Effects of Mung Bean on Serum BUN and Enzyme Activities in Hyperlipidemic Rats Induced by High Cholesterol Diet (고지혈증 유발 흰쥐에서 녹두가 혈청 BUN 및 효소활성에 미치는 효과)

  • The purpose this study was to investigate the influences of 5% mung bean (Phaseolus aureus L.) on BUN and enzyme activities in serum of hyperlipidemic rats. Sprague-Dawley(SD) rats (24 male) were divided into four groups, namely the BD group(normal-nonhyperlipidemic diet), BM group(normal-nonhyperlipidemic diet+5% mung bean), BH group(control-hyperlipidemic diet), and BHM group(hyperlipidemic diet+5% mung bean). Serum concentrations of blood urea nitrogen (BUN) and uric acid were significantly decreased (p<0.05) by mung bean supplementation diet. The activities of AST, ALT, ALP, LDH, amylase and lipase in sera of mung bean diet group were significantly decreased (p<0.05). The catalase activity in serum of mung bean supplementation group was significantly increased than hyperlipidemic diet (p<0.05). In vivo experiment with hyperlipidemic rats showed that ingestion of mung bean were effective in kidney and hepatic functional enzyme activities. Which suggests that mung bean material could be used for further studies as a potential source for nutraceutical foods.

Adsorption of phosphate and mitigation of biofouling using lanthanum-doped quorum quenching beads in MBR

  • The removal of phosphorus, especially phosphate-form phosphorus, is necessary in wastewater treatment. Biofouling induced by the quorum sensing mechanism is also a major problem in membrane bioreactor (MBR), which reduces membrane flux. This study introduces lanthanum-doped quorum quenching (QQ) beads into MBR, confirming their inhibitory effect on biofouling due to Rhodococcus sp. BH4 and their capacity for phosphorus removal through lanthanum adsorption. A batch test was conducted to access the phosphate adsorption of lanthanum-QQ (La-QQ) beads and lab-scale MBR to verify the effect of inhibition. The study aimed to identify distinctions among the MBR, QQ MBR, and La-QQ MBR. In the batch test, the phosphate removal rate increased as the volume of beads increased, while the unit volume removal rate of phosphate decreased. In the lab-scale MBR, the phosphate removal rates were below 20% in the control MBR and QQ MBR, whereas the La-QQ MBR achieved a phosphate removal rate of 74%. There was not much difference between the ammonia and total organic carbon (TOC) removal rates. Regarding the change in transmembrane pressure(TMP), 3.7 days were taken for the control MBR to reach critical pressure. In contrast, the QQ-MBR took 9.8 days, and the La-QQ MBR took 6.1 days, which confirms the delay in biofouling. It is expected that La-QQ can be used within MBR to design a more stable MBR process that regulates biofouling and enhances phosphate removal.

Development of Calcification-Resistant Bovine Pericardium with $PEO-SO_3$ (I) - An implantation study of bovine pericardium at aorta and pulmonary artery in canine model - ($PEO-SO_3$를 이용한 항석회화 조직첨포의 개발 (I) - 잡견을 이용한 대동맥과 폐동맥 이식 실험연구 -)

  • Background: Calcific degeneration is unavoidable in either homo- or heterografts implanted in the human body. We have developed a calcification-resistant cardiovascular tissue patch using a novel technique of anticalcification. Materials and methods: Fresh bovine pericardium was harvested at the slaughter house and transfered to the laboratory in Hank's solution. After trimming and fixing the pericardium, it was embedded in 4$^{\circ}C$ 0.65% glutaraldehyde for a week and then washed by phosphate-buffered saline(PBS) of pH 7.4. This prepared pericardium was then stored in 2.5% sulphonated polyethyleneoxide(PEO-SO3) solution for 2 days at room temperature and reversed by 4$^{\circ}C$ NaBH4 solution for 16 hours. To evaluate the calcification-resistance of surface modified bovine pericardium with PEO-SO3, either glutaraldehyde- treated(GA group, n=4) or PEO-SO3-treated pericardial patch(PEO-SO3 group, n=4) was implanted into adult mongrel dog to reconstruct the main pulmonary artery and the descending aorta using a partial clamp technique. After 1 month follow-up, the implanted patches were retrieved to evaluate the pathologic findings and the content of calcium and phosphorous. Results: The PEO-SO3 group showed substantially less retraction and significantly less calcium deposition than the GA group in both aortic(7.10$\pm$1.05 vs. 13.81$\pm$2.33 mg/g of dried tissue) and pulmonary positions(1.55$\pm$0.29 vs. 6.72$\pm$0.70 mg/g)(p<0.01). Phosphorous contents were also less in the PEO-SO3 group than the GA group significantly, 8.11$\pm$1.07 mg/g vs. 19.33$\pm$4.31 mg/g in the aortic and 2.58$\pm$0.40 vs. 12.60$\pm$3.40 mg/g in thepulmonary position(p<0.01). Conclusions: These findings suggest that PEO-SO3 modified bovine pericardium is highly calcification-resistant but further study is needed to evaluate the long-term biological safety and compatibility of the prosthesis.

