Crossability, Variation in Chromosome Number, and Agronomic Characters in Hybrid of Hexaploid Triticale with Tetraploid Rye (6배체 트리티케일과 4배체호밀 교잡에서의 교잡율, 염색체수 변이 및 작물학적 특성)

  • This experiment was carried out to investigate seed set, variation in chromosome number, and agronomic characteristics of the progeny in the cross between hexaploid triticale variety, Sinkihomil(P$_1$) and tetraploid rye variety, Dooroohomil(P$_2$). Seed set rate obtained was 30.5% in the cross of Sinkihomil with Dooroohomil, whereas 3.26% in reciprocal cross using Dooroohomil as female. Alsoseed set was 8.75% in F$_1$/P$_1$, 7.20% in F$_1$/P$_2$, and 1.53% in F$_2$(=F$_1$ /F$_1$, respectively. Germination rate of crossed seed was 37% in cross of P$_1$ with P$_2$, 39.0% in F$_1$/P$_1$(BC$_1$), 50% in F$_1$/P$_2$(BC$_2$) and 43.0% in F$_1$/F$_1$(F$_2$), and 1,000 grain wight was 20.7g in the cross of P$_1$ with P$_2$, which have 41.9g and 47.7g, respectively, 24.5g in F$_1$/P$_1$, 23.6g in F$_1$/P$_2$, and 24.5g in F$_1$/F$_1$, respectively. In pollen fertility of F$_1$ plant, 69.8% turned out to be abnormal or sterile pollen grains, whereas 30.2% was fertile or normal. In meiosis of pollen mother cell of F$_1$ plant, 13.5 univalents, 8.89 bivalent and 1.24 trivalent were appeared. Somatic chromosome number of 35 in F$_1$, both 32 to 33 and 35 to 36 in F$_2$, 35 to 39 in BC$_1$ and 28 to 36 in BC$_2$ which mean producing female gamate was 14 to 18 chromosome in PMC of F$_1$ plant. Rate of fertile plant turned out to be 100% in F$_1$, 4.5% in F$_2$, 42.9% in BC$_1$, and 50.0% in BC$_2$, respectively. Number of seed set per spike appeared to be 1.17 in F$_1$ plant, 13.3 in F$_2$, 2.36 in BC$_1$, and 3.75 in BC$_2$, respectively. Days to heading of F$_1$ was intermediate, but F$_2$ was later than both parents. Plant height of F$_1$ , BC$_1$ ,and BC$_2$ was shorter than both parent, but F$_2$, longer than both parents.

  • In this paper, a PLHRO(Phase Locked Hair-pin Resonator Oscillator) for K-band is designed with the feedback property of PLL(Phase Locked Loop) using a new tuning mechanism. The proposed PLHRO generates the output power of -0.6 dBm at 24.42 GHz, and has the phase noise of -86.6 dBc/Hz at 100 KHz and -76.5 dBc/Hz at 10KHz offset from carrier frequency, and has suppression characteristics of -23 dBc and spurious noise of -65 dBc. Buffered 24.42 GHz PLHRO generates the output power of 5.6 dBm at 24.42 GHz and has the of a phase noise of -77.34 dBc/Hz at 100 KHz and -72 dBc/Hz at 100 KHz offset from carrier frequency.

Heavy Metal Removal of Acrylic Acid-grafted Bacterial Cellulose in Aqueous Solution (아크릴산으로 그라프트된 미생물셀룰로오스의 수용액 내 중금속 흡착거동)

  • Electron beam-induced grafting polymerization was employed to prepare Acrylic acid-grafted bacterial cellulose (BC-g-AAc). BC-g-AAc as an adsorbent was applied to remove heavy metals (e.g., As, Pb, and Cd). This study examined followings; morphological change of surface, adsorptive behavior of BC-g-AAc, and interpretation of adsorptive kinetics. Specific surface areas of BC and BC-g-AAc were $0.9527m^2g^{-1}$ for BC and $0.2272m^2g^{-1}$ for BC-g-AAc, respectively as measured by BET nitrogen adsorption, revealing the morphological change of the surface of BC-g-AAc. Batch adsorption test was performed to investigate adsorptive behavior of BC-g-AAc in aqueous solution. The amounts of Pb and Cd adsorbed on BC-g-AAc were $69mg\;g^{-1}$ and $56mg\;g^{-1}$, respectively. However, As was not adsorbed on BC-g-AAc due to its neutral nature. Both the Benaissa model and the Kurniawan model were applied in the study to interpret adsorptive kinetics. From the value of correction coefficient ($R^2$), adsorptive kinetics of Pb and Cd were subjected to Kurniawan model referred to pseudo-second-order. Taken together, the results of this study show that BC-g-AAc has potential as a heavy metal (eg., Pb, Cd)-adsorbent made of an environmentally friendly material.

