This study monitored changes in the level of heterotrophic bacteria in the filtrate and investigated the effect of stagnant water on it, using a batch-operated, gravity-driven membrane system for household water treatment. The filtration test was carried out in the presence and absence of stagnant water in the filtrate line. The results showed that stagnant water accelerated the heterotrophic bacteria levels, measured by heterotrophic plate count, even though the heterotrophic plate count of the filtrate finally increased up to $10^5CFU/mL$ regardless of the presence of stagnant water. When the change in heterotrophic plate count of a batch was monitored over filtration time, heterotrophic plate count of the filtrate rapidly decreased within 5 min for each batch filtration. Biofilm formation on the filtrate line was observed in the presence of stagnant water. The biofilm fully covered the filtrate line and contained numerous microorganisms. During storage after filtration, heterotrophic plate count increased exponentially. To improve the filtrate quality of a filtration-based household water treatment system, therefore, the stagnant water in the filtrate line should be minimized, the filtrate produced at the first 5 min is recommended not to be used as potable water, and the storage of filtrate should be avoided.
Objectives: This study aims to provide basic data regarding the bacterial total plate count in the atmospheric environment for related studies. Methods: Total plate count and the identification of culturable bacteria in the atmospheric environment in Incheon took place in 2015 using periodic survey. Correlationship analysis was performed between the number of culturable bacteria and environmental elements. In addition, an estimation of novel bacterial species was undertaken using the similarities and phylogenetic tree based on the 16S rRNA gene. Results: The total plate count of culturable bacteria was on average $176CFU/m^3$, and did not exceed $610CFU/m^3$ in the atmospheric environment. Periodic monthly measuring of total plate count was highest in June at $293CFU/m^3$, while the lowest was in July at $125CFU/m^3$. Furthermore, as a result of the identification of culturable bacteria, the genera Arthrobacter and Kocuria were dominant, while novel bacterial taxa that belong to the genera Chryseobacterium and Herbiconiux were separated. Conclusion: The total number of culturable bacteria from the atmospheric environment in Korea is on average $176CFU/m^3$. In addition, the genera Arthrobacter and Kocuria dominate. The presence of novel bacterial taxa are expected in the atmospheric environment, such as belonging to the genera Chryseobacterium and Herbiconiux.

Forty-eight 40-wk-old Hi-sex laying hens were individually caged in an environmentally controlled house to evaluate the effect of garlic (Allium Sativum) juice administration on egg production, egg quality, and yolk cholesterol. Garlic juice was prepared by blending pealed garlic cloves with distilled water (1:1, w/w). Hens were randomly divided into four equal groups; one served as a control and the other three groups were individually gavaged, 3.75 ml, 7.5 ml, or 15 ml garlic juice, three times a week, which respectively represented 0.25, 0.50 and 1% of body weight. Egg production was recorded on a daily basis; egg weight, albumen height, albumen and yolk pH, Haugh unit, and bacterial count of E. coli-challenged eggs were recorded at day of oviposition (day-1) and after 5 and 10 days of storage at room temperature. Yolk cholesterol content was analyzed for five successive weeks. Garlic juice increased (p<0.05) egg weight and mass with no change in egg production intensity. Garlic juice administration recorded higher (p<0.05) albumen height and improvement in Haugh unit. Also, eggs from garlic-treated hens recorded lower (p<0.05) albumen and yolk pH when compared to eggs collected from control hens. Garlic reduced (p<0.05) the $log_{10}$ of bacterial count in egg contents linearly when challenged with E. coli. Egg-yolk cholesterol content was not influenced by garlic juice administration. It is concluded that garlic juice improved performance characteristics and may increase egg shelf life as indicated by egg quality improvement and lower bacterial count of E. coli-challenged eggs. The levels of garlic juice used in this study were insufficient to influence egg yolk cholesterol.
