Cold pretreatment, washing medium and composition of nutrient media may have marked effects on microspore embryogenesis. When microspores isolated from radish (Raphanus sativus L. cv. Gwanhun) flower buds were washed with Nitsch & Nitsch (NLN) medium liquid medium containing $130g{\cdot}L^{-1}$ sucrose (NLN-13), yields of microspore-derived embryos were greater than when using B5 liquid medium containing $130g{\cdot}L^{-1}$ sucrose. Microspore viability is known to decrease rapidly with storage; however, in this experiment, microspore viability was maintained for 24 h at $4^{\circ}C$ without media. Among the various medium concentrations used ($0.25{\times}$, $0.5{\times}$, $1.0{\times}$, $2.0{\times}$, and $4.0{\times}$ NLN liquid medium), $0.5{\times}$ NLN liquid medium induced the most efficient formation of microspore-derived embryos. In addition, microspore-derived embryos yields were greater when microspores were cultured in $0.5{\times}$ NLN liquid medium supplemented with $0.25{\times}$, $0.5{\times}$, and $1.0{\times}$ NLN microelements, compared to medium not supplemented with microelements. In this study, the highest yield of microspore-derived embryos was observed when the microspores derived from flower buds were washed using NLN-13 liquid medium and then cultured on $0.5{\times}$ NLN liquid medium supplemented with $0.25{\times}$ NLN microelements, followed by incubation at $25^{\circ}C$ for 30 days.
In 2006 to 2007, the potential inoculum source of the anthracnose of sweet persimmon caused by Colletotrichum gloeosporioides was surveyed. The infected twigs, buds, dead twigs, petiole, leaves, dropped fruits were collected and tested for their possibility as overwintering inoculum. The detection rates of the pathogen from various parts of sweet persimmon tree were varied. When the collected samples were examined in April. Over than 93.3% of infected twig samples were harbored mycelia of C. gloeosporioides, and 46.7% of infected buds, 36.7% of dead twigs, 23.3% of petioles, and 16.7% of leaves were beared pathogenic fungus. No pathogenic fungus were detecded from healthy twigs and buds. Infected twigs and bud was important overwintering sites and formed conidia actively in next spring. The infected twigs, leaves, petioles, and fruits in growing season produced great number of conidia and caused active dissemination of the anthracnose disease in sweet persimmon. In growing season, all of the infected parts, such as twigs, leaves, petioles, and fruits produced pathogenic fungus.
This study has been focused on both estrogenic and proliferating activity of genistein (GEN) and bisphenol A (BPA). GEN and BPA enhance the proliferation of estrogen-dependent MCF-7 human breast cancer cells at concentrations as low as 100 nM of GEN and 8 ng/ml of BP A achieving similar effect to that of estradiol at 1 nM. Expression of the estrogen responsive gene, pS2 was also induced in MCF-7 cells by treatment with genistein at dose as low as 1 nM and BPA at dose as low as 4 ng/ml. Using 21 day-old ovariectomized nude mice, we examined end-bud formation and mammary gland development after treatment with bisphenol A or genistein. Compared with untreated control, mammary gland development and end-bud formation were significantly increased in mice fed genistein or bisphenol A (p<0.05). Taken together, it is concluded that GEN and BP A can act as an estrogen agonist resulting in cell proliferation and induction of the estrogen responsive pS2 gene in MCF-7 cells in vitro and in athymic mice in vivo, respectively. Therefore, it is suggested that GEN and BP A might modulate human endocrine system and these compounds might be considered as a endocrine modulator at the low levels of doses.