Compensation of Aethalometer Black Carbon Data Observed at a Gwangju Site (광주 도심지역에서 측정한 Aethalometer 검댕입자 자료의 보정)

  • $PM_{2.5}$ black carbon (BC) concentrations were measured to investigate the filter spot loading effect in raw BC data at 5-minute time-based resolution using a single-wavelength aethalometer at a Gwangju site. Also the elemental carbon (EC) concentrations from 24-hr integrated filter-based measurements of $PM_{2.5}$ particles were determined to compare with the loading compensated BC values. Close examination of the time-series BC data showed clearly the "gaps" when the filter tape advances, suggesting the correction of raw BC data. Therefore, we calculated the average BC concentration in each range of attenuation (ATN) to decide if there was (or was not) an effect on the aethalometer data according to the loading of the filter spot. A consistent decrease of average BC concentration was found with increasing ATN values for every month, suggesting there was a consistent "spot loading effect" in the raw BC data. The loading compensated BC concentration according to a simple compensation model with loading effect was 1.01~1.15 times greater than the raw BC data. The 24-hr average concentration of EC observed during summer sampling period was about 3% higher than the original 24-hr average BC value and 2% lower than the loading compensated BC concentration.

Production and Characteristics of Bacterial Cellulose, an Eco-Friendly Biomaterial, using Different Carbon Sources (탄소원 종류에 따른 환경친화성 생물소재인 세균 섬유소의 생산 및 특성)

  • Production of Bacterial Cellulose (BC) by Gluconacetobacter sp. A5 was studied in shaken culture using different cost-effective carbon sources and its structural and mechanical properties were evaluated. Glycerol showed the highest level (7.26 g/l) of BC production, which was about three times higher than the yield in glucose medium. BC production depended not only on the decrease in pH, but also on the ability of Gluconacetobacter sp. A5 to synthesize glucose from different carbon sources and then polymerize it into BC. All BC produced from different carbon sources exhibited a three-dimensional reticulated structure consisting of ultrafine cellulose fibriles. Carbon sources did not significantly change the microfibrile structure of the resulting BC. BC produced from glucose medium had the lowest water-holding capacity, while BC from molasses medium had the highest. XRD data revealed that all BC were cellulose type I, the same as typical native cellulose. The crystalline strength of BC produced in glucose medium was the highest, and that in molasses medium was the lowest. Our results suggest that glycerol could be a potential low-cost substrate for BC production, leading to the reduction in the production cost, and also to produce BC with different mechanical properties by selecting appropriate carbon source.

Factorial design에 의한 Acetobacter xylinum KJ1의 Bacterial cellulose 생산조건의 최적화

  • Acetobacter xylinum KJ1 efficiently producing bacterial cellulose(BC) in shaking culture was isolated from a rotten grape. The strain was used to investigate optimum operating conditions for increasing BC production and factorial design model was employed for the optimization. The results of experiments were statistically analyzed by SAS program. Reciprocal effects of each factors(carbon source concentration, shaking speeds(rpm), oxygen pressure, and CSL concentration) and culture condition of BC production were examined by getting regression equation of the dependent variable. Comparisons between experimental results and predicted results about BC concentration were done in total 24 experiments by combination of each factors using SAS program, and the correlation coefficients of BC concentration and BC yield were 0.91 and 0.81, respectively. The agitated cultures were performed in various operation conditions of factors which affected considerably to BC production in jar fermentor. The results showed that BC concentration was 11.67g/ L in 80 hours cultivation under the condition of carbon source concentration shaking speeds(rpm) : oxygen pressure: CSL concentration = 4% : 460rpm : 0.28 : 6%. On the other hand BC yield was 0.42g/g in 80 hours cultivation under the condition of carbon source concentration shaking speeds(rpm) : oxygen pressure: CSL concentration = 4% : 564rpm : 0.21 : 2%. The BC production could be enhanced up to more than 65.3% by factorial design. The result of a verifying experiment under the optimal conditions determined by the factorial design to the BC production showed that the model was appropriate by obtaining BC concentration of 11.02g/L in the optimum condition

Yield Improvement by Two Cycles of Mass Selection in Two Sweet Corn Populations

  • Mass selection (MS) is an efficient selection method to directly improve highly heritable traits. In the present study, two cycles of MS for ear length were conducted on two sweet corn populations, BC2-l0 and $BC1-10{\times}Syn-II$ after introgression of exotic germplasm. The improved populations generated from these selections were evaluated in comparison with the base populations at two locations, to determine the genetic gains and performance of the improved populations. The two base populations showed varied average realized responses to MS. In BC2-l0 derived population, the realized responses were $9.1\%$ in BC2-l0 C1 and $1.2\%$ in BC2-l0 C2, whereas in$BC1-10{\times}Syn-II$ derived population, the realized responses were$ 5.6\%$ in $BC1-10{\times}Syn-II$ C1 and $2.9\%$ in $BC1-10{\times}Syn-II$ C2. All the improved populations showed longer ears than their respective base populations and the check varieties. Ear length, which was used as the selection criterion in this study, showed high broad-sense heritability in the BC2-l0 and $BC1-10{\times}Syn-II$ derived populations, while fresh ear yield revealed low heritability, indicating that selection for ear length in these populations would be more effective than direct selection for yield. Results of this study indicate that MS conducted on BC2­10 and $BC1-10{\times}Syn-II$ had significantly increased ear length and fresh ear yield in both populations. The improved populations obtained would serve as better germplasm sources and further selection in these populations could offer better responses.

