Target Identification for Metabolic Engineering: Incorporation of Metabolome and Transcriptome Strategies to Better Understand Metabolic Fluxes

  • Metabolic engineering is now a well established discipline, used extensively to determine and execute rational strategies of strain development to improve the performance of micro-organisms employed in industrial fermentations. The basic principle of this approach is that performance of the microbial catalyst should be adequately characterised metabolically so as to clearlyidentify the metabolic network constraints, thereby identifying the most probable targets for genetic engineering and the extent to which improvements can be realistically achieved. In order to harness correctly this potential, it is clear that the physiological analysis of each strain studied needs to be undertaken under conditions as close as possible to the physico-chemical environment in which the strain evolves within the full-scale process. Furthermore, this analysis needs to be undertaken throughoutthe entire fermentation so as to take into account the changing environment in an essentially dynamic situation in which metabolic stress is accentuated by the microbial activity itself, leading to increasingly important stress response at a metabolic level. All too often these industrial fermentation constraints are overlooked, leading to identification of targets whose validity within the industrial context is at best limited. Thus the conceptual error is linked to experimental design rather than inadequate methodology. New tools are becoming available which open up new possibilities in metabolic engineering and the characterisation of complex metabolic networks. Traditionally metabolic analysis was targeted towards pre-identified genes and their corresponding enzymatic activities within pre-selected metabolic pathways. Those pathways not included at the onset were intrinsically removed from the network giving a fundamentally localised vision of pathway functionality. New tools from genome research extend this reductive approach so as to include the global characteristics of a given biological model which can now be seen as an integrated functional unit rather than a specific sub-group of biochemical reactions, thereby facilitating the resolution of complexnetworks whose exact composition cannot be estimated at the onset. This global overview of whole cell physiology enables new targets to be identified which would classically not have been suspected previously. Of course, as with all powerful analytical tools, post-genomic technology must be used carefully so as to avoid expensive errors. This is not always the case and the data obtained need to be examined carefully to avoid embarking on the study of artefacts due to poor understanding of cell biology. These basic developments and the underlying concepts will be illustrated with examples from the author's laboratory concerning the industrial production of commodity chemicals using a number of industrially important bacteria. The different levels of possibleinvestigation and the extent to which the data can be extrapolated will be highlighted together with the extent to which realistic yield targets can be attained. Genetic engineering strategies and the performance of the resulting strains will be examined within the context of the prevailing experimental conditions encountered in the industrial fermentor. Examples used will include the production of amino acids, vitamins and polysaccharides. In each case metabolic constraints can be identified and the extent to which performance can be enhanced predicted

The isolation of Bacillus subtilis KYS-10 with antifungal activity against plant pathogens (식물 병원균에 대한 항진균 활성을 갖는 Bacillus subtilis KYS-10의 분리)

  • This study was investigated for the purpose of the isolation and identification of antagonistic bacteria with antifungal activity against plant pathogens. This bacteria denominated Bacillus subtilis KYS-10 and the optimum growth condition were 4% sucrose, 1% yeast extract, 0.2% $K_2HPO_4$, pH 7, 150 rpm, $30^{\circ}C$, 8 day. The antifungal activities against nine plant pathogens determined inhibition zone size by diffusion methods. The results, G. zeae (scab) 70 mm and P. grisea KACC 40439 (blast), P. capsici KACC 40177 (phytophthora blight) and C. destructans KACC 41077 (root rot of ginseng) 40~43 mm, and C. gloeosporioides KACC 43520 (ripe rot), C. gloesporioides KACC 40003 (anthracnose), S. shiraiana KACC 41065 (stem rot) and S. shiraiana (mulberry sclerotial disease) 35~39 mm and F. Oxysporum KACC 44452 (bulb rot of ginseng) 28 mm. From these experiment results, author suggest that Bacillus subtilis KYS-10 would be developed as a biological control agent thorough the field experimet in the near future.

An Empirical study on the analysis of the re-using of four-digit personal identification numbers - A university case (네 자리 숫자 비밀번호 재사용 실태 분석 연구 -A대학 사례연구)

  • This research aim is to investigate the rate and pattern of re-using the four-digit personal identification numbers(PINs). 1313 types of PINs were observed by 224 students who took this author's classes from last 2006 to 2011 at A-university. Some students used PINs as few as 3-4 and as many as 12-13. The average is 5.86 per person. The rates of re-using PINs were calculated by each student. 87%(195/224) of students reused PINs and 64% of them reused with just only one type of PINs, 20% reused with 2 types, and about 3% reused with 3-4 types. With respect to PINs, 884 out of the total 1313 PINs were reused, that is around 64.3%. In a broad sense, a pair of slight modification of PINs were also observed, that is, new PINs were partly matched in position or size of numbers of previous PINs. And if the reuse rate falling under the slight modification of PINs, 10.4% is added, about 75% of the PINs were reused in a broad sense. The re-using rate of male students is higher than the one of female students. This paper's results may provide to make plan for hacks of passwords.

