Avicularin Inhibits Lipopolysaccharide-Induced Inflammatory Response by Suppressing ERK Phosphorylation in RAW 264.7 Macrophages

  • Avicularin, quercetin-3-${\alpha}$-L-arabinofuranoside, has been reported to possess diverse pharmacological properties such as anti-inflammatory and anti-infectious effects. However, the underlying mechanism by which avicularin exerts its anti-inflammatory activity has not been clearly demonstrated. This study aimed to elucidate the anti-inflammatory mechanism of avicularin in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophage cells. Avicularin significantly inhibited LPS-induced excessive production of pro-inflammatory mediators such as nitric oxide (NO) and $PGE_2$ and the protein levels of iNOS and COX-2, which are responsible for the production of NO and $PGE_2$, respectively. Avicularin also suppressed LPS-induced overproduction of pro-inflammatory cytokine IL-$1{\beta}$. Furthermore, avicularin significantly suppressed LPS-induced degradation of $I{\kappa}B$, which retains NF-${\kappa}B$ in the cytoplasm, consequently inhibiting the transcription of pro-inflammatory genes by NF-${\kappa}B$ in the nucleus. To understand the underlying signaling mechanism of anti-inflammatory activity of avicularin, involvement of multiple kinases was examined. Avicularin significantly attenuated LPS-induced activation of ERK signaling pathway in a concentration-dependent manner. Taken together, the present study clearly demonstrates that avicularin exhibits anti-inflammatory activity through the suppression of ERK signaling pathway in LPS-stimulated RAW 264.7 macrophage cells.

Analytical Method Development of Avicularin and Quercitrin in Astilbe chinensis Extract Using HPLC (HPLC를 이용한 노루오줌 추출물의 Avicularin 및 Quercitrin 분석법 개발)

  • This study attempted to eatablish a high performance liquid chromatography (HPLC) analysis method for the determination of avicularin, quercitrin as a part of the quality control for the development of functional cosmetic materials from Astilbe chinensis extract. HPLC was performed on a Capcell Pak C18 MGII column with a gradient elution of 0.05% (v/v) trifluoroacetic acid (TFA) and acetonitrile at a flow rate of 1.0 mL/min at 30 ℃. The analyte was detected at 254 nm. The HPLC method was performed in accordance with the international conference on harmonization (ICH) guideline (version 4, 2005) of analytical procedures with respect to specificity, precision, accuracy, and linearity. The limits of detection and quantitation of avicularin and quercitrin were 0.094 and 0.285 mg/mL, 0.031 and 0.095 mg/mL respectively. Calibration curves showed good linearity r2 > 0.99990 for avicularin and r2 > 0.99994 for quercitrin. Precision of analysis was satisfied with less than 0.59% for avicularin and 0.63% for quercitrin. Recoveries of quantified compounds ranged from 100.97 to 101.77% for avicularin and 100.18 to 100.32% for quercitrin. These result indicated that the established HPLC method is very useful for the determination of marker compounds in A. chinensis extracts.

Anti-lipid Peroxidation and Liver Protective Effects of Polygonum aviculare L. (마디풀(Polygonum aviculare L.) 성분의 지질과산화억제 및 간보호에 미치는 효과)

  • The ethylacetate fraction of the overground portion of Polygonum aviculare L. exhibited the anti-lipid peroxidation and the liver protective effect in intoxicated rats. Through silica gel chromatography of the ethylacetate fraction monitered by bioassay, two flavonoids, avicularin and juglanin were isolated as active components. Avicularin and juglanin remarkablely inhibited the lipid peroxidation of rat liver induced by 50% ethanol. Especially avicularin exhibited the stronger anti-lipid peroxidation effect than juglanin. Avicularin as a main principle of Polygonum aviculare L. significantly exhibited liver protective activities by decreasing s-GOT and s-LDH levels which represent for the hepatotoxicity induced by $CCl_4$ in rats. In addition, avicularin significantly decreased not only s-LDH but also s-bilirubin levels in intoxicated rat induced by ${\alpha}-naphthylisothiocyanate\;(ANIT)$. These results suggest that avicularin has the protective effects against the hepatoxicity induced by $CCl_4$ and ANIT in rats.

