영화산업의 생성형 인공지능(Generative AI) 활용 현황과 문제점 (Current Use and Issues of Generative AI in the Film Industry)

  • With the introduction of generative artificial intelligence(AI) tools such as OpenAI's Sora into the global film industry, including Hollywood, there has been a simultaneous emergence of innovations in film production as well as various crises. These changes are spreading throughout the entire film production process, including scriptwriting, casting, editing, and acting. This study analyzes the impact of AI on the film industry, particularly Hollywood, and explores how this technology might bring about changes in Korean cinema. AI technologies applied in the film industry offer benefits such as reducing production time and costs. However, they also pose threats to many filmmakers and actors who rely on the traditional production methods, leading to ethical and legal issues. In Hollywood blockbuster films, AI technology is used to create realistic visual effects, analyze scripts, and suggest optimal shooting angles. While these applications improve the qualitative level of films, they also reduce the human resources required in traditional film production processes. The impact on the Korean film industry is also noteworthy. Some Korean film production companies are leveraging AI to create films in a more creative and efficient manner. Efforts are being made to analyze audience data using AI and develop storylines that appeal to a larger audience. However, these technological changes are controversial among many Korean filmmakers who prefer traditional production methods. This study provides an in-depth discussion on whether the adoption of AI in the film industry can bring about positive innovation or inevitably lead to crises. It analyzes how AI technology is transforming traditional roles in the film industry and what new opportunities and challenges this change generates within the industry. Additionally. This study highlights the differences in technology adoption between Hollywood and Korean film industry and explores how each industry is embracing these technological changes.

Novel Anti-Mesothelin Nanobodies and Recombinant Immunotoxins with Pseudomonas Exotoxin Catalytic Domain for Cancer Therapeutics

  • Recombinant immunotoxins (RITs) are fusion proteins consisting of a targeting domain linked to a toxin, offering a highly specific therapeutic strategy for cancer treatment. In this study, we engineered and characterized RITs aimed at mesothelin, a cell surface glycoprotein overexpressed in various malignancies. Through an extensive screening of a large nanobody library, four mesothelin-specific nanobodies were selected and genetically fused to a truncated Pseudomonas exotoxin (PE24B). Various optimizations, including the incorporation of furin cleavage sites, maltose-binding protein tags, and tobacco etch virus protease cleavage sites, were implemented to improve protein expression, solubility, and purification. The RITs were successfully overexpressed in Escherichia coli, achieving high solubility and purity post-purification. In vitro cytotoxicity assays on gastric carcinoma cell lines NCI-N87 and AGS revealed that Meso(Nb2)-PE24B demonstrated the highest cytotoxic efficacy, warranting further characterization. This RIT also displayed selective binding to human and monkey mesothelins but not to mouse mesothelin. The competitive binding assays between different RIT constructs revealed significant alterations in IC50 values, emphasizing the importance of nanobody specificity. Finally, a modification in the endoplasmic reticulum retention signal at the C-terminus further augmented its cytotoxic activity. Our findings offer valuable insights into the design and optimization of RITs, showcasing the potential of Meso(Nb2)-PE24B as a promising therapeutic candidate for targeted cancer treatment.

청피 에탄올 추출물이 스트레스성 카테콜아민으로 유도한 간암세포의 전이를 억제하는 효과 및 기전 연구 (Inhibition of Adrenergic Agonists-induced Metastatic Ability of Liver Cancer Cells by Ethanol Extract of Premature Citrus Unshiu Peel)

