Bifidobacterium longum HY8001의 섭취가 사람의 장내세균층 및 장내세균 효소에 미치는 영향 (Effect of Bifidobacterium longum HY8001 Administration on Human Fecal Bacterial Enzymes and Microflora)

  • The effects of Bifidobacterium longum HY8001 supplement intake on the fecal microflora and fecal bacterial enzyme activity were studied in ten healthy human volunteers, before, during and after intake (respectively for 3 weeks). During intake of B. longum HY8001 supplement, fecal, $\beta$-glucuronidase and nitroreductase activities significantly decreased 44.6%(p<0.005) and 32.3%(p<0.01), respectively. Although numbers of major bacterial groups of fecal microflora were not affected by B. longum HY8001 intake for 3 weeks, the number of Bifidobacterium was significantly increased (p<0.05). This result indicates that intake of B. longum HY8001 might be potentially beneficial for the prevention and inhibition of colon cancer and improvement of human intestinal microflora composition.

The immune enhancing effects and characteristics of Bifidobacterium longum and Bifidobacterium breve for the probiotic use in humans and animals

  • The purpose of this study was to investigate probiotic characteristics and immune enhancing effects of Bifidobacterium (B.) longum KBB1-26 and BIF-4, B. breve KBB5-22 isolated from human intestine for probiotic use in humans and animals. We measured acid, bile and heat tolerance, antimicrobial activity against pathogenic bacteria, Escherichia (E.) coli, Salmonella (S.) Enteritidis, Staphylococcus (S.) aureus, and Listeria (L.) monocytogenes. Immune enhancing effects of B. longum and B. breve were investigated by measuring nitric oxide (NO), nuclear factor ($NF-{\kappa}b$), $interleukin-1{\beta}$ ($IL-1{\beta}$), interleukin-6 (IL-6), interleukin-12 (IL-12) and tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) in RAW 264.7 cells or RAW BLUE cells. B. longum KBB1-26 was survived at pH 2.0. B. longum KBB1-26 and BIF-4, B. breve KBB5-22 also showed tolerance to 0.3% of oxgall bile salt. B. longum KBB1-26 was able to survive at $70^{\circ}C$ and $80^{\circ}C$ for 20 min. KBB1-26 showed the antimicrobial inhibition zone to pathogenic bacteria such as E. coli (12 mm), S. Enteritidis (14 mm), S. aureus (14 mm) and L. monocytogenes (41 mm). The production of NO ($4.5{\pm}0.00{\mu}M/mL$) and $IL-1{\beta}$ ($39.7{\pm}0.55pg/mL$) of KBB1-26 significantly higher than BIF-4 and KBB5-22, respectively. In addition, KBB1-26 and KBB5-22 induce the production of high level of $TNF-{\alpha}$ and IL-6 in macrophages. Collectively, B. longum KBB1-26 have acid, bile, heat tolerance, antimicrobial activity and immune enhancing effects. These results suggest that KBB1-26 can be used as probiotics for humans and animals.

  • The present study evaluates the immunomodulatory effect of Bifidobacterium longum KACC 91563 in murine primary splenocytes and macrophages. B. longum KACC 91563 regulated T- and B-cell proliferation and inhibited the Th1 (IL-2, IFN-γ)/Th2 (IL-4, IL-10) cytokine imbalance and immune cytokine production. Moreover, immunoglobulin E (IgE) levels were significantly lower after treatment with B. longum KACC 91563. These findings suggest that B. longum KACC 91563 could modulate the systemic immune system toward both IgE production and regulation of the Th1/Th2 balance.

Bifidobacterium longum 산소변이주의 분리와 변이주의 산소내성 (Isolation and Characterization of Oxygen-tolerant Mutant of Bifidobacterium longum.)

