통보릿가루를 첨가한 요구르트의 품질특성 및 항산화 활성 (Quality Characteristics and Antioxidant Activity of Yogurt Added with Whole Barley Floura)

  • 본 연구에서는 보릿가루를 발효식품에 이용하고자 Lactobacillus acidophilus, Lactobacillus delbrueckii subsp. bulgaricus와 Streptococcus thermophilussei (1:1:1)등 3종의 혼합균주를 이용하여 탈지분유와 보릿가루로 요구르트를 제조하고 보릿가루첨가량에 따른 요구르트의 품질특성, ${\beta}$-glucan 분석, 항산화 활성, 관능평가를 측정하였다. 보릿가루를 첨가하지 않은 대조군에 비해 1%와 3% 보릿가루 첨가군에서 pH는 낮았고, 적정산도와 당도는 높았다. 젖산균 수는 대조군에 비해 적었으나 점도는 높았고, 색도의 경우 L값은 낮았고, a값과 b값은 높았다. HPLC에 의한 유기산 분석결과 젖산이 주요 유기산 성분이었으며 3% 보릿가루 첨가군이 대조군에 비하여 높았고 초산의 경우에도 3% 첨가군이 현저한 증가를 나타내었다. ${\beta}$-Glucan 함량과 항산화 활성은 보릿가루 첨가량이 증가할수록 증가하였으며 관능평가 결과 전반적인 기호도는 3% 보릿가루 첨가군이 대조군과 비슷하였고 보릿가루 첨가량이 증가할수록 기호도는 감소하였으며 연령대별 기호도는 보릿가루첨가에 따른 기능성 향상이라는 점에서 높은 연령대에서의 기호도가 높은 것으로 나타났다. 이와 같은 실험결과 보릿가루는 3% 이하로 첨가하는 것이 바람직한 것으로 판단된다.

PREDICTING MALTING QUALITY IN WHOLE GRAIN MALT COMPARED TO WHOLE GRAIN BARLEY BY NEAR INFRARED SPECTROSCOPY

  • Predicting quality traits using near infrared (NIR) spectroscopy on whole grain samples has gained wide acceptance as a non-destructive, rapid and cost effective technique. Barley breeding programs throughout southern Australia currently use this technology as a tool for selecting malting quality lines. For the past 3 years whole grain barley calibrations have been developed at VIDA to predict malting quality traits in the early generation selections of the breeding program. More recently calibrations for whole grain malt have been developed and introduced to aid in selecting malted samples at the mid-generation stage for more complex malting quality traits. Using the same population set, barley and malt calibrations were developed to predict hot water extracts (EBC and IoB), diastatic power, free $\alpha$-amino nitrogen, soluble protein, wort $\beta$-glucan and $\beta$-glucanase. The correlation coefficients between NIR predicted values and laboratory methods for malt were all highly significant ($R^2$ > 0.84), whereas the correlation coefficients for the barley calibrations were lower ($R^2$ > 0.57) but still significant. The magnitude of the error in predicting hot water extract, diastatic power and wort $\beta$-glucan using whole grain malt was reduced by 50% when compared with predicting the same trait using whole grain barley. This can be explained by the complex nature of attempting to develop calibrations on whole grain barley utilizing malt data. During malting, the composition of barley is modified by the action of enzymes throughout the steeping and germination stages and by heating during the kilning stage. Predicting malting quality on whole grain malt is a more reliable alternative to predicting whole grain barley, although there is the added expense of micro-malting the samples. The ability to apply barley and malt calibrations to different generations is an advantage to a barley breeding program that requires thousands of samples to be assessed each year.

검사등급이 다른 맥주보리의 지역에 따른 형질변이 (Variation in Characters by Grades of Malting Barley from Different Locations)

  • 맥주보리 품질관련 형질의 재배지역간 등급별 변이정도를 조사코자 전남 남부 6개군에서 등급별로 원맥 150여점의 시료를 표본하여 분석하였다. 1. 맥주보리의 1등품은 전체 수매량의 25.9%, 2등품 62.7%, 등외품은 11.4%였고, 년차간 변이계수는 1등품과 등외품이 각각 25.0%와 43.4%였다. 2. 원맥의 물리적 특성의 지역간 변이는 천립중 9.1%, 정립율 13.2%, 곡피의 색택 35.7%였고, 발아세 및 발아율은 6.2%와 5.0%였다. 화학적 특성의 지역간 변이는 전분 4.8%, $\beta$-glucan 9.6%, 조단백질 14.0%, 곡피비율 14.7%였다. 3. 농산물검사규격에 적합한 시료는 1등품 75%, 2등품 90%, 등외는 83%였다. 4. 원맥의 등급은 색택, 조단백질, 조섬유, 곡피비율, $\beta$-glucan과 정상을, 정립율, 천립중, 발아세, 발아율, 전분량과는 부상관을 가졌다. 곡피색택은 정립율, 발아세 및 발아율과 부 상관이 있었다.

