토마토 절간(節間)을 이용한 마디삽목 시(時) 삽수의 절위(節位)와 삽수의 소엽(小葉)부착 유무가 묘(苗) 소질에 미치는 영향 (Characteristics of Nursery Plants Influenced by Leaflet and Raising Method for Soft-Nodal cuffing in Cherry Tomato)

  • 토마토 묘(苗)를 한마디씩 4-6마디로 절단하여 절간을 이용한 마디 삽목 시 삽수의 절위와 소엽의 부착유무가 묘소질에 미치는 영향을 검토하기 위하여 본 시험을 수행하였다. 방울토마토 뻬뻬와 꼬꼬 품종을 플러그 묘를 소엽 1엽을 부착하여 삽목한 처리구가 소엽을 모두 제거한 처리보다 초장과 엽수가 많았고, 지상부중 및 지하부중이 약 3-15배 정도 증가되었다. 토마토 마디삽목묘의 발근율은 하배축이 86%였으나 평균 96%의 높은 발근율을 보였다. 토마토 플러그 묘(苗) 1주(株)로부터 4-6개의 삽수가 얻어졌고 삽목 후 20-24일에 정식 가능한 묘가 생산되었으며, 계속적인 증식이 가능하였다. 토마토 삽목묘의 뿌리는 실생묘에 비하여 직근이 적고 뿌리의 수가 많았다. 그리고 실생묘는 뿌리가 하배축의 끝에서 발근되었으나 마디삽목묘는 상토와 접촉된 모든 부위에서 발생되었다. 육묘방법에 따른 소엽의 변화는 실생묘는 자엽으로부터 마디가 생장점으로 이동함에 따라서 약 1엽, 2엽, 3.5엽 5.5엽으로 엽수가 증가되고, 측지삽목묘는 11.3, 11.9, 13.2, 13.3엽으로 첫째 마디부터 엽수가 현저하게 많았다. 한편 마디삽목묘의 엽수는 약 3.5, 3.6, 4.4엽 및 5.8엽으로 증가되어 측지삽목묘보다는 현저하게 적었으나, 실생묘보다는 많았다. 같은 마디삽목묘의 소엽수(小葉數)도 삽수마디가 하배축에서 생장점으로 이동함에 따라서 소엽수(小葉數)가 크게 증가되었다.

다양한 치은 연하 수복물에 대한 치은 섬유아 세포 부착 연구 (Attachment of Human Gingival Fibroblast to Various Subgingival Restorations;A Comparative Study in Vitro)

  • When mucoperiosteal flaps are positioned and sutured to desirable position, the wound contains several interface between tissues which differ fundamentally in composition & biological reaction. Thus the C-T surface of the flap will, on one hand, oppose another vascularized surface, and on the other, the avascular dental material for example, when root resoptions, fractured root, endodontic perforation, deep root carious lesions were filled with amalgam, glass ionomer, resin etc. Recently, a number of case report described the successful treatment of a subgingival root lesion with restorative material & free gingival graft, open flap surgery, but more objective research was needed . Most of study on restorative materials were concerned for cytotoxicity not for actual healing event on that materials and its influencing factors such as biocompatibility, surface wettability, surface topography . The aim of this in vitro study was to evaluate the effect of amalgam, resin modified glass ionomer, composite resin per se, and their surface roughness on the growth of human gingival fibroblast. The cells were obtained and placed on culture flask and incubated for 3 days with the prepared test materials. Then count the attached cell number with hemocytometer,(n=12) and 2 samples were examined with SEM about attachment cell morphology . Another 4 samples were evaluated on their surface roughness with Talysurf and average surface roughness value(Ra) were obtained. Statistical difference in attached cell number, roughness value were analyzed using ANOVA. The number of attached cell was as follows, for root dentin specimen 16.7${\pm}$4.41, resin modified glass ionomer 14.0${\pm}$4.15, resin 8.13${\pm}$3.63, amalgam 0.72${\pm}$3.33(${\times}10^3$). Between root dentin and resin-modified glass ionomer, no significant difference was observed, but resin, amalgam showed a significant less cell numbers than for root dentin, resin modified glass ionomer cement. SEM examination expressed many cell surface attachment apparatus in root dentin and resin modified glass ionomer specimens. For resin specimen, cell attachment was observed but exposed less appratus. The average surface roughness value are following results. Dentin specimen 0.6972${\pm}$ 0.104, resin modified glass ionomer 0.0822${\pm}$0.009, resin 0.0875${\pm}$0.005, amalgam 4.2145${\pm}$0.985(${\mu}m$). Between root dentin, resin-modified glass ionomer, and resin, no significant difference was observed, but amalgam showed a significant more rough surface than other groups. When evlauated the interrelationship between cell attachment and surface roughness, therefore, there was weak reverse correlation.(pearson correlation : - 0.593) These results suggest that resin modified glass ionomer have the favorable healing potential when used for subgingival restoration. And for relationship between cell attachment and surface characteristics, further investigations were needed.

