Effects of Bojeongjeongcheon-tang on Cytokines and Immunoglobulin E in B Cells (보정정천탕의 Cytokine 및 IgE에 대한 조절효과)

  • Objectives : To evaluate experimentally the clinical effect of Bojeongjeongcheon-tang, we observed the cytokines ($IL-1{\beta}$/TEX>, IL-4, IL-5, IL-6, IL-10, TNF-{\alpha},{\;}TGF-{\beta},{\;}IFN-{\gamma}$) and what effect they have on IgE in B cells of a rat. Methods : First of all, we extracted the spleens of healthy Balb/c mice and separated B cells from them. These B cells were cultured with anti-CD40 mAb (500 ng/ml), rmIL-4 (500 U/ml), Bojeongjeongcheon-tang (100 ug/ml, 10 ug/ml, 1 ug/ml). We used rmiL-10 (50 ng/ml) as a control group. Furthermore, we analyzed the expression of IgE, CD23, CD69 and the coherence of HRF in B cells using a flow cytometer. We also analyzed the cytokine gene expression in B cells by reverse transcriptase-PCR. We also measured B cells proliferation using the Liquid Scintillation Counter. Results : In this study, the Bojeongjeongcheon-tang treated group showed a tendency to decrease depending on the density compared with the control group in the expression of IgE+, CD23+, CD69, HRF. All of the Bojeongjeongcheon-tang treated group showed inhibitory effects with $IL-1{\beta}$, IL-4, IL-5 and proliferating effects with IL-6, IL-10, and $IFN-{\gamma}$ on cytokines transcript expression depending on the density. Meanwhile, $TNF-{\alpha}$ increased in all density. In IgE production, there was inhibitory effect on Bojeongjeongcheon-tang (both 100 ug/ml and 10 ug/ml) of significance (p < 0.01, p < 0.05). Also in B cell proliferation, the result revealed an inhibitory effect of Bojeongjeongcheon-tang (both 100 ug/ml and 10 ug/ml), of significance (p < 0.001, p < 0.01). Conclusions : This study shows that Bojeongjeongcheon-tang has an inhibitory effect on the production and activity of B cells. Also it inhibited CD23, IL-4 activity and IgE production and activation. It is obvious that Bojeongjeongcheon-tang treats asthma by inhibiting the production of histamine and HRF, IL-5 and proliferating IL-10. Also Bojeongjeongcheon-tang has some preventive effects on bronchial change by inhibiting $TGF-{\beta}$, which stimulates the bronchial transformation.

PROPERTIES OF GENERALIZED BIPRODUCT HOPF ALGEBRAS

  • The biproduct bialgebra has been generalized to generalized biproduct bialgebra $B{\times}^L_H\;D$ in [5]. Let (D, B) be an admissible pair and let D be a bialgebra. We show that if generalized biproduct bialgebra $B{\times}^L_H\;D$ is a Hopf algebra with antipode s, then D is a Hopf algebra and the identity $id_B$ has an inverse in the convolution algebra $Hom_k$(B, B). We show that if D is a Hopf algebra with antipode $s_D$ and $s_B$ in $Hom_k$(B, B) is an inverse of $id_B$ then $B{\times}^L_H\;D$ is a Hopf algebra with antipode s described by $s(b{\times}^L_H\;d)={\Sigma}(1_B{\times}^L_H\;s_D(b_{-1}{\cdot}d))(s_B(b_0){\times}^L_H\;1_D)$. We show that the mapping system $B{\leftrightarrows}^{{\Pi}_B}_{j_B}\;B{\times}^L_H\;D{\rightleftarrows}^{{\pi}_D}_{i_D}\;D$ (where $j_B$ and $i_D$ are the canonical inclusions, ${\Pi}_B$ and ${\pi}_D$ are the canonical coalgebra projections) characterizes $B{\times}^L_H\;D$. These generalize the corresponding results in [6].

Target Strength Measurements of Live Golden Cuttlefish Sepia esculenta at 70 and 120 kHz