Systematic Review on Researches of Sasang Constitution Diagnosis Using Facial Feature (안면형상을 활용한 사상체질 진단 연구에 관한 체계적 고찰)

  • Objectives : This study proposes developing Sasang Medical Diagnosis Program using Facial form for increase in Sasang Constitution Diagnosis objectivity and putting the Diagnosis Program into practical use. The author presents a review of extant research on Sasang constitution diagnosis utilizing facial feature analysis and suggests an agenda for further research. Methods : For this thesis, a collection of dissertations on the subject of 'Usage of facial form for constitution diagnosis' published until September of 2012 such as RISS4U, OASIS, KISTI, Korean TK were reviewed. The final 33 dissertations were classified into two categories, basic or clinical research and then analyzed. Results : 9 out of 33 dissertations were of basic research and 24 were of clinical research. 1) As result of review of references, a uniform tendency was found in facial form according to Sasang Constitution. 2) In the grade of practical use, facial element is repeatedly used and the facial element of important use has constitutional differences. 3) Standard faces per Sasang Constitution were derived as result of 2-dimensional research. 4) 3-dimensional research focused on improvement of accuracy and reliability of 3D-AFRA, and there has been an attempt to develop a prototype for identification. Conclusions : For practical use of facial feature in Sasang Constitution Diagnosis, 1) Standardization of diagnosis through establishing Sasang Medical Diagnosis clinical protocol must be preceded. After the standardization, practical purpose and direction of facial form in general may be decided. 2) Information on high quality facial form of constitutional and conditional patients must be collected to form extensive database. 3) Subdivided symptomatology, as well as Sasang Constitution must be considered for diagnosis in order for diagnosis technique to acquire clinical practicality.

  • In this paper, we propose a GRNN(: Generalized Regression Neural Network) algorithms for new eyes and face recognition identification system to solve the points that need corrective action in accordance with the existing problems of facial movements gaze upon it difficult to identify the user and. Illite Polytypes: The Characteristics and the Application to the Fault Age Determination (일라이트 폴리타입: 그 특성과 단층 활동연대 결정에의 활용)

  • The 1M and $2M_1$ stacking sequences are the most frequently encountered in the illite species among the possible 6 polytypes. The $1M_d$, derived from the 1M polytype which exhibits a variable degree of disorder in the stacking sequence, is also observed in illite samples. In this paper, the author introduces and reviews the theoretical background of the quantitative analysis method of illite polytypes, and considers the possibility to determine the fault age and its reactivation age using K/Ar age-dating based on the quantification of illite polytypes in the fault system. For the increase of the accuracy and precision of the illite age analysis method, the occurrence, identification, and mineralogical characterization of illite polytypes should be defined in detail. The broadening effect of (hkl) reflections, due to disordering of 1M polytype and the presence of I/S minerals with expandability, are also considered as the main parameters controlling the quantification of illite polytypes using the WILDFIRE(C)simulation.

  • The development of molecular biology has brought many changes in psychiatry. Molecular biology makes us possible to know the cause of mental disorders that provide the way to prevent the disorders, and to develop various accurate diagnostic and treatment methods for mental disorders. The author discusses the concept, cause, and treatment of mental disorders in the aspect of molecular biology. Importing the methods of molecular biology into psychiatry, we can anticipate to get a number of the goals of psychiatric genetics, including identification of specific susceptibility genes, clarification of the pathophysiological processes whereby these genes lead to symptoms, establishment of epigenetic factors that interact with these genes to produce disease, validation of nosological boundaries that more closely reflect the actions of these genes, and development of effective preventive and therapeutic interventions based on genetic counseling, gene therapy, and modification of permissive or protective environmental influences. In addition to their capacity to accelerate the discovery of new molecules participating in the nervous system's response to disease or to self-administered drugs, molecular biological strategies can also be used to determine how critical a particular gene product may be in mediating a cellular event with behavioral importance. Molecular biology probably enables us discover the environmental factors of mental disorders and allow rational drug design and gene therapies for mental disorders, by isolation of gene products that facilitate a basic understanding of the pathogenesis of these disorders. A specific genetic linkage may suggest a novel class of drugs that has not yet been tried. With respect to gene therapy, the hypothetical method would use a gene delivery system, most likely a modified virus, to insert a functional copy of a mutant gene into those brain cells that require the gene for normal function.