약물함유 생체분해성 차폐막의 생채활성도 및 골조직 유도재생 효과 (Cellular activity and guided bone regenerative effect of drug-loaded biodegradable membranes)

  • The purpose of this study was to evaluate the effects of tetracycline(TC}, flurbiprofen, and PDGF-BB loaded biodegradable membranes on the cell-attachment, the activity of loaded PDGF-BB, in vivo release kinetics, and guided bone regenerative potentials. To evaluate the cell attachment to membranes, the number of gingival fibroblasts attached to each membrane(10% TC, 10% flurbiprofen, $200ng/cm^2$ PDGF-BB loaded membranes, drug-unloaded membrane) was counted by coulter counter and the morphologic pattern of attached cells was examined under SEM. To determine whether the activity of loaded PDGF-BB is sustained, the cellular growth and survival rate of gingival fibroblasts was used for both standard PDGF-BB and loaded PDGF-BB. For evaluation of in vivo release kinetics, drug-loaded membranes were implanted on the dorsal skin of the rats. On 1, 3, 7, 10, 14, 21, and 28 days after implantation, the amount of remaining drugs were measured by HPLC assay for TC and flurbiprofen, and by ${\gamma}-scintillation$ counter for $PDGF-BB^{1125}$. For evaluation of guided regenerative potential, the amount of new bone in the calvarial defect(5mm in diameter) of the rat was measured by histomorphometry 1 and 2 weeks after implantation of membranes. The number of cells attached to the PDGF-BB loaded membrane was largest as compared with the other mernbranes.(p< 0.05) The activity of loaded PDGF-BB was not significantly different from the activity of standard PDGF-BB.(p<0.05) After initial burst release of drug during the first 24 hours, drugs were gradually released for 4 weeks. Especially the release rate of PDGF-BB was nearly constant during 4 weeks. PDGF-BB loaded membranes(200, $400ng/cm^2$) were effective in guided bone regeneration as compared with drug-unloaded membrane. These results implicate that drug-loaded biodegradable membranes might be a useful for guided bone regeneration.

저온기 토마토 재배시 적정 측지관리방법 구명 (Optimum management of tomato side shoot removal in winter)

  • 본 실험은 저온기 토마토 재배시 토마토의 생육과 수확량에 최적의 측지관리방법을 구명하고자 수행하였다. 방울토마토인 유니콘(몬산토 코리아, 한국)을 실험에 사용하였다. 배지는 코이어 자루배지를 사용하였다. 모든 측지를 제거하고 모든 화방의 상부 잎 1매까지 제거한 처리(UP-FL), 측지를 전부 제거하고 착과한 화방의 상부 잎 1매까지 제거한 처리(UP-FR), 모든 측지를 제거한 처리(AS-All), 화방 하부 첫 번째 측지의 잎 2매를 유지하는 처리(AS-Part) 등 모두 4가지 방법으로 처리했다. 이상엽의 발생은 UP-FL, UP-FR, AS-All, AS-Part 순으로 많았다. 즉, 주지와 측지의 잎을 많이 제거한 처리일수록 이상엽의 발생이 많았다. 그리고 이상엽은 영양생장과 생식생장이 균형을 균형을 이루는 2~3화방 개화기 이후에는 해소되는 것으로 나타났다. 5화방까지의 수확량은 처리간 차이가 없었다. 따라서 저온기 토마토 재배시 생육초기에는 과도한 측지제거 및 적엽은 지양하는 것이 좋은 것으로 사료된다.

