Effect of Plant Growth Regulators on in vitro Cultured Atractylodes Hybrid 'Dachul' (A. macrocephala x A. japonica) (기내배양 백출 교잡종 '다출'(Dachul, Atractylodes macrocephala x A. japonica)에 미치는 생장조절제처리효과)

  • This study was conducted to establish the tissue culture system for Atractylodes plant which is most frequently used in oriental medicine. Root and auxiliary bud of Dachul cv., which is Atractylodes hybrid (A. macrocephala x A. japonica), were used as target tissues for in vitro culture. In root culture, callus induction rate was higher in the treatment of BAP combined with NAA than others, however, 2-iP was more effective for callus proliferation and root induction. Although calli were effectively induced from the root and proliferated in lower concentration of cytokinin combined with higher auxin, root tissue was inappropriate for shoot regeneration. For plant regeneration with axillary bud, BAP combined with NAA was more effective than 2-iP with NAA or IBA. Number of regenerated plant per bud was 3.8, which was highest, and stem diameters was shown as 5.0mm under the conditions of 1 mg/L BAP combined with 1 mg/L NAA. Although, plant height was tend to be higher in 2-iP than BAP, number of the regenerated plant was lower via versus. Furthermore, root proliferation of regenerated plant was more effective in higher concentration of sucrose (7%) than in lower concentration (3%). In results, auxiliary bud was an efficient target tissue for producing regenerated plant of Atractylodes under the conditions of 1 mg/L BAP combined with 1 mg/L NAA and higher concentration of sucrose was effective for root proliferation of regenerated plants.

  • Here, we describe an efficient and rapid protocol for the micropropagation of Thymus pallidus Cosson ex Batt., a very rare medicinal and aromatic plant in Morocco. After seed germination, we tested the effect of different macronutrients, cytokinins alone or in combination with gibberellic acid (GA3) or auxins, on T. pallidus plantlet growth. We found that Margara macronutrients (N30K) had the best effect on the in vitro development of the plantlets. The addition of 0.93 μM/L 1,3-diphenylurea (DPU), 0.46 μM/L adenine (Ad), and 0.46 and 0.93 μM/L kinetin (Kin) resulted in the best shoot multiplication and elongation. In addition, the combination of 0.46 μM/L Kin, DPU, or Ad with gibberellic acid, in particular, 0.46 μM/L Ad + 0.58 μM/L GA3 and 0.46 μM/L Kin + 1.15 μM/L GA3, led to better bud and shoot multiplication. Moreover, the integration of the combinations of 0.46 μM/L Kin and auxins, namely 0.46 μM/L Kin + 2.85 μM/L indole-3-acetic acid (IAA), 0.46 μM/L Kin + 2.85 or 5.71 μM/L indole-3-butyric acid (IBA), and 0.46 μM/L Kin + 0.3 or 0.57 μM/L 1-naphthaleneacetic acid (NAA), in the culture medium led to better root development and optimized aerial growth. Finally, the in vitro plants from the medium containing N30K + 0.46 μM/L Kin + 2.85 μM/L IAA were successfully acclimatized; these plants served as a source for repeating in vitro culture.

In vitro shoot initiation of Artocarpus heterophyllus Lam. (Jak Fruit) Effect of the explant type and the season of explant collection

