Phytoene, lycopene, ${\beta}-carotene$과 같은 카로티노이드는 식품의 착색제나 영양보조제, 사료첨가제, 화장품의 원료로 사용된다. 이전 연구에서 본 연구진은 분홍색의 색소를 생산하는 새로운 해양 세균인 K. gwangalliensis를 분리 동정하였다. Phytoene desaturase (CrtI) 효소는 crtI 유전자에 암호화되어 있으며, phytoene을 lycopene으로 전환하며, 카로티노이드 합성 초기 단계에 있어서 필수적이다. CrtI는 카로티노이드 생합성 조절의 주요 효소 중 하나이며, 다양한 카로티노이드를 생합성하는 생물들의 카로티노이드 생합성 경로에 있어서 속도 조절 단계에 관련이 있다. 본 논문에서는 K. gwangalliensis로부터 lycopene 생합성을 담당하는 crtI 유전자를 클로닝 하였으며, 이 유전자는 1,584개의 염기서열을 가지며, 527개의 아미노산을 암호화하고 있다. crtI 유전자의 염기 서열을 Kocuria rhizophila와 Myxococcus xanthus를 포함한 다른 종의 염기 서열과 비교한 결과, 진화 과정에서 잘 보존되어 있음을 확인하였다. crtI 유전자를 포함하는 발현 플라스미드를 구축하여 발현시킨 결과, 이 플라스미드를 함유하는 대장균은 약 57 kDa의 재조합 단백질을 생산화였으며, 이는 phytoene desaturase의 분자량에 해당한다. lycopene의 생합성은 lycopene 생합성에 필요한 crtE, crtB 유전자를 포함한 pRScrtEB plasmid를 E. coli에 형질전환 했을 때, Escherichia coli에서 합성되는 것을 확인하였다. 이 연구의 결과는 분자 수준에서 K. gwangalliensis CrtI의 1차 구조에 대한 폭 넓은 지식 기반을 제공할 것이다.
새로 합성된 KC6361 유도체는 기존 디페닐에테르계 화합물에서 보였던 증상(회백색)과는 달리 식물체의 백화를 유기시킨다. 따라서 이들 화합물의 구조상 특징이 무엇이며 식물체에서 어떠한 작용기작을 갖는지를 알기 위하여 본 실험을 수행하였다. 1) 백화를 유기시키기에 비교적 적당한 구조는 B환의 para 위치에 nitro기가 있고 A환의 meta 위치에 탄소수가 3개 이하의 alkyl 또는 allyl기를 가지는 carbamoyl이 치환되는 것이었다. 벼 등은 2kg/ha 수준에서 미미한 약해를 보여 비교적 좋은 선태성을 가졌고 바랭이, 피, 참방동산이, 비름 등은 0.25~0.5kg/ha에서 기타는 0.5~1.0kg/ha에서 거의 방제되었다. 2) 암조건에서 KC6361은 carotenoid 생성을 억제시켰으나 엽록소는 오히려 촉진시켰다. 명조건의 경우 높은 광도에서는 낮은 광도에서 보다, 엽록소보다는 carotenoid가 더욱 저해되었고 $^{4}C$-acetate의 지방성분으로의 혼입 저해정도는 미약하였다. 3) 단은방주의 생장에 미치는 영향이 norflurazon과 동일한 양상을 보였고, KC6361이 처리된 오이, 식용피에 있어서 carotenoid 성분변화를 볼 때 phytoene 및 phytofluene 함량은 증가된 반면 그 이후의 ${\beta}$-carotene은 감소되어 norflurazon과 같은 경향이었다. 따라서 이상의 결과를 종합하여 볼 때 KC6361의 작용점은 norflurazon과 같이 phytoene or/and phytofluene dehydrogenase를 저해하는 것으로 판단된다.
