하지동맥 폐쇄성 질환에서 자가 대복재정맥 정위 이식편의 이용 (The Use of Greater Saphenous Vein In Situ Graft in Arterial Occlusive Disease of Lower Extremity)

  • 배경: 하지동맥의 협착성 질환에서 슬와동맥 및 그 이하 분지들의 협착이 있을 경우 인조혈관과 슬와 동맥 사이의 크기 불일치가 발생하여 원위부 문합이 곤란하게 된다. 본원에서는 자가 대복재정맥 정위 이식편을 이용하여 하지동맥의 혈류를 복원하고자 하였다. 대상 및 방법: 2000년 7월부터 2005년 7월까지 총 26명의 환자에서 대복재정맥 정위 이식편을 이용한 수술을 시행하였으며, 임상 결과들을 환자의 차트 기록에 근거하여 후향적으로 분석하였다. 결과: 술 후 원내 사망이나 조기 사망은 없었으며, 추적기간 동안 총 6명의 만기 사망이 발생하였다. 수술 도중 및 술 후 합병증으로는 5예의 조기 이식편 폐쇄, 2예의 창상 열개, 1예의 이식편 내 동맥류 형성, 1예의 서혜부 창상 장액종 형성, 1예의 이식편 손상이 발생하였으며, 추적기간 동안 이식편 개통률은 69.3%였다. 결론: 대복재정맥 정위 이식편은 슬와동맥 및 그 이하 분지의 협착을 동반한 하지동맥 폐쇄성 질환에서 유용한 혈관 이식편으로 생각된다.

  • MTX는 골육종이나 진행된 비호치킨씨 임파종과 같은 종양성 질환 뿐아니라 건선이나 류마치스 관절염, 유육종증, 웨그너씨 육아종증등의 양성질환에서도 널리 쓰여지는 약물이다. MTX에 의한 폐독성은 약제를 투여받은 환자들 중 0.3-7.5%정도에서 나타나는 것으로 알려져 있다. 급성 또는 만성의 경과로 나타나는 MTX 폐렴의 예가 기술되어 있지만 전형적으로는 MTX를 투여한지 수개월내에 증상이 나타나는 아급성의 임상경과를 취하는 경우가 많다. 조직병리학적으로는 NSIP의 형태로 나타나는 경우가 가장 흔한 것으로 되어 있으나 BOOP나 DAD로 나타나는 경우도 드물게 보고되고 있으며 그 중 육아종의 형성을 보이는 예는 34.7% 정도로 알려져 있다. 우리나라에서는 NSIP나 DAD로 나타난 MTX 폐장염의 예가 보고되어있다. 저자들은 과민성 폐장염의 형태로 발현한 MTX에 의한 폐독성의 증례를 경험하였기에 문헌고찰과 함께 보고하는 바이다.

CCDC26 Gene Polymorphism and Glioblastoma Risk in the Han Chinese Population

  • Background: Glioblastoma (GBM) is an immunosuppressive tumor whose median survival time is only 12-15 months, and patients with GBM have a uniformly poor prognosis. It is known that heredity contributes to formation of glioma, but there are few genetic studies concerning GBM. Materials and Methods: We genotyped six tagging SNPs (tSNP) in Han Chinese GBM and control patients. We used Microsoft Excel and SPSS 16.0 statistical package for statistical analysis and SNP Stats to test for associations between certain tSNPs and risk of GBM in five different models. ORs and 95%CIs were calculated for unconditional logistic-regression analysis with adjustment for age and gender. The SHEsis software platform was applied for analysis of linkage disequilibrium, haplotype construction, and genetic associations at polymorphism loci. Results: We found rs891835 in CCDC26 to be associated with GBM susceptibility at a level of p=0.009. The following genotypes of rs891835 were found to be associated with GBM risk in four different models of gene action: i) genotype GT (OR=2.26; 95%CI, 1.29-3.97; p=0.019) or GG (OR=1.33; 95%CI, 0.23-7.81; p=0.019) in the codominant model; ii) genotypes GT and GG (OR=2.18; 95%CI, 1.26-3.78; p=0.0061) in the dominant model; iii) GT (OR=2.24; 95%CI, 1.28-3.92; p=0.0053) in the overdominant model; iv) the allele G of rs891835 (OR=1.85; 95%CI, 1.14-3.00; p=0.015) in the additive model. In addition, "CG" and "CGGAG" were found by haplotype analysis to be associated with increased GBM risk. In contrast, genotype GG of CCDC26 rs6470745 was associated with decreased GBM risk (OR=0.34; 95%CI, 0.12-1.01; p=0.029) in the recessive model. Conclusions: Our results, combined with those from previous studies, suggest a potential genetic contribution of CCDC26 to GBM progression among Han Chinese.

