Hair Growth Promoting Effect of Radish Crude Saponin Extract on Athymic Nude Mice

  • This study investigates the hair restoration efficacy of selected radish saponin extracts on nude mice. Nude mice genetically predisposed to pattern balding were used in this study. Our study revealed the underlying mechanism of stimulating hair growth in athymic nude mice by repair the nu/nu follicular keratin differentiation defect. Thus, the topical application of radish saponin may represent a novel strategy for the management and therapy of certain forms of alopecia. The term of hair density of PEE treated nude mice were significantly increase as compared with of control nude mice. Histological observation of skin sample showed no hair follicle or only distorted hair follicles were observed in the control samples, in contrast, by the PEE treatment groups showed a fully formed and increased the number of hair follicles up to three times higher than that of control group in terms of the number of hair follicles in nude mouse skin.PEE treated mice the number of BrdU-labeled keratinocytes per anagen follicle increased significantly, especially in the follicular bulbs and outer root sheath compared with the control mice. Moreover, PEE-treated nude mice also exhibited a significant increase in the number of BrdU-labeled epidermal keratinocyte proliferation.

Role of a Burr Hole and Calvarial Bone Marrow-Derived Stem Cells in the Ischemic Rat Brain : A Possible Mechanism for the Efficacy of Multiple Burr Hole Surgery in Moyamoya Disease

  • Objective : This study investigates the role of a burr hole and calvarial bone marrow-derived stem cells (BMSCs) in a transient ischemic brain injury model in the rat and postulates a possible mechanism for the efficacy of multiple cranial burr hole (MCBH) surgery in moyamoya disease (MMD). Methods : Twenty Sprague-Dawley rats (250 g, male) were divided into four groups : normal control group (n=5), burr hole group (n=5), ischemia group (n=5), and ischemia+burr hole group (n=5). Focal ischemia was induced by the transient middle cerebral artery occlusion (MCAO). At one week after the ischemic injury, a 2 mm-sized cranial burr hole with small cortical incision was made on the ipsilateral (left) parietal area. Bromodeoxyuridine (BrdU, 50 mg/kg) was injected intraperitoneally, 2 times a day for 6 days after the burr hole trephination. At one week after the burr hole trephination, brains were harvested. Immunohistochemical stainings for BrdU, CD34, VEGF, and Doublecortin and Nestin were done. Results : In the ischemia+burr hole group, BrdU (+), CD34 (+), and Doublecortin (+) cells were found in the cortical incision site below the burr hole. A number of cells with Nestin (+) or VEGF (+) were found in the cerebral parenchyma around the cortical incision site. In the other groups, BrdU (+), CD34 (+), Doublecortin (+), and Nestin (+) cells were not detected in the corresponding area. These findings suggest that BrdU (+) and CD34 (+) cells are bone marrow-derived stem cells, which may be derived from the calvarial bone marrow through the burr hole. The existence of CD34 (+) and VEGF (+) cells indicates increased angiogenesis, while the existence of Doublecortin (+), Nestin (+) cells indicates increased neurogenesis. Conclusion : Based on these findings, the BMSCs through burr holes seem to play an important role for the therapeutic effect of the MCBH surgery in MMD.

Growth-Suppressing Activity of the Transfected Cx26 on BICR-M1Rk Breast Cancer Cell Line

  • There are accumulating evidences suggesting that connexin (Cx), a gap junction channel-forming protein, acts as a growth suppressor in various cancer cells, and this effect is attributeed to the gap junction-mediated intercellular communication (GJIC). In order to characterize the relationship between the growth-arresting activity of Cx26 and its cytoplasmic localizations after expression, we linked a nuclear export signal (NES) sequence to Cx26 cDNA before transfecting into a rat breast cancer cell line. A confocal fluorescent microscopic observation revealed that the insertion of NES minimized the nuclear expression of Cx26, and increased its cytoplasmic expression, including plasma membrane junctions. Total cell counting and BrdUrd-labeling experiments showed that the growth of the breast cancer cells was inhibited by 74% upon transfection of Cx26-NES, whereas only 9% inhibition was observed with only Cx26 cDNA.

  • The likelihood of failure for the stress corrosion cracking (SCC) of caustic cracking, which affect to a risk of facilities, was analyzed through the risk based-inspection using API-581 BRD. We found that SCC of the caustic cracking was occurred above 5 % NaOH concentration, and the technical module subfactor (TMSF) was maximized for above 50 % concentration. The heat traced and monitoring were not sensitive to the TMSF with NaOH concentration and temperature. But the steam out was more of less affect minimum value of the TMSF. Also, the inspection number, the inspection effectiveness, and the year since inspection were very sensitive to the TMSF with NaOH concentration and temperature. Therefore, the plan of next inspection will be established with compositively considering those at once.

  • A technique is described for producing high resolution G- and R-banded chromosomes in human peripheral lymphocyte cultures. Cultured lymphocyte cells were exposed to ethidium bromide ($10{\mu}g/ml$) and colcemid ($0.02{\mu}g/ml$) each for 2.5h and 0.5h prior to harvest for high resolution G-banded chromosomes. High resolution R-band patterns were obtained by BrdU substitution which was revealed by the fluorochrome-photolysis-Giemsa staining technique. These methods are easy to perform and highly reliable. The data on relative length of chromosomes at the four mitotic stages are presented in units of percentage of haploid autosome length. The characteristic patterns of GTG-bands (G-bands after trypsin and Giemsa) and RBG bands (R-bands after BrdU and Giemsa) were analyzed.

  • For consequence analysis at a gas explosion by vessel release, release rates, demage areas of facilities, and fatality areas were estimated and analyzed at various conditions(release materials, temperatures, pressures, and vessel types) by using the release scenario of API-581 BRD. Simulation results showed that release rates and consequences in the vessel release were higher than those in the pipe release, and the order of release rates and damage areas was as follows; tank>reactor>drum> column.
