Effect of Menhaden Oil Feeding on Protein Kinase C Activity and Membrane Phospholipid Profiles in Mouse Epidermal Cells (지방질원으로서 어유가 백서 상피세포의 인지질 조성 및 Protein Kinase C 활성에 미치는 영향)

  • To investigate the effect of dietary menhaden oil on protein kinase C (PKC) activity and membrane phospholipid composition in epidermal cells, female BALB/C mice were fed either menhaden oil or corn oil with two different levels(5% or 20%) for 6 weeks. Membrane phosphatidycholine(PC) was decreased in menhaden oil-fed group. Eicosapentaenoic acid(EPA) and Docosahexaenoic acid(DHA) were only presented in the acyl chain of membrane phospholipid of menhaden oil-fed mice, so that membrane fluidity of the group could be different from the other group. Both cytosolic and membrane-associated PKC activity in epidermal cells were decreased in menhaden oil-fed mice when compare with corn oil-fed mice. Furthermore, rate of PKC transfer from cytosol to membrane in menhaden oil-fed group was not as fast as in corn oil-fed group. Based on these observations, dietary menhaden oil might act differently from other dietary fat in carcinogenesis.

Consumption of Water-Soluble Egg Yolk Extract on Growth Rate, Changes in Blood Cholesterol Levels, and Immune Modulation in BALB/c Mice

  • Egg consumption has been limited to avoid cardiovascular diseases such as atherosclerosis and hyperlipidemia, because the yolk contains high levels of cholesterol. This study was conducted to evaluate the effect of the water-soluble component of egg-yolk on the growth efficiency, immune modulation, and changes in serum lipid levels in BALB/c mice. A total 5 wk old 120 BALB/c male mice were divided into 4 groups and were fed 0, 2, 10, and 20 mg/d water-soluble egg yolk extract (WSEYE) for 5 wk. Water-soluble egg yolk extract (WSEYE) uptake resulted in a significant reduction in daily weight gain and feed efficiency rate (FER). The mouse groups treated with 2 and 20 mg/d WSEYE showed a significant increase in populations of monocytes at the third wk and B-lymphocyte activity at the fifth wk. In addition, WSEYE uptake did not influence serum immunoglobulin E levels. In serum lipid-profile studies, treatment of WSEYE did not alter total cholesterol and low-density lipoprotein levels; however, blood triglyceride levels were significantly diminished in mice treated with 2 mg/d at the third wk (p<0.05), and the level of high-density lipoprotein was significantly increased in the mice group treated with 2 and 10 mg/d WSEYE after 5 wk (p<0.05). Taken together, the data demonstrate the beneficial effects of WSEYE in the diet on immune modulation and serum lipid profiles in mouse models; therefore, this study suggests that ingestion of water-soluble fraction of egg yolk might not be related to the increased risk of heart disease, but can be an excellent candidate for maintaining health.

  • Toxoplasmn gonnii, an intracellular protozoan infecting many kinds of eukaryotic cells, has been used to experimentally infect macrophages, epithelial cells, fibroblasts, and various cancer cells, but rarely T and B Iymphocytes or granulocytes. The present study was performed to determine the susceptibility of murine (BALB/c or CBA) splenic T and B llrnphocytes, and granulocytes to infection trio T. gondii RH tachyzoites. The ultrastructure of the infected host cells was observed by TEM, and the degree of intracellular parasite proliferation was quantified using 3H-uracil uptake assay. At 24 hrs post-culture, the host cell cytoplasm was found to contain 1 or 2, or a maximum of 7-8 tachyzoites. Infected T Iymphocytes demonstrated a peripherally displaced nucleus, a parasitophorous vacuole enveloping the parasite, and an increased number of mitochondria. In B Iymphocytes infected with tachyzoites, RER was not well developed compared to uninfected B Iymphocytes. Uninfected granulocytes contained many electron dense granules, but T. gondii-infected granulocytes demonstrated a decreased number of granules. Based on the 3H-uracil uptake assay. the susceptibility of T and B Iymphocytes, and granulocytes, to infection with T. gonnii tachyzoites was fairly high irrespective of cell type and strain of mouse. This strongly suggests deterioration in the functioning of infected host immune cells.

