Anti-rheumatiod Arthritis Effects of I-Myo-San Water Extract (이묘산(二妙散)의 항(抗)류마티즘 효능(效能)에 대한 연구(硏究))

  • Rheumatoid arthritis is characterized by focal loss of cartilage due to an up-regulation of inflammatory pathways, which produce pro-inflammatory mediators, such as interleukin-1(IL-1), tumour necrosis factor alpha($TNF-\alpha$), prostaglantin, and nitric oxide(NO). We investigated the anti-arthritic effects of water extracts from Pellodendri cortex and Atractylodis rhizoma in vitro and in vivo. Each extract suppressed the production of inflammatory mediators(NO, $IL-1\beta$, $TNF-\alpha$, and prostaglandin $E_2$) and the expression of inducible NO synthase and cyclooxygenase-2 in lipopolysaccharide-stimulated macrophages in a dose-dependent manner. These suppressive effects were synergistically increased by their combination. The same results were also observed in the rat osteoblast sarcoma cell line ROS17/2.8 stimulated with $IL-1\beta$, $IFN-\gamma,$ and $TNF-\alpha$. Moreover, the combination of these water extracts significantly suppressed collagen-induced mouse arthritis. These results suggest that the combination of water-extractable components of Pellodendri cortex and Atractylodis rhizoma may be useful for therapeutic drugs against rheumatoid arthritis, probably by suppressing the production of inflammatory mediators.

  • Chichul-Tang, a combined preparation of crude drug, which was composed of Atractylodis Rhizoma alba and Ponciri Fructus, has been used widely for digestive disorder. Each herb extract of Atractylodis Rhizoma and Ponciri Fructus, the mixture of both herb extracts were examined for the effects on isolated smooth muscle, intestinal propulsion, anti-cathartic action and gastric secretion. The results were summarized as follows: Chichul-Tang showed the inhibitory effects of the convulsion of smooth muscle. Anti-cathartic effect was shown and gastric secretion was inhibited.

  • A simple and reliable reverse phase HPLC method was developed to determine pharmacologically active marker compounds of Epimedii Herba, Atractylodis Rhizoma Alba, and Polygalae Radix. The stability test of water-extracts of the three natural medicines has been evaluated for six months. However, no significant changes in the content of the marker compounds of each extract were observed during the time of investigation.

Effects of Atractylodis Rhizoma Pharmacopuncture on an Acute Gastric Mucosal Lesion Induced by Compound 48/80 in Rats

  • Objectives: This study was designed to investigate the protective effects of Atractylodis Rhizoma pharmacopuncture (ARP) against acute gastric mucosal lesions induced by compound 48/80 in rats. Methods: The ARP was injected in Joksamni (ST36) and Jungwan (CV12) 1 hr before treatment with compound 48/80. The animals were sacrificed under anesthesia 3 hrs after treatment with compound 48/80. The stomachs were removed, and the amounts of gastric adherent mucus, gastric mucosal hexosamine, thiobarbituric acid reactive substances (TBARS), xanthine oxidase (XO), and superoxide dismutase (SOD) were measured. Also, histological examination were performed. Results: Gastric adherent mucus, gastric mucosal hexosamine and histological defects of gastric mucosa declined significantly after ARP treatment. Changes in gastric mucosal TBARS were also reduced by ARP treatment, but this result was not statistically significant. ARP treatment did not change the XO and the SOD activities. Conclusions: ARP showed protective effects for acute gastric mucosal lesions induced by compound 48/80 in rats. These results suggest that ARP may have protective effects for gastritis.

  • To clarify the botanical origins of oriental medicines which have been argued or confused for plant origins, species of original plants were investigated through the textural research for oriental medicines and the comparison of Chinese, Korean, Japanese and North Korean Pharmacopoeia. Twenty oriental medicines were studied; Angelicae Gigantis Radix, Rhei Rhizoma, Atractylodis Rhizoma Alba, Aconiti Lateralis Radix Preparata, Acanthopanacis Cortex, Osterici Radix, Cnidii Rhizoma, Saposhnikovae Radix, Magnoliae Cortex, Paeoniae Radix, Liriopis Tuber, Zanthoxyli Fructus, Achyranthis Radix, Sinomeni Caulis et Rhizoma, Polygonati Rhizoma, Cinnamomi Cortex, Visci Herba et Loranthi Ramulus, Fritillariae Thunbergii Bulbus, Pogostemonis Herba, and Curcumae Longae Radix.

