}$ in diameter, The other was low electron density granules which were 0.75-1.2${\mu}{\textrm}{m}$ in diameter.cell type III:These cells had fibrous secretory materials and exhibited strongly positive reaction with Toluidine blue.cell type IV:A large number of this type of cells were observed on the ventral region of ospgradial leaflets and positively reacted with periodic acid Schiff reagent. 5)Dark cells contained several electron dense cillaty rootlets and unmerous granules but cellular organelles were not observed.6) Four types basal cells: Four distinci types of basal cells were recognized and arbitrarily designated as Type I, Type II, Type III and Type IV.Cell type I(light cell): These cells exhibited low electuon density and contained short smooth endoplasmic reticula, several vacuoles and granules.

Expression of Bcl-2 in the epithelial lining and clinical findings of keratocystic odotogenic tumor (각화낭치성종양의 이장상피에서 Bcl-2 발현양상과 임상지표)

  • Purpose: The odontogenic keratocysts demonstrated a high recurrence rate and a biologically aggressive nature. This might be due to unknown factors inherent in the epithelium or enzymatic activity in the fibrous wall. Bcl-2 protein is characterized by its ability to inhibit apoptosis. The aim of this study was to evaluate the expression and distribution of bcl-2 in the OKCs, its possible relationship with the tumorous characteristics, such as the aggressive nature and high recurrence rate, and its usefulness to differentiate OKCs from dentigerous cysts. Materials and Methods: Formalin fixed paraffin-embedded tissue sections of 53 OKCs, and 44 dentigerous cyst were immunohistochemically analyzed quantitatively for the immunoreactivity of the bcl-2 protein with i-solution. Results: More Bcl-2 expression was observed in the OKCs (mean34.387%) than dentigerous cyst (mean11.144%) with statistical significance (P<0.001). Seventeen and 15 of the 32 OKCs in this study showed positivity in the basal layer and basal/suprabasal layers, respectively. In dentigerous cyst, 2 of 3 showed positivity in the basal cell layer. Conclusion: Considering that bcl-2 over expression may lead to the increased survival of epithelial cells, this study demonstrated a possible relationship between the aggressive nature of OKC and the intrinsic growth potential of its lining epithelium. Furthermore, the basal/suprabasal distribution of bcl-2 positive cells was observed in some OKCs, which might have a significant impact on the behavior of cysts. The bcl-2 expression of OKCs can be useful for differentiating OKCs from dentigerous cysts.

Classification of Spring Types in the Western Coastal Area of Jeju Island, Korea, Based on the Hydrogeological Characteristics (수리지질 특성을 고려한 제주도 서부 해안지역 용천의 유형 분류)

  • This study aimed to classify spring types based on the hydrogeological characteristics of springs in Yongsu-ri~Hamo-ri coastal area in western part of Jeju Island. The springs in study area can be broadly categorized into three groups: perched groundwatrer springs (soil type), perched groundwater springs (sediment type), and basal groundwater springs. The perched groundwater springs of soil type correspond to springs where groundwater seeps out from the perched aquifer formed in the soil layer due to the development of clayey Kosan Formation beneath the surface. Because of the low hydraulic conductivity of soil layer, the average of spring discharge is less than 1 m3/day. The quality of spring water is significantly influenced by agricultural activities, resulting in high nitrate nitrogen concentrations and electrical conductivity. While the perched groundwater springs (sediment type) of the Suwolbong Tuff, which are located in the upper part of Kosan Formation, exhibited relatively higher discharge rates, their water quality was similar to soil-type springs. Basal groundwater springs are located in the zone of basal groundwater, mostly near the coastline. This type of spring appears to discharge of up to 3,707 m3, and the salinity content varies with the tidal fluctuations, especially increasing significantly during dry seasons.

Sequential Changes in Understory Vegetation Community for 15 Years in the Long-Term Ecological Research Site in Central Temperate Broad-leaved Deciduous Forest of Korea (한반도 온대중부 낙엽활엽수림 장기생태조사지에서 15년간 하층식생 군집의 시계열적 변화)

  • This study aims to provide basic data for the systematic conservation and efficient management of forest ecosystems by analyzing changes in understory vegetation of temperate broad-leaved deciduous forests. One-hectare permanent survey plot, consisting of 100 subplots sized 10 × 10 meters, was installed in Gwangneung forest in Pocheon, Gyeonggi-do in 2003. The state of stands and the understory vegetation in the permanent survey plot were examined at a 5-year interval from 2003 to 2018. The vascular plants found in the survey area were 56 families, 128 genera, 176 species, 18 variants, 4 varieties, and 1 subspecies, for a total of 199 taxa. The number of species in both the shrub layer and the herbaceous layer showed a tendency to decrease with time. The MRPP-tests showed a significantly differing species composition of the shrub layer in all years except 2008-2013, whereas significant differences were found in all years concerning the herbaceous layer. As for the average importance value, Euonymus oxyphyllus (18.23%), Acer pseudosieboldianum (16.48%), and Callicarpa japonica (13.85%) were dominant in the shrub layer, while Ainsliaea acerifolia (23.41%), Disporum smilacinum (9.45%), and Oplismenus undulatifolius (5.62%) were dominant in the herbaceous layer. In the shrub layer, the richness of Smilax china, Lonicera subsessilis, and Philadelphus schrenkii was high when the basal area and the stand density of an upper layer were high. By contrast, smaller basal area and stand density were associated with the richness of Acer pseudosieboldianum, Deutzia glabrata, Morus bombycis, and Cornus kousa. Furthermore, it was found out that the impact of the basal area and the stand density on the herbaceous layer decreased over time, while the herb layer's species composition was greatly affected by cover degrees of Euonymus oxyphyllus and Acer pseudosieboldianum in the shrub layer. In conclusion, the number of species in the understory vegetation in Gwangneung forest is continuously decreasing, thus implying that species diversity, basal area, and stand density of an upper layer can influence the species composition in understory vegetation.