Susceptibility of several strains of mice to Echinostoma hortense infection

  • Susceptibilities of 5 different mice strains, including C3H/HeN, BALB/c, C57BL6, FvB and ICR, to Echinostoma hortense infection, was evaluated. The worm expulsion rate, worm size and egg production were observed from 1 to 8 weeks after infection with 30 metacercariae. C3H/HeN and ICR mice showed the highest worm maturation rates. The worm recovery rate and the number of eggs per gram (EPG) of feces was also higher in C3H/HeN and ICR mice than in BALB/c, C57BL6, and FvB mice. It is suggested that E. hortense is highly infectious to ICR and C3H/HeN mice, but not to the other strains of mice. Based on the results obtained, we believe that the susceptibility of different mouse strains to E. hortense infection is dependent on the genetic and immunologic back-ground of mice.

Measurement of Growth Delay and the Oxygen Enhancement Ratio of Fast Neutron Beam Using Mouse Model System (마우스모델을 이용한 고속중성자선의 성장지연 및 산소증강비의 측정)

  • Neutrons are high LET (linear energy transfer) radiation and cause more damage to the target cells than x-rays or gamma rays. The damage from neutrons is generally considered fatal to a cell and neutrons have a greater tendency to cause cell death through direct interaction on DNA. We performed experiments to measure growth delay ratio and oxygen enhancement ratio (OER) in mouse model system. We inoculated EMT-6 cells to the right hind leg of BALB-c mouse and X-rays and neutron beams were given when the average volume of tumors reached $200-300mm^3$. We irradiated 0, 11, 15.4 Gy of X-ray and 0, 5, 7 Gy of fast neutron beam at normoxic and hypoxic condition. The volume of tumors was measured 3 times per week. In x-ray experiment, growth delay ratio was 1.34 with 11 Gy and 1.33 with 15.4 Gy in normoxic condition compared to in hypoxic condition, respectively. In neutron experiment, growth delay ratio was 0.94 with 5 Gy and 0.98 with 7 Gy, respectively. The OER of neutron beam was 0.97. The neutron beam was more effective than X-ray in the control of hypoxic tumors.

Immunomodulating Activity of Salicornia herbacea Extract (함초 추출물의 마우스 면역 증강 활성)

  • Immunomodulating effect of Salicornia herbacea extract on the mouse splenocytes was investigated. Crude S. herbacea polysaccharide extract (CSP) and other kinds of fine S. herbacea polysaccharides (SPI and SPII) were prepared from S. herbacea by hot water extraction and further ultrafiltration and gel filtration chromatography. In vitro experiment, the mouse splenocytes and separated T cells were treated with CSP, SPI or SPII (0.5, 1, 2, 4 mg/ml). In vivo experiment, three different S. herbacea extracts were orally administrated everyday for one week. For the basic data, body weight and physiological parameters such as organ weight and spleen index were observed. The proliferation of the cells was used as an index for immunemodulating activity and the effect of proliferation was evaluated using MTS assay. The CSP, SPI and SPII directly induced the proliferation of splenocytes and separated T cells in a dose-dependent manner. In results, the proliferation was more increased in the SPI and SPII treated cells than in the CSP treated cells. The best proliferation was shown in the splenocytes cultured with SPI at the concentration of 4 mg/ml for 24 hr. The proliferation of splenocytes and separated T-cells was higher (3.2 and 3.5 times, respectively) than the control. Moreover, when the mouse splenocytes were treated with mitogen, the efficient proliferation was shown in the splenocytes cultured with SPI. In conclusion, polysaccharides from S. herbacea showed a substantial immunomodulating activity in the mouse immune cells.

