Survival and stability of 19 bifidobacterial strains included 13 isolates from Korean infants against acid, bile acid and oxygen exposure were examined. Acid resistance of selected strains at pH values of 4.0, 3.0 and 2.0 was tested. Among the bifidobacterial strains tested, B. bifidum B3, B. longum D6, and B. adolescentis F1 exhibited higher viable cell counts exposed to acid whereas other strains had various results. The abilities of the strains to grow in the MRS broth containing 0.2% thioglycolic acid and 0.2% oxgall were tested and the tolerance of B. bifidum B3 and B. longum D6 to bile acid were higher than that of others. Even though in same species, the tolerance of tested strains to bile acid were variable. Stabilities of tested strains to oxygen exposure were variable and B. bifidum and B. longum strains showed relatively higher viable cell counts after 48 hours exposure to aerobic incubation. These results demonstrated that the survival and stability of bifidobacterial strains to acid, bile acid, and oxygen exposure were variable and strain-dependent. Due to their tolerant ability to environmental factors like acid, bile acid, and oxygen, B. bifidum B3 and B. longum D6 had good potential properties as probiotic cultures and may be useful for industrial application.
Bealmijang is a short-term fermented regional product that is prepared with soybean and extra ingredients. In this study, starter strain candidates were screened from Bealmijang for fermented soybean paste products. Twenty one bacterial strains producing extracellular enzymes (amylase, cellulase, protease, xylanase and lipase) were isolated from Bealmijang, buckwheat sokseongjang. The isolates were assessed for fibrinolytic and antibacterial activities, and salt tolerance. Strain HJ18-4, identified as Bacillus subtilis (AB601598) by biochemical properties (89.6%) and 16S rDNA sequencing (100%), showed the highest enzymatic, fibrinolytic, and antibacterial activities among the isolates. Although the growth of HJ18-4 was inhibited by the increase of NaCl concentration, the growth still exceeded that of B. subtilis KACC 10114 at 5% and 10% NaCl. These results suggest that B. subtilis HJ18-4 is suitable as a starter for soybean paste manufacture.
Purpose: Hepatic allografts from donors with hepatitis B core antibody have been demonstrated to transmit hepatitis B virus (HBV) infection to recipients after liver transplantation (LT). The efficacy of hepatitis B immune globulin (HBIg) to prevent de novo hepatitis B was investigated by comparing active immunization in the early phase to HBIg monotherapy in the late phase of pediatric liver transplants at Samsung Medical Center. Methods: Among pediatric liver transplants, from May, 1996 to June, 2002, 15 recipients who were hepatitis B surface antigen (HBsAg) (-) received an allograft from a donor with hepatitis B core antibody (HBcAb) (+). Except two who died from unrelated causes, eleven of 13 recipients were HBsAb (+), and 2 were naive (HBsAb(-), HBcAb(-)). All patients were vaccinated for HBV before LT. In the early phase (January, 1997~November, 1997, 3 patients), HBsAb (+) recipients received booster vaccination after LT. In the late phase (December, 1997~, 10 patients), all recipients were given booster vaccination and received HBIg therapy in order to maintain HBsAb titer greater than 200 IU/L. Lamivudine was given in one case because of severe side effect of HBIg. We retrospectively analyzed the effect of the preventive therapy for de novo hepatitis B through medical records. Results: De novo hepatitis B developed in three of 13 recipients (23.1%). All of 3 patients who received active immunization in the early phase became HBsAg (+) at 7~19 months after transplantation. One of them was naive before LT and the other two were HBsAb (+). All of 10 recipients who were given HBIg in the late phase remained HBsAg (-) at 7~55 months' follow-up. Conclusion: Passive immunization with HBIg was effective for prevention of de novo hepatitis B in HBsAg (-) recipients of hepatic allografts from HBcAb (+) donors.
Molecular structures of the various conformers for the 1,3-dimethoxy-p-tert-butylthiacalix[4] crown-5-ether (3) were optimized by using DFT B3LYP/6 - 31 + G(d,p) and mPW1PW91/6 - 31 + G(d,p) (hybrid HF-DF) calculation methods. We have analyzed the energy differences and structures of eight in/out orientations (cone_oo, cone_oi, pc_oo, pc_io, pc_oi, pc_ii, 13a_oo, 13a_io) of two methoxy groups in three major conformations (cone, partial-cone and 1,3-alternate). The 13a_oo (out-out orientation of the 1,3-alternate conformer) is calculated to be the most stable among eight different conformations of 3, and in accord with the experimental result. The ordering of relative stability resulted from the mPW1PW91/6 - 31 + G(d,p) calculation method is following: 13a_oo > 13a_io$\sim$pc_io$\sim$cone_oo > cone_oi$\sim$pc_oo$\sim$pc_oi > pc_ii.
The purpose of this study was to examine the microbiological and physicochemical changes on packaged tofu stored at temperatures of 5, 13, 23, and 30℃, and measure the consumable period from the expiry date to ultimately evaluate the microbiological safety on the extension of the consumable period. From the investigation, the pH value of tofu at each storage temperature (5, 13, and 23℃) showed a slight decrease over the storage period, although there was no significant change. The hardness of packaged tofu decreased more rapidly as temperature and storage time increased and the tofu started to show signs of decomposition at the same time. Analysis on the microbial change of tofu at different storage temperature revealed that the number of general bacteria also increased as the temperature increased. It was further found that packaged tofu takes 25 days at 5℃, 7 days at 13℃, and 1 day at 23℃ from the expiry date until the general bacteria count is at least at the early decomposition level which is 10℃ log CFU/g. However, no coliform bacteria was detected from tofu after storing at 5, 13 and 23℃. When packaged tofu was stored at 5℃, the L value changed significantly after 26 days, whereas the a and b values showed no significant change during the storage period (P>0.05). When storing tofu at 13℃ and 23℃ the L value decreased after 8 and 3 days, respectively. However, both a and b values increased (P<0.05).