  • Cuttlefish Sepia esculenta are commercially important in Korea. Assessments of their biomass currently depend on fishery-landings data, which may be biased. Towards fishery-independent acoustic surveys of cuttlefish, target strength (TS) measurements at 70 and 120 kHz were made of 23 live cuttlefish, in early May 2010. The fish were caught by traps in the inshore waters around Geojedo, Korea. The TS were measured using split-beam echosounders (Simrad ES60 and EY500, respectively). The cuttlefish mantle lengths (L) ranged from 15.6 to 23.5 cm (mean L=17.8 cm) and their masses (W) ranged from 335 to 1020 g (mean W=556.1 g). Their mean TS values at 70 and 120 kHz were -33.01 dB (std=1.39 dB) and -31.76 dB (std=2.15 dB), respectively. The mean TS at 70 kHz was 0.17 dB higher than the TS-length relationship resulting from a least-squares fit to the data ($TS=24.67{\log}_{10}L(cm)-64.03$, $r^2$ = 0.52, N=23). The mean TS at 120 kHz was 0.45 dB higher than the fitted TS-length relationship ($TS=40.59{\log}_{10}L(cm)-82.96$, $r^2$ = 0.58, N=23). The differences between the mean TS values and an equation regressed from all of the TS measurements at both frequencies ($TS=24.92{\log}_{10}L(cm)-4.92{\log}_{10}{\lambda}(m)-22.82$, $r^2$ = 0.86, N=46) was 0.22 dB at 70 kHz and 0.31 dB at 120 kHz, respectively.

Radial Distribution of Calcium, Phosphorus, Iron, Thiamine and Riboflavin in the Degermed Brown Rice Kernel (현미입(玄米粒) 내의 칼슘, 인, 철, 비타민$B_1$$B_2$의 분포에 관한 연구)

  • Degermed brown rice of Akibare (short grain) and Milyang 23 (medium grain) was abraded fiveconsecutive times to remove outer $5{\sim}6%$ of the kernel per milling. Samples were analyzed for calcium, phosphorus, iron, thiamine and riboflavin. Milled fraction I (about $5{\sim}6%$ of the kernel) contained 8 times as much calcium and phosphorus as did the original kernel; iron, $4{\sim}5$; thiamine, 3; and riboflavin, 4. Contents of fraction I were much greater than those in the residual kernel; 18 times as great for calcium; $32{\sim}36$ times for phosphorus; $5{\sim}10$ times for iron 5 times for thiamine; and $19{\sim}30$ times for riboflavin. Milyang 23 showed a steeper concentration gradient of calcium and riboflavin, but more even distribution of iron than did Akibare. There were no significant differences in phosphorus and thiamine gradients between the two rices.

HspBP1 Is the Negative Regulator of the Bovine Progesterone Receptor

  • We have investigated whether HspBP1, a Hsp70 binding protein, could have effect on the assembly of the bovine progesterone receptor (bPR) with a chaperone complex consisting of bovine Hsp90 (bHsp90), bovine Hsp70 (bHsp70), Hop, Ydj-1, and p23. The bPR, isolated in its native conformation, loses its function to interact with progesterone hormone in the absence of this protein complex. However, in the presence of bHsp90, bHsp70, Hop, p23 and Ydj-1, its function could be restored in vitro. Our findings here indicate that the inclusion of HspBP1 to five-protein system prevented the proper assembly of progesterone receptor-chaperone complex and induce the loss of bPR ability to interact with hormone. Immunoprecipitation assays of bPR with HspBP1 show that the presence of HspBP1 did not have any effect on the assembly of Ydj-1 and bHsp70 with the progesterone receptor. However, further assembly of Hsp90, Hop and p23 was completely prevented and the function of the bPR was lost. In vitro competition and protein folding assays indicated that the binding of HspBP1 to bHsp70 prevented the ternary complex formation of bHsp70, bHsp90, and Hop. These results indicate that HspBP1 is a negative regulator of the assembly of Hsp90, Hop and Hsp70, and thus, prevent the proper maturation of unliganded bPR with chaperones assembly system.

TMA Study on Phase Evolution During Hydrogen-assisted Disproportionation of Nd-Fe-B Alloy

  • Phase evolution during the hydrogen-assisted disproportionation of $Nd_{12.5}Fe_{80.6}B_{6.4}Ga_{0.3}Nb_{0.2}$ alloy was investigated mainly by using a magnetic balance-type thermomagnetic analyser (TMA). In order to avoid any undesirable phase change in the course of heating for TMA, a swift TMA technique with very high heating rate (around 2 min to reach $800^{\circ}C$ from room temperature) was adopted. The hydrided $Nd_{12.5}Fe_{80.6}B_{6.4}Ga_{0.3}Nb_{0.2}$ alloy started to be disproportionated in hydrogen from around $600^{\circ}C$, and the alloy after the early disproportionation (from 600 to $660^{\circ}C$) has been partially disproportionated. The partially disproportionated alloy consisted of a mixture of $NdH_x$, $Fe_3B$, ${\alpha}$-Fe, and the remaining undisproportionated $Nd_2Fe_{14}BH_x$-phase. During the subsequent heating to $800^{\circ}C$ in hydrogen, two additional phases of $Fe_{23}B_6$ and $Fe_2B$ were formed, and the material consisted of a mixture of $NdH_x$, $Fe_{23}B_6$, $Fe_3B$, $Fe_2B$, and ${\alpha}$-Fe phases. During the subsequent isothermal holding at $800^{\circ}C$ for 1 hour, the phase constitution was further changed, and one additional unknown magnetic phase was formed. Eventually, the fully disproportionated $Nd_{12.5}Fe_{80.6}B_{6.4}Ga_{0.3}Nb_{0.2}$ alloy consisted of $NdH_x$, $Fe_{23}B_6$, $Fe_3B$, $Fe_2B$, ${\alpha}$-Fe, and one additional unknown magnetic phase.