2-deoxy-D-Glucose Synergizes with Doxorubicin or L-Buthionine Sulfoximine to Reduce Adhesion and Migration of Breast Cancer Cells

  • Background: Cancer metastasis depends on cell motility which is driven by cycles of actin polymerization and depolymerization. Reactive oxygen species (ROS) and metabolic oxidative stress have long been associated with cancer. ROS play a vital role in regulating actin dynamics that are sensitive to oxidative modification. The current work aimed at studying the effects of sub-lethal metabolic oxidative stress on actin cytoskeleton, focal adhesion and cell migration. Materials and Methods: T47D human breast cancer cells were treated with 2-deoxy-D-glucose (2DG), L-buthionine sulfoximine (BSO), or doxorubicin (DOX), individually or in combination, and changes in intracellular total glutathione and malondialdehyde (MDA) levels were measured. The expression of three major antioxidant enzymes was studied by immunoblotting, and cells were stained with fluorescent-phalloidin to evaluate changes in F-actin organization. In addition, cell adhesion and degradation ability were measured. Cell migration was studied using wound healing and transwell migration assays. Results: Our results show that treating T47D human breast cancer cells with drug combinations (2DG/BSO, 2DG/DOX, or BSO/DOX) decreased intracellular total glutathione and increased oxidized glutathione, lipid peroxidation, and cytotoxicity. In addition, the drug combinations caused a reduction in cell area and mitotic index, prophase arrest and a decreased ability to form invadopodia. The formation of F-actin aggregates was increased in treated T47D cells. Moreover, combination therapy reduced cell adhesion and the rate of cell migration. Conclusions: Our results suggest that exposure of T47D breast cancer cells to combination therapy reduces cell migration via effects on metabolic oxidative stress.

Factors Influencing the Acrylamide Content of Fried Potato Products

  • BACKGROUND: Acrylamide (CAS No. 79-06-1) is known to be a carcinogenic compound, and is classified as a Group 2A compound by the International Agency for Research on Cancer (IARC, 1994). Acrylamide can be generated during the browning process via the non-enzymatic Maillard reaction of carbohydrates such as reducing sugars and of amino acids such as asparagine, both of which occur at a temperature above $120^{\circ}C$. Potato tubers contain reducing sugars, and thus, this will affect the safety of processed potato products such as potato chips and French fries. In order to reduce the level of acrylamide in potato processed products, it is therefore necessary to understand factors that affect the reducing sugar content of potatoes, such as environmental factors and potato storage conditions, as well as understanding factors affecting acrylamide formation during potato processing itself. METHODS AND RESULTS: Potatoes were cultivated in eight regions of Korea; For each of these different environments, soil physico-chemical characteristics such as pH, electrical conductivity, total nitrogen, available phosphate, and exchangeable cation content were measured and correlations with potato reducing sugar content and potato chip acrylamide levels were examined. The reducing sugar content in potato during storage for three months was determined and acrylamide level in potato chip was analyzed after processing. The storage temperature levels were $4^{\circ}C$, $8^{\circ}C$, or $10^{\circ}C$, respectively. The acrylamide content of chips prepared from potatoes stored at $10^{\circ}C$ or $20^{\circ}C$ for one month was analyzed and the different frying times were 2, 3, 5, and 7 min. CONCLUSION: This study showed that monitoring and controlling the phosphate content within a potato field should be sufficient to avoid producing brown or black potato chips. For potatoes stored at low temperatures, a reconditioning period ($20^{\circ}C$ for 20 days) is required in order to reduce the levels of reducing sugars in the potato and subsequently reduce the acrylamide and improve chip coloration and appearance.

냉동 시 잠재용융열에 의한 피해를 최소화할 수 있는 이상냉동 곡선 (Ideal Freezing Curve Can Avoid the Damage by Latent Heat of Fusion During Freezing)

