Studies on the synthesis and bactericidal activity of formamidines (Formamidine류의 합성 및 살균성)

  • Six novel compounds of N,N-dimethyl-N'-(6-substituted-2-benzothiazolyl) formamidines nad six novel compounds of N, N-dimethyl-N'-(substituted-phenyl)formamidines were synthesized. They were evaluated fro their bactericidal activities aginst Salmonella typhoso, Escherichia coli, Vibrio cholera, Staphyloccus aureus, Sarcina lutea and for their fungicidal activities against Saccharomyces cereviseae, Candida albicans. It was found that these compounds were considerably more active than phenol, especially against Vibrio cholera, and N, N-dimethy-N'-(4-methyl-phenyl_formamkidine, N, N-dimethyl-N'-(2-methyl-4-bromo-phenyl)formanidine showed most potent bactericidal activities.

  • The physical and chemical characteristics of bacteriostatic water filter media are compared to granular activated carbon upon which a silver impregnated. Silver ion that has been chemically impregnated the activated carbon was the bactericidal agent in water filter media. Also, silver Ion that has teen chemically impregnated onto the activated acetate resin was the bactericidal agent in ultrasonic humidifier. Silver impregnated active carbon and silver cartridge will be able to the bactericidal agents for statical water.

Bactericidal activities of LB20304, a new fluoroquinolone

  • The time-kill curves of LB20304, a novel fluoroquinolone that has potent antibacterial activity against gram-positve and gram-negative bacteria, were calculated at the concentrations of 1/4-, 1/2-, 1-, 2- and 4-times the MIC against Staphylococcus aureus 77, Escherichia coli 3739E, Pseudomonas aeruginosa 1912E. The bactericidal activity of LB20304 for these strains was very rapid and comparable to that of ciprofloxacin. LB20304 produced a decrease in the $log_10$ CFU per milliliter of${\geq}$3 within 2 h at 4-times the MIC for all strains and consitently prevented regrowth of bacteria after 24 h of incubation. The MBCs (Minimal Bactericidal Concentration) of LB20304 against test organisms were equal to or at most four-times higher than the MICs.

  • Water-soluble phosphate glasses containing Ag or Cu ion were prepared through melting process. Then the powdered glass samples were dissolved in D.I. water at room temperature with changing the dissolution time. In terms with the glass composition, dissolution characteristics, bactericidal effects and cytotoxicities were investigated. Dissolved amounts increased uniformly with dissolution time, and the dissolution rate was higher for ternary glass than for binary glass and with less metal oxide amount. And the dissolution rate of the glass with Ag ion was higher than that with Cu ion, and the bactericidal effect of the glass with Ag ion was also greater. Solution with more than 25 ppm of Ag was observed to have strong cytotoxicity to L929, and solutions of lower Ag concentration or with Cu seemed to have little cytotoxicity.

  • The purpose of this study is to determine the effect of concentration and temperature on the bactericidal action of sodium hypochlorite by means of comparing the killing time of several kinds of microorganism on each different concentration and temperature of sodium hypochlorite. The results were as follows: 1. As the concentration of sodium hypochlorite was increased, the bactericidal action of sodium hypochlorite was increased in all specimens. 2. The bactericidal action of sodium hypochlorite at $37^{\circ}C$ was more potent than that of sodium hypochlorite at $21^{\circ}C$. 3. Among the 3 experimental microorganisms, Pseudomonas aeruginosa was the most resistant to sodium hypochlorite, then comes staphylococcus aureus, and the least resistant microorganism was Streptococcus mutans.

  • In order to observe the bactericidal effect of Ranunculus species on pathogenic bacteria, the minimal inhibitory concentration and bactericidal effect was tested Throughout the studies, the following experimental results were obtained and summarized. 1. Minimal inhibitory concentration of Ranunculus species extracts on E. coli was observed in the medium in which 1% Ranunculus species extracts added to brain heart infusion agar. 2. Minimal inhibitory concentration of Ranunculus species extracts on Salmonella species observed in the medium in which 1% Ranunculus species extracts added to brain heart infusion agar. 3. Minimal inhibitory concentration of Ranunculus species extracts on Staphylococcus and Streptococcus was observed in the medium in which 1.5% Ranunculus species extracts added to brain hrart infusion blood agar. 4. The Bactericidal effect of Ranunculus species extracts on E. coli and S. typhi was observed in 30 minutes. 5. The Bactericidal effect of Ranunculus species extracts on staphylococcus aureus was obserded in 40 minutes.

  • Fungal perylenequinones have photodynamic activity and are promising photosensitizers for photodynamic therapy (PDT). Here, we investigated the bactericidal and antitumor activities of phleichrome from the fungal perylenequinone family in vitro. Photodynamic bactericidal activity of phleichrome was analyzed by agar-well diffusion method under dark and illuminated conditions. The photodynamic antitumor activity of phleichrome was analyzed in MCF-7, HeLa, SW480, and HepG2 human cancer cell lines using in vitro cytotoxicity assays. Photodynamic bactericidal activities against Gram-negative and Gram-positive bacteria were species-specific. Antitumor activity against all tumor cell lines increased under the illuminated condition. Depending on the results of the analyses, Phleichrome has potential for further drug development related to its antibacterial and antitumor activities.

Analyzing the mechano-bactericidal effect of nano-patterned surfaces by finite element method and verification with artificial neural networks

  • The study investigated the effect of geometric structures of nano-patterned surfaces, such as peak sharpness, height, width, aspect ratio, and spacing, on mechano-bactericidal properties. Here, in silico models were developed to explain surface interactions with Escherichia coli. Numerical solutions were performed based on the finite element method and verified by the artificial neural network method. An E. coli cell adhered to the nano surface formed elastic and creep deformation models, and the cells' maximum deformation, maximum stress, and maximum strain were calculated. The results determined that the increase in peak sharpness, aspect ratio, and spacing values increased the maximum deformation, maximum stress, and maximum strain on E. coli cell. In addition, the results showed that FEM and ANN methods were in good agreement with each other. This study proved that the geometrical structures of nano-patterned surfaces have an important role in the mechano-bactericidal effect.

Bactericidal action by complement of fish serum (어류혈청의 보체에 의한 살균작용)

  • In order to know the defense mechanism of fish, bactericidal activity was examined into the sera of carp Cyprinus carpio, tilapia Oreochromis niloticus and flounder Paralichthys olivaseus. Each examined normal serum of fishes showed the bactericidal activity against avirulent Escherichia coli but it wasn't appeared against virulent Edwardsiella tarda. When the normal serum of each fish was treated with zymosan, it lost the bactericidal activity completely. After addition of EGTA into the normal serum of each fish, the sera of tilapia and flounder still exhibited the bactericidal activity but the serum of carp lost it. In the presence of specific antibody and complement, bactericidal activity of each antiserum was exhibited high level with in one hour incubation. On the other hand, heat inactivated antiserum showed bacteriostatic reaction. From the above results, although the activity of alternative or classical pathway of each fish complement is different, it is an important function in fish defense mechanism.
