Effects of Heat Treatment on Soybeans With and Without the Gene Expression for the Kunitz Trypsin Inhibitor: Chick Growth Assays

  • A total of 864 broiler chicks were used at Kansas State University and the University of Nebraska to determine the effects of heat treatment of two soybean genotypes on the growth performance. The soybeans were Williams 82 variety with (+K) and without (-K) gene expression for the Kunitz trypsin inhibitor. Heat treatment (autoclaving at $121^{\circ}C$ and $1.1kg/cm^2$) was applied for 0, 3, 6, 12, 18, and 24 min, resulting in a $2{\times}6$ factorial arrangement of treatments. Station and station treatment effects occurred, indicating that response in nutritional value of the soybean genotypes to heat treatment varied from year to year and location to location. However, the interactions were in magnitude of response rather than direction of response, with greater reductions in trypsin inhibitor concentrations for the soybeans heat processed at the Nebraska location. Pooled data indicated that -K supported greater (p<0.001) ADG, ADFI and gain/feed than the +K genotype. As the length of heat treatment increased, the ADG, ADFI, and the gain/feed ratio increased for chicks fed both soybean genotypes (p<0.0001). However, heating the -K soybeans resulted in a greater response in ADG, ADFI, and gain/feed than heating the +K soybeans (genotype heat treatment interaction, p<0.001). Pancreatic weights (mg pancreas/g of BW) of chicks fed -K soybeans were reduced compared to those from chicks fed +K (p<0.001). Increasing heat treatment decreased pancreas weights in chicks fed both soybean genotypes (p<0.001). Chicks fed heated soybeans in the Nebraska experiment had lower pancreatic weights than chicks fed heated soybeans in the Kansas experiment (station heat treatment interaction, p<0.0001). Chick growth performance was improved and pancreatic weights decreased by feeding raw -K soybeans versus raw +K soybeans, and by increasing heat treatment of both soybean genotypes. However, the response to heat treatment was not independent of genotype. Both +K and -K soybeans heated for 24 min supported similar ADG, ADFI, gain/feed, and pancreas weights, although chicks fed raw +K soybeans had lower growth performance than chicks fed -K soybeans. In conclusion, raw -K soybeans supported greater growth performance in broiler chicks than raw +K soybeans, although this advantage was lost when both soybean genotypes were heated for 24 min. Heat treatment of +K soybeans supported similar growth performance to heated -K soybeans, even though +K soybeans supported lower rates and efficiencies of gain than -K soybeans when fed raw.

Reconstruction with Extracorporeally Radiated Autogenous Bone Graft After Wide Resection of Bone Tumors (골종양 절제후 방사선 조사한 자가골을 이용한 재건술)

  • PURPOSE : For the reconstruction of large bone defect after tumor resection, it is possible to reuse the bone involved by tumor with some treatment to it. Several bone-reusing methods have been reported such as autoclaving, low-heat treatment(pasteurization) and intraoperative radiotherapy. We have used extracorporeally radiated autogenous bone graft for reconstruction after tumor resection, and analyzed the periods for junctional union, functional results and complications to know the indications of this method. METHODS : From Dec. 1993 to Sept. 1995, nine patients had taken autogenous bone graft with extracorporeal irradiation. Eight cases were osteosarcoma and 1 giant cell tumor. The graft sites were 5 in femur, 3 proximal tibia and 1 femur and tibia. Stage 3 was 1 case(GCT), Stage IIB 3 and Stage IIIB 5. After wide resection, surrounding soft tissue and intramedullary and extramedullary portion of the tumor were removed. Radiation was done in 5000cGy to the resected bone. Ender nails and bone cement were inserted and filled into the medulla to prevent fracture. RESULTS : Average follow-up period was 12.3(4 to 21) months. Average junctional union period in simple X-ray was 6.5 months in 4 cases. Average functional score following Enneking's criteria was 19(12-27). Complications were as follows ; condylar fractures and femur neck fracture in 4 cases, subluxation of the knee joint 3 and infection 1. Although local recurrence was detected in 1 case, the site of recurrence was not in the radiated bone but surrounding soft tissue. At final follow-up, no recurrence was found in one case(GCT), CDF 2, AWD 2, DOD 3, and died of chemotherapy related sepsis 1. CONCLUSIONS : Extracorporeally radiated bone autograft is considered to be a method for reconstruction of the large bone defect made by tumor resection, especially in the reconstruction around the joint.

Effect of Cooking Methods on S-Allyl-L-Cysteine Content of Garlic (마늘의 조리방법에 따른 S-Allyl-L-Cysteine 함량 변화)

  • This study investigated changes in the S-allyl-L-cysteine (SAC) content of garlic using different cooking methods. Methods for determining SAC were validated by determining specificity, linearity, limit of detection (LOD), limit of quantification (LOQ), precision, and accuracy using an high-performance liquid chromatography-fluorescence detection system. Results showed high linearity in the calibration curve with a coefficient of correlation ($R^2$) of 0.9999. The LOD and LOQ values for SAC were 0.15 and $0.47{\mu}g/mL$, respectively. The relative standard deviations for intra- and inter-day precision of SAC were less than 5%. The recovery rate of SAC was in the range from 97.35% to 97.47%. The SAC content of raw garlic was 2.77 mg/g, and there was no significant difference in SAC content according to blanching and microwave treatment. However, SAC content upon boiling and autoclaving at $110^{\circ}C$ and $121^{\circ}C$ increased from 3.50~9.16 mg/g, 6.52~16.21 mg/g, and 14.15~50.24 mg/g with increasing cooking temperature and time, respectively.