  • Previous studies have highlighted the pivotal role of the β-adrenergic receptor (β-AR) signaling pathway in stimulating cancer metastasis induced by chronic stress. According to the theory of traditional Korean medicine, chronic stress can induce Qi stagnation. Based on the traditional role of premature citrus unshiu peel in moving Qi, we hypothesized that an ethanol extract of premature citrus unshiu peel (EPCU) can attenuate chronic stress-induced cancer progression. In this study, we investigated the potential role of EPCU on modulating the adrenergic agonists-induced metastatic properties of liver cancer cells. Our findings revealed that adrenergic agonists, including norepinephrine (NE), epinephrine (E), and isoproterenol (ISO), augmented the migratory capacity of Hep3B human hepatocellular carcinoma cells, which was completely abrogated by EPCU treatment in a concentration-dependent manner. Consistently, EPCU inhibited the E-induced invasive property of Hep3B cells in a dose-dependent manner. These results suggest that EPCU efficiently attenuates adrenergic agonists-induced metastatic abilities of liver cancer cells. As a molecular mechanism, EPF suppressed the phosphorylation of major components of β-AR signaling pathway, including Src, signal transducer and activator of transcription 3 (STAT3) and ERK, induced by E treatment. Taken together, our results demonstrate that EPCU impedes the adrenergic agonists-driven metastatic potential of cancer cells by inhibiting β-AR signaling pathway. This study provides basic evidence supporting the probable use of premature citrus unshiu peel to prevent metastasis in liver cancer patients under chronic stress.

  • 최근의 전 세계적으로 주목받고 있는 증감현실(增强現實, Augmented Reality, AR)과 가상현실(假想現實, virtual reality, VR), 그리고 이들을 섞은 혼합현실(mixed reality, MR)등은 과학의 테크놀로지 범주를 넘어 대중문화 전반에 큰 영향을 끼치고 있다. 구글, 애플, 삼성, 마이크로소프트, 소니, LG등 세계 굴지의 IT회사들은 대중을 위한 AR. VR 기술개발에 주력하고 있으며 크고 작은 관련 회사들도 해당 하드웨어, 소프트웨어, 콘텐츠 개발에 박차를 가하고 있다. 특정한 플랫폼이나 프로그램을 이용해 인간의 인지력을 인위적으로 조작 통해 특정한 장소나 상황을 경험하거나 보이지 않는 것을 보이게 해준다는 의미에서 AR, VR, MR은 모두 가상의 공간의 현실화라는 공통적인 기술을 포괄하고 있다. 특히, 기존의 평면적 구도의 한계성을 드러낸 입체영상에서 벗어나 180도, 360도 영상으로 객관적 시야와 감각과 같은 주관적 현상을 동시에 제공하고 참가자들이 이를 선택 할 수 있어 참가와 몰입을 크게 유도 할 수 있는 VR 기술은 업계뿐만 아니라 일반대중에게도 초유의 관심을 이끌어내고 있다. 2015년 선댄스 영화제의 뉴 프론티어 프로그램에서는 10개 이상의 관련 작품이 소개 되었고 열풍이 되어버린 게임인 '포켓몬 고(PoKetmon GO)'는 세계 게임시장을 석권하고 있으며 의료, 건축, 쇼핑, 영화, 애니메이션 등 관련 콘텐츠가 등장하고 있고 관련 모바일 어플리케이션도 이미 수천 개 이상 상용화 되어있다. 또한 시판되는 360도 카메라를 통해 개인이 VR 영상을 제작/공유 할 수 있어 유저간의 쌍방형 터널이 가능해 지고 있다. VR 기술의 적용범위 확대와 다양한 현실화로 앞으로 가능성도 희망적으로 여기고 있다. 이는 세계적인 추세며 우리나라 역시 후발주자로 그 추세를 따르고 있다. 그럼에도 불구하고 일부 학자들은 VR, 즉, 가상공간의 현실화가 주는 윤리적 퇴행과 가치관의 혼란이 내재 되어있음을 지적하고 있다. 4K혹은 HUD, 위치추적, 동작 센서, 연산능력, 그리고 뛰어난 3D그래픽, 촉감, 냄새 등의 4D기술, 3차원 오디오 기술 등이 그 어느 때 보다 발전해 리얼리티에 최대한 접근하고 있고 그에 따른다. 윤리적 퇴행, 정체성, 세대갈등, 현실도피 등이 우려되기 때문이다. 리얼리티를 추구하는 애니메이션 역시 이 범주 안에 든다. 미학적 이미지와 환영성의 특정한 구조를 살펴본다면 오히려 영상이라는 유사점을 가지고 있음에도 불구하고 순수한 애니메이션이 VR 콘텐츠 제작에 가장 뒤쳐져 있는 요인이 될 수도 있다. 하지만 VR기술과 플랫폼이 게임과 오락성에 치중 해 있지만 그 안에는 결국 시각적인 VR영상으로 구성 되어 있다는 점을 감안한다면 평면상에 머물고 있는 애니메이션에도 새로운 모색점을 맞이하게 될 것이 분명하다. 결국 어떻게 VR기술을 이용한 가상공간에서 만들어지는 리얼리티가 애니메이션에 적용 할 수 있을까? 그렇다면 방법과 수단이 무엇이 될 것인가 하는 문제에 대한 연구가 공통된 관심이 될 것이다. 그동안 평면적인 화면에 시간과 공간의 연속성에 제한을 받아온 애니메이션은 VR기술을 통해 제한에서 벗어나고자 하는 움직임이 일어나고 있다.