  • Rabbitfish hatchlings were given a mixed food of rotifers (Brachionus rotundiformis) and trochophore larvae of oyster. Only the oyster-trochophore larvae were found in the gut of 62-h old fish larvae. The fish larvae, fed on rotifer and ciliate alone did not survive. However, their survival increased to 3.3 % on the 10th day after hatching, when trochophore was supplemented. Corresponding with the accelerated growth, the number of rotifers consumed increased from 11 in a 5-day old fish to 165 in a IS-day old fish. In a field ecosystem containing live diatom, Nannochloropsis oculata, rotifers and copepods, fish larvae were shocked and the oyster's trochophore larvae were fed from 2 to 7 days after hatching. A total of 76,000 seedling were produced after 50 days of hatching with 12.7 % survival. Mean total length and body weight were 65.6 mm and 3.4 g, respectively. Growth of body length (BL), body height (BH), body weight (BW) and head length (HL) as a function of the total length (TL) showed regressional relationships as follows; BL=0.8565 TL+0.0852 ($t^2$=0.9996); BH=0.3207 TL - 0.5052 (($t^2$=0.9641) BW=0.0652 TL2.3508 (($t^2$=0.9925); HL=0.2595 TL - 0.1898 (($t^2$=0.9901)

Influence of Ca Reduction Process on the Properties of Nanocrystalline Nd-Fe-B Powders Prepared by a Thermochemical Process (열화학공정으로 제조된 나노결정형 Nd-Fe-B 분말의 특성에 미치는 Ca환원 공정의 영향)

  • Nanocrystalline Nd-Fe-B powder was synthesized by a new thermochemical process that combined with past reduction-diffusion process and spray-dry process. In this process, Ca reduction process is vary important due to formation of hard magnetic$Nd_{2}Fe_{14}B$ phase from various oxides by Ca powder. Therefore, the final products are essentially affected a shape, size, and composition etc. of the Ca reduced powders. Ca reduction was performed to way that raw powders just mixed with Ca powder in proper ratio unlike to compress into compact. The powders after mixture-type Ca reduction mainly composited with $Nd_{2}Fe_{14}B$ phase even relativily low reaction temperature ($800^{\circ}C$) and all particle size of powder were distributed less than 1 ${\mu}m$ except for powder after Ca oxides as magnetic properties of powders after cake-type Ca reduction, with the consequence that high magnetic properties has been expected. The magnetic properties of powders prepared by mixture-type Ca ruduction, with the conseqence that high magnatic properties has been expected. The magnetic properties of powders prepared by mixture-type Ca reduction process showed $_iH_c$ = 5.9 kOe, $B_r$ = 5.5 kG, (BH)max = $Nd_{2}Fe_{14}B{\to}Nd_{2}Fe_{17}B$ decomposition by violent exothermic reaction during washing.