This study investigated the effects of temperature on the shoot growth and flowering of potted kumquat [Fortunella margarita ( Lour.) Swingle] trees grown in subtropical conditions of I-Lan County in Taiwan. Temperature treatments included T 25-32, T 17-25, T 22, and T 18. The T 25-32 treatment trees were to the day/night temperatures of $25/18^{\circ}C$ for 2 weeks, followed by 28 weeks at $32/25^{\circ}C$. T 17-25 was exposed for 4 weeks to $17/10^{\circ}C$ followed by 26 weeks at $25/18^{\circ}C$. T 22 and T 18 were exposed at $22/18^{\circ}C$ and $18/13^{\circ}C$, respectively, for the entire duration of the experiment. Control trees were placed in a plastic greenhouse under conditions similar to the natural environment. The kumquat trees exposed to high-temperature environment of $32/25^{\circ}C$ showed more frequent and speedy sprouting of new buds, but induced the earlier termination of shoot elongation growth, resulting in decreased vegetative growth. The temperature treatments lower than $22^{\circ}C$ suppressed the new shoot production but increased the shoot growth period, resulting in increased shoot length and diameter. Temperatures higher than $25/18^{\circ}C$ readily induced flowering, with flowering being advanced under the higher temperature conditions such as $32/25^{\circ}C$. However, flowering was substantially inhibited under temperature conditions lower than $22/18^{\circ}C$, indicating the negative role of relatively lower temperatures on flowering of kumquat trees.

Culture conditions for plant regeneration of Camellia japonica were achieved by organogenesis in explants of cotyledon. Seed cotyledons were cultured on MS medium containing various auxin, 2,4-D or NAA and cytokinins BA. The adventitious shoot buds were efficiently formed without embryogenesis on the basal region of cotyledon cultured on MS medium supplemented with $0.1\;{\mu}g/m{\ell}$ 2.4-D and $1\;{\mu}g/m{\ell}$ BA. Seed cotyledons could be used as a source of explants in experiments of genetic transformation of the genotypes evaluated for improving the efficiency of production of transgenic Camellia plants.
Recently, increasing interests in local foods have been highlighted along with active efforts and activities from autonomy organizations to develop local cultures and promote local economies by generating value added products and profits through the development of local foods. Subsequently, useful effects might result by making local foods attractive to tourists, such as by using indigenous Gimpo products to make menu items that incorporate the social, economic, and cultural aspects of the Gimpo area, and contributing to its food culture, the development and use of its indigenous products, and promoting local restaurant businesses, etc. The items of the survey used to select the local food menu items to be developed were limited to ideas offered by expert groups, and were implemented by a new menu development process. In order to derive the menu items to develop, the following method was applied: a brainstorming session with experts to generate and draft ideas, a questionnaire to chefs and cooks in special grade hotels is Seoul to select the menu items to develop from those drafted, and sensory evaluations by experts to evaluate the developed items. Frequency analysis and technical statistical analysis were performed using the SPSS 12.0 program package, where 13 kinds of local foods were ultimately developed using indigenous products of Gimpo. The developed local food menu items were rice & grape sujaebi, grape sikhae, fried rice embryo bud, grape yanggaeng, rice pancakes, rice spaghetti, grape seed oil dressing, grape sauce, rice pizza, grape pie, rice & grape ice cream, grape chocolate, and rice roll cakes.
