Breast Milk-Transmitted Cytomegalovirus Infection in Preterm Infants

  • Purpose: The purpose of this study is to describe the rate of cytomegalovirus (CMV) virolactia, and the prevalence of breast milk (BM)-transmitted postnatal CMV infection among premature infants after freeze-thawing (FT) and Holder pasteurization (HP) of breast milk. Methods: This is a single-center, retrospective study of 312 infants born at less than 32 weeks of gestation, or with a birth weight less than 1,500 g from January 2013 to June 2017. All infants were screened for CMV-specific immunoglobulin (Ig) G and IgM at birth. Initial CMV specific polymerase chain reaction (PCR) and CMV culture were performed on mothers' BM and babies' urine within the first 21 days of life. FT and HP of BM was used to prevent the transmission of CMV. For the surveillance of postnatal CMV infection, CMV culture and CMV specific PCR of urine from babies were repeated one to two months after the initial screening. Screening for viremia and viruria was performed if postnatal CMV infection was suspected. Results: Among 178 BM samples obtained from mothers of CMV-IgG-seropositive infants, 80 (44.9%) were CMV PCR positive. CMV deoxyribonucleic acid (DNA) was detected in five of the 22 BM samples (22.7%) obtained from the mothers of CMV-IgG seronegative infants. When CMV DNA load in BM was measured before and after HP, various results were shown. Sixty-three infants out of 232 (27.2%) were evaluated for postnatal CMV infection and four infants out of 63 (6.3%) were infected. Conclusion: Interventions to prevent BM-transmitted CMV infection can reduce the chance of postnatal CMV infection, but not completely eliminate it.

A Detection Algorithm Study of the Victim Signal for the DAA Regulation in MB-OFDM UWB System (MB-OFDM UWB 시스템에서 DAA 기술 기준 적용을 위한 피 간섭 신호 검출 방안 연구)

  • The purpose of this paper is to propose a detection algorithm and a tracking algorithm based on silent time using MB-OFDM UWB(Multi-Band Orthogonal Frequency Division Multiplexing Ultra Wide Band) receiver in order to satisfy DAA(Detect And Avoid) regulation of Korea to permit UWB in 3.1~4.8 GHz. In DAA regulation of Korea, if UWB device receives a signal more than -80 dBm/MHz from the victim system during UWB operation, the UWB system should avoid the collision within 2 sec. In this paper, we proposed the detection algorithm to detect the victim signal received by -80 dBm/MHz for the avoidance process that changes the operating UWB frequency to other UWB frequency and the subcarrier tracking algorithm to follow up the subcarrier positions of the victim signal for the tonenulling avoidance process that decreases the TX power of subcarriers occupied by the victim signal by -70 dBm/MHz. The performance of the detection algorithm and the tracking algorithm suggested in this paper is verified in simulation results considering various conditions.

Production of the BmCecB1 antimicrobial peptide in transgenic silkworm

  • This peptide has antibacterial activity against several Gram-positive and Gram-negative bacteria. Bombyx mori cecropinB1(BmCecB1) is antimicrobial peptides from Bombyx mori and belongs to cecropin family. Antimicrobial peptides are important components of the innate immune systems in all living organism. To produce the BmCecB1 antimicrobial peptide, we constructed transgenic silkworm that expressed BmCecB1 gene under the control BmA3 promoter using piggyBac vector. The use of the 3xP3-driven EGFP cDNA as a marker allowed us to rapidly distinguish transgenic silkworm. Mixtures of the donor vector and helper vector were micro-injected into 600 eggs of bivoltin silkworms, Baegokjam. In total, 49 larvae (G0) were hatched and allowed to develop into moths. The resulting G1 generation consisted of 22 broods, and we selected 2 broods containing at least 1 EGFP-positive embryo. The rate of successful transgenesis for the G1 broods was 9%. We identified 9 EGFP-positive G1 moths and these were backcrossed with wild-type moths. With the aim of identifying a BmCecB1 as antimicrobial peptide, we investigated the Radical diffusion Assay (RDA) and then demonstrated that BmCecB1 possesses high antibacterial activities against Gram-negative bacteria.