본 연구는 체중과다나 비만에서 총항산화능의 변화를 관찰하고, 총항산화능과 평소의 식사섭취상태와는 어떤 관계가 있는가를 관찰하고져 실시하였다. 20대의 젊은 남성을 대상으로 정상체중군(n = 17, BMI 20∼25, 비만지수 90∼110% 체지방 16∼20%)과 체중과다군(n = 13, BMI > 25, 비만지수 120%이상, 체지방 25%이상)으로 분류한 후, 인체계측, 생화학적 검사(총콜레스테롤, glucose, albumin, creatinine, GOT, GPT, uric acid), 식이섭취 조사(24회상법)를 실시하였다. 총항산화능은 체중과다군에서 유의적으로(p <0.01) 높게 나타났으며 , 측정된 생화학적 지수들에서는 uric acid의 함량이 체중과다군에서 유의하게(p<0.05) 높았다 평소의 식이섭취 상태는 두 군 모두 칼슘과 비타민 B2의 섭취량이 권장량에 비해 부족한 것으로 나타났으며, 체중과다군에서 비타민 C(p <0.05)의 섭취량이 유의적으로 낮게 나타났다. U-carotene과 비타민 E 섭취량에는 두 군간 차이가 없었고, 채소와 과일의 섭취량은 체중과다군이 적게 섭취한 반면, 음주와 흡연량은 체중과다군이 더 높게 나타났다. 두 군 모두 식사의 질은 양호하였다. 총항산화능 및 uric acid 함량과 측정지표들간의 상관관계를 살펴본 결과에서는 두 군 모두에서 총항산화능 및 uric acid 함량에서 서로 다른 경향을 보였다. 체중과다군에서는 총항산화능과 음주량이 부( - )의 상관 관계를 보였으며, uric acid 함량과 BMI, 비만지수, 및 총항산화능은 유의적인 정 (+)의 상관관계를 보였다. 이상의 결과에서 체중과다군은 정상체중군에 비해 총항산화능에 유의적인 차이가 있었으며 식이섭취상태와의 관련성은 나타나지 않았다. Uric acid 함량의 측정은 총항산화능과 정 (+)의 상관관계를 보여 또 다른 항산화능을 나타내주는 좋은 지표임을 알 수 있었다.
This study was conducted to identify dietary factors that may affect the occurrence of gastric cancer in Koreans. Preoperative daily nutrition intake and diet quality of patients diagnosed with gastric cancer were evaluated. Collected data were comparatively analyzed by gender. The results were then used to prepare basic materials to aid in the creation of a desirable postoperative nutrition management program. The subjects of this study were 812 patients (562 men and 250 women) who were diagnosed with gastric cancer and admitted for surgery at Soonchunhyang University Hospital between January 2003 and December 2010. Nutrition intake and diet quality were evaluated by the 24-hr recall method, the nutrient adequacy ratio, mean adequacy ratio (MAR), nutrient density (ND), index of nutritional quality (INQ), dietary variety score (DVS), and dietary diversity score (DDS). The rate of skipping meals and eating fast, alcohol consumption, and smoking were significantly higher in males than those in females. The levels of energy, protein, fat, carbohydrate, phosphorous, sodium, potassium, vitamin $B_1$, vitamin $B_2$, niacin, and cholesterol consumption were significantly higher in males than those in females. Intake of fiber, zinc, vitamin A, retinol, carotene, folic acid were significantly higher in females than those in males. MAR in males was significantly higher (0.83) than that in females (0.79). INQ values were higher in females for zinc, vitamin A, vitamin $B_2$, vitamin $B_6$, and folic acid than those in males. The average DVS was 17.63 for females and 13.19 for males. The average DDS was 3.68 and the male's average score was 3.44, whereas the female's average score was 3.92. In conclusion, males had more dietary habit problems and poor nutritional balance than those of females. Our findings suggest that proper nutritional management and adequate dietary education for the primary prevention of gastric cancer should be emphasized in men.
