더덕을 첨가하여 속성시킨 된장형 제품의 품질에 미치는 영향 (Effects of Deodeok Contents on the Qualities of Quick Fermented Doenjang Type Product)

  • 본 연구는 된장 제조에 더덕의 함량을 5, 10, 15 및 20% (w/w)로 달리하여 첨가 제조한 된장의 이화학적 및 관능검사 등의 품질에 미치는 영향을 조사하였다. 된장 제조에 사용한 균주는 재래식 메주에서 $\alpha$-amylase, $\beta$-amylase 및 산성 protease 활성이 우수한 것으로 선정된 Bacillus sp. B-3과 Aspergillus sp. M-9이다. Bacillus sp. B-3을 starter로 제조한 된장의 숙성 중 아미노태질소 함량을 조사한 결과 10%(w/w) 더덕 함유 된장이 다른 처리구에 비해 가장 많이 생성되고 있었다. 또한 Aspergillus sp. M-9를 starter로 제조한 된장의 경우 Bacillus sp. B-3로 제조한 된장과 시판재래식된장보다 월등히 많은 아미노태질소를 생성하고 있었다. 한편, 더덕 함량이 15%(w/w) 이상 함유된 더덕된장에서는 아미노태질소 함량이 감소하는 경향을 보여 더덕 함량이 된장 중의 Bacillus sp.와 Aspergillus sp.의 생육에 미치는 영향이 있는 것으로 나타났다. 숙성이 끝난 된장에 대한 관능검사에서는 Bacillus sp. B-3로 제조한 더덕된장의 경우 더덕 10%(w/w) 함유 된장이 가장 높은 점수를 얻었으며, Aspergillus sp. M-9로 제조한 더덕된장에서는 더덕 20% (w/w) 함유 된장이 가장 높은 점수를 얻었다. 시중에서 구입한 된장과 더덕된장을 비교한 결과, Bacillus sp. B-3 및 Aspergillus sp. M-9로 제조한 된장이 우수한 것으로 나타나 Bacillus sp. B-3로 제조할 경우 더덕 10%(w/w)을, Aspergillus sp. M-9로 제조할 경우는 더덕 20%(w/w)를 첨가하는 것이 기호도 측면에서 우수한 것으로 나타났다.

Quality Characteristics of Kochujang Prepared with Different Meju Fermented with Aspergillus sp. and Bacillus subtilis

  • For preparation of high quality kochujang by the traditional fermentation method, 4 types of kochujang were prepared with brick- or grain-shaped meju fermented with different strains (Aspergillus sojae, Aspergillus oryzae+Bacillus subtilis). After 100 days of fermentation at $25^{\circ}C$, the moisture, pH, salt, and ethanol content of kochujang were 40.52-43.20%, 4.71-4.82, 8.7-9.1%, and 0.75-0.94%, respectively, showing slight differences according to the strains and shapes of meju. Titratable acidities were slightly increased for up to 60 days of fermentation. The amino-type nitrogen content of kochujang prepared with brick-shaped meju (A. oryzae+B. subtilis) was the highest (164.20 mg%) among all of the kochujang types. The redness (a) value of kochujang prepared with brick-shaped meju (A. sojae) were higher (19.08) than those of other treatments (18.37-18.59). Sensory evaluation of kochujang prepared with grain-shaped meju (A. sojae) showed the highest scores for color and overall acceptability, 'at $6.43{\pm}1.87$ and $6.29{\pm}1.44$, respectively. It was estimated that high quality kochujang could be made by using meju fermented with selected strains.

  • To standardize a manufacturing method and improve the quality of traditional kochujang, eight-types of meju with different shapes (brick, grain) were prepared using Aspergillus oryzae (A.o) or Aspergillus sojae (A.s) alone or in combination with Bacillus subtilis (B.s). The physicochemical characteristics and enzyme activities of the various meju were compared during fermentation for 12 days at $28^{\circ}C$. The moisture content of both the brick- and grain-shaped meju were gradually decreased from an initial content of 50.47 to 54.89% to a content of 12.91 to 16.25% on day 12 of fermentation. The neutral protease activities of the brick-shaped meju ranged from $1.19{\pm}0.12$ to $1.25{\pm}0.28\;unit/mL$, and were similar for all treatments. The ${\alpha}$-amylase activities in A.s+B.s treatment of brick-shaped and grain-shaped meju were the highest, $11{\pm}0.6$ and $9{\pm}0.7\;unit/mL$, respectively. The ${\beta}$-amylase activities ranged from $1.53{\pm}0.01$ to $1.56{\pm}0.02\;unit/mL$, and were similar for all treatments. The amino type nitrogen content of A.o+B.s brick-shaped meju was the highest, $0.39{\pm}0.03%$. We confirmed that the brick-shaped meju prepared with A. oryzae and B. subtilis could be used to prepare traditional kochujang to improve the quality of the product.

Microbial Diversity during Fermentation of Sweet Paste, a Chinese Traditional Seasoning, Using PCR-Denaturing Gradient Gel Electrophoresis

  • The aim of this study was to elucidate the changes in the microbial community and biochemical properties of a traditional sweet paste during fermentation. PCR-denaturing gradient gel electrophoresis (DGGE) analysis showed that Aspergillus oryzae was the predominant species in the koji (the fungal mixture), and the majority of the fungi isolated belonged to two Zygosaccharomyces species in the mash. The bacterial DGGE profiles revealed the presence of Bacillus subtilis during fermentation, and Lactobacillus acidipiscis, Lactobacillus pubuzihii, Lactobacillus sp., Staphylococcus kloosi, and several uncultured bacteria were also detected in the mash after 14 days of main fermentation. Additionally, during main fermentation, amino-type nitrogen and total acid increased gradually to a maximum of $6.77{\pm}0.25g/kg$ and $19.10{\pm}0.58g/kg$ (30 days) respectively, and the concentration of reducing sugar increased to $337.41{\pm}3.99g/kg$ (7 days). The 180-day fermented sweet paste contained $261.46{\pm}19.49g/kg$ reducing sugar and its pH value remained at around 4.65. This study has used the PCR-DGGE technique to demonstrate the microbial community (including bacteria and fungi) in sweet paste and provides useful information (biochemical properties) about the assessment of the quality of sweet paste throughout fermentation.