  • The two school $M{\bar{a}}dhyamika$ and $Yog{\bar{a}}c{\bar{a}}ra$ act as a representative of $Mah{\bar{a}}y{\bar{a}}na$ Buddhism in India. But the two school disputed with each other insisting ${\acute{s}}{\bar{u}}nyat{\bar{a}}-v{\bar{a}}da$ and $vij{\tilde{n}}aptim{\bar{a}}trav{\bar{a}}da$ separately. To introduce the disputation shortly is as follow. $N{\bar{a}}g{\bar{a}}rjuna$ explained the world and truth by two truth theories(二諦說) which carry out truth of a word and the dimension(spiritual enlightenment) which is absolute(ultimate) to it being lokasaṁvṛtisatya(世俗諦) about the truth which can be expressed verbally, and which is phenomenon-like (everyday) at paramaarthasatya(勝義諦). By the way, lokasaṁvṛtisatya and paramaarthasatya are actually distinction of the recognition which is not an ontological distinction. That is, lokasaṁvṛti(世俗) is paramaartha(勝義) as it is the time of seeing by the eyes of those who have realized. The two truth theories of $N{\bar{a}}g{\bar{a}}rjuna$ was developed logical more precisely by his successors. With an everyday language, the position of Candrakīrti(月稱) that it cannot be expressed as the position of $Bh{\bar{a}}vaviveka$(淸辨) that paramaarthasatya can be expressed logically is opposed to each other, and dissociates by $Sv{\bar{a}}tantrika$(自立論證派) and $P{\bar{a}}rsagika$(歸謬論證派). Confrontation of $Sv{\bar{a}}tantrika$ and $P{\bar{a}}rsagika$ is the dispute about the ability of s which is the highest truth to be proved logically. The $P{\bar{a}}rsaga$ of Candrakirti thinks that people exist truly, and is because it claims not existing in the world where a favorite thing is actually actual. However, $Bh{\bar{a}}vaviveka$ proved Sunyata(空性)을 positively based on the reliance to language and logic. Also the mokṣa of $M{\bar{a}}dhyamika$ is not recovery of original condition of $vij{\tilde{n}}apti$ which is pure in itself as $Yog{\bar{a}}c{\bar{a}}ra$ saying, as well as obtaining a thing which is dravya-sat as $Sarv{\bar{a}}stiv{\bar{a}}din$ saying. The mokṣa of $M{\bar{a}}dhyamika$ means a condition of liberated from karma and pains through extinction of $prapa{\tilde{n}}ca$ and discrimination by realizing the real aspect of all dharma which is said by pratītyasamutpāda, $praj{\tilde{n}}apti$, niḥsvabhāva, ${\acute{s}}{\bar{u}}nya$, $madhyam{\bar{a}}pratipad$.

The Patterns of Change in Arterial Oxygen Saturation and Heart Rate and Their Related Factors during Voluntary Breath holding and Rebreathing (자발적 호흡정지 및 재개시 동맥혈 산소포화도와 심박수의 변동양상과 이에 영향을 미치는 인자)

  • Background : In sleep apnea syndrome, arterial oxygen saturation($SaO_2$) decreases at a variable rate and to a variable degree for a given apneic period from patient to patient, and various kinds of cardiac arrythmia are known to occur. Factors supposed to affect arterial oxygen desaturation during apnea are duration of apnea, lung voulume at which apnea occurs, and oxygen consumption rate of the subject. The lung serves as preferential oxygen source during apnea, and there have been many reports related with the influence of lung volume on $SaO_2$ during apnea, but there are few, if any, studies about the influence of oxygen consumption rate of an individual on $SaO_2$ during breath holding or about the profile of arterial oxygen resaturation after breathing resumed. Methods : To investigate the changes of $SaO_2$ and heart rate(HR) during breath holding(BH) and rebreathing(RB) and to evaluate the physiologic factors responsible for the changes, lung volume measurements, and arterial blood gas analyses were performed in 17 healthy subjects. Nasal airflow by thermistor, $SaO_2$ by pulse oxymeter and ECG tracing were recorded on Polygraph(TA 4000, Gould, U.S.A.) during voluntary BH & RB at total lung capacity(TLC), at functional residual capacity(FRC) and at residual volume(RV), respectively, for the study subjects. Each subject's basal metabolic rate(BMR) was assumed on Harris-Benedict equation. Results: The time needed for $SaO_2$ to drop 2% from the basal level during breath holding(T2%) were $70.1{\pm}14.2$ sec(mean${\pm}$standard deviation) at TLC, $44.0{\pm}11.6$ sec at FRC, and $33.2{\pm}11.1$ sec at RV(TLC vs. FRC, p<0.05; FRC vs. RV, p<0.05). On rebreathing after $SaO_2$ decreased 2%, further decrement in $SaO_2$ was observed and it was significantly greater at RV($4.3{\pm}2.1%$) than at TLC($1.4{\pm}1.0%$)(p<0.05) or at FRC($1.9{\pm}1.4%$)(p<0.05). The time required for $SaO_2$ to return to the basal level after RB(Tr) at TLC was not significantly different from those at FRC or at RV. T2% had no significant correlation either with lung volumes or with BMR respectively. On the other hand, T2% had significant correlation with TLC/BMR(r=0.693, p<0.01) and FRC/BMR (r=0.615, p<0.025) but not with RV/BMR(r=0.227, p>0.05). The differences between maximal and minimal HR(${\Delta}HR$) during the BH-RB manuever were $27.5{\pm}9.2/min$ at TLC, $26.4{\pm}14.0/min$ at RV, and $19.1{\pm}6.0/min$ at FRC which was significantly smaller than those at TLC(p<0.05) or at RV(p<0.05). The mean difference of 5 p-p intervals before and after RB were $0.8{\pm}0.10$ sec and $0.72{\pm}0.09$ sec at TLC(p<0.001), $0.82{\pm}0.11$ sec and $0.73{\pm}0.09$ sec at FRC(p<0.025), and $0.77{\pm}0.09$ sec and $0.72{\pm}0.09$ sec at RV(p<0.05). Conclusion Healthy subjects showed arterial desaturation of various rates and extent during breath holding at different lung volumes. When breath held at lung volume greater than FRC, the rate of arterial desaturation significantly correlated with lung volume/basal metabolic rate, but when breath held at RV, the rate of arterial desaturation did not correlate linearly with RV/BMR. Sinus arrythmias occurred during breath holding and rebreathing manuever irrespective of the size of the lung volume at which breath holding started, and the amount of change was smallest when breath held at FRC and the change in vagal tone induced by alteration in respiratory movement might be the major responsible factor for the sinus arrythmia.