창상치유목적의 골수기질세포 동종이식을 위한 고분자막의 조건 (Optimal Condition of Microporous Membrane for Bone Marrow Stromal Cell Allotransplantation to Stimulate Wound Healing in Vitro)

  • Purpose: Major drawbacks of conventional bone marrow stromal cells (BSCs) transplantation method are mainly caused by direct transplanted cell to host cell interactions. We hypothesized that separation of the transplanted cells by a microporous membrane might inhibit most of the potential adverse effects and induce superior effect. The purpose of the study is to determine the optimal condition of the microporous membrane. Methods: First, BSCs were placed in polyethylene terephthalate (PET) transwell inserts with 3, 8, or $12{\mu}m$ pore size, and cultured in 24 well culture plates. After 5 days, bottoms of the plates were observed for presence of attached BSCs in monolayer and cell numbers were evaluated. Second, BSCs were placed PET, polycarbonate (PCT), and mixed cellulose esters (MCE) transwell inserts with 3 and $8{\mu}m$ pore size, and cultured in 24 well culture plates. After 3 days, the supernatants of the media left in culture plate were analyzed for collagen, vascular endothelial growth factor (VEGF), platelet derived growth factor BB (PDGF-BB), and basic fibroblast growth factor (bFGF). Third, BSCs were placed in 15% and 70% of the PET membrane with $3{\mu}m$ pore size. All the experimental conditions and methods were same as the second study. Results: The optimal pore sizes to prevent BSC leakage were $3{\mu}m$ and $8{\mu}m$. The amounts of type I collagen and three growth factors tested did not show significant differences among PET, PCT, and MCE groups. However, the collagen, VEGF, and bFGF levels were much higher in the high (70%) density group than in the low (15%) density group. Conclusion: This study revealed that the optimal pore size of membrane to prevent direct BSC to recipient cell contact is in between $3{\mu}m$ and $8{\mu}m$. Membrane materials and pore sizes do not influence the collagen and growth factor passage through the membrane. The most striking factor for collagen and growth factor transport is pore density of the membrane.

W 및 Ti 박막 위에서 나노결정질 다이아몬드의 성장 거동 (Growth of Nanocrystalline Diamond on W and Ti Films)

  • The growth behavior of nanocrystalline diamond (NCD) film has been studied for three different substrates, i.e. bare Si wafer, 1 ${\mu}m$ thick W and Ti films deposited on Si wafer by DC sputter. The surface roughness values of the substrates measured by AFM were Si < W < Ti. After ultrasonic seeding treatment using nanometer sized diamond powder, surface roughness remained as Si < W < Ti. The contact angles of the substrates were Si ($56^{\circ}$) > W ($31^{\circ}$) > Ti ($0^{\circ}$). During deposition in the microwave plasma CVD system, NCD particles were formed and evolved to film. For the first 0.5h, the values of NCD particle density were measured as Si < W < Ti. Since the energy barrier for heterogeneous nucleation is proportional to the contact angle of the substrate, the initial nucleus or particle densities are believed to be Si < W < Ti. Meanwhile, the NCD growth rate up to 2 h was W > Si > Ti. In the case of W substrate, NCD particles were coalesced and evolved to the film in the short time of 0.5 h, which could be attributed to the fact that the diffusion of carbon species on W substrate was fast. The slower diffusion of carbon on Si substrate is believed to be the reason for slower film growth than on W substrate. The surface of Ti substrate was observed as a vertically aligned needle shape. The NCD particle formed on the top of a Ti needle should be coalesced with the particle on the nearby needle by carbon diffusion. In this case, the diffusion length is longer than that of Si or W substrate which shows a relatively flat surface. This results in a slow growth rate of NCD on Ti substrate. As deposition time is prolonged, NCD particles grow with carbon species attached from the plasma and coalesce with nearby particles, leaving many voids in NCD/Ti interface. The low adhesion of NCD films on Ti substrate is related to the void structure of NCD/Ti interface.