Changes of Mouse Gut Microbiota Diversity and Composition by Modulating Dietary Protein and Carbohydrate Contents: A Pilot Study

  • Dietary proteins influence colorectal cancer (CRC) risk, depending on their quantity and quality. Here, using pyrosequencing, we compared the fecal microbiota composition in Balb/c mice fed either a normal protein/carbohydrate diet (ND, 20% casein and 68% carbohydrate) or a high-protein/low-carbohydrate diet (HPLCD, 30% casein and 57% carbohydrate). The results showed that HPLCD feeding for 2 weeks reduced the diversity and altered the composition of the microbiota compared with the ND mice, which included a decrease in the proportion of the family Lachnospiraceae and Ruminococcaceae and increases in the proportions of the genus Bacteroides and Parabacteroides, especially the species EF09600_s and EF604598_s. Similar changes were reported in patients with inflammatory bowel disease, and in mouse models of CRC and colitis, respectively. This suggests that HPLCD may lead to a deleterious luminal environment and may have adverse effects on the intestinal health of individuals consuming such a diet.

Molecular Characterization and Mitogenic Activity of a Lectin from Purse Crab Philyra Pisum

  • A lectin from the hemolymph of purse crab, Philyra pisum, was found to have anti-proliferative activity on human lung cancer cells by our laboratory. In this study, P. pisum lectin (PPL) was molecularly characterized including molecular mass, amino acid sequences, amino acid composition, and the effects of metal ions, temperature, and pH on the activity. We found that PPL showed mitogenic activity on human lymphocytes and BALB/c mouse splenocytes. The mitogenic activity (maximum stimulation index, $SI=9.57{\pm}0.59$) of PPL on human lymphocytes was higher than that of a standard well-known plant mitogen, concanavalin A (maximum $SI=8.80{\pm}0.59$). The mitogenic activity mediated by PPL is required for optimum dosing, and higher or lower concentrations caused decreases in mitogenic response. PPL also induced mitogenic activity on mouse splenocytes, however, the maximum SI ($1.77{\pm}0.09$) on mouse splenocytes of PPL was lower than that ($2.14{\pm}0.15$) of concanavalin A. In conclusion, PPL is a metal ion-dependent monomer lectin with mitogenic activity, and could be used as a lymphocyte or splenocyte stimulator.

Characterization of Mouse Hybridoma Producing Monoclonal Antibody Paragonimus westermani (폐디스토마에 대한 Mmonoclonal Antibody생산 세포주개발에 대한 연구)

  • In this study, hybridoma techniques were applied to produce monoclonal antobodies to Paragonimus westermani, commonly known as lung distoma. Balb/c mice were immunized intraperitoneally every week with increasing doses of purfied protein of Paragonimus westermani adult worms begining with 0.3/mg/mouse/7 days and ending with 0.5mg at 28th day. Spleen cells from these immunized mice were hybridized with myeloma cells (NS-1) and the hybridized cells were selected in HAT media. The antibody secreting cells among the hybrid cells were initially selected by ELISA. Those initially selected cells were further screened by the criteria of antibody producing activity, and seneral cell lines among them were further tested with immunodiffusion method. One hybridoma clone produced IgM and another clone produced IgG1. The supernatant of the hybridoma clone producing IgM had titer 1:64 and the hybridoma clone producing IgG1 had titer 1:256 measured by immunofluorescence technique.

  • Objectives : This study was carried out to investigate the latest study tendency in mouse model of skin pruritus published since 2005 and to arrange various experimental methods. Methods : We examined the journal(such as Experimental dermatology and British journal of dermatology) and in Pubmed since 2005, regarded pruritus and scratching behavior model as key words. Results and Conclusion : 1. BALB/c, hairless, NC/Nga and ICR mice were the most used in scratching behavior model. According to scratching-induced agents. we need to select experimental mice. 2. There are various methods inducing skin pruritus; by cohabitation with NC/Nga mice having severe skin lesions, by injection intradermally(or subcutaneously) with agent inducing inflammation, by inducing contact dermatitis with TNCB, TNFB, destruction of skin barrier and by transgenic mice. 3. Injection intradermally(or subcutaneously) with agent inducing inflammation is the most used out of methods inducing skin pruritus. Compound 48/80, histamine, substance P and others(chloroquine, serotonin, carrageenin, TPA etc.) were included in agents inducing inflammation and pruritus in skin pruritus model. 4. Video camera, SCLABA system, MicroAct and acoustic evaluation system were included in evaluation methods of scratching behavior mouse model.