  • 배경: 액체질소에 의한 냉동방법은 생물학과 의학에서 세포와 조직의 장기보존으로는 성공적인 방법이다. 잘 조절된 냉동속도와 해동의 방법과 함께 글라이세롤이나 디메칠설폭사이드 같은 냉동보존제의 사용으로 얼음결정이 생기는 것을 방지하여 구조의 유지와 생존율을 모두 향상시킬 수 있으며 반영구적으로 보존할 수 있다. 여러 조직의 초저온냉동에는 조직에 맞는 냉동속도가 있다. 대상 및 방법: 가장 적합한 냉동곡선과 이를 위한 냉동챔버온도를 찾기 위해서 우리들은 조직의 열역학적 계산을 두 가지 방법으로 하였다. 하나는 직접계산방법으로 모든 냉동 대상물의 열물리학적 특성, 잠재용융열, 면적, 농도와 체적을 알아야 계산할 수 있다. 이러한 방법은 매우 복잡하고 어떠한 경우에는 실제 값을 알 수 없다. 다른 방법은 간접계산방법으로 우선 기존의 냉동곡선으로 조직을 냉동시켜 조직의 실제 냉동곡선을 얻은 다음 시간상수로 냉동곡선을 분석한 다음 온도반응을 계산하고 적합한 x차방정식을 대입시켜 냉동 시 온도상승을 막고 이것을 거꾸로 냉동챔버온도를 산출하는 방식이다. 결과: 이 냉동 프로그램을 중배엽줄기세포, 연골세포와 골아세포에 적용시켜 검사하였다. 조직의 온도는 온도상승과정 없이 이상적인 냉동곡선을 따라 감소하였다. 그러나 세포의 양과 수용액의 양이 적어 냉동곡선간의 생존력이 통계학적으로 차이가 있지는 않았다. 만약 더욱 부피가 큰 조직을 냉동시키거나 프로그램을 순차적으로 계속한다면 이상곡선에 더욱 근접하게 되어 차이가 있을 것으로 판단된다. 결론: 이 프로그램은 이상적인 냉동곡선으로 조직을 냉동시키기 위한 냉동챔버온도를 쉽게 찾을 수 있도록 도움이 될 것이다.

바이오차의 시용이 채소 유묘 생장 및 양분 흡수량에 미치는 영향 (Effects of Biochar on Early Growth and Nutrient Content of Vegetable Seedlings)

  • BACKGROUND: Biochar is used in various environmental fields, such as water quality and soil restoration, and affects soil fertility and nutrient cycling. Also, when crops are grown on biochar-applied soil, their characteristics may be affected. Biochar is used especially with commercial vegetable seedlings. METHODS AND RESULTS: The objective of this study was to determine the effects of biochar content in seeding mixes on early growth of lettuce (Lactuca sativa L.), Chinese cabbage (Brassica rapa L.), and red pepper (Capsicum annuum L.). Treatments consisted of a control (0: 10, ratio of biochar to seeding mixes (w/w)), 1: 9 (biochar 10%), 3: 7 (biochar 30%), 5: 5 (biochar 50%), and 7: 3 (biochar 70%). The biochar was made from risk husk and had a C/N ratio of 104. As the mixing ratio of biochar increased, pH increased whereas EC and nitrogen content decreased. The highest phosphorus content was with the treatment of 30% biochar, while there were significant increases in the weight of lettuce seedlings and concentrations of T-N, P2O5, K2O, MgO, and Na with the treatments of 30% and 50% biochar. Although the weight of Chinese cabbage seedlings increased with the treatment of 10% biochar, the increase was not statistically significant. Also, there was an increase in the weight of red pepper seedlings with the treatment of 30% biochar, but the increase was not statistically significant. With increases in the biochar mixing ratio, the K2O concentration of red pepper seedlings increased, but the concentrations of P2O5, CaO, MgO, and Na decreased. It was believed that this was because of absorption inhibition by calcium-phosphate formation in the seeding mixes owing to increased pH. CONCLUSION: In conclusion, adding biochar to seeding mixes is considered to be an important mean for growing healthy vegetable seedlings. More field experiments are needed to verify the effect of biochar on vegetable crop growth over the entire growing season.

Effect of Amniotic Fluid Stem Cells and Amniotic Fluid Cells on the Wound Healing Process in a White Rat Model

  • Background Amniotic-fluid-derived stem cells and amniocytes have recently been determined to have wound healing effects, but their mechanism is not yet clearly understood. In this study, the effects of amniotic fluid stem cells and amniocytes on wound healing were investigated through animal experiments. Methods On the back of Sprague-Dawley rats, four circular full-thickness skin wounds 2 cm in diameter were created. The wounds were classified into the following four types: a control group using Tegaderm disc wound dressings and experimental groups using collagen discs, amniotic fluid stem cell discs, and amniocyte discs. The wounds were assessed through macroscopic histological examination and immunohistochemistry over a period of time. Results The amniotic fluid stem cell and amniocyte groups showed higher wound healing rates compared with the control group; histologically, the inflammatory cell invasion disappeared more quickly in these groups, and there was more significant angiogenesis. In particular, these groups had significant promotion of epithelial cell reproduction, collagen fiber formation, and angiogenesis during the initial 10 days of the wound healing process. The potency of transforming growth factor-${\beta}$ and fibronectin in the experimental group was much greater than that in the control group in the early stage of the wound healing process. In later stages, however, no significant difference was observed. Conclusions The amniotic fluid stem cells and amniocytes were confirmed to have accelerated the inflammatory stage to contribute to an enhanced cure rate and shortened wound healing period. Therefore, they hold promise as wound treatment agents.

