Alkaline .alpha.-amylase expressed in the transformant, Baciollus subtills MB809, containing alkaline amylase gene cloned from alkalophilic Bacillus sp. AL-8, was purified through for step separation processes. The purified alkaline .alpha.-amylase had molecular weight of app[roximately 59, 000 daltons on SDS-PAGE and Sephaex G-100 gel filtration. Amino acid sequence of terminal portion of the enzyme was analyzed with pure amylase eluted form the SDS-PAGE gel. N-terminal amino acid sequence of .alpha.-amylase was determined by the Edman degradation method and resulted in $NH_{2}$-ser-thr-ala-pro-ser-(ile)-lys-ala-gly-thr-(ile)-leu. For C-terminal amino acid sequencing, purified .alpha.-amylase was digested with carboxypuptidase A and B, and reverse-phase HPLC gradient elution system resulted in -thr-trp-pro-lys-COOH.
The aceB gene, encoding for malate synthase, one of the key enzymes of glyoxylate bypass, was isolated from a pMT1-based Corynebacterium glutamicum gene library via complementation of an Escherichia coli aceB mutant on an acetate minimal medium. The aceB gene was closely linked to aceA, separated by 598 base pairs, and transcribed in divergent direction. The aceB expressed a protein product of Mr 83, 000 in Corynebacterium glutamicum which was unusually large compared with those of other malate synthases. A DNA-sequence analysis of the cloned DNA identified an open-reading frame of 2, 217 base pairs which encodes a protein with the molecular weight of 82, 311 comprising 739 aminoo acids. The putative protein product showed only limited amino acid-sequence homology to its counteliparts in other organisms. The N-terminal region of the protein, which shows no apparent homology with the known sequences of other malate synthases, appeared to be responsible for the protein s unusually large size. A potential calciumbinding domain of EF-hand structure found among eukaryotes was detected in the N-terminal region of the deduced protein.

For the DRT satellite transponder system, BER and total SNR to be required in the earth terminal are evaluated under the condition of HPA nonlinearity in the FBJ(full-band jamming) or PBJ(partial-band jamming) of uplink and downlink. In case that the satellite Inter bandwidth( Ws) is same to the earth terminal bandwidth($W_r$),in conditions of uplink JSR 10[dB], downlink JSR 10[dB] and processing gain 30[dB], linear transponder system shows that uplink SNR needs to be 14.2[dB] to achieve the total SNR 10[dB] requirement in downlink SNR 14[dB]. However, Nonlinear transponder system with OBO(output backoff) 2[dB] requires 20.1 [dB] uplink SNR. From the above results, the nonlinearity of HPA in the satellite transponder causes the degradation of BER performance so that it is of interest to consider the power increase.
M2PI is an active single chain mini-proinsulin with a 9-residue linker containing the turn-forming sequence 'YPGDV' between the B- and A-chains, but which retains about 50% of native insulin receptor binding activity. The refolding efficiency of M2PI is higher than proinsulin by 20-40% at alkaline pH, and native insulin is generated by the enzymatic conversion of M2PI. The solution structure of M2PI was determined by NMR spectroscopy. The global structure of M2PI is similar to that of native insulin, but the flexible linker between the B- and A-chains perturbed the N-terminal A-chain and C-terminal B-chain. The helix in the N-terminal A-chain is partly perturbed and the ${\beta}$-turn in the B-chain is disrupted in M2PI. However, the linker between the two chains was completely disordered indicating that the designed turn was not formed under the experimental conditions (20% acetic acid). Considering the fact that an insulin analogue, directly cross-linked between the C-terminus of the B-chain and the N-terminus of the A-chain, has negligible binding activity, a flexible linker between the two chains is sufficient to keep binding activity of M2PI, but the perturbed secondary structures are detrimental to receptor binding.
Following the introduction of containerized sea transportation, the size of container ships has progressively increased. While the primary objective of this enlargement is to achieve economies of scale, it significantly affects the operational environment of ports where these ships dock, raising concerns about the validity of efficiency gains from ultra large container vessels from a port operational perspective. This study evaluates the efficiency of various ship sizes from January to December 2022, utilizing internal data from the container terminals A and B located in the Busan New Port. The analysis encompasses ships affiliated with The Alliance at Container Terminal A and those associated with 2M at Container Terminal B. Key input variables considered for the study include ship size (TEU), total berth time, and loading/unloading time, with cargo handling performance serving as the only output variable. The findings indicate that larger ship sizes exhibit higher efficiency compared to smaller container ships.
Seven soybean varieties and 21 F 2 hybrids of a diallel cross among these varieties were used as the materials, and genetic studies were conducted to evaluate the frequency and distribution of genes governing the characters of soybean leaf shapes. Partial dominance was exhibited by petiole length of trifoliates, petiole length of terminal leaflets, length of terminal leaflets, length of lateral leaflets, width of terminal leaflets Mean values of length of terminal leaflet were higher than those of lateral leaflets and length of leaflets were higher than width of leaflets, but mean values of width of terminal leaflets were shorter than those of lateral leaflets.
Against environmental stresses, many bacteria utilize the alternate sigma factor RpoS that induces transcription of the specific set of genes helpful in promoting bacterial survival. Intracellular levels of RpoS are determined mainly by its turnover through proteolysis of ClpXP protease. Delivery of RpoS to ClpXP strictly requires the adaptor protein RssB. The two-component-type response regulator RssB constantly interacts with RpoS, but diverse environmental changes inhibit this interaction through modification of RssB activity, which increases RpoS levels in bacteria. This review discusses and summarizes recent findings on regulatory factors in RssB-RpoS interactions, including IraD, IraM, IraP anti-adaptor proteins of RssB and phosphorylation of N-terminal receiver domain of RssB. New information shows that the coordinated regulation of RssB activity in controlling RpoS turnover confers efficient bacterial defense against stresses.
Ankyrins are a ubiquitously expressed family of intracellular adaptor proteins involved in targeting diverse proteins to specialized membrane domains in both the plasma membrane and the endoplasmic reticulum. We described here that the C-terminal domain of ankyrin-B interact specifically with Z-line portion of titin in yeast two-hybrid analysis, in vitro pull-down assays and localization experiments in COS-7 cells. In this study we provide the first experimental evidence that Z-line portion of titin is necessary for the localization of ankyrin-B and ankyrin-B links between the sarcolemma and the myofibril in costameres.

In this paper, the I/Q mismatch problem between the transmission line and antenna switch is solved by a new CPW matching circuit for a VoWLAN terminal with 54 Mbps communication speed through the IEEE 802.11 b/g CCK/OFDM. I/Q performances with the new matching circuit are analyzed and improved by the output spectrum mask, the characteristics of the EVM(Error Vector Magnitude) due to the nonlinear CCK/OFDM demodulation data, and receiver's sensitivity.
