A Novel Inhibitor of Translation Initiation Factor eIF5B in Saccharomyces cerevisiae

  • The eukaryotic translation initiation factor eIF5B is a bacterial IF2 ortholog that plays an important role in ribosome joining and stabilization of the initiator tRNA on the AUG start codon during the initiation of translation. We identified the fluorophenyl oxazole derivative 2,2-dibromo-1-(2-(4-fluorophenyl)benzo[d]oxazol-5-yl)ethanone quinolinol as an inhibitor of fungal protein synthesis using an in vitro translation assay in a fungal system. Mutants resistant to this compound were isolated in Saccharomyces cerevisiae and were demonstrated to contain amino acid substitutions in eIF5B that conferred the resistance. These results suggest that eIF5B is a target of potential antifungal compound and that mutation of eIF5B can confer resistance. Subsequent identification of 16 other mutants revealed that primary mutations clustered mainly on domain 2 of eIF5B and secondarily mainly on domain 4. Domain 2 has been implicated in the interaction with the small ribosomal subunit during initiation of translation. The tested translation inhibitor could act by weakening the functional contact between eIF5B and the ribosome complex. This data provides the basis for the development of a new family of antifungals.

Retrieval Model Based on Word Translation Probabilities and the Degree of Association of Query Concept (어휘 번역확률과 질의개념연관도를 반영한 검색 모델)

  • One of the major challenge for retrieval performance is the word mismatch between user's queries and documents in information retrieval. To solve the word mismatch problem, we propose a retrieval model based on the degree of association of query concept and word translation probabilities in translation-based model. The word translation probabilities are calculated based on the set of a sentence and its succeeding sentence pair. To validate the proposed method, we experimented on TREC AP test collection. The experimental results show that the proposed model achieved significant improvement over the language model and outperformed translation-based language model.

Functional Equivalence of Translation Factor elF5B from Candida albicans and Saccharomyces cerevisiae

  • Eukaryotic translation initiation factor 5B (eIF5B) plays a role in recognition of the AUG codon in conjunction with translation factor eIF2, and promotes joining of the 60S ribosomal subunit. To see whether the eIF5B proteins of other organisms function in Saccharomyces cerevisiae, we cloned the corresponding genes from Oryza sativa, Arabidopsis thaliana, Aspergillus nidulans and Candida albican and expressed them under the control of the galactose-inducible GAL promoter in the $fun12{\Delta}$ strain of Saccharomyces cerevisiae. Expression of Candida albicans eIF5B complemented the slow-growth phenotype of the $fun12{\Delta}$ strain, but that of Aspergillus nidulance did not, despite the fact that its protein was expressed better than that of Candida albicans. The Arabidopsis thaliana protein was also not functional in Saccharomyces. These results reveal that the eIF5B in Candida albicans has a close functional relationship with that of Sacharomyces cerevisiae, as also shown by a phylogenetic analysis based on the amino acid sequences of the eIF5Bs.

Marine birnavirus (MABV)'s 5' terminal region of segment A acts as internal ribosome entry site (IRES)

  • Eukaryotic translation is initiated by either cap-dependent or cap-independent way, and the cap-independent translation can be initiated by the internal ribosomal entry site (IRES). In this study, to know whether the 5'UTR leader sequence of marine birnavirus (MABV) segment A and segment B can act as IRES, bicistronic vectors harboring a CMV promoter-driven red fluorescent gene (mCherry) and poliovirus IRES- or MABV's leader sequence-driven green fluorescent gene (eGFP) were constructed, then, transfected into a mammalian cell line (BHK-21 cells) and a fish cell line (CHSE-214 cells). The results showed that the poliovirus IRES worked well in BHK-21 cells, but did not work in CHSE-214 cells. In the evaluation of MABV's leader sequences, the reporter eGFP gene under the 5'UTR leader sequence of MABV's segment A was well-translated in CHSE-214 cells, indicating 5'UTR of MABV's segment A initiates translation in the cap-independent way and can be used as a fish-specific IRES system. However, the 5'UTR leader sequence of MABV's segment B did not initiate translation in CHSE-214 cells. As the precise mechanism of birnavirid IRES-mediated translation is not known, more elaborate investigations are needed to uncover why the leader sequence of segment B could not initiate translation in the present study. In addition, further studies on the host species range of MABV's segment A IRES and on the screening of other fish-specific IRESs are needed.

