Using semi-selective (SS-) and selective (Jee-) medium, we identified the pathogens isolated from the symptomatic plants and soils collected from different locations, such as Suwon, Andong, and Youngju, as P. drechsleri, which is Phytophthora root rot causal agent of A. macrocephala. At $25^{\circ}C$, these isolates were grown faster on 10% V8A (V8 juice agar) medium than on PDA (potato dextrose agar) with hyphal swelling, but no growing was observed at below $5^{\circ}C$ and over $40^{\circ}C$. In order to identify the pathogenicity of each isolate, seedlings of A. macrocephala were inoculated with mycelium -zoospore suspended inoculum, which was prepared by culturing on 10% V8A medium and homogenizing in distilled water. By this method, wide ranges of pathogenicity were observed as follows; $5.0%{\sim}26.4%$ of disease severities concerning the lesion areas of the top plants and $23.5%{\sim}72.2%$ of disease incidences. Therefore, this was considered as a efficient method to identify the pathogenicity of P. drechsleri in large scale screening. P-A200073, isolated from soils in Andong, and P-9755, from the root of symptomatic plant of A. macrocephala in Suwon, showed the highest degree of pathogenicity to the seedlings. By these isolates, lesion areas and disease incidences of the inoculated seedlings were occurred $26.4%{\sim}63.2%$ and $25.1%{\sim}72.2%$, respectively. However, no symptoms were observed in uninoculated control. Same pathogens were reisolated from roots and lower stems of the inoculated plants, but not from leaves.

Background: Atractylodes radix is a well-known medicinal crop having many physiological effects. This study was conducted to select useful Atractylodes japonica ${\times}$ Atractylodes macrocephala (AJM) cultivars by comparing anti-oxidative and anti-inflammatory efficacies. Methods and Results: Seven extracts from AJM cultivars were used to treat lipopolysacchride (LPS)-treated BV2 cells, and the effects on cell viability and inhibition on reactive oxygen species (ROS) and nitric oxide (NO) production were analyzed. In vitro scavenging activities of 2,2-diphenyl-1-picrylhydrazyl (DPPH) and peroxynitrite ($NOO^-$) radicals were also investigated. Contents of total phenol, atractylenolide I, and atractylenolide III in the AJM extracts were measured using high performance liquid chromatography (HPLC) or spectrophotometry. The experiments show that none of the seven extracts was cytotoxic above 89.2% at $20-250{\mu}g/m{\ell}$. Extracts of Gowon, Dawon, Sangchul, and Huchul inhibited ROS generation in a dose-dependent manner, and Sangchul extract showed the highest inhibition on ROS production. All the AJM extracts showed effective inhibitory activity after on NO release in the LPS-treated BV2 cells, and Sangchul extract showed the highest activity. Sangchul extract had the most potent scavenging activities for $NOO^-$ and had some DPPH radical scavenging effect. Sangchul extract also had the highest content at total phenol and atractylenolide I content. Atractylenolide III was not detected in the AJM extracts. Conclusions: The results suggested that Sangchul was the most useful anti-oxidative and anti-inflammatory resource among the AJM cultivars.
To investigate the germanium content in paddy soil in Gyeongnam province, 310 paddy soil samples were collected at 19 areas in Gyeongnam, Ulsan and Busan. Contents of germanium in paddy soils were analyzed in different topographies, soil types and soil textures. Average content of germanium in Gyeongnam was 0.24 mr/kg, those of Masan, Jinju and Hadong were above 0.30 mg/kg. Germanium content with different topographies were no difference. Germanium contents in different soil types were 0.27 mg/kg in well adapted soil and 0.23 mg/kg in poorly drained soil. Germanium contents in different soil textures were 0.27 mg/kg in silt loam and 0.23 mg/kg in sandy loam. To determine germanium content on agricultural product in the field, content of germanium in cereals, vegetables and fruits were analyzed. Germanium content of agricultural product was high in the order of cereals>vegetables>fruits. In case of vegetables, germanium contents were high in the order of leaf vegetables>root vegetables>fruit vegetables. Germanium contents were high with 62 and $65{\mu}g/kg$ in lettuce and young radish, respectively. To analyze the germanium content in medicinal plant, samples were collected from 19 medicinal plants at Hamyang areas. Germanium contents in Angelica keiskei, Ligusticum chuanxiong, Panax ginseng and Atractylodes macrocephala were relatively high with $100{\mu}g/kg$ above.

This study was carried out to determine the functional works and basic ingredients of Sagoonja-Tang on sponge cake. The experimental groups consisted of 6% mixtures of five (Ed-confirm the number) kinds powders : S1 for Sagoonja-Tang powder, S2 for Panax ginseng, S3 for Poria cocos, Koidz, S4 for Atractylodes macrocephala, and S5 for Glycyrrhiza uralensis Fisch. The volume of sponge cake somewhat decreased after adding the ingredients, and the volume differences in order from highest to lowest, were control>S1>S3>S2>S4>S5. In the microbiological quality test, viable cell counts were high in control groups of sponge cake, and the numbers of viable cell for control sponge cake reached to $9{\times}10^7CFU/g$ on the 7th day of storage, and decreased to $2.5{\times}10^5CFU/g$ on the 10th day of storage. All sponge cakes added with ingredient's powder showed pretty low viable cell counts. Especially, the group S5 showed the lowest counts of $1.2{\times}10^2CFU/g$ on the 7th day of storing. In the texture analyses of sponge cake, All groups showed higher degree of hardness, gumminess and chewiness than SC. The antioxidative activity of the Sagoonja-Tang's ingredients was measured. The POV value measured was S5>S4>S3>S2 >S1>control in order of highest to lowest. In sensory test, the overall acceptability of sponge cake was from highest to lowest S3>SC S2>S1>S5>S4.

