Various biosensors were evaluated for identifying and detecting foodborne pathogens in a rapid and effective manner. First, five strains of Escherichia coli and six strains of Salmonella were identified using Fourier transform infrared spectroscopy and a statistical program. For doing this, lipopolysaccharides (LPSs) and outer membrane proteins (OMPs) were extracted from a cell wall of each bacterial strain. As a result, each strain was identifed at the level of 97% for E. coli and 100% for Salmonella. Second, E. coli O157:H7, S. Enteritidis, and Listeria monocytogenes were identified by multiplex PCR products from four specific genes of each bacteria using a capillary electrophoresis (CE). Also, ground beef for E. coli O157:H7, lettuce for S. Enteritidis, and hot dog for L. monocytogenes were used to determine the possibility of detecting pathogens in foods. Foods inoculated with respective pathogen were cultivated for six hours and multiplex PCR products were obtained and assessed. The minimum detection levels of tested bacteria were <10 cells/g, <10 cells/g, and $10^4$ cells/g for E. coli O157:H7, S. Enteritidis, and L. monocytogenes, respectively. Third, it was possible to detect S. Typhimurium in a pure culture and lettuce by a bioluminescence-based detection assay using both recombinant bacteriophage P22::luxI and a bioluminescent bioreporter. In addition, bacteriophage T4 was quantitatively monitored using E. coli including luxCDABE genes.
정족수 인식 체계라 불리는 세균들의 세포간 의사교환 전략은 다양한 유전자의 발현조절을 통해, 생물막 성숙, 세포 외 고분자물질의 생산, 병원성 발현 및 항생제 생산 등과 같은 다양한 표현형을 조절하는 세균의 다세포성 행동 양식을 제어한다. 다수의 연구에 의하면 많은 종류의 그람 (Gram)음성 세균들이 정족수 인식체계에 필요한 신호전달 물질로 acyl-homoserine lactones (AHLs)를 사용하고 있으며, 이들은 생물막 형성에 중요한 인자로 작용함을 시사하였다. 이러한 정족수 인식체계에 의한 생물막의 형성은 물이 존재하는 모든 표면환경에서 불필요한 바오매스 축적이라는 심각한 기술적, 경제적 문제를 초래하고 있다. 최근 정족수 인체 체계를 교란하는 다수의 물질들이 다양한 미생물로부터 발견되어, 그들의 정족수 인식 체계와 관련된 주요 기능과 기작들이 밝혀지고 있다. 이러한 정족수 제어 물질들은 최근 다양한 산업에서 발생하는 생물 부착현상들을 제어할 수 있는, 환경 친화적이며 세균의 항생제 다재 내성을 완화 시킬 수 있는 새로운 방법으로 대두되고 있다. 따라서 본 논문은 세균의 정족수 인식 체계와 관련된 최근 정보, 정족수 인식 신호를 제어할 수 있는 정족수 제어 효소와 이러한 기술을 이용한 생물 부착 저해 방법 등을 논의하고자 한다.

Aromatic hydrocarbons are of major concern among genotoxic chemicals due to their toxicity and persistence. Some microorganisms can utilize aromatic hydrocarbons as carbon and energy sources by inducing expression of catabolic operon(s). The XylR regulatory protein activates transcription of the catabolic enzymes to degrade BTEX (benzene, toluene, ethylbenzene, and xylene) from its cognate promoters, Pu and Ps upon exposure of the cells to the aromatic hydrocarbons. The activity of XylR on the promoters was previously monitored using luciferase luc reporter system. The xylR, its promoter Pr and the promoter Po for the phenolic compound catabolic operon were introduced upstream of firefly luciferase luc in the pGL3b vector to generate about 7.1 kb of pXRBTEX. Here E. coli harboring the plasmid was freeze-dried under various conditions to fin,d optimal conditions for storage and transport. The cell viability and luciferase activity were maintained better, when the cells were freeze-dried at -7$0^{\circ}C$ in the addition of the 10% skim milk or 12% sucrose. However, coaddition of protectants such as 10% skim milk plus 10% glucose or 12% sucrose plus 10% glucose, resulted in much better viability and bioluminescence activity compared with the effect of single addition of each protectant. In addition, it was shown that the freeze-dried cells maintained almost intact bioluminescent activities and cell viability for at least 1 week after freeze-drying. This work demonstrated that the properly freeze-dried recombinant bacterial cells could be utilized as a whole-cell biosensor for simple and rapid monitoring of BTEX in the environment.