  • Bifidobacterium의 효과적인 산업적 이용을 위해서는 산소에 내성이 강한 균주를 선발하는 연구뿐만 아니라, 산소 존재하에서 비피도박테리아의 내성기작을 규명하는 기초적인 연구도 필요하다. 산소에 민감한 B. adolescentis 균주와 산소내성 B. longum JI-1균주의 용존산소 제거능을 비교한 결과산소 민감성 균주는 총존산소를 전혀 제거하지 못하였으나 산소내성 균주는 10분 이내에 3%이상의 용존산소를 제거하여 총존산소 제거능과 Bifidobacterium의 산소내성과는 밀접한 관계가 있음을 알 수 있었다. 이 B. longum JI-1 균주를 미호기성 조건에서 배양하여 내산소성 돌연변이 균주인 B. longum ADJ-1 균주를 분리하였다. B. longum ADJ-1과 모균주의 특성을 당자화능, NADH 산화효소 및 세포지방산등으로 비교하였을 때 차이점을 발견하지 못했으며, 균생육은 B. longum ADJ-1이 모균주의 80% 정도를 보여 주었다. 그러나 B. longum ADJ-1는 매우 두꺼운 slime layer를 형성하였는데 confocal scanning laser microscopy에 의한 분석 결과 돌연변이 균주는직경이 약 6 $\mu\textrm{m}$에 이르는 층을 형성한 반면 모균주는 약 3 $\mu\textrm{m}$인 층을 형성하였다. 그리고 돌연변이 균주는 산소에 24시간 이상 노출되었을 경우 모균주에 비하여 더 큰 산소내성을 보여주었다. 그러므로 돌연변이 균주의 산소내성의 증가는 slime layer 차이에서 유래된 것으로 생각되며 이는 Bifidobacterium의 내산소성 기작중의 하나인 것으로 보인다.

High Cell Density Cultivation of Bifidobacterium longum Using a Calcium Carbonate-Alginate Beads System

  • A $CaCO_3$-alginate beads system was developed for high cell density cultivation of Bifidobacterium longum and the cost-effective media were also screened. In batch process with $CaCO_3$, beads, two strains of B. longum showed both the highest viable cells and optical density in TPY medium, resulting in maximum optical density and viable cell counts of 12.40, $2.22{\times}10^10$ cfu/ml for B. longum ATCC 15707 and 13.71, $3.93{\times}10^10$ cfu/ml for B. longum HLC 3742. Released size distribution, according to $CaCO_3$-alginate bead size preparation, was smaller than others. These results were also examined by observing their morphology. The skim milk-based medium was most adequate to cultivate B. longum as the cheapest medium, and $10\%$ skim milk supplemented with $2\%$ glucose and $1\%$ yeast extract was a suitable medium, supporting the growth to $5.57{\times}10^10$ cfu/ml for ATCC 15707 and $6.82{\times}10^9$ cfu/ml for HLC 3742. During the long-term storage at $4^{\circ}C\;and\;-20{\circ}C$, B. longum cultivated with $CaCO_3$ beads had the highest stability. Consequently, $CaCO_3$-alginate beads buffer was found to be useful not only to cultivate B. longum but also to preserve cultures.

Improvement of Bifidobacterium longum Stability Using Cell-Entrapment Technique

  • A cell-entrapment technique using compressed air was applied to Bifidobacterium longum KCTC 3128 for the improvement of bifidobacteria viability. The main cell-entrapment matrix used was alginate, and viability improvement of the B. longum entrapped in alginate lattices was monitored along with the effects of other additional biopolymers. A prerequisite for acquiring consistent results was the uniformity of bead size and cell distribution which was achieved by using compressed air and mixing the cell suspension with sterilized alginate powder, respectively. The viability losses of the B. longum entrapped in alginate beads in the presence of three different substances logarithmically increased in relation to the reaction time, and proportionately decreased with an increased alginate concentration and bead diameter. The strongest improvement in B. longum viability was exhibited with a bead containing 3% alginate and 0.15% xanthan gum.

산양유를 섭취한 성인 여성의 분변에서 분리한 Bifidobacteria와 Lactobacilli의 탄수화물 발효특성 (Carbohydrate Fermentation Character of Bifidobacteria and Lactobacilli isolated from Feces of the Adult Women supplied with Goat Milk)