Objectives : The purpose of this study was to examine the effects of herbal medicines on pacemaker potentials of large intestinal interstitial Cells of Cajal (ICC) in mice. Methods : We made the ICC culture in large intestine in mice and used the electrophysiological method to record pacemaker potentials. Also we used MTT assay to check cell viability and examined the ICC protein expression by western blot. Results : 1.Glycyrrhiza uralensis Fischer (GF) (50-150 ㎍/ml) induced pacemaker depolarization and decreased frequency with concentration-dependent manners. EC50 is 58.95 ㎍/ml. Angelica gigas (AG) (50-200 ㎍/ml) induced pacemaker depolarization and decreased frequency with concentration-dependent manners. EC50 is 77.22 ㎍/ml. Poncirus fructus (PF) (10-100 ㎍/ml) induced pacemaker depolarization and decreased frequency with concentration-dependent manners. EC50 is 13.39 ㎍/ml. Citrus unshiu S. Marcov. (CU) (10-500 ㎍/ml) induced pacemaker depolarization and decreased frequency with concentration-dependent manners. EC50 is 139.80 ㎍/ml. Gardenia jasminoides J. Ellis (GJ) (100-500 ㎍/ml) induced pacemaker depolarization and decreased frequency with concentration-dependent manners. EC50 is 78.70 ㎍/ml. Coptis chinensis (CC) (100-1000 ㎍/ml) induced pacemaker depolarization and decreased frequency with concentration-dependent manners. EC50 is 138.10 ㎍/ml. Scutellaria baicalensis (SB) (10-100 ㎍/ml) had no effects on pacemaker potentials and decreased frequency with concentration-dependent manners. IC50 is 18.34 ㎍/ml. Atractylodes macrocephala koidzumi (AM) (10-100 ㎍/ml) induced pacemaker hyperpolarizations and decreased frequency with concentration-dependent manners. IC50 is 18.54 ㎍/ml. 2. PF, SB and AM had no effects on cell death in large ICC. 3. PF increased the ANO1 and c-kit protein expression and SB and AM increased the c-kit protein expression in large ICC. Conclusions : These results suggest that PF, SB, and AM are likely to be the optimal combination of herbal medicines that can be used to treat diseases such as gastrointestinal motility disorders such as irritable bowel syndrome.
Homogenization of quality was important in order to use herbal medicines as pharmaceuticals. To solve this problem, it was important to establish quality standards. Atractylodis Rhizoma Alba has no quantitative method in the Korean Pharmacopoeia. Thus, we have researched to improve the quality evaluation method of Atractylodis Rhizoma Alba with an HPLC. Atractylenolide III and atracylodin were selected as potential marker compounds. This analytical procedure was subject to validation. According to validation guideline of South Korea's Ministry of Food and Drugs Safety, the specificity, linearity, precision, range, quantitative limits, detection limits and accuracy were measured. Because the specificity, linearity, precision, range, quantitative limits, detection limits and accuracy meet criteria of the guideline, the analytic method was validated. With this analysis, Atractylodis Rhizoma Alba and Atractylodis Rhizoma analyzed. As a result, both atractylenolide III and atracylodin appear to be suitable standard compounds. it confirmed that tractylodes Rhizome, similar to Atractylodes Rhizome Alba, could be distinguished.
We studied concentration of heavy metals in Sagunjatang. In this experiment was analyzed the concentration of heavy metals of boiled Sagunjatang, decoction and its ingredient herbal medicines. The concentration of heavy metals(As, Pb, and Cd) were analysed using ICP-AES, and Hg was analysed by mercury analyzer. The average concentration of heavy metals in Sagunjatang were as follows : In all ingredient herbal medicines (Glycyrrhizae Radix, Atractylodes Macrocephala, Poria Cocos and Jinseng) of Sagunjatang, As(arsenite) contents in ail samples was in the range of 0.369-0.723ppm, Cd(cadmium) was in the range of 0.000-0.085ppm, Pb(lead) was in the range of 0.059-0.871ppm and Hg(mercury) was in the range of 0.001-0.004ppm. In boiled Sagunjatang, the concentration of heavy metals(As, Pb, Cd and Hg) was in the range of 0.000-0.016ppm, respectively. In the decoction of herbal medicines after boiled, the concentration of heavy metals was in the range of 0.004-0.387ppm. These results suggest that Sagunjatang which we take is less harmful than herbal medicine itself, and there are more significant for using the decoction of herbal medicines.

For white pan bread, pH of control, B3, and B5 decreased on the 2nd day during storage, but increased on the 4th and 7th days. In the texture analyses of white pan bread, B4 and B5 showed a higher degree of hardness than that of control, while B2, and B3 were the lowest. In sensory test, the overall acceptability of regular white pan bread in order from highest to lowest was control>B3>B2>B1 ${\cdot}$ B5>B4.