  • A method for rapid propagation of mature Jack fruit was developed. Four types of explants (mature embryos, apical meristems of young seedlings, apices from mature plants and nodal segments) were used. It has been found 88% of young apical meristems produced shoots in Campbell and Durzan (CD) medium compared to 60% in Murashige and Skoog (MS) medium. Only 1/3 of them produced multiple shoots. Shoot initiation from nodal segments was very rare. Mature apices produced callus. Although removal of the sheathing cover around mature buds enhanced the shoot initiation but success rate was low in growth regulator free medium. Embryos respond to the CD medium but not to the MS medium. Embryos from seeds soaked in water for 24 hours produced shoots after 8 weeks of incubation and the success rate was 70% while embryos from dry seeds only produced roots. There was no significant effect of cold storage (refrigeration) for 7 days on shoot initiation from mature embryos (65%) but the ability for shoot induction declines with storage time (55% after 21 days of cold storage). Mature axillary buds were established in Modified Campbell and Durzan (CD) medium supplemented with 0.5mg/1 and IBA. There was a significant difference in the growth performance of shoots according to the period of the year in which explants were collected. Highest (60%) was observed in November-January period. It was only 30% when the explants were collected in February-April or May-July and decreased to 20% in August-October. The shoots produced in November-January showed a higher vigor than those produced in other months. Since Jak fruit show seasonal changes in fruit bearing and shedding of leaves, it can be suggested that the difference in growth performances of tissues cultured in artificial culture media would have been affected by endogenous rhythms.

  • 최근 호두 기내 소비량은 급증하나 대부분 실생묘로 조림되어 결실까지 장기간 소요되며, 무성 번식률이 낮아 보급확대가 어렵다. 본 연구는 호두나무 관행 묘목 번식법 개선이 가능한 우량 국산묘 대량증식 기술 개발로 최적 배양조건을 구명하고자 수행하였디. 호두나무 등 목본류는 식물 조직배양 기본배지에서 산화방지제를 첨가하여 사용하는 경우, 배양체에서 생기는 페놀 화합물(Phenolic compound)의 작용을 억제하여 배양 식물체의 갈변(Browning) 또는 흑변(Blackening) 현상을 감소시키는 효과가 있다. 페놀 추출물은 배지로 빠져나와 갈변시키며 생존율을 저하시키는 원인이 되고 이를 제거하는 산화방지제는 배양 식물체의 생산효율을 높이는 효과를 나타내는 강력한 항산화제(Antioxidant) 역할을 한다. 호두나무의 액아를 배양하게 되면 조직 및 배지의 갈변화가 심하게 일어나는데, 휴면 정도가 깊을 수록 정단에서 추출되는 페놀성 물질의 함량이 증가하며, 식물 조직 내 페놀성 물질의 함량 정도와 배양의 고사율과는 상관관계가 있다. 이러한 식물 배양체에서 생기는 페놀 화합물에 의해 발생되는 배양상의 문제점을 해결하기 위하여 계대배양을 자주해 주거나 활성탄(Activated charcoal), Ascorbic Acid 등의 첨가물을 배지에 추가하는 방법이 있다. 이에 따라 호두 '신령' 품종의 액아배양의 페놀집적을 제거하기 위해 배지 내에 산화방지제 첨가물질 등을 농도별로 처리하였다. 호두나무 액아 배양 후 8주차 산화화방지제 첨가물질의 농도별 처리 효과는 생존율은 85.3%를 보였으며, 신초 형성율 76.4%, 신초수 1.9개/주, 신초장 27mm/주의 양호한 결과를 나타냈으며, 페놀 제거 및 유식물체 고사 억제 효과를 보였다.

Micorpropagation of Corylopsis coreana by Thidiazuron Treatment (Thidiazuron 처리에 의한 히어리나무의 기내번식)

  • This study was conducted to evaluate the effect of thidazuron(TDZ) on shoot proliferation and growth from axillary buds of 20-years-old Corylopsis coreana. Shoots proliferation was effectively achieved on WPM(Woody Plant Medium) supplemented with 0.03∼0.1mg/L TDZ. The highest shoot number(6.5$\pm$0.7) was obtained on 0.1mg/L TDZ treatment. On the TDZ medium shoots formed as clusters less than 1cm in height and therefore needed to subculture on GA$_{3}$ containing medium to induce elongation. In consecutive cultures, phenolic compounds were excreted at the proximal part of the explants and inhibited growth of the explants. Growth inhibition by the compounds was overcome using liquid and paper bridge culture system. About 60% of the elongated shoots rooted on half- strength MS medium containing IBA. Generally, IBA was mire effective on in vitro rooting than NAA with optimal range of 0.5mg/L to 1.0mg/L. Rooted plantlets were transferred in an artificial soil(vermculite) and acclimatized in high humidity greenhouse condition. Survival rate differed greatly depending on rooting types of the explants. Two types of rooting were observed. The first type was direct rooting from the explants. The second type was callus formation followed by rooting from the callus. The explants showing the 1st type rooting survived can be multiplicated in vitro by TDZ treatment followed by elongation with GA$_{3}$ and rooting with IBA.