랫트의 실험적 동맥경화증에 대한 $\beta$-Glucan의 예방 효과 (Preventive Effect of $\beta$-Glucan on the Experimental Atherosclerosis in Rats)

  • The present studies were undertaken in attempt to investigate the preventive effect of $\beta$-glucan from barley ad diltiazem on cholesterol and vitamin D2 induced-atherosclerotic rat. The results obtained were summerized as follows. 1.The group, fed only the mixture of cholesterol and vitamin D2, showed significant increase of calcium, inorganic phosphorus, total cholesterol, lipid LDL-cholesterol and phospholipid in serum, and total lipid in the liver (p<0.05) as comparing with normal group. The aorta showed severe damage of disorganization, necrosis and lipid deposition in the elastic membrane. 2. The group fed mixture of cholesterol and vitamin D2 plus diltiazem simultaneously, showed significant increase of total cholesterol, total lipid and phospholipid in serum, and total lipid and triglyceride n the liver (p<0.05) as comparing with normal group, but the significant decrease of calcium and inorganic phosphorus in serum(p<0.05) as comparing with the atherogenic control group. The aorta showed slight damage of elastic membrane and lipid deposition as comparing with the atherogenic control group. 3.The group, fed mixture of cholesterol and vitamin D2 puls $\beta$-glucan simultaneously, showed significant decrease of total cholesterol, LDL and VLDL-cholesterol, total lipid, phospholipid and triglyceride in serum, and total lipid in the liver as comparing with the atherogenic control group(p<0.05), but the significant increase of calcium in serum as comparing with the normal group(p<0.05). The aorta showed no changes in elastic fiber and no lipid deposition in comparing with the atherogenic control group.

발아조건별 당화용 엿기름의 분해효소 활성도 (Activities of Hydrolytic Enzymes in Barley Malts Prepared under Different Germination Conditions)

  • 엿기름의 적정 제조조건을 설정하기 위하여 발아조건를 달리하여 제조한 엿기름의 ${\beta}-glucanase$, ${\alpha}-amylase$${\beta}-amylase$의 활성을 조사하였다. 보리는 발아가 진행됨에 따라 ${\beta}-glucanase$의 활성이 증가하였으며, 이로 인해 세포벽의 주요 구성다당류인 ${\beta}-glucan$이 분해되어 감소하였다. ${\beta}-Glucanase$ 활성은 $15^{\circ}C$의 발아온도에 비해 $18^{\circ}C$$21^{\circ}C$에서 발아초기에 급격히 증가하였으며 발아 6일경에는 최고값에 달하였다. 6조보리인 올보리의 ${\beta}-glucanase$ 활성이 2조보리인 진양보리에 비해 높게 나타났다. 올보리의 ${\alpha}-amylase$ 활성 역시 진양보리에 비해 현저히 높았으며 6일간의 발아일수에서 증가하는 경향이 있었다. 발아보리의 ${\alpha}-amylase$ 활성은 발아온도 $15^{\circ}C$에서 가장 낮았고 $18^{\circ}C$에서 가장 높았으며 $21^{\circ}C$에서는 약간 감소하였다. ${\beta}-Amylase$는 원맥에서 상당한 활성이 있었으며 발아과정중에 지속적으로 증가하는 경향을 보였다. $15^{\circ}C$에 비해 $18^{\circ}C$$21^{\circ}C$에서 발아일수에 따른 ${\beta}-amylase$ 활성의 증가폭이 컸으며 발아 5일 이후부터는 효소활성도에 있어서 크게 변화를 보이지 않았다. 엿기름의 당화력(diastatic power)은 올보리가 진양보리에 비해 $1.4{\sim}1.9$배 정도 높았으며 $18^{\circ}C$에서 $5{\sim}6$일의 발아가 당화력이 가장 우수한 것으로 나타나 발아조건으로서 적합한 것으로 제시되었다.