The objectives of this study were to investigate 1) the foods intake as well as nutrients intake, 2) the nutritional knowledge, and 3) the relations between the foods and nutrients intakes and nutritional knowledge in adolescent rhythmic gymnasts (RGs). The results are summarized as follows. Average daily intakes of energy, calcium, iron, vitamin A, vitamin B$_1$ vitamin B$_2$ and niacin were in 45-74% of the RDAs for Koreans. Average daily energy intake was only about 50% of energy expenditure by physical activities. Average daily intakes of each food group were generally lower, but intakes of breads and confectionaries, sugar and sweets, milks and dairy products, and instant foods were higher in the RGs compared to the nonathletic students of the same age from the National Health and Nutrition Suvey. The contribution of empty-calorie foods such as breads and confectionaries, beverage and instant foods to the major energy nutrients were high. Mean of total nutritional knowledge score of RGs was 28.4 of total score 50. In categorical score of nutritional knowledge, the RGs make a best score on food sources and they get the lowest marks for food exchange. Among the nutrients, intakes of crude fiber, animal Fe, Na, K, vitamin A, carotene, vitamin B$_1$ niacin and vitamin C and, among the food groups, intakes of mushrooms, fruits and meats had relations with one of the total or categorical nutritional knowledge scores in the RGs. In conclusion, nutritional status of adolescent RGs was poor due to their unbalanced diet composed of empty-calorie foods, and their undesirable food intake pattern was supposed to be related to the low nutritional knowledge score of them. These results indicate that the RGs should be given more nutritional knowledge to improve their nutritional status and the exercise performance.
Biomass and community composition of microphytobenthos in the Saemankeum tidal flat were studied by HPLC analysis of the photosynthetic pigments from November 2001 to November 2002. The environmental factors of sediment were also investigated to examine the relationship between microphytobenthos biomass and sedimentary environments. The detected photosynthetic pigments of microphytobenthos were chlorophyll a, b, c, fucoxanthin, 19'-hexanoyloxyfucoxanthin, violaxanthin, diadinoxanthin, alloxanthin, diatoxanthin, zeaxanthin+lutein, peridinin and beta-carotene. Pheophytin a, the degradation product of chlorophyll a, was also detected. The results of pigmen analysis suggest the presence of diatom (fucoxanthin), euglenophytes (chlorophyll b), chlorophytes (chlorophyll b + lutein), cyanobacteria (zeaxanthin), cryptophytes (alloxanthin), chrysophytes (fucoxanthin + violaxanthin), prymnesiophytes (19'-hexanoyloxyfucoxanthin) and dinoflagellates (peridinin). Chlorophyll a concentration in the top 0.5 cm of sediment was in the range of $0.24\;mg{\cdot}m\^{-2}\;-32.11\;mg{\cdot}m\^{-2}$ in the study area. The increase of chlorophyll a concentration in the spring indicates the occurrence of a microphytobenthic bloom. In the summer, there was a sharp decrease of the chlorophyll a concentration which was probably due to high grazing activity by macrobenthos. The annual mean chlorophyll a concentration in the study area was low compared to that in most of other tidal flat areas probably due to active resuspension of microphytobenthos and high grazing activity by macrobenthos. There was no clear relationship between microphytobenthos biomass and sedimentary environments because of a large variety of physical, chemical and biological factors, Pigment analysis indicated that while diatoms were dominated in the microphytobenthic community of the Geojon tidal flat, euglenophytes and/or chlorophytes coexisted with diatoms in the Mangyung River tidal flat.
To better investigate the antioxidative property of thermal treatment yam (Dioscorea batatas DECNE.) in Korea, some established methods were used. 70% Methanol, 70% ethanol and chloroform-methanol (CM, 2:1, v/v) extracts were collected. 70% Methanol extract exhibited stronger antioxidative activity evaluated by ferrous ion chelating activity, NO radical scavenging activity and ${\beta}$-carotene bleaching assays. On the contrary, CM extract was the most effective in inhibiting linoleic acid peroxidation. Yam available in Korea was also analyzed for its bioactive compounds such as lycopene, chlorophyll a, b, tannin, phytic acid and total saponin contents. Total saponin was abundant in thermal treatment, which determined to be $42.52{\pm}1.88mg/g$. Based on the results obtained from this study, thermal treatment yam could be used as natural antioxidant source due to its high antioxidant activity and bioactive compound contents.