인터랙티브 제품 디자인을 위한 프로토타이핑 도구: MIDAS의 활용 사례 및 유용성 연구 (Investigation into a Prototyping Tool for Interactive Product Design: Development, Application and Feasibility Study of MIDAS (Media Interaction Design Authoring System))

  • 본 논문은 인터랙션 디자인 관련 디자이너들이 기능적 프로토타입(Working Prototype)을 제작할 때 쉽게 활용할 수 있는 저작 도구 MIDAS(Media Interaction Design Authoring System)를 제안하며, 그에 관련된 조사 연구와 사례 연구를 통해 검증된 유용성에 관해 기술한다. MIDAS는 기술적 구현 경험이 부족한 디자이너들이 디지털 아날로그 입 출력 제어, 소프트웨어-하드웨어 통합, 혼합 현실 기술 등을 디자이너들이 사용하는 기존 멀티미디어 제작 환경 내에서 간편히 응용할 수 있게 한다. 디자이너들에게 필요한 인터랙티브 제품 프로토타이핑 도구의 요구 사항을 도출하기 위해 인터랙션 디자인 과정 고찰, 관련 연구 분석, 실무 디자이너 인터뷰를 수행 하였다. 이를 바탕으로 널리 활용되고 있는 멀티미디어 저작 도구인 디렉터와 플래시를 기반으로 프로토타이핑 도구를 개발하였다. 이 도구를 활용하여 다양한 인터랙션 디자인 프로젝트 사례 연구를 진행하였다. 사례연구를 통하여 디자이너들이 기존 디자인 구현 과정에서 겪던 기술적인 어려움들이 MIDAS를 통해 효과적으로 해소함을 알 수 있었다. 본 연구에서 제안하는 프로토타이핑 도구 및 방법은 인터랙티브 제품 디자인 영역 이외에도 인터랙티브 미디어아트, 탠저블 인터페이스와 연관된 인간 컴퓨터 상호작용 (Human Computer Interaction)분야의 교육 및 연구에도 기여 할 수 있을 것으로 기대된다.

Parecoxib: an Enhancer of Radiation Therapy for Colorectal Cancer

  • Background: To study the effect of parecoxib, a novel cyclooxygenase-2 selective inhibitor, on the radiation response of colorectal cancer (CRC) cells and its underlying mechanisms. Materials and Methods: Both in vitro colony formation and apoptosis assays as well as in vivo mouse xenograft experiments were used to explore the radiosensitizing effects of parecoxib in human HCT116 and HT29 CRC cells. Results: Parecoxib sensitized CRC cells to radiation in vitro with a sensitivity enhancement ratio of 1.32 for HCT116 cells and 1.15 for HT29 cells at a surviving fraction of 0.37. This effect was partially attributable to enhanced apoptosis induction by parecoxib combined with radiation, as illustrated using an in vitro apoptosis assays. Parecoxib augmented the tumor response of HCT116 xenografts to radiation, achieving growth delay more than 20 days and an enhancement factor of 1.53. In accordance with the in vitro results, parecoxib combined with radiation resulted in less proliferation and more apoptosis in tumors than radiation alone. Radiation monotherapy decreased microvessel density (MVD) and microvessel intensity (MVI), but increased the hypoxia level in xenografts. Parecoxib did not affect MVD, but it increased MVI and attenuated hypoxia. Conclusions: Parecoxib can effectively enhance radiation sensitivity in CRC cells through direct effects on tumor cells and indirect effects on tumor vasculature.