  • To investigate the mode of bactericidal action for antimicrobial peptide, pediocin, synthetic and mutant pediocins were prepared by direct chemical synthesis. Native pediocin was purified from Pedio-coccus acidilactici M and its conformational structure and bactericidal functions were analyzed and compared to synthetic pediocin. Schematic mode of pediocin actions, how pediocin binds on the target cell membrane, penetrates and makes tunnel are proposed. For these purposes, primary and secondary structures of pediocin was analyzed and disulfide bond assignment was also done. The pediocin purified from P. acidilactici M had high effective bactericidal ability against gram positive bacteria, especially Listeria monocytogenes and was very stable at extreme pHs and even at high temperatures such as autoclaving temperature (121$^{\circ}C$). Pediocin was consisted of 44 amino acids with four cysteines. Novel synthetic peptides were achieved by solid phase peptide synthesis(SPPS) method. To explain the function of cysteine in C-terminal region, mutant pediocin, Ped[C24A+C44A], was synthesized and their structural and biological functions were analyzed. Second mutant pediocin, Ped[KllE], was prepared to explain the function of lysine at 11 of N-terminal part of pediocin, especially loop of $\beta$-sheet, and to predict the initial binding site of pediocin. The native and synthetic pediocins was showed random coil conformation by spectropolarimetry in moderate conditions. This conformation was observed in extreme conditions such as high temperature and low and high pHs, also. Circular dichroism(CD) data also showed the existence of $\beta$-turn structure in N-terminal part both native and synthetic pediocins. A structural model for pediocin predicts that 18 amino acids in the N-terminal part of the peptide assume a three-strand $\beta$-sheet conformation. This random coil in C-terminal part of pediocin was converted to folding structure, helix structure, in nonpolar solvents such as alcohol and TFE. The disulfide bond between $^{9}$ Cys and $^{14}$ Cys was concrete and inevitable, however, evidences of disulfide bond between $^{24}$ Cys and $^{44}$ Cys was not. Data of Ped[C24A+C44A], pediocin mutant showed that $^{44}$ Cys was required during killing the target cells but not inevitable, since Ped[C24A+C44A] still have bactericidal activity but much less than native pediocin. Another pediocin mutant, Ped[KllE], had still bactericidal activity, was controversial to propose that positive charge like as $^{11}$ Lys in loop or hinge in bacteriocin bound or helped to binding to microorganism with electrostatic interaction between cell membrane especially teichoic acid and positive amino acid nonspecifically. The conformation of pediocin among native, synthetic and mutant pediocins did not show big difference. The conformations between oxidized and reduced pediocin were almost similar regardless of native or synthetic.

  • The persistence of butachlor and nitrofen in different soil conditions applied organic matter, lime, and other pesticides was studied under submerged and field moisture capacity. Degradation of the herbicides in soil was significantly retarded by autoclaving the soil and half-life of nitrofen was much longer than that of butachlor under this condition. Submerging the soil enhanced degradation of the herbicides, in particular that of nitrofen. On the other hand, half-life of nitrofen under field moisture capacity was twice longer than that of butachlor. Increased amendment of rice straw to the soil shortened the half-life of nitrofen under submerged soil, however it prolonged that of butachlor when the amendment was exceeded 1000kg/10a level. Liming the soil stimulated herbicide decomposition in the soil, which appears to be pH independent. Butachlor degradation in submerged soil was slightly stimulated by simultaneous application of fungicides and insecticides, but nitrofen persistence was not influenced.

Investigation of the Transfer Rate of Zearalenone in Herbal Medicines to Their Decoction (한약재 중 제랄레논의 탕액으로 이행률 조사)

  • The objective of this study was to establish an analytical method to detect zearalenone (ZEA) in herbal medicines and their decoctions and investigate the ZEA transfer rate from raw materials of herbal medicines to their decoctions. Herbal medicines (Trichosanthis Semenm, Eucommiae Cortex, Rubi Fructus) spiked with a known concentration of ZEA were presoaked or unsoaked (as a pretreatment) and boiled for 3 h at $100^{\circ}C$ or autoclaved for 1 h at $121^{\circ}C$. The decoction and the remnants were separated, cleaned up with an immunoaffinity column, and analyzed using HPLC. Recoveries for decoctions and remnants were 68.39-83.68% and 72.91-80.25%, respectively. ZEA was not detected in the decoction, whereas it was found in the remnants. Although ZEA in the raw material of herbal medicines was not transferred into the decoction during heating and autoclaving, the continuous monitoring for ZEA in raw herbal medicines should be carried out for the safe ingestion and utilization of herbal medicines.

