The Ultrastructure of Spermatozoa of the Ussurian Bullhead, Leiocassis ussuriensis (Teleostei, Siluriformes, Bagridae) with Phylogenetic Considerations (대농갱이 Leiocassis ussuriensis 정자의 미세구조와 계통적 고찰 (경골어류, 메기목, 동자개과))

  • The fine structure of spermatozoa of Leiocassis ussuriensis was examined with scanning and transmission electron microscopies. The spermatozoon of L. ussuriensis is approximately $68.8\;{\mu}m$ in length and a relatively simple cell with a spherical nucleus, a short midpiece and a tail. The ultrastructure of spermatozoa of L. ussuriensis is characterized by the following features. The nuclear fossa, the length of which is about two-thirds of the nuclear diameter, contains two centrioles. The centrioles are orientated approximately $180^{\circ}$ to each other. The mitochondria are arranged in two layers and their number is 12 or more. The axoneme is the 9+2microtubular pattern and has inner but no outer dynein arms as in other bagrids. The two axonemal fins are in the same plane with the two central microtubules, the doublets 3 and 8. The axonemal fins and the inner dynein arm are shared in Bagridae and the deep nuclear fossa is shared in Siluriformes. The axonemal finsobse observed in Bagridae and Amblycipitidae of Siluriformes might be the apomorphic character in Ostariophysi. They are not reported in Cyprinidae and Characiformes.

  • Catfish (S. asotus) and bullhead (P. fulvidraco), economically important aquacultural species, belonging to order Siluriformes including two family Siluridae and Bagridae, respectively, are considered to be closely related to each other by various authors.   • A study on the diel activities and feeding habits of Pseudobagrus brevicorpus living in the Daegacheon Stream of Nakdonggang River was conducted in September 2017. Diel activity took place during the night, just after sunset and just before sunrise. Pseudobagrus brevicorpus swam in the middle depths, and when they found prey organisms, approached and consumed them immediately. Fish were divided by age into three groups. Prey organisms whose index of relative importance in the stomach contents exceeded 1% belonged to the orders Ephemeroptera (80.8%), Diptera (8.6%), and Trichoptera (8.5%). An index of relative importance by age showed that the Ephemeroptera are very important prey organisms, and the sizes of prey organisms increase with fish growth.

  • The skin of the Korean bullheads, Pseudobagrus brevicorpus and P. koreanus, is composed of epidermis, dermis and subcutis. The epidermis has three layers, the outermost layer, middle layer and stratum germinativum. The epidermis consists of two types of gland cells, an unicellular mucous cell of sulfomucin and a large club cell having sometimes two nuclei. The epidermis has numerous intraepithelial blood vessels in P. brevicorpus but not at all in P. koreanus. Lymphatic spaces containing lymphocytes are well developed in mainly the stratum germinativum. The dermis lacks scales and consists mostly of bundles of coarse collagen fibers. The collagen bundles are arranged in parallel to each other in the dorsum and lateral region toward the dorsum, but vertically at intervals in the abdomen and lateral region toward the abdomen. Considering this unique skin structure, the two species are likely to exercise cutaneous respiration as a dual respiratory system to overcome hypoxic conditions which frequently occurs in their habitats.

  • The early life history of Korean bullhead, Pseudobagrus fulvidraco was studied to obtain some information required in aquaculture and reinforcement of natural population. The fertilized eggs were almost spherical in shape and demersal. The egg membranes were transparent with minute folds on the surface, causing them to stick to other substrates. Yolk is yellowish without oil droplets. The eggs just after fertilization were measuring $1.4{\pm}0.03mm$(1.3~1.5mm, n=10) and expanded to $1.7{\pm}0.08mm$(1.6~1.8mm, n=10) in diameter in 1.5 hr. The blastodisc was formed in 30 min and cleavage started in 1 hr after fertilization, and the intervals of each stage of cleavage was about 30 min at $25.0^{\circ}C$. The yolk from 32-cell stage to gastrula stage partly depressed and the depressed part moved clockwise. Hatching occurred in 53 hr after fertilization and hatched embryos had 18~19+20~21(38~40) myomeres measuring 4.2~4.3mm in total length. At the age 7 d after hatching, 4 pairs of barbels were already formed ; 1 pair on the posterior part of the nostril, 1 pair on the upper jaw, and 2 pairs on the lower jaw. And the posterior margin of caudal fin changed into two folds. The lateral band and the form of all fins were completed in 3 weeks.

