이산화탄소의 성질 실험 장치 개선 방안 탐색 (A Study about Improvement of Experimentation on Carbon Dioxide Properties)

  • One of the problems with testing for the presence of $CO_2$ is that the results are not visible. In order to over-come this weak point, a new testing method was developed with BTB indicator equipment, that made the gas visible. After that, the experiment was performed and tested the effects of the new visible equipment. The visible equipment could be adapted to regular class use successfully. Also, it was effective in that it reduced waste of gas, minimized danger potential through use of $CO_2$ canisters and candle-sticks, and also increased knowledge about indicators. The new experimental method and equipment affected the students' interest. It is possible that the students' positive participation was due to their interest in the new apparatus and application of the visual senses. The new system was tested fer its effects on teaching content and helping to produce sustained memory of the content. There were no significant differences between the groups in terms of content learning on initial content memory. However, when students in both groups were tested 4 months later, the visual experiment group sustained memory performance, while the other group showed a significant decrease. Generally, boys score higher than girls in terms of interest and participation in experimental activities. In this case, however, there were no difference between groups. It may have been due to introduction of new equipment and different methods from the textbooks. So, this could increase participation in science using various experiments.

Expression characterization and transcription regulation analysis of porcine Yip1 domain family member 3 gene

  • Objective: The Yip1 domain family (YIPF) proteins were proposed to function in endoplasmic reticulum (ER) to Golgi transport and maintenance of the morphology of the Golgi, which were homologues of yeast Yip1p and Yif1p. YIPF3, the member 3 of YIPF family was a homolog of Yif1p. The aim of present study was to investigate the expression and regulation mechanism of porcine YIPF3. Methods: Quantitative realtime polymerase chain reaction (qPCR) was used to analyze porcine YIPF3 mRNA expression pattern in different tissues and pig kidney epithelial (PK15) cells stimulated by polyinosine-polycytidylic acid (poly [I:C]). Site-directed mutations combined with dual luciferase reporter assays and electrophoretic mobility shift assay (EMSA) were employed to reveal transcription regulation mechanism of porcine YIPF3. Results: Results showed that the mRNA of porcine YIPF3 (pYIPF3) was widely expressed with the highest levels in lymph and lung followed by spleen and liver, while weak in heart and skeletal muscle. Subcellular localization results indicated that it expressed in Golgi apparatus and plasma membranes. Upon stimulation with poly (I:C), the level of this gene was dramatically up-regulated in a time- and concentration-dependent manner. pYIPF3 core promoter region harbored three cis-acting elements which were bound by ETS proto-oncogene 2 (ETS2), zinc finger and BTB domain containing 4 (ZBTB4), and zinc finger and BTB domain containing 14 (ZBTB14), respectively. In which, ETS2 and ZBTB4 both promoted pYIPF3 transcription activity while ZBTB14 inhibited it, and these three transcription factors all played important regulation roles in tumorigenesis and apoptosis. Conclusion: The pYIPF3 mRNA expression was regulated by ETS2, ZBTB4, and ZBTB14, and its higher expression in immune organs might contribute to enhancing ER to Golgi transport of proteins, thus adapting to the immune response.

전처리과정과 분석방법에 따른 막걸리의 총산 함량 비교 (Comparative Study of Total Acidity Content of Makgeolli with Pre-Treatment and Analysis Methods)

  • 막걸리의 총산 함량은 함유된 부유물과 발효과정에서 발생한 탄산가스의 제거 여부 및 분석방법들에 의해 영향을 받는다. 시중 유통되는 막걸리를 여과하여 부유물을 제거하고 magnetic bar로 stirring 하여 탄산가스를 제거한 후 총산을 분석한 결과 살균막걸리(4종)의 여과 전 0.379~0.477% 총산이 여과 후 0.167~0.225%로, 탄산 제거 후에는 0.132~0.170%로 유의적으로 감소하였고(P<0.05), 생막걸리(5종)는 여과 전 0.412~0.467%의 총산이 여과 후에는 0.157~0.365%로(P<0.05), 탄산 제거 후에는 0.143~0.280%로 유의적으로 감소하였다(P<0.05). 총산 분석방법을 비교한 결과 BTB+NR 지시약을 사용한 경우 살균막걸리와 생막걸리의 총산 함량은 각각 0.105~0.123%와 0.105~0.200%로써 막걸리의 총산 기준 규격인 0.5(acetic acid%) 이하로 분석되었다. 반면 AOAC 분석법인 phenolphthalein 용액으로 적정한 경우 살균막걸리와 생막걸리의 총산 함량은 각각 0.129~0.154%와 0.130~0.255%였고, pH 8.2를 적정점으로 한 경우 0.137~0.162%와 0.140~0.279%로 나타나 총산 함량이 높게 나타났다. BTB 용액으로 적정한 경우 BTB+NR 지시약을 사용한 경우보다는 높고 AOAC법보다는 낮은 것으로 확인되었다. 막걸리에 함유된 유기산은 oxalic acid, malic acid, citric acid, lactic acid, acetic acid+succinic acid였으며, malic acid는 H와 I를 제외한 모든 막걸리에서 0.030~0.134 mg/mL, acetic acid+succinic acid는 0.056~0.372 mg/mL, lactic acid는 0.127~0.776 mg/mL로 살균막걸리보다 생막걸리에 비교적 많이 함유되었고, citric acid는 ND~1.159 mg/mL로 분석되었다.

저항형초전도한류기 과도특성을 고려한 EMTDC 모델개발 (Development of EMTDC model for Resistance type Fault Current Limiter considering transient characteristic)

  • Nowadays, one of the serious problems in KEPCO(Korea Electric Power Co-Operation) system is the more higher fault current than the SCC(Short Circuit Capacity) of circuit breaker. There are many alternatives to reduce the increased fault current such as isolations of bus ties, enhancement of SCC of circuit breaker, applications of HVDC-BTB(High Voltage Direct Current-Back to Back) and FCL(fault current limiter). But, these alternatives have some drawbacks in viewpoints of system stability and cost. As the superconductivity technology has been developed, the HTS-FCL(High Temperature Superconductor -Fault Current Limiter) can be one of the attractive alternatives to solve the fault current problem. Under this background, this paper presents the EMTDC(Electro-Magnetic Transient Direct Current) model for resistance type HTS-FCL considering the nonlinear characteristic of final resistance value when quenching phenomena occur.
