Effects of Culture Type and Inoculation Quantity in Bioreactor on Production of Potato Plantlets

  • Potato (Solamum tuberosum 'Dejima') plantlets were investigated on culture type and initial quantity of inoculation in bioreactor and survival rate by hydroponics for mass production. rode stems (1 to 1.5cm in length) of potato plantlets multiplied in vitro were grown for 3 weeks in liquid Murashige and Skoog (MS) medium with sucrose $30 g\; L^{-1}$. When plantlets (80-node inoculation) were raised in 10L balloon type bubble (BB) bioreactor, the healthiest growth of plantlets was obtained from explants cultured in ebb & flow culture with medium supplied periodically 12 times per day. The suitable inoculation quantity of 20L BB bioreactor was 120 pieces of stem segments (mean 2.2g fresh weight) in ebb & flow culture. Number of nodal shoot was eight on the average. In controlled culture room, survival rate of plantlets at 7 days after stem cutting was above $70\%$ when they were acclimatized by hydroponics grown in deep flow and solid medium culture. The highest survival rate of the stem cutting plantlets was in nutrient solution adjusted to EC $1.4dS{\cdot}m^{-1}$. Stem cutting plantlets through one culture could be obtained $670\~900$, when plantlets were grown in ebb & flow culture during 3 weeks using a 20L bioreactor with initial 120 pieces of nodal segments. 11 is possible In do mass production of seedlings cultured in bioreactor and hydroponics.

Effects of Rice Straw Incorporation by Cutting Methods on Soil Properties and Rice Yield in a Paddy Field (볏짚 혼입이 논 토양개선 및 쌀수량에 미치는 영향)

  • This study was to investigate the effects of rice straw incorporation by cutting height on paddy soil fertility. The average residual amount of rice straw by cutting height were showed 1,420 kg $ha^{-1}$, 1,850 kg $ha^{-1}$, and 2,400 kg $ha^{-1}$ for depths of 10 cm, 15 cm, and 20 cm, respectively. For soil physical properties, soil hardness and bulk density were decreased while porosity was increased by rice straw incorporation. But soil organic matter (SOM), available silicate content, and cation exchange capacity (CEC) were significantly decreased when rice straw was removed from the field. These results indicated that the SOM as residual amount of rice straw was influenced by level of cutting height. Milled rice yield was increased by 28% and 32% for cutting heights of 15 cm and 20 cm, compared with that of control, respectively. The number of spikelets per square meter and the percentage of ripeness were increased with increasing incorporation by lower level of cutting height of rice straw. Therefore, incorporation of rice straw practices under cutting method influenced soil improvement and rice yield in paddy field.

Effect of in vitro Culture Condition and Lines on Growth Pattern of Lateral Bud from Nodal Cutting of Phalaenopsis Flower Stalk (팔레높시스 기내 화경 배양조건 및 계통이 액아의 발육형태에 미치는 영향)

  • This study was carried out to investigate the effects of in vitro culture condition and among lines on growth pattern of lateral buds from nodal cuttings of Phalaenopsis flower stalks. The ratio of bud growing into shoot from nodal cuttings of flower stalks were 90.9% and 54.4% on MS and hyponex medium, respectively. The number of buds grown vegetatively were increased remarkably on the MS medium containing 5 mg/L BA. The rate of buds grown vegetatively was higher in basal and middle parts than in upper part of flower stalks. The flower stalk sections cultured at 25~28$^{\circ}C$ showed the highest ratio of vegetative growth. Medium contamination was decreased by pretreatment of etiolation to the flower stalk. However, the pretreatment did not show specific effects on shoot development and reduction of phenolic compound. Average shoot number which was formed from flower stalk segments in 27 of 30 accessions were 3.17, while high number of shoots were obtained from Phal. 3020 and Phal. 3039. The growth pattern of lateral buds in F$_1$hybrids was similar to that of their parents.