Improvement of osteogenic potential of biphasic calcium phosphate bone substitute coated with two concentrations of expressed recombinant human bone morphogenetic protein 2

  • Purpose: The aim of this study was to determine whether biphasic calcium phosphate (BCP) bone substitute with two different concentrations of Escherichia coli-expressed recombinant human bone morphogenetic protein 2 (ErhBMP-2) enhances new bone formation in a standardized rabbit sinus model and to evaluate the concentration-dependent effect of ErhBMP-2. Methods: Standardized, 6-mm diameter defects were made bilaterally on the maxillary sinus of 20 male New Zealand white rabbits. Following removal of the circular bony windows and reflection of the sinus membrane, BCP bone substitute without coating (control group) was applied into one defect and BCP bone substitute coated with ErhBMP-2 (experimental group) was applied into the other defect for each rabbit. The experimental group was divided into 2 subgroups according to the concentration of ErhBMP-2 (0.05 and 0.5 mg/mL). The animals were allowed to heal for either 4 or 8 weeks and sections of the augmented sinus and surrounding bone were analyzed by microcomputed tomography and histologically. Results: Histologic analysis revealed signs of new bone formation in both the control and experimental groups with a statistically significant increase in bone formation in experimental group 1 (0.05 mg/mL ErhBMP-2 coating) after a 4-week healing period. However, no statistically significant difference was found between experimental group 1 and experimental group 2 (0.5 mg/mL ErhBMP-2 coating) in osteoinductive potential (P<0.05). Conclusions: ErhBMP-2 administered using a BCP matrix significantly enhanced osteoinductive potential in a standardized rabbit sinus model. A concentration-dependent response was not found in the present study.

VR 영상이 신체 안정성에 미치는 영향 : 손 안정성을 중심으로 (Effects of Virtual Reality Images on Body Stability : Focused on Hand Stability)

  • 본 논문의 목적은 영상자극이 신체 안정성에 미치는 영향을 개념 모델(Conceptual Model)로 제시하고, 시청자에게 제공되는 영상의 종류(2D, VR)에 따라 신체 안정성(손 안정성)에 미치는 영항을 실험을 통해 알아보는 것이다. 최근에 VR과 같은 입체 영상이 스마트폰이나 운동기구 등과의 결합이 활발해지고 있고, 영상자극이 제거된 후 일시적으로 신체 균형 및 손 안정성에 영향을 미침에 따라 안전사고나 인적오류의 가능성도 높아지고 있다. 개념모델은 기존 연구결과들을 바탕으로 제시되었고, 실험결과를 기반으로 뇌에서 일어나는 인간정보처리과정 및 인지적 자원모델과 결합되어 설명되었다. 20명의 피실험자는 2D, VR 자극에 노출 된 후 영상피로가 Cybersickness 질문지를 통해, 손 안정성은 Hand Steadiness Tester를 이용해 측정되었다. 실험결과 2D보다 VR 영상이 높은 영상피로, 낮은 손 안정성을 유발하였다. 본 연구는 아직 시도되지 않은 영상 종류 및 영상피로 수준에 따른 손 안정성을 개념모델과 실험을 통해 밝혔다는데 의의가 있다.