This study investigated the efficacy of ultraviolet-C (UV-C) irradiation in enhancing the quality of raw bovine milk by targeting microbial populations and lipid peroxidation, both of which are key factors in milk spoilage. We categorized the raw milk samples into three groups based on initial bacterial load: low (<3 Log 10 CFU/mL), medium (3-4 Log 10 CFU/mL), and high (>4 Log 10 CFU/mL). Using a 144 W thin-film UV-C reactor, we treated the milk with a flow rate of 3 L/min. We measured the bacterial count including standard plate count, coliform count, coagulase-negative staphylococci count, and lactic acid bacteria count and lipid peroxidation (via thiobarbituric acid reactive substances assay) pre- and post-treatment. Our results show that UV-C treatment significantly reduced bacterial counts, with the most notable reductions observed in high and medium initial load samples (>4 and 3-4 Log 10 CFU/mL, respectively). The treatment was particularly effective against coliforms, showing higher reduction efficiency compared to coagulase-negative staphylococci and lactic acid bacteria. Notably, lipid peroxidation in UV-C treated milk was significantly lower than in pasteurized or untreated milk, even after 72 hours. These findings demonstrate the potential of UV-C irradiation as a pre-treatment method for raw milk, offering substantial reduction in microbial content and prevention of lipid peroxidation, thereby enhancing milk quality.
Purpose: This study was done to examine the effects of meatal care with essential oil on meatal E-coli and pH of inpatients in geriatric hospital. Methods: The participants were 40 patients admitted to J geriatric hospital in G city, Korea. Twenty patients were assigned to the experimental group and 20 to the control group. Participants in the experimental group received meatal care with essential oil (application of essential oil mixture consisting of lavender, tea tree, and frankincense). The control group received meatal care with saline. The meatal care was performed twice daily for one week in both groups. The scores for meatal odor, meatal pH and bacterial count for E-coli were measured before and after the treatment. Results: The score for meatal odor were significantly lower in the experimental group compared to the control group. The meatal pH and bacterial count for E-coli significantly decreased in the experimental group compared to the control group. Conclusion: Findings indicate that meatal care with essential oil is an effective nursing intervention to reduce meatal odor, meatal pH and bacterial count for E-coli for elderly women inpatients in geriatric hospitals.
To develop a functional Kimchi utilizing onion, 5 different Kimchi with onion used as a major ingredient were formulated. The changes in pH. titratable acidity, reducing sugar. total bacterial count, and the number of lactic acid bacteria in the process of fermentation were studied A. onion Kimchi Control. B : onion Kimchi added with oyster, C : onion Kimchi added with salted shrimp, D : onion Kimchi added with oyster, cucumber, and a bit of radish, E : onion Kimchi added with salted shrimp, cucumber, and a bit of radish. pH of onion Kimchi decreased during storage, but titratable acidity increased. The pH values of onion Kimchi were not significantly different among groups, the changes in pH during fermentation were the lowest in A, and changes in B and D were lower than those of C and E. Salt concentration tended to decrease during the fermentation process, and the changes in salt concentration were lower in D and E than in B and C. Reducing sugar content maximized at 4 days of fermentation and decreased after 12 days. The number of lactic acid bacteria increased during first 4 days of fermentation and decreased after 12 days. Total lactic acid bacterial count were the most lowest in A.

Changes in the soil bacterial community of a coniferous forest were analyzed to assess microbial responses to wildfire. Soil samples were collected from three different depths in lightly and severely burned areas, as well as a nearby unburned control area. Direct bacterial counts ranged from $3.322.6\times10^8\;cells/(g{\cdot}soil).$ In surface soil, direct bacterial counts of unburned soil exhibited a great degree of fluctuation. Those in lightly burned soil changed less, but no significant variation was observed in the severely burned soil. The fluctuations of direct bacterial count were less in the middle and deep soil layers. The structure of the bacterial community was analyzed via the fluorescent in situ hybridization method. The number of bacteria detected with the eubacteria-targeted probe out of the direct bacterial count varied from $30.3\;to\;84.7\%,$ and these ratios were generally higher in the burned soils than in the unburned control soils. In the surface unburned soil, the ratios of $\alpha,\;\beta\;and\;gamma-proteobacteria,$ Cytophaga-Flavobacterium group, and other eubacteria groups to total eubacteria were 9.9, 10.6, 15.5, 9.0, and $55.0\%,$ respectively, and these ratios were relatively stable. The ratios of $\alpha,\;\beta\;and\;gamma-proteobacteria,$ and Cytophaga-Flavobacterium group to total eubacteria increased immediately after the wildfire, and the other eubacterial proportions decreased in the surface and middle layer soils. By way of contrast, the composition of the 5 groups of eubacteria in the subsurface soil exhibited no significant fluctuations during the entire period. The total bacterial population and bacterial community structure disturbed by wildfire soon began to recover, and original levels seemed to be restored 3 months after the wildfire.