형질 전환 기법을 이용한 인체 간암의 마우스 모델 제작 및 특성 규명 (Production and Characterization of a Transgenic Mouse Model of Human Liver Cancer)

  • 본 연구에서는 SV40 Tag을 마우스 albumin 유전자의 promoter/enhancer 조절 하에 발현하도록 설계된 재조합 유전자를 마우스 1세포기 수정란에 미세 주입하여 형질 전환마우스를 제작하고 이들의 인체 간암 모델로써의 적합성을 조사하였다. 형질 전환이 확인된 총 11개체의 founder 생쥐들 중 4개체가 간암을 일으켰고, 두 개체는 신장암을, 한 개체는 피부 및 뇌에서 종양을 각각 일으켰다. 이들로부터 외래 유전자를 계대 유전하는 3가계를 얻었다(#1-2, #1-6, #1-11). 이들 가계의 자소들에서 8주령(#1-2, #1-6)혹은 10주령(#1-11)시부터 간암이 반복적으로 발생되었으며, #1-11 founder개체에서 폐로 암세포가 전이된 것 외에는 다른 조직에서의 형태학적 변이가 발견되지 않았다. 간암 발생은 조직학적 변화에 따라 3단계로 나눌 수 있었다. 즉, 출생에서 3주령까지는 간세포의 과량증식을 보이나 세포핵의 이상은 관찰되지 않았으며, 4주령부터 7주령(#1-2, #1-6) 혹은 9주령(#1-11)까지는 diffuse liver cell dysplasia를 나타내지만 tumor nodule은 발견되지 않았고, 그 이후에는 liver dysplasia를 배경으로 간암이 발생하였다. 본 연구에서 작출한 간암 모델 마우스는 인체 간암과 일부 유사한 소견을 보였는바 인체 간암 기전 연구를 위한 유용한 동물 모델로 이용할 수 있을 것으로 생각된다.

Stem Cells and Cell-Cell Communication in the Understanding of the Role of Diet and Nutrients in Human Diseases

  • The term, "food safety", has traditionally been viewed as a practical science aimed at assuring the prevention acute illnesses caused by biological microorganisms, and only to a minor extent, chronic diseases cause by chronic low level exposures to natural and synthetic chemicals or pollutants. "food safety" meant to prevent microbiological agents/toxins in/on foods, due to contamination any where from "farm to Fork", from causing acute health effects, especially to the young, immune-compromised, genetically-predisposed and elderly. However, today a broader view must also include the fact that diet, perse (nutrients, vitamins/minerals, calories), as well as low level toxins and pollutant or supplemented synthetic chemicals, can alter gene expressions of stem/progenitor/terminally-differentiated cells, leading to chronic inflammation and other mal-functions that could lead to diseases such as cancer, diabetes, atherogenesis and possibly reproductive and neurological disorders. Understanding of the mechanisms by which natural or synthetic chemical toxins/toxicants, in/on food, interact with the pathogenesis of acute and chronic diseases, should lead to a "systems" approach to "food safety". Clearly, the interactions of diet/food with the genetic background, gender, and developmental state of the individual, together with (a) interactions of other endogenous/exogenous chemicals/drugs; (b) the specific biology of the cells being affected; (c) the mechanisms by which the presence or absence of toxins/toxicants and nutrients work to cause toxicities; and (d) how those mechanisms affect the pathogenesis of acute and/or chronic diseases, must be integrated into a "system" approach. Mechanisms of how toxins/toxicants cause cellular toxicities, such as mutagenesis; cytotoxicity and altered gene expression, must take into account (a) irreversible or reversal changes caused by these toxins or toxicants; (b)concepts of thresholds or no-thresholds of action; and (c) concepts of differential effects on stem cells, progenitor cells and terminally differentiated cells in different organs. This brief Commentary tries to illustrate this complex interaction between what is on/in foods with one disease, namely cancer. Since the understanding of cancer, while still incomplete, can shed light on the multiple ways that toxins/toxicants, as well as dietary modulation of nutrients/vitamins/metals/ calories, can either enhance or reduce the risk to cancer. In particular, diets that alter the embryo-fetal micro-environment might dramatically alter disease formation later in life. In effect "food safety" can not be assessed without understanding how food could be 'toxic', or how that mechanism of toxicity interacts with the pathogenesis of any disease.
