Experimental Studies on Antitumor Activity of Herb Drugs (I)-Effectiveness on Rat Natural Killer Cell Activity- (수종(數種)의 생약(生藥)에 대(對)한 항암효과(抗癌效果)의 실험적(實驗的) 연구(硏究)(I) -백서(白鼠)의 자연살해세포활성(自然殺害細胞活性)에 미치는 영향(影響)-)

  • Natural Killer cells are considerd to play an important role in antitumor immune surveilance mechanism. In this study, 21 putative anticancer drugs selected from reference were assessed by evaluating the effect on rat Natural Killer cell activity (NKCA). All 21 herb drugs were extracted in boiling water, lyophilized, autoclaved, and then used for experiment. Culture supernatant of concanavalin-A (Con-A)-stimulated rat spleen cells as a source of lymphokine was also used as a control of comparison. Rat spleen cells were used as effector and NKCA was measured in 4hr $^{51}Cr-release$ assay against Yac-1 mouse lymphoma cell line. In order to determine the optimal conditions for NKCA augmentation, effector cells were treated with 3 different concentrations of each drug for 24, or 48 hrs before testing of NKCA, In optimal conditions determined from previous results, the effect of herb drugs on NKCA were assessed in 3 to 5experiments. NKCA was significantly enhanced by treatment with 4 herb drugs(Ponciri Fructus, Houttuyniae Herba, Aurantii Pericarpium, Nepetae Herba). Culture supernatant of Con-A-stimulated spleen cells also augmented the rat NKCA more significantly. The results show that 4 of the herb medicines supposed to display anticancer effect may have activity as a biological response modifier through augmentation of NKCA.

Effect of Herbal Chokong Pill on the Microstructure of Liver Cell in Rats Fed High Fat Diet (고지방식이 섭식 횐쥐의 간조직에 미치는 한방초콩환의 영향)

  • The purpose of this study is to investigate the effects of the herbal Chokong pill (hereafter HCKP) on the liver cell and enzyme activities of serum in rat. HCKP were mixed with pickled black soybeans and five different kinds of medicinal herbs (Rhynchosia nulubilis, Glycyrrhiza uralensis, Zizyphus vulgaris, Atractylodes macrocephala K., Astragalus membranaceus and Cornus officinalis). Four groups of male Sprague-Dawley rats were fed by different diets for 9 weeks: normal diet (Nor), high-fat diet (HF), high-fat diet supplemented with 1% (T1) and 5% (T5) HCKP powder, respectively. Depending on the presence of HCKP in high fat diet, the activities of the blood serum GOT and GPT were decreased. GOT and GPT activities of T1 and T5 were decreased 6.1%, 17.8% and 25.4%, 32.4% compared with HF. On microstructure observing through the transmission electron microscope (TEM) of liver cell, in normal group, a normal large and clear nucleus, rough endoplasmic reticulum (RER) and mitochondria possessing well-defined double outer-limiting membranes were found. However, in HF, it was hard to observe the microstructures in cytoplasm, because of too many fat granules. It showed severely damaged cell, pyknotic nucleus, swollen disintegrating RER and mitochontria loosing the cristae. In T1, there were more repaired liver cells and less fat granules than HF. In T5, there were much less numbers and smaller size of the fat granules than T1, and the morphology was similar to normal cell.

Taxonomical and Ecological Study on the Vegetation of the Seungbong Island (승봉도 식물상에 관한 분류 및 생태학적 연구)

  • Seungbong Island is situated about 100km away Inchon Port in the western sea of the Korean Peninsula. Field investigations were conducted for two days each from June 30 through July 1, 1981 and from August 20 through 21, 1981. The results obtained are summarized as follows: Plants growing in this island consist of a total of 193 species (70 families, 151 genera, 170 species, 22 varieties and 1 forma). Pinus thunbergii was confirmed to be the dominant species. The natural degree observed in this island was high, as a whole. The coefficient of ferny plants (Pte-Q) is 0.95, lower than grass. Atractylodes japonica Koidz., Miscanthus sinensis Anders. var. purpurascense Rendle, and Rhododendron mucronulatum Turz. are distributed mainly on the forest bed of Pinus thunbergii. The poor distribution of Chenopodium album var. centrorubrum Makino, Setaria viridis (L.) Beauv., Amaranthus mangostanus Linne is considered attributable to the phenomenon of allellopathy caused by a chemical substance secreted from the leaves of Pinus thunbergii. The component ratio of species in this island is lower than that of other islands due probably to the small area of arable land and grassland. The poor growth of plants in the forest of Pinus thunbergii, the dominant species in this island, seems to be attributed to the low relative light intensity of the forest. Seaside plants consisting of a total of 7 species were distributed mainly in the vicinity of sandy beaches. Naturalized plants comprising a total of 11 species were relatively diversified in the number of species.