무전해 Ni-P UBM과 95.5Sn-4.0Ag-0.5Cu 솔더와의 계면반응 및 신뢰성에 대한 연구 (A study on the interfacial reactions between electroless Ni-P UBM and 95.5Sn-4.0Ag-0.5Cu solder bump)

  • Even though electroless Hi and Sn-Ag-Cu solder are widely used materials in electronic packaging applications, interfacial reactions of the ternary Ni-Cu~Sn system have not been known well because of their complexity. Because the growth of intermetallics at the interface affects reliability of solder joint, the intermetallics in Ni-Cu-Sn system should be identified, and their growth should be investigated. Therefore, in present study, interfacial reactions between electroless Ni UB7f and 95.5Sn-4.0Ag-0.5Cu alloy were investigated focusing on morphology of the IMCs, thermodynamics, and growth kinetics. The IMCs that appear during a reflow and an aging are different each other. In early stage of a reflow, ternary IMC whose composition is Ni$_{22}$Cu$_{29}$Sn$_{49}$ forms firstly. Due to the lack of Cu diffusion, Ni$_{34}$Cu$_{6}$Sn$_{60}$ phase begins growing in a further reflow. Finally, the Ni$_{22}$Cu$_{29}$Sn$_{49}$ IMC grows abnormally and spalls into the molten solder. The transition of the IMCs from Ni$_{22}$Cu$_{29}$Sn$_{49}$ to Ni$_{34}$Cu$_{6}$Sn$_{60}$ was observed at a specific temperature. From the measurement of activation energy of each IMC, growth kinetics was discussed. In contrast to the reflow, three kinds of IMCs (Ni$_{22}$Cu$_{29}$Sn$_{49}$, Ni$_{20}$Cu$_{28}$Au$_{5}$, and Ni$_{34}$Cu$_{6}$Sn$_{60}$) were observed in order during an aging. All of the IMCs were well attached on UBM. Au in the quaternary IMC, which originates from immersion Au plating, prevents abnormal growth and separation of the IMC. Growth of each IMC is very dependent to the aging temperature because of its high activation energy. Besides the IMCs at the interface, plate-like Ag3Sn IMC grows as solder bump size inside solder bump. The abnormally grown Ni$_{22}$Cu$_{29}$Sn$_{49}$ and Ag$_3$Sn IMCs can be origins of brittle failure.failure.

  • 관비농법(灌肥農法)에 의한 사질계농지(砂質系農地)자원 개발을 목표로 동 농법에 의한 모래재배를 실시하고 본 농법의 생산성(生産性)을 높이기 위한 기초적 문제로써 칼슘 기타 양(陽)"이온"이 이십일(二十日)무의 생육(生育)과 근권환경(根圈環境)에 관련하여 하나의 생산성요인(生産性要因)을 이루고 있는 것으로 보이는 근계(根系)와 매지(媒地)의 상호작용(相互作用)(응립성(凝粒性))에 어떠한 영향(影響)을 미치는가를 실험하였다. 1. 고수준의 칼슘 및 고수준의 양이온(가리, 칼슘, 마그네슘)을 가진 관비(灌肥)는 모래에서 재배한 그 이일(二日)무우의 생육(生育) 특히 지하부(地下部) 생육(生育)을 촉진하고 수량(收量)을 증대하였다. 2. 고수준 칼슘 공급 및 고수준양이온 공급은 근계(根系)에 모래알을 응립(凝粒)케 하는 응립성을 현저히 증대시키고 있다. 3. 고수준칼슘 및 고수준양이온구에서 응립된모래와 세근(細根)의 Ca함량이 타(他)기관 및 재배상(栽培床)에 비하여 증대되고 있어 응립성에 Ca의 관여를 시사한다. 4. 일반적으로 지상부(地上部)의 Ca, Mg, Mn 함량은 지하부(地下部)보다 높으나 N.P.K는 양기관 사이 큰 차이가 없다. 5. 관비농법하 모래재배가 고도의 생산성을 나타내고 특히 고수준 칼슘 및 양이온공급이 생체중(生體重)과 수량(收量)을 증대하고 있는 것은 칼슘이 관여하는 근계(根系)와 매지(媒地)와의 상호작용 즉 응립성에 의하여 조성된 근권환경(根圈環境)에 연유하는 것으로 보았다.