  • In order to study the change of nitrogeneous compounds during the "Natto" preparation, the contents of insoluble protein, water soluble protein and amino nitrogen were determined and the pattern of peptides and amino acids was investigated by paper chromatography for the fractions resulting from molecular sieving. The results are summarized as follows: 1. Insoluble protein nitrogen which was increased to 84% by autoclaving the native soybean decreased to 44%, whereas the trichloroacetic acid soluble nitrogen increased from 8% to 45% during Natto preparation, But the soluble protein nitrogen showed a slight increase. 2. Fractionation of the peptides using Dowex-50 resins showed that they consisted mostly of lower molecular weight peptides which increased in accordance with the progress of fermentation, especially after 30-hour period. 3. Sixteen known and two unknown amino acids, and three peptides with different Rf values were identified during the Natto preparation. Their appearance showed some difference in that phenylalanine appeared after 10 hours, methionine, after 20 hours and proline, after 30 hours, respectively. The three peptides appeared at the different stage of fermentation.

THE EFFECT OF NAOCL TREATMENT AND STERILIZATION PROCEDURES ON THE CORROSION OF ENDODONTIC FILES (차아염소산나트륨 처리와 멸균법이 근관 치료용 파일의 부식에 미치는 영향에 관한 연구)

  • A variety files made of stainless steel (S-S) or nickel-titanium (Ni-Ti) are used during endodontic treatment. The purpose of tt)is study was to evaluate the corrosion susceptibility of S-S and Ni-Ti endodontic files. Three brands of files were used for this study: $K-flex^{(R)}$ S-S files (Maillefer, USA), $Profile^{(R)}$ Ni-Ti files (Maillefer, USA), $K-3^{(R)}$ Ni-Ti files (SybronEndo. USA). 120 files of each brands (21mm, ISO size $\#20$) were divided into 12 groups according to 1) sterilization methods using Autoclave or Ethylene Oxide (E-O) gas, 2) Irrigation solutions using $5.25\%$ NaOCl or Saline, 3) the number of sterilization (1, 5, 10 times), After above procedures, each of the files was inspected by three examiners with a light microscope and camera at X25. Each file was judged and ranked according to the following criteria: 0;, no corrosion, 1; mild corrosion, 2; moderate corrosion, and 3; severe corrosion. The files of high score were examined under the Scanning Electron Microscope. Data were statistically analyzed with the Kruskal-Wallis test (p < 0.05). Most of the ten time-autoclaved files had showed mild to moderate corrosion. But, one or five time-autoclaved files did not show corrosive surface. NaOCl treatment and E-O gas sterilization did not influence on corrosion. There was a significant difference in corrosion susceptibility between sterilization methods and the number of autoclaving. However, there was no significant difference between brands and file materials.

Synthesis of Na-A Type of Zeolite from Funnel-Glass Waste (브라운관의 후면유리 폐기물을 이용한 제올라이트 합성)

  • Through alkaline hydrothermal activation processes, Na-A type zeolite was synthesized as a single phase with funnel-glass waste from a television tube factory. The autoclaving was performed in a closed teflon vessel in the range of 80~95$^{\circ}$C. The silica-rich solution as a starting material was hydrothermally synthesized with quartz in IN NaOH by heating 350uC under the pressure of 1,500 atm. $NaAlO_2$ was made from NaOH and Al(OHh by heating 95$^{\circ}$C for 2-3 hours and the molar ratios of it were $Na_2O/Al_2O_3$ = 1.4 and $H_2O/Na_2O$=8. The equi-dimensional A type zeolite (1-2 11) was formed by the simple mixing of the silica-rich solution, glass waste and $NaAlO_23$ for 1-3 hours-heating at $80^{\circ}C$. The characterization of the reaction product shows Na-A as a single phase. The synthesized zeolite has cuba-dodecahedral form and $Ca^{2+}$ ion exchange capacity of the Na-A was in the range of 215-220 mequiva1entilOO g.

Preparation Conditions of Agar Oligosaccharides with Organic Acids (한천 올리고당 제조를 위한 유기산 처리 조건)

  • The optimum organic acid and temperature conditions were investigated for the preparation of oligosaccharides from agar. The tested organic acids were acetate, citrate, lactate, malate, and succinate and the conditions for oligosaccharides preparation were $0.3\%,\;0.5%;and\;0.7\%$ organic acid concentrations at $80\~120^{\circ}C.$ The low concentration of organic acid below $0.3\%$ decreased the degrading ratio and the high concentration up $0.5\%$ could not changed the degrading ratio. Conditions below $100^{\circ}C$ was not good for degrading agar. But $100^{\circ}C\;or\;120^{\circ}C$ was optimal temperature conditions for agarooligosaccharides according to the organic acid type and concentration. The organic acid concentration was $0.5\%$ and organic acid was the citrate or malate. The treatment time considered optimum was 120$\~$180 min. The maximal degrading ratio giving optimum conditions such as $100^{\circ}C\;and\;120^{\circ}C\;was\;35.5\%\;and\;38.7\%,$ respectively. The agarooligosaccharides prepared by autoclaving at $100^{\circ}C\;and\;120^{\circ}C$ were 2$\~$7 species oligomer.