  • This study was undertaken to know the possibility of in vitro propagation of Stevia through axillary bud culture and the results indicated that: (1) Addition of NAA (0.01-0.05 mg/l) alone on Murashige-Skoog basal medium promoted shoot differentiation and growth rate. And also additional of kinetin of 0.5-1.0 mg/1 alone showed the same trend as that of NAA: (2) Addition of both NAA (0.01-0.05 mg/l) and kinetin (0.5-1.0mg/l) to MS medium promoted better shoot formation. (3) Shoot differentiation and growth were better on the full salt strength of MS medium (1X MS) than that of half strength ( $\frac{1}{2}$MS), while their effects were reversed for root differentiation

  • These studies were designed to define the effects of Benzyladenine and 2, 4-D on the induction and growth of callus tissue from embryos and plant segments of Korean ginseng. 0.5PPM was the minimum concentration of 2, 4-D for the induction of callus tissue from embryos and plant segments of ginseng. Best callus induction occurred at a 2, 4-D concentration of 5 mg/liter but growth of this callus was best at a 2, 4-D concentration of about 1.0 to 2.0 mg/liter and benzyladenine was ineffective as callus inducer. When the embryos were grown on the media containing 0.5 mg/liter of 2, 4-D, 5 to 6 axillary buds were formed at the basal part of epicotyle.

An Effective Selection of PAT Gene Transformed Populus alba $\{times}$ Populus glandulosa No.3 using Herbicide Basta Treatment (제초제 Basta를 이용한 Phosphinothricin Acetyltransferase 유전자로 형질전환된 현사시 3호의 효율적인 선발)

  • This study was conducted to simple transformants selection by herbicide Basta treatment after transformation with Agrobacteium tumefaciens MP90/PAT in hybrid poplar(Populus alba ${\times}$ P. glandulosa No. 3). In preliminary study, we determined that the lethal concentration of herbicide Basta was 1.0mg/L in callus culture, adventitious bud formation and axillary bud elongation experiment. By the treatment of 1.0mg/L Basta, we could be selected the transformed shoots effectively from the various cultures. In addition, the treatment was useful on selection of transformants which are growing in soil pot. Finally, we also confirmed the transformation by PAT assay, Above results show that the herbicide Basta treatment and PAT assay can be a very simple and effective method for the identification of PAT gene transformed hybrid poplar.

In vitro plantlet regeneration of "dwarf" Indian olive (Elaeocarpus robustus Roxb.): a fruit plant of Bangladesh

  • A plantlet regeneration protocol was developed on pot-grown mature plants of Elaeocarpus robustus Roxb. cv. Dwarf from nodal and leaf explants. The best yield of adventitious shoots was achieved from the leaf-derived calli in a modified MS ($MMS_1$, half strength of major salts, full strength of minor salts, and vitamins) medium containing $4.0{\mu}M$ BA + $4.0{\mu}M$ Kn + $0.5{\mu}M$ NAA + 15% coconut water (CW). The shoot multiplication rate was amplified about twofold per culture after the addition of 15% CW to the medium. The rate of shoot multiplication reached maximum at the 5th subculture, and it maintained this rate throughout the 3 subsequent subcultures. The best rooting in vitro was investigated by subculturing the microcuttings in an $MMS_2$ (half strength of both major salts and minor salts and full strength of vitamins) medium containing $1.0{\mu}M$ IBA in the dark for one initial week at $30^{\circ}C$, followed by subculturing them in a plant-growth regulator (PGR)-free medium in the light. The plantlets raised in vitro were successfully established under ex vitro conditions.