These studies were carried out to identify Bacillus cereus group 1..5-] strain isolated from 5uyeong Bay. This strain was differentiated from B. cereus group using conventional, API system and fatty acid composition analysis. Colony characteristics were opague. mucoid, entire margin. convex. circular and non hemolysis on sheep blood agar plates, and were observed with central spore forming positive bacilli in a Gram stained preparation. and had no motility. The carbohydrates tested; glucose.maltose, and sucrose were assimilated but neither trehalose nor salicin were assimilated. This strain ultilized gelatin and was also inhibited by 6.5% NaCI. The results of biochemical examination were differented from B. cereus group LS-1 compared with others B. cereus group. The fatty acid composition contained major amounts of branched chain acids. iso $C_{15}$ and iso $C_{13}$ and the range of chain length was $C_{12}$ to C"$C_{17}$ and n$C_{15}$, acid was not detected. Automated fatty acid computer profile indicated "B. mycoides GC subgroup B of 0.312 similarity index." The results agreed with other research cases. On the other hand. A TB computer prolile index of API system (API 50 CHB & API 20E) identified" Doubtful profile of 99.7% B. firmus" . These results were presented with considerable discrepancies between API system and fatty acid analysis. With 67 biochemical characters. the similarity matrix of B. mycaides (KCTC 1033). B. thuringiensis (KCTC 1033). B. cereus (5-3) and B. mycoides (S-12) showed 42%. 42%. 59%, and 52%. respectively. Through the key tests and fatty acid analyses. we could notice the appearance of B. mycoides of the B. cereus group and this leads us to suspect the existence of a new biotype B. mycoides.
We have examined the effect of ball-milling on the superconducting properties of $MgB_2$ doped with C. The ball-milling of pre-reacted $MgB_2$ powder was carried out in dry or wet state using C or diethylenetriamine ($C_{4}H_{13}N_3$) as additives. The diethylenetriamine, whose chemical formula contains no oxygen, was chosen to avoid an excess oxidation during doping. The superconducting transition temperature (Tc) of the ball-milled or doped $MgB_2$ powders was only slightly smaller than that of undoped $MgB_2$. The critical current density (Jc) of the highly ball-milled $MgB_2$ was higher than that of C-doped $MgB_2$. The addition of diethylenetriamine was detrimental to Jc, although Tc was almost unchanged.
1-(Fluoren-2-yl)-benzo[d][1,2,3]triazoles 5a-b were synthesized starting from 2-nitrofluorene. 2-Nitrofluorenes 1a-b were reduced by catalytic hydrogenation, reacted with 2,4-dinitrofluorobenzene followed by catalytic hydrogenation to afford 2-(N-2,4-diaminophenyl)aminofluorenes 4a-b. Diazotization of 4a-b with $NaNO_2/H_2SO_4$ followed by treatment with $H_3PO_2$ gave 5a-b. Sulfonation of 5a-b yielded 7-benzotriazol-1-yl-fluorene-2-sulfonic acids 6a-b. The mode of action of mastoparan B, an antimicrobial cationic tetradecapeptide amide isolated from the hornet Vespa basalis, toward phospholipid bilayers was studied with synthetic mastoparan B and its analogs with individual Ala instead of hydrophobic amino acids (1-Ile, 3-Leu, 6-Leu, 7-Val, 9-Trp, 13-Val, 14-Leu) in mastoparan B. Mastoparan B and its analogs were synthesized by the solid-phase method. Circular dichroism spectra showed that mastoparan B and its analogs adopted an unordered structure in buffer solution. In the presence of neutral and acidic liposomes, most of the peptides took an α-helical structure. The calcein leakage experiment indicated that mastoparan B interacted strongly with neutral and acidic lipid bilayers than its analogs. Mastoparan B also showed a more or less highly antimicrobial activity and hemolytic activity for human erythrocytes than its analogs. These results indicate that the hydrophobic face in the amphipathic α-helix of mastoparan B critically affect biological activity and helical contents.
The results of geoarchaeological studies of the prehistoric cultural complexes on the Russian Far East (Primorye, or Maritime Province; the Amur River basin; and Sakhalin Island) are presented. Upper Palaeolithic sites are dated to ca. 40,000-10,500 B.P. They existed during the mild climate of the Chernoruchie interstadial (ca. 40,000-21,000 B.P.); during harsh climate at the Last Glacial Maximum, ca. 20,000-18,000 B.P., in several places on the Russian Far East (Primorye, Amur River basin, and Sakhalin); and during climatic amelioration in the Late Glacial time, ca. 16,000-10,500 B.P. The earliest Neolithic sites, represented by Osipovka and Gromatukha cultures, existed at ca. 13,000-10,000 B.P. in the environment of coniferous forests with admixture of broadleaved taxa. Since ca. 8000 B.P., Neolithic cultures appeared in all of the Russian Far East. They existed until ca. 3000 B.P., first during the Holocene Climatic Optimum, ca. 8000-5000 B.P., in the environment of coniferous-broadleaved forests, and later, at ca. 5000-3000 B.P., in the environment of birch-oak and coniferous forests.