This study was carried out to investigate the check shelling distribution of Lindera erythrocarpa Makino in Kanghwa Island, MT. Suri, Kyunggido and MT. Kaya, Chungchongnamdo. The results were as follows : The total distribution area of Linedra erythrocarpa where are Jeondeung temple and Jugsu temple Kanghwa Island, MT. Suri, Kyunggido and MT. Kaya, Chungchongnamdo was 344.4ha and the total number of Linedra erythrocarpa was 3,224. The total number of femle Linedra erythrocarpa was625(22.2%) except the seedlings under 2cm of DBH(Diameter breast of height). For the estimation of tree age by counting of tree rings, the regression equation was Y=1.79X+9.47($R^2$=0.83, DBH=cm). The soil acidity of stands studied was $pH4.6{\sim}pH5.8$. And the soil acidity of Seoul was $pH4.2{\sim}pH4.5$ So for the Planting Linedra erythrocarpa in Seoul area, the soil acidity of planting area should be changed to $pH4.6{\sim}pH5.8$. The flower size of and female of Linedra erythrocarpa was 5.96mm, 3.66mm, respectively. The length of petiole of male and female of Linedra erythrocarpa was 5.96mm, 3.66mm, respectively. This paper documents the defining morphological characteristics of the larval stages of Chionoecetes tanneri Rathbun, 1893, the grooved Tanner crab, from specimens reared in the laboratory. Chionoecetes tanneri larval stages include two zoeae and one megalopa. The first zoea is characterized by: six setae on the posterior margin of the carapace; postero-lateral spines on abdominal somites 3 and 4, extending beyond the posterior margin of adjacent somites and bearing 9-10 spinnules; 12 plumose setae and one stout distal plumose seta present on the margin of the scaphognathite of the maxilla; and one fused lateral spine and one articulated dorso-medial spine on each fork of the telson. The second zoea is characterized by: 9 setae on the postero-lateral margin of the carapace; a serrated mandible molar; a mandibular palp bud; 25-26 plumose setae on the margin of the scaphognathite of the maxilla; pereiopods with well-developed gills and buds; and four pairs of stout setae on the posterior margin of the telson. For the megalopal stage, the distinguishing characteristics include: a rostral spine equal in length to the supraorbital spine; six setae on the exopod of the uropod; and a single spine on the ischium of the second pereiopod. This study allows C. tanneri larvae to be distinguished from the larvae of known sympatric congeners. This information provides a basic taxonomic tool for researchers in fisheries management and zooplankton ecology who are addressing issues related to trophic interactions, metapopulation dynamics and ecosystem impacts in the evolving marine resource management strategies in the North Pacific, and those related to Chionoecetes species in particular.
Recently, we identified a domain, termed MIRC3-4, for the protein-protein interaction between yeast chitin synthase 3 (CHS3) and chitin synthase 4 (CHS4). In this study, the functional roles of MIRC3-4 were examined at the G1 phase and cytokinesis of the cell cycle by Calcofluor staining and FISH. Some mutations in MIRC3-4 resulted in disappearance of the chitin ring in the early G1 phase, but did not affect chitin synthesis in the cell wall at cytokinesis. The chitin distribution in chs4 mutant cells indicated that CHS4 was involved in the synthesis of chitinring in the G1 phase and in the synthesis of cell wall chitin after cytokinesis, suggesting that Chs4p regulates chitin synthase 3 activity differently in G1 and cytokinesis. Absence of the chitin ring could be caused either by delocalization of Chs3p to the bud-neck or by improper interaction with Chs4p. When mutant cells were immunostained with a Chs3p-specific antibody to discriminate between these two alternatives, the mutated Ch3p was found to localize to the neck in all MIRC3-4 mutants. These results strongly irdicate that Chs4p regulates Chs3p as an activator but not a recruiter.

The intense drought affecting olive production in Northern Chile underscores the need to research non-traditional irrigation strategies to obtain the best crop performance. Accordingly, this study aimed to obtain preliminary data to guide future research on this topic. Different water replenishment levels on crop evapotranspiration ($ET_c$ ; 13.5, 27.0, 40.5, and 54%) were established in a young orchard, cv. Arbequina, from the end of fruit drop (EFD) to full bloom in the next season. We evaluated the influence of plant water status (${\Psi}_{stem}$ ) and crop load, considered as function of fruit number divided by trunk cross-sectional area, on reproductive and productive variables using multiple linear regressions. Our results show that crop load and ${\Psi}_{stem}$ measured from EFD to harvest affected yield components. Nevertheless, ${\Psi}_{stem}$ had the strongest influence on fruit size, pulp development, oil accumulation, and yield. Oil content and yield were reduced by 54% and 50% for each MPa, respectively, from ${\Psi}_{stem\;EFD-H}$ -1.8 MPa, an effect that intensified as crop load increased. During the period of flower development (September-November), the number of flowers per inflorescence and percentage of perfect flowers were reduced when ${\Psi}_{stem}$ was less than -2.0 MPa. These preliminary results showed that bud differentiation, inflorescence and flower formation are highly sensitive to water deficit.