  • It is generally accepted that parasite-specific IgE plays a crucial role in host defense against helminthic parasites. However, the role of high levels of nonspecific IgE in helminthic infections is still controversial. To investigate the role of nonspecific IgE in primary infections with P. westemani the effect of anti-lgE mAb treatment on serum IgE, $Fc{\varepsilon}RII/CD23$ expression and worm burden in Parcgonimus-infected mice were examined. In mice treated with anti-lgE antibody, the total IgE levels were not detectable ($1{\;}{\mu\textrm{g}/ml}$) throughout the experiment compared with untreated infected mice. The mean percentages of $Fc{\varepsilon}RII/CD23$ positive splenic B cells in anti-lgE treated mice (ridge: 20.3 - 30.5) were also decreased throughout the experiment compared with untreated infected mice (range: 35.7-44.4). Reduction of the total IgE and expression of $Fc{\varepsilon}RII/CD23$ on splenic B cells resulted in decreased worm burden six weeks post infection. These results suggest that high levels of nonspecific IgE in mice with primary infections of P. westemnni play a harmful, rather than beneficial, role for the host, perhaps by interfering with CD23-dependent cellular pathways.

Effects of UV-B and Growth Inhibitor on Overgrowth Retardation and Growth and ield after Planting in Fruit-Vegetable Plug Seedlings (UV-B와 생장억제제 처리가 과채류 플러그 묘의 도장 억제와 정식 후 생육 및 수량에 미치는 영향)

  • This experiment was performed to investigate the effect of UV-B (4 kJ${\cdot}m^{-2}{\cdot}d^{-1}$) irradiation and growth inhibitor (50 mg${\cdot}L^{-1}$ diniconazole, 500 mg${\cdot}L^{-1}$) treatment on the overgrowth retardation and the growth and yield afterplanting in plug-grown cucumber, tomato, and hot pepper sedlings. Stem length of UV-B-irradiated and iniconazole-treated seedlings was shortened by 38 and 35%^ in cucumber, 37 and 41% in tomato, and 23 and 23% in hot pepper, respectively, compared with non-treated seedlings. While retarding effect ofhexaconazole waslower that that of UV-B or diniconazle. Leaf area and dry weight alson decreased but ldaf thickness increased in UV-B irradiated or growth-inhibitor-treated seedlings. Even thouth the plant height and leaf area of UV-B-irradiated seedlings ant seedling stage were shorter and smaller that those ofnon-treatedd seedlings, they were recovered to the similar level to the growth of non-treated seedlings 20to 30days after trasplantin. The diconazole-and hexaconazole-treated seedlings were delayed recovery to their normal growth, Fruit yiedl of UV-V-irradiated hot pepper and tomato slightly increased but UV-B irradiated cucumer had similar yield to growth-inhibitor-treated one. It suggested that the use of UV-B irradiation could become a reliable tool of overgrowth retardation of plug-grown vegetable seedlings in greenhouse.

The Hearing Ability of Coralfish Chromis notatus to Low Frequency Sound 2. The Auditory Critical Ratio and Hearing Index (저주파음에 의한 자리돔의 청각 능력 2. 청각 임계비 및 청각능력지수)

  • In order to obtain the fundamental data on the auditory thresholds of fishes for catching method using low frequency sound, the auditory thresholds of coralfish Chromis notatus were measured in the presence of masking noise in the spectrum level range of 73~83dB re l$\mu$Pa/√Hz by heartbeat conditioning technique using pure tones coupled with a delayed electric shock. Critical ratios were about 23~41dB at measurement frequency, The critical ratio increased almost linearly with increasing frequency from 500Hz. The noise spectrum level at the start of masking was about 60~65dB. This suggests that hearing of coralfish is masked in the natural environment with the noise spectrum level above 60dB. The sound pressure level of which the signal sound of 300Hz is recognized by coralfish under the ambient noise is above 88dB and the critical ratio of them is above 23dB. The hearing index of coralfish with ambient noise was 81.
