Syntheses of New Film-Forming Aromatic Poly(amide-imide)s Containing Isoindoloquinazolinedione Unit in the Backbone: Poly(biphenylphthalicdianhydride-oxydianiline-4,4-diamino-3-carbamoyl-benzanilide) (Poly(BPDA-ODA-DACB))

  • New film forming aromatic poly(amide-imide)s containing isoindoloquinazolinedione (IQ) unit in the backbone chain (polymer XIV) have been successfully synthesized by preparing prepolymers of poly(amic acid-carbonamide). followed by subsequent thermal cyclization of the prepolymers. 4,4-Diamino-3-carbamoylbenzanilide (DACB) V has been synthesized by reduction of 3-carbamoyl-4-amino-4-nitrobenzanilide IV. The prepolymers of poly(amic-acid-carbonamide) (polymers VII and VIII) which exhibit viscosities ranging from 1.4 to 1.7 dl/g have been prepared by a condensation polymerization of monomers such as BPDA, ODA, and DACB. Polymer XIV has been obtained by thermal cyclization of the polymers VII and VIII. During the thermal cyclization reaction, imide ring structure was first introduced and then transformed to the structure of IQ unit. The thermal degradation rate of the resultant polymers were influenced by the cleavage of amide bond but the final char yield was comparable to that of poly(BPDA-ODA).

Backbone Cyclization of Flavin Mononucleotide-Based Fluorescent Protein Increases Fluorescence and Stability

  • Flavin mononucleotide-binding proteins or domains emit cyan-green fluorescence under aerobic and anaerobic conditions, but relatively low fluorescence and less thermostability limit their application as reporters. In this work, we incorporated the codon-optimized fluorescent protein from Chlamydomonas reinhardtii with two different linkers independently into the redox-responsive split intein construct, overexpressed the precursors in hyperoxic Escherichia coli SHuffle T7 strain, and cyclized the target proteins in vitro in the presence of the reducing agent. Compared with the purified linear protein, the cyclic protein with the short linker displayed enhanced fluorescence. In contrast, cyclized protein with incorporation of the long linker including the myc-tag and human rhinovirus 3C protease cleavable sequence emitted slightly increased fluorescence compared with the protein linearized with the protease cleavage. The cyclic protein with the short linker also exhibited increased thermal stability and exopeptidase resistance. Moreover, induction of the target proteins in an oxygen-deficient culture rendered fluorescent E. coli BL21 (DE3) cells brighter than those overexpressing the linear construct. Thus, the cyclic reporter can hopefully be used in certain thermophilic anaerobes.

New Radiolytic Cyclization Products, Phlorocyclin and Isophlorocyclin Exhibit Anti-inflammatory Effects in LPS-stimulated Macrophages

  • Phlorocyclin (PC) and isophorocyclin (IPC) are rare benzofuran derivaitves obtained from the representive dihydrochalcone glucoside, phloridzin (PZ) and are a type of neolignan backbone with a potential anti-glycative agents. However, research related to the enhancement of biological functionallites to inflammation of the newly converted products is very limited. This research was directed with the purpose of discovery more effective anti-inflammatory agents in macrophages of newly radiolysis products PC and IPC. The anti-inflammatory capacities of the characterized products in RAW 264.7 and DH82 macrophages treated with lipopolysaccharide (LPS) to stimulate an inflammation response were examined. The pro-inflammatory factors such as prostaglandin E2 (PGE2), tumor necrosis factor-α (TNF-α), nitric oxide (NO), interleukin-6 (IL-6), and IL-10, without cytotoxicity in LPS-stimulated macrophages, were significantly inhibited after treatment with PC and IPC, when compared to PZ. Moreover, PC and IPC decreased the appearance of cyclooxygenase-2 (COX-2) and inducible NO synthase (iNOS) proteins in macrophages. The cyclization products modified by radiolysis showed the greatest anti-inflammatory effects in macrophage cells, indicating PC and IPC are a potential candidate for use in anti-inflammatory agents.

  • Synthesis of an optically active azabicyclo[3.2.1]octane skeleton as a backbone of the tropane alkaloids has been achieved by employing intramolecular Mannich reaction. Utilizat ion of (R)-${\alpha}$-methylbenzylamine as a chiral auxiliary provided an excellent cyclization of amino dioxolane precursor. However, this auxiliary did not afford high asymmertic induction for the preparation of the optically active cyclization precursor.