Sorption and Desorption Kinetics of Naphthalene and Phenanthrene on Black Carbon in Sediment (퇴적물내 Black Carbon에 대한 Naphthalene과 Phenanthrene의 수착 및 탈착동력학)

  • Black carbon (BC), a kind of high surface area carbonaceous material (HSACM), was isolated from Andong lake sediment. Sorption and desorption kinetics of naphthalene (Naph) and phenanthrene (Phen) in organic carbon (OC) and BC in the Andong lake sediment were investigated. Several kinetic models such as one-site mass transfer model (OSMTM), two-compartment first-order kinetic model (TCFOKM), and a newly proposed modified two-compartment first-order kinetic model (MTCFOKM) were used to describe the sorption and desorption kinetics. The MTCFOKM was the best fitting model. The MTCFOKM for sorption kinetics showed that i) the sorbed amounts of PAHs onto BC were higher than those onto OC, consistent with BET surface area; ii) the equilibration time for sorption onto BC was longer than those onto OC due to smaller size of micropore ($11.67{\AA}$) of BC than OC ($38.18{\AA}$); iii) initial sorption velocity of BC was higher than OC; and iv) the slow sorption velocity in BC caused the later equilibrium time than OC even though the fast sorption velocity was early completed in both BC and OC. The MTCFOKM also described the desorption of PAHs from the OC and BC well. After desorption, the remaining fractions of PAHs in BC were higher than those in OC due to stronger PAHs-BC binding. The remaining fractions increased with aging for both BC and OC.

Physicochemical and Adsorptive Properties of Black Carbon for Radioactive Cesium under Various Combustion Conditions and Tree Species (연소 조건과 수종을 달리한 블랙카본의 물리화학적 성질 및 세슘의 흡착 특성)

  • This study was carried out to investigate the physicochemical and adsorptive characteristics of black carbon (BC) materials for cesium in case of severe nuclear accidents. The BC was prepared with a xylem of oak and pine trees incompletely combusted with different ramp rate and final temperature. Carbon (C), hydrogen (H) and oxygen (O) atomic ratios, BET, pore structure, and zeta potential were characterized for the produced BC. A low cesium concentration ($C_w{\approx}10^{-7}M$) was used for sorption batch experiments. The H/C and O/C ratios of BC decreased with the increase of final temperature, which indicates a carbonization of the wood materials regardless of ramp rate and tree species. However, SEM images showed different pore structures depending on tree species such as steric and plate-like for oak-BC and pine-BC, respectively. The greatest sorption distribution coefficients of $K_{d,Cs}{\approx}1,200{\sim}1,800L\;kg^{-1}$ were observed for the oak-BC produced at $400^{\circ}C$, while comparatively low $K_{d,Cs}$ < $100L\;kg^{-1}$ for pine-BC. In addition, the sorption capabilities of BC declined with the increase of combustion temperature up to $600^{\circ}C$, because high temperature destroyed surface functionalities with the rise of ash components in the BC. Therefore, the sorption processes of BC for radioactive cesium are predominantly controlled by final production temperature of BC as well as raw materials (e.g., tree species).

Exposure Assessment of Diesel Engine Exhaust among Door-to-door Deliverers in Daegu (대구지역 택배서비스업 종사자의 디젤엔진배출물 노출 평가)

  • Objectives: This study evaluated the diesel engine exhaust (DEE) exposure levels of door-to-door deliverers in Daegu from July to September. Methods: We measured exposure levels of DEE surrogates for the same door-to-door deliverers who joined the particulate matter 2.5 exposure study previously published in this journal. Black carbon(BC) concentrations were measured using real-time BC monitoring devices with 1 minute interval. $NO_2$ concentrations were monitored using passive badges. DEE exposure data were analyzed using the same characteristics and GPS information as the first study. Results: A total of 40 measurements of BC concentrations and $NO_2$ concentrations were collected during delivery of parcels. The average exposure levels to BC, and $NO_2$ were $2.23{\mu}g/m^3$ ($0.001-350.85{\mu}g/m^3$) and 21.26 ppb(3.3-61.37 ppb), respectively. Exposure levels to BC according to the day of a week and coverage areas were not significantly different(p>0.05). Delivery trucks manufactured before 2006 caused significantly higher exposure to BC than the trucks manufactured after 2006(p<0.05). Exposure levels of BC integrated for each time in residential area and roadsides were $1.96{\mu}g/m^3$ and $3.46{\mu}g/m^3$, respectively, and the difference was statistically significant(p<0.001). The Pearson correlation coefficients between the ambient $PM_{2.5}$ and BC was significant, r=0.26(p<0.01); however, the correlations between $PM_{2.5}$ and ambient $PM_{2.5}$, and between BC of DEE and $PM_{2.5}$ of DEE did not show a significant correlation Conclusions: BC and $NO_2$ exposure levels were significantly lower when door-to-door deliverers drove newer trucks. BC exposure levels of deliverers were higher in roadsides than in residential area. DEE from nearby vehicles through open windows might be the main source of BC exposure.