  • The chemical structure of glycolipid of Selenomonas ruminantium cell wall was to be elucidated. The bacterial cells were treated in hot TCA and the glycolipid fractions were extracted by the solvent $CHCl_3\;:\;CH_3OH$ (1 : 3). The extracted glycolipids fraction was further separated by acetone extraction. The acetone soluble fraction was named as the spot A-compound. The acetone insoluble but ether soluble fraction was named as the spot B-compound. These two compounds were examined for elucidation of their chemical structure. The results were as follows: 1. The IR spectral analysis showed that O-acyl and N-acyl fatty acids were linked to glucosamine moiety in the spot A-compound. However in the spot B-compound in addition to O and N-acyl acids phosphorus was shown to be attached to glucosamine. 2. It was recognized by gas liquid chromatography that spot A compound contained beta-OH $C_{13:0}$ fatty acid in predominance in addition to the fatty acid with beta-OH $C_{9:0}$, whereas the spot B compound was composed of the predominant fatty acid of beta-OH $C_{13:0}$ with small amount of beta-OH $C_{9:0}$. 3. According to the paper chromatographic analysis of hydrazinolysis products of the spot A compound, a compound of a similar Rf value as the chitobiose was recognized, which indicated a structure of two molecules glucosamine condensed. The low Rf value of the hydrazinolysis product of the spot B-compound confirmed the presence of phosphorus attached to glucosamine. 4. The appearance of arabinose resulting from. ninhydrin decomposition of the acid hydrolyzate of the spot A compound indicated that the amino group is attached to $C_2$ of glucosamine. 5. The amount of glucosamine in the N-acetylated spot A compound decreased in half of the original content by the treatment. with $NaBH_4$, indicating that there are two molecules of glucosamines in the spot A compound. The presence of 1, 6-linkage between two molecules of glucosamine was suggested by the Morgan-Elson reaction and confirmed by the periodate decomposition test. 6. By the action of ${\beta}-N-acetyl$ glucosaminidase the N-acetylated spot A compound was completely decomposed into N-acetyl glucosamine, whereas the spot B compound was not. This indicated the spot A compound has a beta-linkage. 7. When phosphodiesterase or phosphomonoesterase acted on $^{32}P-labeled$ spot B compound, $^{32}P$ was not released by phosphodiesterase, but completely released by phosphomonoesterase. This indicated that one phosphorus is linked to glucosamine moiety. 8. The spot A compound is assumed to have the following chemical structure: That is glucosaminyl, ${\beta}-1$, 6-glucosamine to which O-acyl and N-acyl fatty acids are linked, of which the predominant fatty acid is beta-OH $C_{13:0}$ fatty acid in addition to beta-OH $C_{9:0}$ fatty acid 9. The spot B compound is likely to have the linkage of $glucosaminyl-{\beta}-1$, 6-glucosamine to which phosphorus is linked in monoester linkage. Furthermore both O-acyl and N-acyl fatty acids contained beta-OH $C_{13:0}$ fatty acid predominantly in addition to beta-OH $C_{9:0}$ fatty acid.

