Evaluation of Selective Media for Isolation of Foodborne Bacteria (주요 식중독균 분리용 선택배지의 타당성 연구)

  • This study was conducted to evaluate the selective media listed in currently available Food Code in Korea. The 29 different types of media of five different types of foodborne bacteria including Salmonella spp., Bacillus cereus, Listeria monocytogenes, Vibrio parahaemolyticus, Staphylococcus aureus were tested in the broth and food. The recovery test for five different types of foodborne bacteria was performed in the artificially inoculated into chicken, rice, pork and mackerel. There was no significant differences in isolation capabilities among twenty nine different types of isolation selective media for five different types of foodborne bacteria in broth condition, while there was significantly a little differences in isolation capabilities among those on foods (P<0.05). The higher number of foodborne pathogens were isolated from conventional selective media approved in Food Code than newly developed selective media such as chromogenic media. This results suggest that there was differences of selectivities among currently available isolation selective media in many countries and further studies are needed to be approved by Korean Food and Drug Administration.

Control efficacy of BtPlus against two mosquitoes, Aedes koreicus and Culex vagans (한국숲모기와 줄다리집모기에 대한 비티플러스 방제 효과)

  • Two mosquito species were collected in still-water near farming area in Andong, Korea. Based on morphological characters, these two mosquitoes were identified as Aedes koreicus and Culex vagans, respectively. DNA barcode analyses supported the identification. An entomopathogenic bacterium, Bacillus thuringiensis subsp. israelensis (BtI), exhibited insecticidal activities against the two mosquito species and its virulence was more potent than that of B. thuringiensis subsp. kurstaki. It has been known that the bacterial metabolites of Xenorhabdus spp. suppress insect immunity and enhance pathogenicity of B. thuringiensis. This study tested the effect of the bacterial culture broth of Xenorhabdus spp. on enhancing BtI pathogenicity. Among three Xenorhabdus spp., culture broth of X. ehlersii (Xe) was relatively effective to enhance BtI pathogenicity against both mosquito species. Indeed, organic extracts of Xe culture broth suppressed the hemocyte-spreading behavior, suggesting the presence of immunosuppressant in the culture broth. These results suggest a formulation of BtPlus by mixing BtI spore and Xe culture broth to be applied to control the two mosquito species.

Isolation of Chitinolytic Bacteria from the Viscera of Korean Bony Fishes and Optimization of the Enzyme Production

  • In order to produce functional chitin oligosaccharides, a chitinolytic bacterium was newly screened from the viscera of Korean bony fishes, and identified as Bacillus sp. LJ-25. For the production of chitinolytic enzymes, $1.0\%$ nutrient broth and $0.3\%$ colloidal chitin were used as nitrogen and carbon source, respectively. The optimal temperature, initial pH and concentration of NaCl for the enzyme production by Bacillus sp. LJ-25 were $30^{\circ}C$ 6.5-7.0 and $1.0\%$, respectively. The enzyme activity of Bacillus sp. LJ-25 increased until the incubation time of 168 hr, followed by a decrease in activity.

Production of Agarooligosaccharides using of Agarase from marine Bacterium Bacillus cereus ASK202 (해양세균 Bacillus cereus ASK202가 생산하는 Agarase를 이용한 Agarooligosaccharides의 생산)

  • An agarase was partially purified from the culture broth of marine bacterium Bacillus cereus ASK202. Optimal pH and temperature of this agarase were found to be 7.0 and 40$^{\circ}C$, respectively. The maximum productivity of agarooligosaccharides was obtained from 0.3 %(w/v) agar by using of 1 unit agarase. As the results of TLC and HPLC analysis, these oilgosaccharides consisted of neoagarobiose, neoagarotetraose and neoagarohexaose. Under the optimal reaction conditions, 77.5 %(w/v) neoagarobiose and 6.2 %(w/v) neoagarotetraose were produced from agar and the conversion yield of total agarooligosaccharides was 83.7 %(w/v) after for 2 h reaction at 40$^{\circ}C$.

  • The strain which produced highly viscous exopolysaccharides (EPS) in liquid culture was selected from soil. The strain was supposed to Bacillus sp. from the results of mophological, biochemical and physiological tests. The medium composition for EPS production was trypton 0.75%, sucrose 4%, CaCO3 0.01%, Winogradsky's nitrogen free mineral medium 5ml/l and pH 7.0. In 2-l jar fernenter, the viscosity of culture broth after 120-hr cultivation time was very high (60, 000 cps) and the amount of EPS was 6.2g/l.