Anti-oxidant activity of avicularin and isovitexin from Lespedeza cuneata

  • This study evaluated the anti-oxidant activity of avicularin and isovitexin from Lespedeza cuneata using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and hydroxyl (OH) scavenging assays. The results showed that among the four fractions, the highest OH radical scavenging activity was exhibited by the n-butanol fraction. The DPPH radical scavenging activities of avicularin and isovitexin were higher than 50% at a concentration of $100{\mu}g/mL$. Moreover, one hundred micrograms per liter of avicularin and isovitexin exhibited effective anti-oxidant activity, with the OH radical scavenging rates being 87.54 and 91.48%, respectively. These results suggest that Lespedeza cuneata would be useful anti-oxidant agents from natural sources.

Anti-inflammatory Constituents of Polygonum bistorta (권삼의 소염활성성분(I))

  • Five compounds were isolated from the BuOH fraction of Polygonum bistorta (Polygonaceae). On the basis of spectral data, these compounds were established as caffeic acid, quercimeritrin, avicularin, gallic acid and protocatechuic acid. The inhibitory activies on $3{\alpha}-hydroxysteroid$ dehydrogenase $(3\;{\alpha}-HSD)$ of isolated compounds were compared. $IC_{50}$ value of isolated compounds were $133.57\;{\mu}g/ml\;(caffeic\;acid)$, $89.1\;{\mu}g/ml\;(quercimeritrin)$, $189.85\;{\mu}g/ml\;(avicularin)$, $140.69\;{\mu}g/ml\;(gallic\;acid)$ and $165.27\;{\mu}g/ml\;(protocatechuic\;acid)$ respectively. Although all compounds showed lower inhibition activities than BuOH fraction $(IC_{50}<50\;{\mu}g/ml)$ of Polygonum bistorta, it showed higher inhibition activities than aspirin $(IC_{50}\;246.81\;{\mu}g/ml)$.

  • Euphorbia humifusa Willd (EuH), called Ttang-Bin-Dae in Korea, is a traditional medicinal plant widely used for its anti-inflammatory and antiviral activity. Ethyl acetate (EA) extracts of EuH (EA/EuH) inhibit invasion and metastasis by inhibiting tumor necrosis factor TNFá-induced matrix metalloproteinases (MMP)-9 expression in human breast cancer cells. However, the bioactive components of EA/EuH mediating the inhibition of MMP-9 expression have not been identified. In the present study, three bioactive constituents of EA/EuH were isolated using high-performance liquid chromatography. Nuclear magnetic resonance spectroscopy revealed isoquercetin, avicularin, and astragalin as the bioactive compounds responsible for preventing TNFα-induced MMP-9 mRNA expression in breast cancer cells. These findings suggest that isoquercetin, avicularin, and astragalin could be used as valuable anti-metastatic agents against metastatic cancers.

Simultaneous Determination of Avicularin and Quercitrin in Astilbe chinensis by HPLC

  • This study developed a HPLC analysis method for the determination of avicularin (AL) and quercitrin (QT) in Astilbe chinensis by HPLC. HPLC was performed on a Capcell Pak C18 MGII column ($4.6{\times}250mm$, $5{\mu}m$) with a gradient elution of 0.05% (v/v) trifluoroacetic acid (TFA) and acetonitrile at a flow rate of 1.0 mL/min at $30^{\circ}C$ and a UV detection wavelength of 254 nm. The analytical method was validated for its specificity, precision, accuracy, and linearity. The limits of detection and quantitation were AL 0.09 and 0.28 mg/mL, QT 0.03 and 0.09 mg/mL respectively. Calibration curves showed good linearity (AL r2 > 0.99990 and QT r2 > 0.99994), and the precision of analysis was satisfied (less than AL 0.59%, QT 0.63%). Recoveries of quantified compounds ranged from AL 100.57 to 102.08%, QT 99.94 to 100.64%. These results indicate that the developed HPLC method was applied successfully to the detection of AL and QT in A. chinensis.