항암제(抗癌劑) Mitomycin C와 수종(數種) 보익제(補益劑)의 병용투여(倂用投與) 효과(效果)에 대한 연구(硏究) (A Study on the Combined Effects of Several Kinds of Tonifying Prescriptions and Mitomycin C)

  • The studies were conducted to investigate the combined effects of Tonics and Mitomycin C(MMC). The effects of Tonics and MMC on the proliferation of Molt-4 cells, human leukemic cell line, and activation of human lymphocytes were estimated by MTT colorimetric assays. Selected medicines among 9 kinds of Tonics by results of MTT assays were treated with MMC in mice. The Tonics itself enhanced the proliferation of Molt-4, but the anti-proliferative effect of MMC was not intercalated by the combined treatment of Tonic and MMC. Inhibitory action of MMC was augmented by Sa Kun Ja Tang(SKT). This result was due to the inhibition of DNA synthesis. Among 9 kinds of Tonics, Sip Jean Dae Bo Tang(SDT), Saeng Maek San(SMS) and Kwi Bi Tang(KBT) did not inhibit the action of MMC, but activated lymphocytes. When the mice were treated by MMC, the number of leukocytes was decreased significantly at the 1st day, but recovered at the 7th day. In the groups of MMC treated with SDT or KBT, the number of leukocytes was increased significantly than the group of MMC treated only at the 3rd day. Combined treatment of the Tonics(SDT, SMS) and MMC retained the body weight of mice at the level of normal mice. SDT, SMS and KBT did not change the number of plaque forming cells(PFC), but MMC treated group decreased the number of PFC. The combined treatment of MMC and SDT increased the number of PFC significantly than the MMC treated group. SDT, SMS and KBT did not influence the proliferation of T cells, but MMC treated group decreased the proliferation of T cells. The combined treatment of MMC and those tonics increased the T cell proliferation significantly than the MMC treated group. In conclusion, the results presented in this paper suggest that SDT, SMS and KBT can recover the side effects of MMC, such as weight loss, leukopenia and immunosuppresion, without any intercalating the anti-proliferative action of MMC in vivo.

Contradictory Effects of Superoxide and Hydrogen Peroxide on $K_{Ca}3.1$ in Human Endothelial Cells

  • Reactive oxygen species (ROS) are generated in various cells, including vascular smooth muscle and endothelial cells, and regulate ion channel functions. $K_{Ca}3.1$ plays an important role in endothelial functions. However, the effects of superoxide and hydrogen peroxide radicals on the expression of this ion channel in the endothelium remain unclear. In this study, we examined the effects of ROS donors on $K_{Ca}3.1$ expression and the $K^+$ current in primary cultured human umbilical vein endothelial cells (HUVECs). The hydrogen peroxide donor, tert-butyl hydroperoxide (TBHP), upregulated $K_{Ca}3.1$ expression, while the superoxide donors, xanthine/xanthine oxidase mixture (X/XO) and lysophosphatidylcholine (LPC), downregulated its expression, in a concentration-dependent manner. These ROS donor effects were prevented by antioxidants or superoxide dismustase. Phosphorylated extracellular signal-regulated kinase (pERK) was upregulated by TBHP and downregulated by X/XO. In addition, repressor element-1-silencing transcription factor (REST) was downregulated by TBHP, and upregulated by X/XO. Furthermore, $K_{Ca}3.1$ current, which was activated by clamping cells with 1 ${\mu}M$ $Ca^{2+}$ and applying the $K_{Ca}3.1$ activator 1-ethyl-2-benzimidazolinone, was further augmented by TBHP, and inhibited by X/XO. These effects were prevented by antioxidants. The results suggest that hydrogen peroxide increases $K_{Ca}3.1$ expression by upregulating pERK and downregulating REST, and augments the $K^+$ current. On the other hand, superoxide reduces $K_{Ca}3.1$ expression by downregulating pERK and upregulating REST, and inhibits the $K^+$ current. ROS thereby play a key role in both physiological and pathological processes in endothelial cells by regulating $K_{Ca}3.1$ and endothelial function.