Bioassay using the marine bacteria, Vibrio fischeri and rotifer, Brachionus plicatilis, and chemical analyses were conducted to assess the toxicity of the various sewage sludges, one of the major ocean dumped materials in the Yellow Sea of Korea. Sludge elutriates extracted by filtered seawater were used to estimate the ecotoxicity of the sludge. Chemical characterization included the analyses of organic contents, heavy metals, and persistent organic pollutants in sludge. Bacterial bioluminescent inhibition (15 min), rotifer mortality (24 hr) and rotifer population growth inhibition (48 hr) assay were conducted to estimate the sludge toxicity. EC50 15 min (inhibition concentration of bioluminescence after 15 minutes exposed) values by Microtox(R) bioassay clearly revealed different toxicity levels depending on the sludge sources. Highest toxicity for the bacteria was found with the sludge extract from dyeing waste and followed by industrial waste, livestock waste, and leather processing waste. Clear toxic effects on the bacteria were not found in the sludge extract from filtration bed sludge and rural sewage sludge. Consistent with Microtox(R) results, rotifer neonate mortality and population growth inhibition test also showed highest toxicity in dyeing waste and low in filtration bed and rural sewage sludge. High concentrations of persistent organic pollutants (POPs) and heavy metals were measured in the samples from the industrial wastes, leather processing plant waste sludge, and urban sewage sludge. However, there was no significant correlation between pollutant concentration levels and the toxicity values of the sludge. This suggests that the ecotoxicity in addition to the chemical analyses of various sludge samples must be estimated before release of potential harmful waste in the natural environment as part of an ecological risk assessment.
자연미생물, KP964에 luxAB 유전자를 주입하여 발광기능을 부여함으로써 그들의 발광정도가 스트레스에 대한 적응, 생존 및 활성도를 나타내는 색인이 될 수 있는가, 그리고 특히 non-culturable but viable(NCBV) 상태로 존재하는 미생물의 활성에 대한 정보를 제공할 수 있는가를 알아보았다. 영양결핍조건 하에서 그들의 총 세균수, 배양가능세균(colony-forming-unit; CFU)수, viable cell 수, 그리고 발광정도(relative light unit; RLU)의 변화를 조사하였다. 형광현미경으로 확인된 총 수는 변화가 없었으며, 첨가된 yeast extract에 의해 성장하는 viable cell 수는 완만하게 감소하였으나 CFU 수는 급격히 줄어들어 37일 째 이후에는 탐지한계 이하로 감소하였다. RLU의 경우, 처음 7일의 영양결핍동안 0.016%까지 감소하였으며, 그 이후에는 탐지한계수준 이하로 검출되었다. 아울러 스트레스를 받는 4시간 동안 CFU수와 RLU의 관계를 알아보기 위하여 발광 KP964 균주에게 독성유해물질, acid shock, osmotic shocks를 접촉시킨 후 시간별로 CFU와 RLU를 측정한 결과, KP964 CFU 당 RLU는 9.1-26% 까지 감소하였다. 이는 스트레스의 종류에 따라 접촉시간이 길수록 CFU 당 나오는 빛의 양이 줄어듬을 보인 것이다. 이러한 결과는 영양결핍조건 시 RLU의 감소율이 CFU의 감소율보다 더 크며 영양결핍 이외의 다른 스트레스 조건 하에서도 RLU의 감소율과 CFU의 감소율의 연관성을 찾을 수 없음을 보이므로, 발하는 빛의 정량만으로는 NCBV 상태는 물론 배양가능한 세균의 양이나 활성을 나타내지 못하는 듯 하였다. 그러나 스트레스를 받은 KP964의 활성 정도와 잠재발광능력과의 관계는 좋은 연관성을 보여주었다. 즉, p-xylene, hypo-osmotic stress, 또는 영양결핍에 노출된 세균으로부터 각 스트레스를 제거하였을 시 나타나는 발광증가 유형을 조사하였는데, 증가가 시작하기까지의 lag period는 각각 32, 26, 22분으로 나타났다. 즉 미생물에 강력한 스트레스로 작용한 경우일수록 lag time이 길고, 또한 발광의 증가율이 느리며 도달하는 최대 RLU값도 낮음이 관찰되었다. 따라서 스트레스 하의 세균으로부터 직접적인 RLU의 측정보다는 이들이 스트레스 조건으로부터 재성장 또는 소생되는 과정에서의 잠재 발광능력의 분석이 자연세균의 적응, 활성 및 생존의 정도를 나타내주었다.