홍조류 무절석회조, 납작돌잎 (Lithophyllum yessoense) 사분포자체의 포자방출과 생장 (Tetraspore Release and Growth of a Crustose Coralline Alga, Lithophyllum yessoense (Rhodophyta, Corallinaceae))

  • 부산의 기장군 지역 조하대에 폭넓게 분포하는 무절석회조류의 한 종인 납작돌잎 (Lithophyllum yessoense)을 대상으로 하여 사분포자의 배양환경에 따른 생장과 사분포자 방출량을 정량화 함으로써 포자방출 양상을 구명하고자 하였다. 포자의 배양실험은 2000년 3월에 채집한 납작돌잎 사분포자체로부터 사분포자를 받아 수행하였다. 배양조건은 온도는 5조건 (5, 10, 15, 20, $25^{\circ}C$), 광량은 4조건 (0, 20, 50, 100$\mu$mol${\cdot}m^{-2}$${\cdot}s^{-1}$)과 염분농도는 6조건 (0, 9, 17, 25, 34, 43 ppt) 하에서 각각 포자의 상대생장률을 측정하였다. 월별 포자방출의 정량화는 2000년 8월부터 2001년 7월까지 매월 시료를 채집하여 50${\mu}mol\;m^{-2}s^{-1},\;15^{\circ}C$과 16:8h (L:D) 조건하에서 48시간 동안 방출된 포자의 총 수를 계수하였다. 배우체의 최적 생장 조건은 $20^{\circ}C$, $20{\mu}mol m^{-2}. s^{-1}$과 34ppt조건이었으며, 최대 상대생장률은 $20^{\circ}C$ 조건에서 0.1232였다. 포자의 방출량은 9월에 사분포자체 단위면적 ($cm^2$) 당 266개로 최고치를 보였고, 연중 1월부터 3월까지의 시기를 제외한 모든 시기에 포자가 방출되는 것으로 나타났다.

광학(光學) 및 전자현미경기술(電子顯微鏡技術)에 의(依)한 Mycoplasma pneumoniae의 분열(分裂)과 연속분화(連續分化)에 관(關)한 연구(硏究) (Replication and Sequential Development of Adherent Mycoplasma Pneumoniae Studied by Light and Electron Microscopies)

  • Mycoplasma pneumoniae의 형태적(形態的) 분화과정(分化過程)을 광학(光學) 및 Carbon replication과 critical point drying 전자현미경 기술(技術)을 이용하여 관찰(觀察)하였다. 단세포(單細胞)로 부터 시작하는 전분화(全分化) 과정에서 이 세균(細菌)은 구형(球形)과 방추형(紡錘形)의 세포형태(細胞形態)로 특유(特有)한 분화(分化)를 하며 이 과정은 배양액(培養液)의 pH변화(變化)와 일치(一致)하여 진행(進行)되었다. 본(本) 연구(硏究) 결과(結果)로 부터 M. pneumoniae의 단세포(單細胞)에서 시작되는 새로운 생활회로(生活回路)를 다음과 같이 제기(提起)하였다. 단세포기(單細胞期)를 지나 제1분열기(第1分裂期)에는 구형세포(球形細胞)가 주로 대수증식기(對數增殖期)의 초기에 이분법(二分法)에 의하여 분열하고 그 후에 방추형 세포(細胞)가 분절법(分節法)에 의하여 분열하는 제2분열기(第2分裂期)가 뒤 따른다. 방추형(紡錘形)의 긴 세포(細胞)는 극성(極性)을 갖고 세포 한쪽 끝으로 부터 성장(成長)하여 수 많은 방추형세포에 의한 망상기(網狀期)가 된다. 이와 같은 생장회로(生長回路)는 좋은 생장(生長) 환경에서는 반복되지만 pH가 6.8 이하로 떨어지거나 다른 조건(條件)들이 나빠지면 세포표면(細胞表面)이 조악(粗惡)한 구형(球形)으로 변형(變形)되어 사멸기(死滅期)에 들어가 모든 세포(細胞)는 생명력(生命力)을 상실하게 된다.