Geologic and Geochemical study on the rock sequences containing oily materials in Southwestern Coast Area of Korea (한국서남해안지역(韓國西南海岸地域)에 분포(分布)하는 함유질물층(含油質物層)에 대(對)한 지질학적(地質學的) 및 지구화학적연구(地球化學的硏究))

  • This study has been made for the enlargement of a previous work of 1964 which was carried out by an author of this work emphasizing the stratigraphy, micropaleontology, depositional environment, and structural tectonics of the studied area. The stratigraphic sequences of the area are groupped into four units: (1) basement of Pre-Cretaceous, (2) lower sediments of Late Cretaceous, (3) upper sediments of Late Cretaceous and (4) igneous rocks of Late Cretaceous and Tertiary (?). The oldest rocks consisting of schists and gneisses of Pre-Cambrian and schistose granite' of Jurassic age are exposed at the base of this area on which the thick Cretaceous sediments were deposited. These old rocks are unconformably overlain by the lower sedimens of Late Cretaceous composed of three members, an alternation of black shale and tuffaceous sediments, fine tuff and rhyollite flow in ascending order. The oily material was found from the black shales of the alternation m"ember as semi-solid greaselike material, oily order and microscopic granular spherical material and oily stain. The lower sediments are also overlain, in low-angleunconfromity, by the 'upper sediments having three members, an alternation of volcanic conglomerate and andesitic tuff, rhyollitic tuff and andesite flow in the same order. The igneous suit of diabase, diorites, biotite granite, porphyritic granite and porphyries of the latest Cretaceous and small exposure of pitchstone of Tertiary (?) intruded into the pre-existed rocks above mentioned. Considerable amount of ostra- coda microfossils have been chemically extracted from the black shales of the lower sediments and the identification of the fossils suggests that the depositional environment of the sediments were under fresh or brackish water condition. The distribution of the geology and its tectonic data also suggest a combination of dome and basin structures in the area of San-i peninsula and Jin-do as shown in fig. 8. Between these two units an anticlinal structure was constructed. As a result of this study, a seismic survey in a district between U-su-yong and north coast of Jin-do is recommended to determine the underground features.

  • The author studied to determine the sex and age of 8 ancient human skeletal remains, which had excavated from ancient tombs located in Yeanri, Kimhae. Some kinds of personal identification methods their skulls and teeth were used for this study and the results were obtained as follows : 1. Sex determination was possible in ancient teeth from 4th to 5th century, using detection of X-Y homologous amelogenin gene by polymerase chain reaction. 2. DNA analysis proved that the materials examined were all male, but which always did not coincide with the results from other methods for sex determination including comparison of sexual differentiation of cranium and teeth and use of discriminant functions in the dental measurement. 3. There was little difference of the estimated ages between the methods by regression of pulp cavity and attrition in teeth. The ages from these two methods always did not coincide with ones from evaluation closure of cranial and palatal sutures. 4. Sex and age of the materials were determined as follows ; $\cdot$ Y9 was estimated to be male in early sixties. $\cdot$ Yl2 was estimated to be male in late twenties to early thirties. $\cdot$ Y37 was estimated to be male in early forties. $\cdot$ Y70 was estimated to be male in early sixties. $\cdot$ Y87 was estimated to be male in late forties. $\cdot$ Y109 was estimated to be male in early forties. $\cdot$ Yl29 was estimated to be male in late thirties to early forties. $\cdot$ Yl42 was estimated to be male in late fifties to early sixties.

A Survey on Microbial Contamination of Currently-Sold Drugs (I) -Bacterial contamination of marketed liquid- (시장의약품(市販醫藥品)의 미생물(微生物) 오염도(汚染度) 조사(調査) (제1보)(第1報) -시판내복액제(市販內服液劑)의 세균오염도(細菌汚染度)-)

  • An investigation was carried out on a basis of the bacteriological examination with a view to detecting the degree of bacterial contamination for the 77 samples collected from the locally-sold liquid specialties. It's test period was 50 days from July 10 to August 30, 1971. Specially, the survey has put emphasis on the population of general bacteria and the identification of coli-form group, staphylococcus species, streptococcus species, bacillus species, fungi, and yeast species from liquid samples. The results obtained are summarized as follows; (1) For the 77 samples tested, the contamination of general bacteria was found out as minimun 0, i,e., maximum, $12{\times}10^4$ and the total average $45{\times}10^2$ per milliliter. (2) Although streptococcus species could not be detected with the samples, the contamination of the coli-form and staphylococcus species means the strong suggestion of the possibility of pathogenic bacterial contamination. (3) Specially, the products which stay in the neutral pH range and use suspending agents need to care for the microbial contamination in the manufacturing crocess. (4) It is thought necessary to perform the microbiological quality control in the liquid preparations only at least. (5) As the microbial contamination degree in the liquid decreases according to the elapse of time, the microbiological quality control will have to be carried out immediately after the completion of the manufacturing process in order to know the accurate degree. (6) The author thinks that the main reason of the microbial contamination in the liquid is the contamination during the manufacturing process. (7) For the purpose of prevention of the microbial contamination in liquid, therefore, it is more important to make efforts for the rationalization of manufacturing process, the improvement of equipment and environment, the specific training of workers for hygienic knowledges, etc. rather than the use of preservatives for the preparations.