ON TRANSLATION LENGTHS OF PSEUDO-ANOSOV MAPS ON THE CURVE GRAPH

  • We show that a pseudo-Anosov map constructed as a product of the large power of Dehn twists of two filling curves always has a geodesic axis on the curve graph of the surface. We also obtain estimates of the stable translation length of a pseudo-Anosov map, when two filling curves are replaced by multicurves. Three main applications of our theorem are the following: (a) determining which word realizes the minimal translation length on the curve graph within a specific class of words, (b) giving a new class of pseudo-Anosov maps optimizing the ratio of stable translation lengths on the curve graph to that on Teichmüller space, (c) giving a partial answer of how much power is needed for Dehn twists to generate right-angled Artin subgroup of the mapping class group.

  • We prove in this paper the absolute continuity of the representing measures of the hypergeometric translation operators $\mathcal{T}_x$ and $\mathcal{T}_x^W$ associated respectively to the Cherednik operators and the Heckman-Opdam theory attached to the root system of type $B_2$ and $C_2$ which are studied in [9].

  • We study translation surfaces in the Euclidean 3-space ��3 and the Gauss map N with respect to the so-called Cheng-Yau operator ☐. As a result, we prove that the only translation surfaces with Gauss map N satisfying ☐N = AN for some 3 × 3 matrix A are the flat ones. We also show that the only translation surfaces with Gauss map N satisfying ☐N = AN for some nonzero 3 × 3 matrix A are the cylindrical surfaces.

Construction and Characterization of Vector Expressing Low Level of Translation Factor eIF5B (단백질합성인자 eIF5B의 저 발현 효모벡터의 제조 및 특성)

  • eIF5B is a translation initiation factor that delivers Met-$tRNA^{Met}$ to AUG start codon and subsequently joins the small and large ribosomes. In order to study the function of eIF5B encoded by FUN12, we constructed FUN12 which lacked 5' end of its sequence. We found that this construct lacking almost all of its promoter in pRS plasmid partially complemented slow growth phenotype of fun12 deletion strain. Interestingly, this construct expressed N-terminally truncated eIF5B and its expression level was about 5% of that of wild type eIF5B. Low amount of the eIF5B expressed additionally in fun12 deletion strain played a direct role as a limiting factor for its growth. This limiting factor eIF5B in those strains also modulates activities of overall translation in vitro.

EVALUATION FORMULA FOR WIENER INTEGRAL OF POLYNOMIALS IN TERMS OF NATURAL DUAL PAIRINGS ON ABSTRACT WIENER SPACES

  • In this paper, we establish an evaluation formula to calculate the Wiener integral of polynomials in terms of natural dual pairings on abstract Wiener spaces (H, B, 𝜈). To do this we first derive a translation theorem for the Wiener integral of functionals associated with operators in 𝓛(B), the Banach space of bounded linear operators from B to itself. We then apply the translation theorem to establish an integration by parts formula for the Wiener integral of functionals combined with operators in 𝓛(B). We finally apply this parts formula to evaluate the Wiener integral of certain polynomials in terms of natural dual pairings.

Probabilistic Part-Of-Speech Determination for Efficient English-Korean Machine Translation (효율적 영한기계번역을 위한 확률적 품사결정)

  • Natural language processing has several ambiguity problems, and English-Korean machine translation especially includes those problems to be solved in each translation step. This paper focuses on resolving part-of-speech ambiguity of English words in order to improve the efficiency of English analysis, which is in part of efforts for developing practical English-Korean machine translation system. In order to improve the efficiency of the English analysis, the part-of-speech determination must be fast and accurate for being integrated with machine translation system. This paper proposes the probabilistic models for part-of-speech determination. We use Penn Treebank corpus in building the probabilistic models. In experiment, we present the performance of the part-of-speech determination models and the efficiency improvement of the machine translation system by the proposed part-of-speech determination method.