Comparative Analysis of Common Herbs for Pain Related Disease between Yak-Jing and Textbook of Herbology (약징(藥徵)과 본초학 교과서의 통증 질환 관련 공통 약물에 대한 비교 분석)

  • Objectives The purpose of this study is to compare the common herbs for pain related disease between Yak-Jing and textbook of herbology. Methods We searched the formation of "Koho" and theory of Todo yoshimasu. We selected the common herbs that is used for pain related disease in Yak-Jing and textbook of herbology. These herbs were compared with each other. Result 1. Common Herbs for pain related disease are Ephedrae Herba (麻黃), Aconiti Lateralis Preparata Radix (附子), Euphorbiae Kansui Radix (甘遂), Euphorbiae pekinensis Radix (大戟) Daphnis Genkwa Flos (芫花), Atractylodis Rhizoma (朮), Paeonia Radix Rubra (芍藥), JujubaeFructus (大棗), Gypsum (石膏), Anemarrhenae Rhizoma (知母), Cinnamomi Ramulus (桂枝), Poria (茯苓), Rhei Rhizoma (大黃), Natrii Sulfas (芒草), Pueraiae Radix (葛根). There are some difference between textbook of herbology and Yak-Jing about Aconiti Lateralis Preparata Radix (附子), Paeonia Radix Rubra (芍藥), Atractylodis Rhizoma (蒼朮), Pueraiae Radix (葛根). 2. Preparata Radix (附子) excrete water poisons (水毒) in Yak-Jing, but it has the effect of restore yang and tonify fire (回陽補火) in textbook of herbology. 3. Paeonia Radix Rubra (芍藥) is effective for pathological condition caused by the formation of the band and spasm (結實拘攣) in Yak-Jing, but it is to cure heat pattern in textbook of herbology. 4. Atractylodis Rhizoma (蒼朮) has the effect of water-draining (利水) in Yak-Jing, but it has the effect of dispersing (發散) in textbook of herbology. 5. Pueraiae Radix (葛根) is used in dyspnea and sweating (喘而汗出) in Yak-Jing, but it is to cure engender fluid (生津), skin disease, diarrhea in textbook of herbology. Conclusion There are some difference between textbook of herbology and Yak-Jing in the herb medicine that is used in pain related disease.

Anti-obesity Effect of Rhizoma Atractylodis Herbal Acupuncture in High Fat Diet-induced Obese ICR Mouse (고지방 식이로 유도된 비만 생쥐에서 창출약침의 항비만 효과)

  • Objectives : The aim of this study was to investigate the anti-obesity potential and mechanisms of action of Rhizoma Atractylodis(RA) herbal acupuncture in high fat diet- induced obese ICR mice. Methods : Sample solutions for herbal acupuncture were prepared from the Rhizoma Atractylodis water extract powder at concentration of 150mg/kg and 300mg/kg with distilled water. Five week-old ICR mice acclimatized to the laboratory environment for 1 week were allocated into four groups: regular diet group (RD), high fat diet group(HFD), groups fed HFD with 150mg/kg RA herbal acupuncture treatment (RAE 150) and with 300mg/kg RA herbal acupuncture treatment(RAE 300). Herbal acupuncture groups were injected with either 150mg/kg or 300mg/kg of Rhizoma Atractylodis(RA) subcutaneously onto both Sinsu($BL_{23}$) alternately on the same time everyday for 30days. Body weight, gross appearance of epididymal fat area, blood glucose, insulin, insulin resistance(HOMA-IR), non-esterified fatty acid, cholesterol, triglyceride, AST, ALT, histological analysis of white adipose tissue, gene expression responsible for adipocyte differentiation and AMPK activation were analyzed. Results : RA herbal acupuncture inhibited the development of weight gain, hyperglycemia, hyperinsulinemia, hyperlipidemia, increases of AST and ALT, and the enlargement of fat cell size induced by HFD. Also, RA herbal acupuncture inhibited the expression of PPAR-${\gamma}$, C/$EBP{\alpha}$, aP2, LPL, FAS, SCD-1 and enhanced the activation of AMP-activated protein kinase. Conclusions : The results of this study demonstrate that RA herbal acupuncture can exert the anti-obesity effect and it is partially mediated by activation of AMPK and inhibition of the gene expressions responsible for adipocyte differentiation. Further studies will be required to ascertain the nti-obesity effect and mechanisms of action of RA herbal acupuncture in animal models and human for aclinical application.