  • This experiment is conducted fer method cf bring to perfectly from defective cutting sapling 25 a production method of sapling which is to substitute for conventional graf ting. 1. In green wood cutting root comes out after 15 days of cutting with nearly straight development of root system; after 40 days of cutting, roots with total length of 1119 mms 43 roots, and 5.4 grs a root in total weight obtained. 2. Survival percentage of green wood cutting ranges between 56% and 90%, average 73% of that and it varies with natural characteristics of varietes. The results show variety of Gaeryang-Suban and Iljiroe with 80% as a best ones in contrast with the variety of Shipyung. 3. The varietics or Gaeryang-Suban. Iljiroe, Suwon-Sang No.4, Rosang makes of much more roots than Yongchonchuwoo and Shipyung do. 4. Root ability made good number of roots commercially, when cutting is conducted soaking in 0.01 % NAA solution or 0.02% NAA solution for 2 or 3 seconds as a chemical promoter. 5. Economical measures for increase of scion adapted 1) 2 scions from a green wood 50 cm long should be taken and basal pan of scion at its middle portion should be cut right under the node. 2) Scions below 50cm long may be used. Small scions or growthceased shoots h3ve also considerable root ability enough to be used. 3) Thus far, up 100,000 scions might be produced in 10 a. 6. We can find number of root increased, when cutting the opposite side in obliquity manner at 450. 7. When 110,000 saplings in 10 a. for the production of bring to perfectly from defective cutting sapling planted, quality of stocks does not go to bad condition without any obstacles in practical use. 8. Although the times or grafting delayed until middle of July, quality of sapling goes just a little down. Grafting may be conducted from early June to middle of July separately in several times, and the green wood of prunned mulberry in spring is available for a scion after end of June. 9. 10 grs weight of defective cutting sapling makes 95% of complete sapling, otherwise 5 or 10 grs in weight of one becomes 80% of complete sapling with its quality as similar as grafted one. 10. When the sapling planted, its branches should be cut, leaving 3 or 4 buds at the bottom of new branches. 11. In view of economical stand point, production cost of bring to perfectly from defective cutting sapling obtains 52% of grafting cost.

Effects of Cutting Time, Auxin Treatment, and Cutting Position on Rooting of the Green-wood Cuttings and Growth Characteristics of Transplanted Cuttings in the Adult Prunus yedoensis (왕벚나무 성목 녹지삽목에서 삽목시기, 옥신처리 및 삽수부위가 발근에 미치는 영향과 이식 삽목묘의 생육특성)

  • This study was conducted to develop an efficient mass propagation method for the mature $Prunus$ $yedoensis$ Matsumura (43 to 58 years old). Cutting was conducted depending on cutting time, auxin treatments (IBA and NAA treatments mixed with talc powder), and cuttings position on shoots in a plastic house equipped with a fog system without heating. Rooted cuttings were transplanted to a nursery bed, and their growth characteristics were investigated in order to check whether the cuttings are successful or not for roadside tree planting. The average rooting rate was highly significant ($P$ < 0.0001) in all treatments: cutting on June 1st (61.4%) was more than two times greater in rooting rate than that on August 1st (23.6%); IBA 1,000 $mg{\cdot}L^{-1}$ (90.8%) and IBA 500 $mg{\cdot}L^{-1}$ (89.2%) showed much greater rooting rates than those of the other treatments; upper part of the cuttings treated with IBA 1,000 $mg{\cdot}L^{-1}$ showed the highest rooting rate, 96.7%. The interactions among treatments in the average rooting rate were also significant. There were significant differences ($P$ < 0.0001) among the auxin treatments in the survival rate of leafed cuttings transplanted to a nursery bed. The average survival rate was 46.5%, and IBA 1,000 $mg{\cdot}L^{-1}$ treatment was the highest in leafed cuttings 79.2%, but most of leafless cuttings were dead. There were significant differences ($P$ < 0.0001) among the cuttings, grafts, and in the seedlings height, diameter at root collar, the number of roots, branches, and leaves, etc., and the cuttings was the best. We can expect a possibility of mass propagation of improved $P.$ $yedoensis$ Matsumura and a high planting survival rate through the transplanting of cuttings to a nursery bed in which the cuttings should be the following conditions: cutting in June to July, use of the upper part of cuttings, IBA treatment, and rooting in August in a cutting-greenhouse equipped with a fog system.