Biological Control of Fusarium Wilt of Carnation Plants by Antagonistic Microorganism in Greenhouse (카네이션의 시설재배에서 길항성 세균을 이용한 Fusarium Wilt 의 생물학적 방제)

  • This study was carried out to screen and select the effects of antifungal bacterial strains which inhibit the growth of plant pathogen, Fusarium oxysporum(fusarium wilt) occurred in carnation plants in greenhouse. We isolated an effective bacterial strains and investigated into the antifungal activity of the antagonistic microorganism and it's identification. Twenty bacterial strains which strongly inhibited Fusarium oxysporum were isolated from roots of carnation plants and the soil in greenhouse, and the best antifungal bacteria designated as C121, was finally selected. Antagonistic bacterial strain, C121 was identified to be the genus Bacillus sp. based on the morphological, biochemical and cultural characterizations. The Bacillus sp. C121 showed 58.1% of antifungal activity against the growth of Fusarium oxysporum. By the bacterialization of the cultural broth and the heat bacterialization culture filtrate of it, Bacillus sp. C121 was shown 92.1% and 21.0% of antifungal activity, respectively.

Biological Control of Fusarium Wilt of Tomato Plants by Antagonistic Microorganism in Greenhouse (온실재배 토마토에서 발생하는 위조병의 미생물학적 제어)

  • This study was conducted to screen the antagonistic bacteria which inhibit the growth of plant pathogen, fusarium wilt(Fusarium oxysporum) occurred in tomato plants in greenhouse. We isolated an effective bacterial strains and investigated into the antifungal activity of the antagonistic microorganism and it’s identification. Ten bacterial strains which strongly inhibited Fusarium oxysporum were isolated from the nature, and the best antagonistic bacterial strain designated as KC175, was selected. The antagonistic strain KC175 was identified to be the genus Bacillus sp. based on the morphological and biochemical characterization. The Bacillus sp. KC175 showed 58.2% of antifungal activity against the growth of Fusarium oxysporum. By the bacterialization of the culture broth and the heat bacterialization culture filtrate of it, Bacillus sp. KC175 showed 91% and 18% of antifungal activity, respectively.

Production of main Taste Components in Traditional Korean Soy Sauce by Bacillus licheniformis (Bacillus licheniformis를 이요한 한국 재래식 간장의 주요맛 성분)

  • This study investigated conditions necessary for factory production of traditional Korean soy sauce flavors, using Bacillus licheniformis SSA3-2M1. We determined whether the flavors were good or bad by comparing sensory evaluation values and the contents of the main taste components of traditional Korean soy sauce with those of the manufactured soy sauce. The soy broth was cultured by Bacillus licheniformis SSA3-2M1. By providing from 1/3 vvm to 2/3 vvm of air, and a culturing time of 412 hours at 30$^{\circ}C$, we produced the taste of traditional Korean soy sauce; moreover its PH was in the PH range of traditioal Korean soy sauce. The distributions of the main taste components and the amino acids, free sugars, and organic acids in the manufactured soy sauce were similar to tradituioal Korean soy sauce.

Production and Characterization of Crystalline Cellulose-Degrading Cellulase Components from a Thermophilic and Moderately Alkalophilic Bacterium

  • A moderately thermophilic, alkalophlic and powerful crystalline cellulose-digesting bacterium, Bacillus K-12, was isolated from filter paper wastes and found to be similar to Bacillus circulans or Bacillus pumilis, except for its ability to grow at a moderately high pH and temperature. The isolate grew at a pH ranging from 6 to 10 and at a temperature ranging from 35 to $65^{\circ}C$ and produced a large amount of cellulase components containing avicelase, xylanase, CMCase, and FPase when grown in avicel medium for 5 to 7 days at $50^{\circ}C$. The crude enzyme preparation from the culture broth hydrolyzed xylan, raw starch, pullulan and ${\beta}-1,3$ glucan such as laminarin. Furthermore, the enzyme hydrolyzed crystalline cellulose to cellobiose and glucose and had a broad pH activity curve (pH 6~9). The enzyme was stable up to $70^{\circ}C$.

  • An antifungal bacterium was isolated to inhibit of the growth of Asp. flavus and Asp. parasiticus, and its antifungal compounds were purified from lyophilized culture broth using chromatographic methods. Antifungal bacterium #19 which was shown a higher inhibitory activity on the growth of aflatoxin producing fungi was identified as Bacillus subtilis. The purified antifungal compound(1 mg) was demonstrated strong antifungal activity against the aflatoxin producing fungi.