A philological study on adverse effect of restoratives for invigorating qi(補氣藥) (보기약(補氣藥)의 부량반응(不良反應)에 관한 문헌적 고찰)

  • Objectives & Method:We investigated adverse symptoms, toxicity, treatment and prevention against adverse effects of restoratives for invigorating qi(補氣藥) in order to use herbal medicines accurately. Result:Ginseng Radix(人參), Codonopsis Pilosulae Radix(黨參), Panacis Quinquefolii Radix(西洋參), Astragali Radix(黃芪), Atractylodis Rhizoma Alba(白朮), Dioscoreae Rhizoma(山藥), Dolichoris Semen(白扁豆), Glycyrrhizae Radix(甘草), Jujubae Fructus(大棗) and Mel(蜂蜜) may give rise to some side effects, allergic reaction or toxic symptoms in restoratives for invigorating qi(補氣藥). The representative methods of poisoning treatment in western medicines are stopping medication, washing out the stomach, promotion of vomiting, causing diarrhea, supplies of grape sugar and symptomatic treatment, etc. The representative methods of poisoning treatment in oriental medicine take advantage of various herbs. And Oriental medical doctor should meet symptoms as patients call for attention. In order to prevent against poisoning of restoratives for invigorating qi(補氣藥), the patients should keep usage, dosage and notes. Conclusion:We should pay attention to clinical using of Ginseng Radix(人參), Codonopsis Pilosulae Radix(黨參), Panacis Quinquefolii Radix(西洋參), Astragali Radix(黃芪), Atractylodis Rhizoma Alba(白朮), Dioscoreae Rhizoma (山藥), Dolichoris Semen(白扁豆), Glycyrrhizae Radix(甘草), Jujubae Fructus(大棗) and Mel(蜂蜜) in restoratives for invigorating qi(補氣藥).

  • Low Back Pain Prescriptions of Jin, sa tak are peculiar to using Atractylodis Macrocephalae Rhizoma(AMR). All low back pain prescriptions which are enlisted in Byunjeunggimoon, Byunjeungrok and Byunjeungokham include AMR. Whereas low back pain prescriptions which is enlisted in Seoksilbirok are 7 of 10. Preexistence of low back pain prescriptions are not necessarily used AMR. But Jin, sa tak who lived at Ming and Ching era presented AMR in low back pain treatment. AMR is able to get rid of dampness between the kidney functional area and umbilicus. The results are as follows : It is made much of the malicious dampness which is in the kidney. There are not used cold but warm and eliminating dampness herbs to invigorate kidney. It shows that Jin, sa tak who was a Taoist used invigorating and warming kidney herbs. Atractylodis Macrocephalae Rhizoma is mainly used in treating low back pain from Jin, sa tak. Jin, sa tak shows concrete prognosis to treat a disease.

Inhibitory effects of ethanol extract of Atractylodis Rhizoma on melanogenesis in B16/F10 melanoma cells

  • The inhibitory effect of Atractylodis Rhizoma alba extract(AM) on melanogenesis was studied by using B16/F10 melanoma in culture. Cells were cultured in the presence of various concentrations of AM for 48 hrs, and the experiment of total melanin content as a final product and activity of tyrosinase, a key enzyme, in melanogenesis. AM significantly inhibited tyrosinase activity, and melanin content in a dose-dependent manner. These results show that Atractylodes macrocephala extract could be developed as skin whitening components of cosmetics.