Characterization and Evaluation of Worker s Exposure to Airborne Glass Fibers in Glass Wool Manufacturing Industry (유리섬유 단열재 제조업 근로자의 공기중 유리섬유 폭로 특성 및 평가 방법에 관한 연구)

  • To characterize worker's exposure to glass fibers, to find the correlation between airborne total dust concentrations and fiber concentrations and to recommend an appropriate evaluation method for worker's exposure to fibrous dusts in glass wool industry, we carried out this study. Average respirable fiber levels at five factories were 0.013-0.056 f/cc, and fairly below the OSHA PEL, 1 f/cc. A factory showed the lowest airborne fiber level, 0.013 f/cc, which was different significantly from those of other factories of which average fiber concentration was 0.046 f/cc. The cutting and grinding operations of insulation products resulted in higher airborne fiber cocentrations than any other processes(p<0.05). To characterize airborne fiber dimension, fiber length and diamter were determined using phase contrast microscope. The geometric means of airborne fiber lengths were $42-105 \mu m$. One factory had airborne fibers whose length distribution(GM = $105 \mu m$) was different from those of other factories(GM = $42-50 \mu m$). The percentages of respirable fibers less thinner than 3 gm were 38.9-90.9% at four factories, and two factories of them had the higher percentages than others. The findings explain for variation of airborne fiber diameters between factories. On the other hand, between the processes were the difference of fiber-length distributions observed. The cutting and grinding operations showed shorter fiber-length distributions than the fiber forming one. However, fiber-diameter distributions or respirable fiber contents were similar in all processes. The airborne fiber concentrations and the dust concentrations had relatively weak correlation(r=0.25), thus number of fibers couldn't be expected reliably from dust amount. Fiber count is appropriate for assessing accurate exposures and health effects caused by fibrous dusts including glass fibers. Ministry of Labor have established occupational exposure limit to glass fibers as nuisiance dust, but should establish it on the basis of respirable fiber concentration to provide adequate protection for worker's health

Influence of Polyethylene Film Mulch and Seedling Types on Growth and tuber Yield of Fall-Grown Potato (폴리에틸렌필름 멀칭 및 종묘종류가 가을감자의 생육과 수량에 미치는 영향)

  • Normal seed tubers and plug seedlings raised from stem cutting and mini-tubers ($10\pm3$ g) were planted or transplanted on August 20,2001 in bare soil or under transparent and black polyethylene film mulches to evaluate the effects of mulch material and seedling type on the growth and tuber yield of fall-grown potato (Solanum tuberosum L. cv. Dejima) in Jeju region. Total tuber number per plant was 4.6 regardless of mulch treatment, and direct planting seed potatoes resulted in the greatest tubers (5.14)/plant and followed by transplanting seedlings raised from cutting and mini-tubers (4.15 and 5.15 tubers/plant). Transparent and black polyethylene film mulches increased total tuber yield by 16 and 8%, respectively, and average tuber weight by 14 and 12%, respectively, compared with no mulch (21.16 t/ha and 72.2 g/tuber). Transplanting plug seedlings raised from cutting resulted in the greatest tuber yield, followed by transplanting plug seedlings raised from mini-tubers and direct planting seed potatoes.

