Effect of Cultivation Time on the Incidence of Brown Fruit Stem of Glasshouse Sweet Pepper (Capsicum annuum L.) (재배시기가 착색단고추의 과병무름증 발생에 미치는 영향)

  • The objectives of this study were to determine the effect of cultivation time on the incidence of brown fruit stem (BFS) in glasshouse sweet pepper (cv. Special) and to investigate environmental causes of this disorder. The plants transplanted on 31 August (T1) showed more incidence of BFS than those on 24 November (T2) (6.1% vs.2.9%; P<0.01). The BFS symptom began to appear after completion of fruit enlargement, more often around fruit coloring period. Comparing the environmental factors between T1 and T2, with their data collected for 3 weeks around fruit coloring period, the factor that was most likely responsible for BFS incidence was found to be the night-time humidity deficit (HD) ($1.9g{\cdot}m^{-3}\;vs\;2.9g{\cdot}m^{-3}HD$). These results were reconfirmed as T1 was compared to the plants (T3) that were transplanted at a similar time of the following year to T1, but designed to reduce BSF by increasing air HD via heating at night. That is,T3 had much higher night-time HD than T1 ($5.9g{\cdot}m^{-3}\;vs\;1.9g{\cdot}m^{-3}HD$), and showed no incidence of BFS. These results indicated that, to prevent BFS incidence in the winter-harvesting sweet pepper plants, air humidity at night should be controlled low, especially for the fruit coloring period after fruit enlargement period is completed.

Cloning and Characterization of a Gene for Fibrinolytic Enzyme from Bacillus subtilis BB-1 Isolated from Black Bean Chung-kuk (흑두로 제조한 청국에서 분리된 Bacillus subtillus BB-1으로 부터 혈전용해효소 유전자 크로닝 및 특성규명)

  • A bacterium producing five fibrinolytic isozymes was isolated from black bean chung kuk. The bacterium was identified as Bacillus subtilis BB-1 by 16s rDNA sequence homology search. A gene out of five fibrinolytic genes in the Bacillus subtilis BB-1 was cloned by shot-gun method. A Cla I DNA fragment of B. subtilis BB-1 chromosome was cloned in to pBluescript II SK(-) and showed the fibrinolytic activity to bacterial cells. The Cla I DNA fragment was sequenced and the sequences did not show homology with gene for protease or fibrinolytic enzyme genes in other organisms. The Cla I DNA fragment was reduced to 2,142 bp by activity-guided PCR cloning method. The optimum pH and temperature of the enzyme were 5.0 and $35^{\circ}C$, respectively. Substrate specificity of the fibrinolytic enzyme was detected in skim milk, casein, gelatin and blood agar plates. The activity of the enzyme was not detected with these substrates. Taken together, this enzyme is a new fibrinolytic enzyme and may be used to prevent thrombosis and arteriosclerosis.

Effect of Band Spotty Fertilization on the Yield and Growth of Peanut(Arachis hypogaea L.) in Plastic Film Mulching Cultivation (비닐피복 땅콩 재배시 생육 및 수량에 미치는 국소시비 효과)

  • This study was carried out to establish low-input fertilization and seeding technique using the simultaneous with seeding and fertilizer application machine and band spotty applicator which were manufactured for experiment during cultivation of mulching for peanut(Arachis hypogaea L.). The labor hour for seeding by simultaneous with seeding and fertilizing machine was appeared over 90% reduction effect compared with control plot($17.3hr\;10a^{-1}$). In band spotty fertilization plots, the emergence date was delayed about 4 days and the seedling stand rate was decreased 11~18% compared with control plot(man power). The content of total nitrogen of soil after experiment was increased while the contents of organic matter, available phosphate and exchangeable potassium were decreased than before experiment. The content of nitrogen forming nitrate was increased in band spotty fertilization(BSF) plots by increasing the amount of applied fertilizer from early growth stage till the middle growth stage. Growth rate was increased in band spotty fertilization plots and the absorbed amount of phosphate and potassium for peanut were increased in 70% band spotty fertilization plot compared with control plot. Yield of peanut was increased 70% in band spotty fertilization plot due to high pod kernel ratio and ripened pod rate compared with control plot($3,150kg\;ha^{-1}$). It was found that 70% band spotty fertilization was more effective as fertilization method to reduce both environmental pollution and chemical nitrogen fertilizer in plastic film mulching cultivation of peanut.