Gelatinase, a Possible Etiologic Factor of Photoaging, is Present in Healthy Human Facial Skin and is Inhibited by Turmeric Extract

  • Influence of gelatinase on basement membrane (BM) structure was investigated by using a skin equivalent (SE) model. The results showed that (1) gelatinase produced by cells degraded the BM and (2) the addition of matrix metalloproteinase-specific inhibitor to the SE medium accelerated the formation of BM structure, indicating that gelatinase is involved in BM impairment. The activity of gelatinase was also studied in healthy human facial skin tissues. The result of in situ zymography revealed gelatinase activity around the basal layer of the epidermis, where BM integrity was severely compromised. Therefore, this enzyme was suggested to be associated with BM decomposition in human facial skin. To assess the behavior of gelatinase in stratum corneum (SC) non-invasively, an immunological study was performed. Since positive immunostaining of pro-gelatinase B was observed in SC stripped from sun-exposed skin, whereas no positive staining detected in SC of non-irradiated skin, gelatinase in the epidermis could be non-invasively detected by measuring gelatinase in SC. Gelatinase in SC of healthy female volunteers was monitored using a special film that sensitively and conveniently detects gelatinase. Ninetr percent of SC from facial skin (l00 women, 40's-50's) was gelatinase-positive. On the other hand, SC from non-irradiated skin was negative. These results strongly suggest that (1) gelatinase is constantly produced in the facial epidermis of most middle-aged woman during their daily life, and (2) the enzyme might be involved in the aging-related degeneration of both BM and the matrix fibers of the upper layer of the dermis, acting as a very important aging factor. Strong inhibitory activity against gelatinase was found in turmeric extract and identified curcumin as the major ingredient. Topical application of cream containing turmeric extract significantly decreased the number of gelatinase-positive SC clusters in human facial skins. These results indicated that turmeric is an effective ingredient to prevent skin from photo aging by suppressing chlonically upregulated gelatinase activity by UV and to improve skin condition.

Mutation of Cellulose Synthase Gene Improves the Nutritive Value of Rice Straw

  • Rice straw is an important roughage resource for ruminants in many rice-producing countries. In this study, a rice brittle mutant (BM, mutation in OsCesA4, encoding cellulose synthase) and its wild type (WT) were employed to investigate the effects of a cellulose synthase gene mutation on rice straw morphological fractions, chemical composition, stem histological structure and in situ digestibility. The morphological fractions investigation showed that BM had a higher leaf sheath proportion (43.70% vs 38.21%, p<0.01) and a lower leaf blade proportion (25.21% vs 32.14%, p<0.01) than WT. Chemical composition analysis showed that BM rice straw was significantly (p<0.01) higher in CP (crude protein), hemicellulose and acid insoluble ash (AIA) contents, but lower in dry matter (DM), acid detergent fiber (ADFom) and cellulose contents when compared to WT. No significant difference (p>0.05) was detected in neutral detergent fiber (NDFom) and ADL contents for both strains. Histological structure observation indicated that BM stems had fewer sclerenchyma cells and a thinner sclerenchyma cell wall than WT. The results of in situ digestion showed that BM had higher DM, NDFom, cellulose and hemicellulose disappearance at 24 or 48 h of incubation (p<0.05). The effective digestibility of BM rice straw DM and NDFom was greater than that of WT (31.4% vs 26.7% for DM, 29.1% vs 24.3% for NDFom, p<0.05), but the rate of digestion of the slowly digested fraction of BM rice straw DM and NDF was decreased. These results indicated that the mutation in the cellulose synthase gene could improve the nutritive value of rice straw for ruminants.

Harmful Materials treatment in Shipboard sewage by SBR process with BM (BM 미생물제제를 이용한 선박 오·폐수 내 유해물질처리)

  • Lab scale experiment study was carried out for biological treatment process development in shipboard. SBR(Sequence Batch Reactor) process with BM(Beneficial Microorganisms) was investigated for practical application on shipboard sewage treatment. From the results it was suggested that SBR process with BM might be a suitable process in terms of harmful materials removal. By adding BM to SBR system, the useful species of microorganisms and EPS(Extracellar Polymetric Substances) in sludge was increased. It was found that the biodegrability and harmful organic compounds like VOCs and harmful inorganic compounds like heavy metals. was reduced over 70%. As far as reclamation water is considered, this process is very advantageous to special environments such as cruise ships, because the method of adding BM makes it unnecessary to add other facilities on the SBR system.