Stability Analysis for the Pohang Deep Geothermal Borehole (포항 심부 지열 시추공의 안정성 분석 연구)

  • This paper presents the analysis about the stability of the Pohang deep geothermal borehole drilled in 2006. Severe wellhole instability problems such as collapse and tight hole occurred in weak rocks while drilling. Optimal mud pressure (mud window) required to prevent instability problems during drilling is obtained from analysis on in-situ stress and rock strength. The window is bounded by vertical stress in its upper limit and by either collapse pressure or pore pressure in its lower limit. Mud window varies with different types of rocks. In the top-most semi-consolidated mudstone formation, no mud window can secure borehole stability. In some weak rock types (basic dyke and crystal tuff), the borehole pressure needs to be higher by $50{\sim}60%$ than hydrostatic pressure. That means a mud density of 1.5 g/$cm^3$ or higher should be applied during drilling in order to prevent excessive collapse around the borehole.

Effect of Au content on the electro-catalytic activity of Pt catalyst for Pt-Au/C composite catalyst (Pt-Au/C 복합촉매에 있어서 Au 혼합비가 Pt 촉매의 활성에 미치는 영향)

  • 고분자 전해질막 연료전지(Polymer Electrolyte Membrane Fuel Cell; PEMFC)는 수소를 이용하여 전기를 발생시키는 친환경적이고 이상적인 발전장치로 고효율과 높은 전류밀도를 가지며 그 응용분야가 다양하다. 저온에서 작동하는 PEM fuel cell은 전극에서 효과적인 산화환원반응을 위해 그 촉매로 활성이 우수한 Pt(Platinum)을 사용하고 있으나, Pt의 높은 가격은 연료전지의 상용화에 걸림돌이 되고 있다. 본 연구에서는 연료전지의 Pt/C 촉매 층에서 Pt의 분산성을 높여 Pt의 담지량을 줄이고 작동 중 발생하는 Pt의 응집 현상을 방지하여 Pt의 수명을 연장시킬 목적으로, Au(gold) 나노입자를 첨가한 Pt-Au/C 복합나노촉매를 제조하였다. 본 발표에서는 합성된 Pt-Au/C 복합촉매 중 Au 첨가량이 Pt 촉매의 활성에 미치는 영향을 조사하기 위하여, 복합촉매 중에 금속(Pt+Au)의 총 함량이 30 wt.%와 40 wt.% 인 Pt-Au/C 촉매에 대하여 각각 Au 첨가량을 변화시켜, cyclic voltammetry 법에 의해 Au 첨가 효과를 조사한 결과에 대하여 보고하고자 한다. Au 나노입자를 제조하기 위한 출발 물질로는 $HAuCl_4{\cdot}4H_2O$를 이용하였고 trisodium citrate와 $NaBH_4$를 환원제로 하여, 입경이 5~8 nm 인 Au 콜로이드를 제조하였다. Pt-Au/C 복합나노촉매를 제조하기 위하여 먼저 Au/C 복합분체가 제조되었다. 0.03g의 carbon이 첨가된 carbon 현탁액에 합성된 Au 콜로이드 수용액을 첨가한 후 24시간 동안 교반하여 Au/C 복합분체를 제조하였다. 이 Au/C 복합분체에 $H_2PtCl_6{\cdot}6H_2O$ 수용액을 현탁하고 methanol 을 환원제로 사용해 Pt를 환원 석출시켜 Pt-Au/C 복합촉매를 제조하였다. Pt-Au/C 복합 나노촉매에서 Pt와 Au를 다양한 비율(3:1, 2.5:1.5, 2:2)로 합성하였으며 Pt-Au/C 복합촉매 중 금속(Pt+Au) 촉매의 총 함량은 30 wt.%와 40 wt.%로 각각 제조되었다. Au 나노입자 콜로이드의 분산성은 UV-visible spectrum의 흡광도에 의해 관찰되었고, Pt-Au/C 복합 나노촉매의 형상 및 분산성 분석은 transmission electron microscopy(TEM)에 의해 이루어졌다. 또한, 촉매의 전기화학적 특성평가는 cyclic voltammetry(CV)에 의해 조사되었다.