할맥과 압맥의 칠반 및 호화특성 (Cooking and Pasting Properties of Split and Pressed Barley)

  • 국내 시판 할맥과 압맥제품의 성분 및 취반특성을 알아보고자 보리쌀 24제품(할맥 9개, 압맥 15개)을 수집하여 단백질, $\beta$-glucan, 아밀로오스 함량, 백도, 취반 및 호화특성 등을 조사하였다. 또한 일본산 가공 보리쌀 7제품(할맥 3개, 압맥 4개)도 구입하여 비교 분석하였다. 국내산 할맥과 압맥제품들의 단백질, $\beta$-glucan, 아밀로오스 함량은 비슷하였으며, 일본산 제품들의 경우에도 같은 경향을 나타내었다. 국내 제품의 경우 일본 제품에 비하여 단백질함량이 높고 백도가 낮은 특징을 보였으며 국내산과 일본산 모두 백도는 압맥이 할맥보다 높은 경향을 나타내었고 단백질과 백도는 부의 상관관계(r=-0.5112, p<0.0l)를 나타내었다. 국내산 할맥제품의 흡습성은 342~436, 퍼짐성은 할맥제품이 449~608%로 높고 용출고형물은 낮아 압맥제품보다 좋은 취반특성을 나타내었다. 특히, 찰성보리쌀을 가공한 제품이 더 좋은 취반특성을 나타내었다. 일본산 제품의 경우에도 할맥의 퍼짐성이 431~518%로 압맥보다 높은 경향을 나타내었다. 국내산 할맥과 압맥 제품의 호화개시온도는 압맥과 할맥제품에서 비슷하였으며 찰성보리쌀을 가공한 할맥 제품이 다른 제품에 비하여 낮은 호화개시온돈 높은 강하점도와 낮은 치반점도를 나타내었다. 일본 제품은 국내산 제품보다 낮은 호화개시온도와 높은 치반점도를 나타내었다.

유화액 시스템에서 옥테닐 호박산 베타글루칸의 계면 특성 (Interfacial Properties of Octenyl Succinyl Barley ${\beta}$-Glucan in Emulsion System)

  • The synthesis of octenyl succinyl ${\beta}$-gucan (OSA-${\beta}$-glucan) was carried out and its interfacial properties at the oil-water interface and in emulsion systems were investigated. An aqueous ethanol system as a reaction media was used to facilitate the synthesis process; 10% (w/w) ethanol found to be the best as it showed a maximum degree of substitution (DS: 0.0132). FT-IR showed a characteristic absorption spectrum at $1736cm^{-1}$, indicating the esterification of octenyl succinyl groups to ${\beta}$-glucan backbone. As for interfacial tension measurements, it was decreased with increasing concentration of OSA-${\beta}$-glucan in the aqueous phase and when NaCl was added to aqueous OSA-${\beta}$-glucan solution in the range of 0.01 M to 0.1 M and also when pH was raised (pH 3 ~ pH 9). In systems of emulsion stabilized with OSA-${\beta}$-glucan, fat globule size found to decrease with increasing concentration of OSA-${\beta}$-glucan, showing a critical value of about $0.32{\mu}m$ at 0.5 wt%. When the OSA-${\beta}$-glucan emulsions were stored, it was found that fat globule size was increased with storage time and particularly pronounced increase was observed in emulsion with 1% OSA-${\beta}$-glucan, possibly due to depletion flocculation. Results of creaming stability evaluated by light scattering technique showed that it was more stable in emulsions containing smaller fat globule size. Surface load of OSA-${\beta}$-glucan in emulsions increased with increasing concentration of OSA-${\beta}$-glucan, suggesting a multilayer adsorption.

동맥경화증의 실험동물 모델화와 식이섬유의 동맥경화 방어기전에 관한 연구 (Studies on the Laboratory Animal Modelling of Atherosclerosis and the Preventive Mechanisms of Dietary Fiber against Atherosclerosis)

  • mechanisms for the hypocholesterolemic effects of $\beta$-glucan remain unclear. Rats were divided into 3 groups ; normal control group, atherogenic group(oral administration of cholesterol 40 mg/kg/day plus vit. $D_2$ 320,000 IU/kg/day), $\beta$-glucan treatment group(atherogenic treatment plus $\beta$-glucan 0.135 g/kg/day). The $\beta$-glucan treatment group showed moderate increases of serum lipids concentration compared with atherogenic group. In histopathological examination, aortas showed no critical lesions. The total fecal neutral sterols and bile acids excreted for 6 days was increased compared with both normal and atherogenic group. To compare effects of soluble fiber and insoluble fiber extracted from barley on postprandial lipemia, 5 healthy male adults ingested on separate days a low-fiber(total dietary fiber 2.61g) control meal or dietary fiber-enriched(12.61g) meals. Fasting and postprandial blood samples were obtained for 6.5h and serum lipids were analyzed. The serum total lipids, total cholesterols, LDL & VLDL-cholesterol were markedly reduced with soluble fiber-enriched meals, but no decrease with insoluble fiber-enriched meals. These results suggest that mechanisms for the hypocholesterolemic effect of $\beta$-glucan on rats were due to the inhibition of cholesterol absorption in the intestinal lumen and acceleration of cholesterol catabolism in the liver. And the soluble dietary fiber($\beta$-glucan) has the hypocholesterolemic effect by dropping serum LDL & VLDL-cholesterol in the clinical study.