Soluble expression, purification and the role of C-terminal glycine residues in scorpion toxin BmK AGP-SYPU2

  • The existence of glycine residues in long-chain scorpion toxins has been well documented. However, their role as analgesics has not been evaluated. To address this issue, we investigated the functional role of glycines in the C-terminal end of Chinese-scorpion toxin from Buthus martensii Karsch (BmK AGP-SYPU2) using site-directed mutagenesis and analgesic activity assays. Recombinant BmK AGP-SYPU2 and its mutants were efficiently expressed in E. coli and purified to homogeneity using immobilized metal ion affinity chromatography (IMAC) and cation exchange chromatography. The mouse-twisting test was used to detect the analgesic activity of BmK AGP-SYPU2 and its mutants. As a result, we identified glycines at the C-terminal end that, when altered, significantly affected analgesic activity. Also, Mut6566 was significantly decreased compared to BmK AGP-SYPU2. These data indicate that the glycines at the C-terminal end are important for the analgesic activity of BmK AGP-SYPU2.

Parameter Estimation and Fitting Error Analysis of the Representative Spectrums using the Wave Spectrum off the Namhangjin, East Sea (남항진 파랑 스펙트럼 정보를 이용한 대표 스펙트럼 매개변수 추정 및 분석)

  • The parameters of the modified BM and JONSWAP spectra are estimated using spectral data set off Namhangjin, located in the east coast of Korea, collected during high wave events. The parameters of the modified BM spectrum were estimated to be 1.04 and 0.27, which were similar to the conventional values of 1.098 and 0.30, but showed significant differences in statistical terms. On the other hand, the peak enhancement factor of JONSWAP spectrum was estimated to be 1.4, which was substantially small compared to the conventional value of 3.3. The RMSE differences from the fitted results of the two spectra were small, approximately 0.2. In the frequency range greater than the peak frequency, however, the spectral energy density showed relatively mild decrease with increase of the frequency, compared to the standard forms of the modified BM and JONSWAP spectra.

A 77GHz MMIC Transceiver Module for Automotive Forward-Looking Radar Sensor

  • A 77GHz MMIC transceiver module consisting of a power amplifier, a low noise amplifier, a drive amplifier, a frequency doubler and a down-mixer has been developed for automotive forward-looking radar sensor. The MMIC chip set was fabricated using $0.15{\mu}m$ gate-length InGaAs/InAlAs/GaAs mHEMT process based on 4-inch substrate. The power amplifier demonstrated a measured small signal gain of over 20dB from $76{\sim}77GHz$ with 15.5dBm output power. The chip size is $2mm{\times}2mm$. The low noise amplifier achieved a gain of 20dB in a band between $76{\sim}77\;GHz$ with an output power of 10dBm. The chip size is $2.2mm{\times}2mm$. The driver amplifier exhibited a gain of 23dB over a $76{\sim}77\;GHz$ band with an output power of 13dBm. The chip size is $2.1mm{\times}2mm$. The frequency doubler achieved an output power of -16dBm at 76.5GHz with a conversion gain of -16dB for an input power of 10dBm and a 38.25GHz input frequency. The chip size is $1.2mm{\times}1.2mm$. The down-mixer demonstrated a measured conversion gain of over -9dB. The chip size is $1.3mm{\times}1.9mm$. The transceiver module achieved an output power of 10dBm in a band between $76{\sim}77GHz$ with a receiver P1dB of -28dBm. The module size is $8{\times}9.5{\times}2.4mm^3$. This MMIC transceiver module is suitable for the 77GHz automotive radar systems and related applications in W-band.

Implementation of the Vehicle Antenna for the AM/FM/TDMB/GPS Receiver (AM/FM/TDMB/GPS 수신용 전장 안테나 제작)

  • In this paper, a vehicle antenna for AM, FM, TDMB, GPS systems was designed and implemented. AM antenna loaded into small space of shark shape was designed by ferrite turn style antenna. For the FM and TDMB antenna, folded monopole antenna which helical is folded was applied. GPS antenna for achieving characteristics both bandwidth and gain improvement was designed microstrip patch that has air substrate and fractal structure. Receiving signal strengths by the measurement were presented for the AM, FM and TDMB antenna. AM signal strength was -65.7dBm, this strength is almost as same conventional pole antenna as -63.4dBm. It can be replaced conventional pole or glass antenna by the studied antenna. Signal strengths for FM and TDMB were -55.66 and -43.50dBm at center frequency, they are 5~10dB higher than conventional antenna. Measurements of bandwidth and gain for the GPS antenna showed 135 MHz under VSWR 2 : 1 and 4.31dBi, gains over GPS band were 3~5dB higher than ceramic patch antenna.