Effective synthetic antioxidants such as butylated hydroxyanisole(BHA) and butylated hydroxytoluene(BHT) have been widely used in the food industry, but they are suspected to be toxic and carcinogenic effects. Therefore, the development of safely available natural antioxidants such as ascorbic acid, ${\alpha}$-tocopherol, ${\beta}$-carotene, flavonoids and selenium is essential. In particular, flavonoids, 2-phenyl-benzo-${\alpha}$-pyrones, are polyphenolic compounds that occur ubiquitously in food of plant origin. flavonoids occur in foods generally as O-glycosides with sugars bound usually at the C\ulcorner position. And variations in their heterocyclic ring gibes rise to flavones, flavonols, flavanones, flavanols, catechins, anthocyanidins, chalcone and isoflavones. Vegetables, fruits, and beverages are the main dietary sources of the flavonols, primarily as quercetin, kaempferol, and myricetin and the corresponding flavones, apigenin and luteolin. These flavonoids have biological activity such as antioxidant, anti-inflammatory, antithrombotic, antimutagenic, anticarcimogenic antiallergic and antimicrobial activity effects in vitro and in vivo. Flavonoids posses strong antioxidant activities acting as oxygen radicals scavenger, metal chelators and enzyme inhibitor. The antioxidant activity of flavonoids is determined by their molecular structure and more specially, by the position and degree of hydroxylation of the ring structure. All flavonoids with the 3`, 4`-dihydroxy(ortho-dihydroxy) posses marked antioxidant activity. And antioxidant activity increases with the number of hydroxyl groups substituted on the A-and B-rings. There is as yet no certainty about the effect of the presence of a double bond between C\ulcorner and C\ulcorner on the antioxidant activity of flavonoids.
Sa-Sang, one of the Korean traditional medicines, classifies people's constitution into 4 types, which are referred to as Tae-Yang-In, Tae-Eum-In, So-Yang-In and So-Eum-In. The purpose of this study was to examine demographic and dietary factors and to test any significant differences between Sa-Sang constitution and related factors. The participants were 483 college students and the data of 428 students were finally analyzed. Constitution was determined identically through both the questionnaire on Sa-Sang constitution and an inquiry by a Korean traditional medical doctor. Tae-Eum-In showed statistically significant higher BMI and higher smoking frequency. Tae-Eum-In also had significantly higher intakes in iron, potassium, sodium, vitamin B$_1$, niacin, $\beta$-carotene and vitamin E. So-Eum-In had significantly lower intakes in protein, phosphorous, and folate. From this study, we could expect to get some scientific, objective and accurate diet information on the types of constitution. Considering the fact that most chronic degenerative diseases could be developed by any lifestyle factors, it is necessary to conduct educational programs about lifestyles including the dietary habit for maintaining good health. Therefore, it will be better to continue a long-term follow-up study on any chronic degenerative disease based on the types of Sa-Sang constitution in the future. We investigated the quality and sensory characteristics of model tomato sauces which was made from fresh Vitaking tomatoes and American canned tomatoes in the mixture ratios of 0:100% ($S_1$), 25:75% ($S_2$), 50:50% ($S_3$), 75:25% ($S_4$) and 100:0% ($S_5$). Soluble solids, reducing sugars, vitamin C and organic acids increased as the contents of fresh tomatoes increased. pH and contents of $\beta$-carotene and lycopene decreased as the contents of fresh tomatoes increased. Lightness(L value) and yellowness (b value) increased as the contents of fresh tomatoes increased whereas redness (a value) didn't show any significant differences among samples. In the preference test, $S_3$ was the most preferred in red color, taste and overall preference. In the descriptive test, $S_1$ ranked the highest in redness, viscosity, palatability and flavor except for sweet and sour taste. $S_5$ was the lowest in redness, viscosity and palatability, being the highest in sour taste. From this result, we found that the mixture ratio of half fresh tomatoes and canned tomatoes was the best condition to make tomato sauce with preferred red color.