  • The early life history of black bullhead, Pseudobagrus koreanus, endemic to Korea was investigated to get biological information needed in artificial production of seedlings and in recovering natural resources. The fertilized eggs showed some characteristics in having heavy sticky material and minute folds which is formed radical pattern on the egg membrane. The shape of egg was spherical and $2.59{\pm}0.08$(2.45~2.70, n=10)mm in diameter. The yolk had not oil globule. The first cleavage was observed 2 hrs after insemination at $21{\sim}23^{\circ}C$, and the progressive cleavage were done about 30 min. interval. The characteristic changing of the yolk surface started at morula stage and continued to the end of gastrula. Hatching was started 72 hrs and completed 90 hrs after fertilization. The size of the larvae were 5.41~5.81mm in total length and 2.76~2.94mm in preanal length, and the number of so mites was 15-16+33~34(48~50). The barbels and swimbladder were completed and all the fins except second dorsal were appeared 1 week after hatching. The larvae attained 9.67~10.52mm in total length and 5.20~5.65mm in preanal length. All the fin sets and color pattern were completed 2 weeks after hatching and body mucus was secreted at that stage. The juvenile attained 14.59~16.02mm in standard length, 3.31~4.16mm in head length and 8.07~9.31mm in prenal length.

Egg Development of the Ussurian Bullhead Fish, Leiocassis ussuriensis (Pisces: Bagridae) and Morphological Development of Its Larvae and Juveniles

  • This study was examined the ovogenesis of Ussurian bullhead, Leiocassis ussuriensis and the morphological development of its larvae and juveniles and to use the results as basic information for the preservation of species and resource enhancement. For artificial egg collection, human chorionic gonadotropin (HCG) was injected at a rate of 10 IU per gram of fish weight. During breeding period, water temperature maintained at $24.5{\sim}26.5^{\circ}C$ (mean $25.0{\pm}0.05^{\circ}C$). The process of ovogenesis reached the two-cell stage in 50 minutes after fertilization. In 73 hours of fertilization the movement of the embryoid body became active state and the larvae began to hatch from the tail through the oolemma. Length of prelarvae were 6.33~6.50 mm long (mean $6.40{\pm}0.06mm$) just after hatching having yolk with their mouth not opened. After thirty eight days of hatching, juveniles were 30.6~32.5 mm long (mean $31.5{\pm}0.65mm$). The color was dark yellowish brown throughout the entire body, and the number of caudal fin rays developed to thirty six perfectly.

  • Comparative study of retinal structure in two Korean endemic freshwater fishes, Zacco koreanus and Pseudobagrus koreanus, was carried out by light and scanning electron microscopy. In the visual cell layer comprised of cone cells and rod cells, the cone cells showed a distinct difference between both species. Z. koreanus had two types of cone cells, single cone cells including a long single and short single cone cells, and double cone cells, whereas P. koreanus had only one type of single cone cells having no its short single and long single cones. Meanwhile, the cone cells of Z. koreanus were arranged in more compact-rowed pattern, but it was more loose and irregular in P. koreanus. Based on field observation, Z. koreanus is mainly a epipelagic or sometimes benthopelagic and a diurnal fish with a round and large eye, while P. koreanus is a demersal and nocturnal fish with an oval and small eye. Therefore, the difference in the retinal structure of two species seems to be closely related to their ecological habitats.