PURPOSE. To evaluate the efficacy of sodium hypochlorite (1 : 10) and iodophor disinfectants on alginate impressions along with their effect on the survived bacterium count on the gypsum cast. MATERIALS AND METHODS. Four alginate impression on each dentate patients were made, of which Group I were not washed or disinfected, Group II impressions were merely washed with water, Group III were disinfected by spraying with sodium hypochlorite (1 : 10), Group IV were disinfected with iodophor (1 : 213). Gypsum cast (type III) were made from all the impression. Impressions and gypsum cast were swabbed in mid palatal region for bacterial culture. Bacterial colony counting done after 3 days of incubation at $37^{\circ}C$ in blood agar media. The data obtained was analyzed by one way ANOVA test at a significant difference level of 0.05. RESULTS. Group I and Group II showed significantly more bacteria compared to Group III and Group IV. Bacterial colonies on the alginate impression and gypsum cast in group disinfected with Sodium hypochlorite (1 : 10) were 0.18, 0.82 respectively compared to group treated with iodophor (1 : 213). There was an increase in bacterial count on dental cast compared to source alginate impressions. CONCLUSION. Sodium hypochlorite (1 : 10) was found to be better disinfectant for alginate impression. There was an indication of increase in number of bacteria from alginate impression to making of dental cast. Additional gypsum cast disinfectant procedures need to be encouraged to completely eliminate cross infection to dental laboratory.
Objectives: The purpose of this study was to identify the stains causing infections in dental clinics by analyzing bacterial contamination, as well as to suggest improvements for infection control in dental clinics. Methods: In this study, a questionnaire survey of 47 dental hospitals and clinics located in Gyeonggi-do and Incheon, South Korea was administered from June 2013 to September 2013 and used to investigate the practice rates of infection control by dental hygienists and to analyze the bacterial contamination levels in dental offices. Results: In the studied institutions, the bacterial contamination levels of water lines were $20.9{\times}10^3$ colony forming units (CFU)/mL for three-way syringes, $12.7{\times}10^3CFU/mL$ for high-speed handpieces and $9.8{\times}10^3CFU/mL$ for gargling water. The bacterial contamination levels of surfaces were $44.9{\times}10^3CFU/mL$ in cuspidors, higher than in unit chairs ($2.9{\times}10^3CFU/mL$) and light handles ($6.7{\times}10^3CFU/mL$). The mean bacterial cell count of water lines and surfaces was relatively high in all establishments founded 11 years ago or more, and the mean bacterial cell count of waterline handpieces was $6.27{\times}10^3CFU/mL$ in establishments founded between one and five years ago, $11.16{\times}10^3CFU/mL$ six to ten years ago and $20.04{\times}10^3CFU/mL$ 11 years ago or more, which suggests that earlier foundation is associated with higher bacterial contamination levels with a statistical difference (p<0.01). Similarly, the mean bacterial cell count of cuspidors using water from water lines was also $70.16{\times}10^3CFU/mL$ in at least 11-year-old establishments, statistically significantly higher among in one- to five-year-old ($4.61{\times}10^3CFU/mL$) and six- to ten-year-old clinics ($47.89{\times}10^3CFU/mL$) (p<0.05). Conclusion: This study may be utilized to improve the bacterial contamination levels in dental offices by controlling the characteristics and environmental factors of dental offices that affect the microbial contamination of waterlines and surfaces in such institutions.
The effects of acetic acid (AA) on aerobic plate counts (APC), gram-negative bacterial counts (GNC), and generation time (GT) in chicken wings stored at 4* were assessed. Chicken wings were treated with 0.5-1.5% (v/v) AA at exposure times of 5 min. Treatments of AA for 5 min significantly (P<0.05) reduced aerobic plate counts (APC) and gram-negative bacterial counts (GNC) on the surface of chicken wings for 8 days, respectively. After 4 days of storage, treatments of 1.0% AA and 1.5% AA for 5 min completely (P<0.05) inhibited APC and GNC compared to initial controls. Based on these results, treatments of 1.0% AA and 1.5% AA for 5 min prolonged the microbiological shelf-life for 8 days compared to those of 0.5% AA and the controls. All treatments of AA increased the lag phase and GT of aerobic microorganisma.