  • Isoprenoids represent the most diverse group of metabolites, which are functionally and structurally identified in plant organism to date. Ginsenosides, glycosylated triterpenes, are considered to be the major pharmaceutically active ingredient of ginseng. Its backbones, categorized as protopanaxadiol (PPD), protopanaxatriol (PPT), and oleanane saponin, are synthesized via the isoprenoid pathway by cyclization of 2,3-oxidosqualene mediated with dammarenediol synthase or beta-amyrin synthase. The rate-limiting 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR), which is the first committed step enzyme catalyzes the cytoplasmic mevalonate (MVA) pathway for isoprenoid biosynthesis. DXP reductoisomerese (DXR), yields 2-C-methyl-D-erythritol 4-phosphate (MEP), is partly involved in isoprenoid biosynthesis via plastid. Squalene synthase and squalene epoxidase are involved right before the cyclization step. The triterpene backbone then undergoes various modifications, such as oxidation, substitution, and glycosylation. Here we will discuss general biosynthesis pathway for the production of ginsenoside and its modification based on their subcellular biological functions.

Synthesis of Cyclic Antifreeze Glycopeptide and Glycopeptoids and Their Ice Recrystallization Inhibition Activity

  • Until now, few groups reported the antifreeze activity of cyclic glycopeptides; however, the tedious synthetic procedure is not amenable to study the intensive structure activity relationship. A series of N-linked cyclic glycopeptoids and glycopeptide have been prepared to evaluate antifreeze activity as a function of peptide backbone cyclization and methyl stereochemical effect on the rigid Thr position. This study has combined the cyclization protocol with solid phase peptide synthesis and obtained significant quantities of homogeneous cyclic glycopeptide and glycopeptoids. Analysis of antifreeze activity revealed that our cyclic peptide demonstrated RI activity while cyclic glycopeptoids showed no RI activity. These results suggest that the subtle changes in conformation and Thr orientation dramatically influence RI activity of N-linked glycopeptoids.

Identification of Korean Ginseng Cytochrome P450 gene and Its Characterization by Transformation System (고려인삼 유래 Cytochrome P450 유전자의 동정 및 형질전환에 의한 특성검정)

  • Triterpenoid saponins were synthesized in Panax ginseng C.A. Meyer via the isoprenoid pathway by cyclization of 2,3-oxidosqualene to give primarily oleanane (beta-amyrin) or dammarane triterpenoid skeletons. The triterpenoids are backbone and undergoes various modifications (oxidation, substitution and glycosylation), mediated by cytochrome P450 (CYP)-dependent monooxygenases, glycosyltransferase and other enzymes. This is likely to be due in part to the complexity of the molecules and the lack of pathway intermediates for biochemical studies. A cDNA clone encoding a putative CYP gene was isolated from flower bud of ginseng and transformed into the plant(Nicotiana tabacum cv. Xanthi) and confirmed by PCR analysis. The CYP gene (PgCYP) contained an open reading frame(ORF) encoding mature protein of 500 amino acids. The expression of PgCYP were investigated in transgenic tobacco by reverse transcriptase-polymerase chain reaction (RT-PCR).

Synthesis and Thermal Properties of Aromatic Poly(o-hydroxyamide)s Containing Phenylene Diimide Unit (Phenylene Diimide 단위를 포함한 방향족 Poly(o-hydroxyamide)s의 합성 및 열적 특성)

  • In this study we attempt to modify the backbone structure and improve processibility of PBO having high melting and glass transition temperature. A series of aromatic poly(o-hydroxyamide)s(PHAs) were synthesized by direct polycondensaton of diacides containing diimide unit with two types of bis(o-aminophenol)s including 3,3'-dihydroxybenzidine and 2,2-bis(3-amino-4-hydroxyphenyl)hexafluoropropane. PHAs were studied by FT-IR, $^1H$-NMR, DSC and TGA. PHAs exhibited inherent viscosities in the range of 0.34~0.65 dL/g at $35^{\circ}C$ in DMAc solution. The PHA 1 and 6F-PHA 6, introducing o-phenylene unit in the main chain showed excellent solubilities in aprotic solvents such as NMP etc. However, the PHA 3, having p-phenylene unit was not even dissolved perfectly with LiCl salt. 6F-PHAs were readily soluble at room temperature in aprotic solvents except 6F-PHA 3. But they showed better solubility than that of PHAs. The polybenzoxazoles(PBOs) were quite insoluble in other solvents except partially soluble in sulfuric acid. PBOs exhibited relatively high glass transition temperatures(Tg) in the range of 306~$311^{\circ}C$ by DSC. The maximum weight loss temperature and char yields of PHA3 and 6F-PHA3 showed the highest values of $658^{\circ}C$ and $653^{\circ}C$, 62.6 % and 62.1 %, respectively.