Natural Regeneration of Tree Species after Clear-cutting in a Coniferous Plantation (침엽수(針葉樹) 인공조림지(人工造林地) 개벌(皆伐) 후의 교목류(喬木類) 천연갱신(天然更新))

  • To study the regeneration process of tree species after clear cutting, we investigated the density of seedling occurred after 1 year in the coniferous forest of Sikotuko Hokkaido, northern Japan that was clearcut after windthrow damage due to typhoon. As the results, 25 species of tree seedlings were growing and the density of seedlings and sprouts was $9.8trees/m^2$ ($1.25tres/m^2{\sim}54.44trees/m^2$) in the area of clear cutting. The 87% ($8.6/m^2$) of all seedlings was current seedlings and non-animal dispersal seedlings (average density $7.2tree/m^2$) were about 5 times more than animal-dispersal seedlings ($1.4tree/m^2$). The seedling density was beyond $6tree/m^2$ within 10m and $2{\sim}9tree/m^2$ in 150~250m from a natural forest. Number of non-animal dispersal seedlings were decreased along the distance from a natural forest but there was not such a tendency in animal dispersal seedlings. The variation in seedling density was higher in non-animal dispersal seedling than in animal dispersal seedling. In natural regeneration of tree species after clear-cutting, the possibility that pioneer species like Betula spp. etc. will be composed of the major species is high. Therefore, in order to maintain the species diversity, the nurture work for reducing competition among the individuals is necessary.

Selection of a New Allium tuberosum R. Cultivar 'Dongjanggun' for protected horticulture in winter (동계 시설재배용 재래부추 '동장군' 선발)

  • ‘Dongjanggun’ was developed at the Gyeongbuk Agricultural Research and Extension Services, Taegu in 2000. It was selected among the local varieties gathered korean native chinese chive from 1993 to 1996. It has longer and narrower leaves, stronger flavor than check variety ‘Greenbelt’. And also, contents of ascorbic acid in the leaves of ‘Dongjanggun’ was higher than the ‘Greenbelt’s. Number of tillers per plant was greater in ‘Dongjanggun’ than that in ‘Greenbelt’ during the winter in unheated plastic house. Its growth and regrowing velocity after cutting of top leaves were vigorous. Number of flowers and seeds per umbel were fewer in ‘Dongjanggun’ than in ‘Greenbelt’. Average yield was 6% higher than ‘Greenbelt’s during the thee years.

  • The purpose of this study is to understand women's wear manufacturing industries. First, the study was to investigate the present production systems and how much the automatic facility are by comparing them. This study enhanced more efficient, stable, and suitable work line. This intern will direct the way in which automatic facilities will be created. Second, through this study on the general character of the inspectors, the ratio of impaired goods, and the reasons for unsatisfactory goods, I intended to find out a way to decrease the impaired goods and to produce competitive and high quality goods. The results of the survey can be summarized as follows; 1. The result of the research on the automatic industrial facilities shows that the majority of the factories (77.4%) are 40% below the automatic facility rate. The reasons for this according to order are that was a deficit in money, no reason for expensive machines, and lack of the technique and the number of workers required to handle the machines. 2. At this time, the most required equipments are shown according to its importance; automatic sewing machine, automatic cutting machine, automatic spreading machine, and finishing & pressing machine. So in the women's wear manufacturing industries, they think that they need more automatic cutting machine, automatic spreading machine in the cutting field rather than high price automatic machine in the sewing field such as pattern former, pocket welting, automatic sleeve connecting machine and automatic label connecting machine. 3. The result of the research in the goods quality shows that the average impaired rate is 12.7% at the first inspection. In addition the average rate for complete impaired rate is 1.52%. The line system shows that it has a impaired rate that is double the rate of the pair system. Because of this, the industries plan to combine the line system and pair system to create an improved and suitable production system which can boost the quality and productivity of the goods. 4. The fabric is the main point of the impaired goods. The factors of the impaired goods in manufacturing are the lack of mental abilities of the worker, impaired fabrics and a lack of cooperation in the working system. Furthermore, there is a lack of technique for new material. 5. To prohibit impaired goods in manufacturing, there need to be a way to educate the workers and to enhance the workers' mind on the productive goods. Also there need to increase in the investments of automatic production machines. Finally there need to be a standardized working line. Therefore, there need to be an improvement on the management of the production of goods, the development of technique and an increase in the education for the workers, with this there will be a decrease in impaired goods, and an increase in better quality of goods to enforce the domestic apparel industries.