  • 산양유을 섭취한 성인의 분변 내 bifidobacteria의 증식 효과 및 분변에서 분리된 bifidobacteria와 lactobacilli의 당 발효 능력을 조사하였다. 1. 산양유를 섭취한 처리군이 대조군보다 bifidobacteria 세균수가 상대적으로 높았으며, 8주에서 95% 신뢰한계에서 유의성이 있었다. 2. 분변에서 분리한 균주들을 13 균주의 16S rRNA 염기서 열로 동정한 결과, 산양유를 섭취한 처리구는 B. adolescentis, B. longum, B. pseudocatenulatum, B. dentium, L. sakei 순으로 분리되었다. 대조군의 12 균주는 B. adolescentis, B. longum, L. ruminis, L. sakei, B. pseudocatenulatum 순으로 분리되었다. 3. 산양유 올리고당과 lactulose는 모든 bifidobacteria 및 lactobacilli 균주가 발효하였다. Fructooligosaccaride는 B. adolescentis 7 균주 모두 발효하였다. 조사한 B. pseudocatenulatum 4 균주, L. sakei 3 균주, B. longum 7 균주 중에서 각각 3, 2, 1 균주가 fructooligosaccharide를 발효하였다.

빵의 저장성 증진을 위한 종균배양 방법 (Development of Starter Cultures for the Extension of the Shelf Life of Bread)

  • Starter cultures were developed for the extension of the shelf life of bread. Lactobacillus plantarum and Bifidoacterium longum were selected as the mixed culture with Propionibacterium freudenreichii. P. fredenreichii with B. longum and L. plantarum produced 2.261% and 1.715% total acid, and pH value of cultured solutions was near 3.95. Propionic acid was produced 14 ~16 mg/ml by P. freudenreichii with L. plantarum and B. longum during 17 days, which was more than that of L. brevis. The pH values of the bread were 4.83 and 4.89 in respect to B. longum and L. plantarum when the fermented products were used to make the bread. Mold was not found for 10 days of storage of the bread at room temperature when the fermented products of B. longum were used for making bread. 37$^{\circ}C$, pH 6.0 and the lowest agitation were optimum conditions for the production of propionic acid in the scaled up fermentation.

산, 담즙산, 산소 노출에 대한 비피도박테리아의 생존에 관한 연구 (Viability of Bifidobacterial Strains against Acid, Bile Acid, and Oxygen Exposure)

  • 한국인 유아에서 분리한 13개 균주를 포함한 19개 bifidobacteria 균주를 대상으로 산, 담즙산 및 산소 노출시의 생균수의 변화를 측정하였다. 내산성은 pH 4.0, 3.0, 2.0에서 2시간까지 측정하였는데, 실험 균주마다 매우 상이한 결과를 나타내었으며, 실험 균주 중에서 B. bifidum B3, B. longum D6, B. adolescentis Fl이 우수한 생존력을 나타내었다. 실험 균주를 0.2% thioglycolic acid와 0.2% oxgall이 함유된 MRS 배지에서 생장을 측정한 결과, B. bififdum B3과 B. longum D6이 담즙산에 우수한 저항성을 나타냈으며, 균종간의 경향은 관찰되지 않았다. 호기 상태에서 48시간 노출시킨 후에 생균수의 변화를 측정한 결과, 실험 균주마다 상이한 결과를 나타내었으며 상대적으로 B. bifidum과 B. longum균주들의 생존력이 우수하였다. 산, 담즙, 산소 노출에 대한 생존력은 균종간의 경향을 나타내지 않고 균주마다 상이한 결과를 나타내었는데, 실험 균주 중에는 B. bifidum B3과 B. longum D6 균주가 모두 우수한 생존율을 나타내었다. 이들 균주는 현재 시판되는 상업용 종균과 비교해서도 동등한 성적을 나타내어 상업용 종균으로서의 사용 가능성을 시사하였다.

Fermentation Conditions for the Production of Cell Mass and Comparison of Saccharide Utilization in Bifidobacterium longum and B. breve

  • Saccharide utilizations for the growth by Bifidobacterium longum and Bifidobacterium breve were compared. B. longum fermented glucose more rapidly than lactose as a carbon source whereas B. breve fermented lactose at a rate higher than that of glucose. The highest cell concentration, in the case of B. longum, was obtained when cultivated in a jar fermentor that contained modified MRS medium that half the beef extract was replaced by the same amount of tuna extract, and that pH was controlled at 6.0. B. breve showed the best growth when grown in a jar fermentor containing the MRS medium with lactose instead of glucose, controlled at pH 6.0. The optimal concentration of peptone in MRS medium for the growth of B. breve was 5 g/l.