Anti-wrinkle Effect of Oriental Medicine Cosmetics Containing Black Ginseng (흑삼을 함유한 한방소재 화장품의 주름개선 효과)

  • This study evaluated anti-wrinkle effect using toner and lotion containing ginsenoside Rg3 enriched black ginseng in oriental medicine like Liriope platy phyllla, Inonotus obliquus, Glycyrrhiza glabra and nourishing of the blood and energy(Atractylodes japonica, Poria cocos Wolf, Angelica gigas, Rehmanniae Radix Preparata, Cnidii Rhizoma, Paeonia lactiflora). 6 female adults on average at the age of 42.5 applied lotion after toner under the normal conditions of use on the face including crow's feed area, twice a day (in the morning and evening) for 4weeks. After 4 weeks, the replica of eye wrinkle was made and analyzed the image. The result was decreased in R1($3.15{\pm}0.35$ to $2.28{\pm}0.22$), R2($2.99{\pm}0.37$ to $2.13{\pm}0.21$), R3($2.09{\pm}0.31$ to $1.56{\pm}0.18$), R4($0.37{\pm}0.05$ to $0.29{\pm}0.03$), and R5($0.89{\pm}0.1$ to $0.75{\pm}0.06$)(P<0.05). Therefore, oriental medicine cosmetics containing black ginseng may play a role anti-wrinkle.

Isolation of the Phytophthora Root Rot Pathogen of Atractylodes macrocephala, Phytophthora drechsleri, and Bioassay of the isolates with Seedlings (백출의 역병균 분리 및 유묘를 이용한 병원성 정정 방법의 확립)

  • Using semi-selective (SS-) and selective (Jee-) medium, we identified the pathogens isolated from the symptomatic plants and soils collected from different locations, such as Suwon, Andong, and Youngju, as P. drechsleri, which is Phytophthora root rot causal agent of A. macrocephala. At $25^{\circ}C$, these isolates were grown faster on 10% V8A (V8 juice agar) medium than on PDA (potato dextrose agar) with hyphal swelling, but no growing was observed at below $5^{\circ}C$ and over $40^{\circ}C$. In order to identify the pathogenicity of each isolate, seedlings of A. macrocephala were inoculated with mycelium -zoospore suspended inoculum, which was prepared by culturing on 10% V8A medium and homogenizing in distilled water. By this method, wide ranges of pathogenicity were observed as follows; $5.0%{\sim}26.4%$ of disease severities concerning the lesion areas of the top plants and $23.5%{\sim}72.2%$ of disease incidences. Therefore, this was considered as a efficient method to identify the pathogenicity of P. drechsleri in large scale screening. P-A200073, isolated from soils in Andong, and P-9755, from the root of symptomatic plant of A. macrocephala in Suwon, showed the highest degree of pathogenicity to the seedlings. By these isolates, lesion areas and disease incidences of the inoculated seedlings were occurred $26.4%{\sim}63.2%$ and $25.1%{\sim}72.2%$, respectively. However, no symptoms were observed in uninoculated control. Same pathogens were reisolated from roots and lower stems of the inoculated plants, but not from leaves.

The effect of enhancing eye-wrinkle applying traditional herb medicine cosmetics (한방화장품의 눈가 주름 개선 효과)

  • This study evaluated anti-wrinkle effect for eye using essence and cream containing extract of Liriope platy phyllla, Inonotus obliquus, Glycyrrhiza glabra, black ginseng, Atractylodes japonica, Poria cocos Wolf, Angelica gigas, Rehmanniae Radix Preparata and Cnidii Rhizoma, Paeonia lactiflora. 6 female adults on average at the age of 44.8, applied cream after essence under the normal conditions of use on the crow's feet area, twice a day (in the morning and evening), 2mg/$1cm^2$ for 4weeks. After 4 weeks, the replica of eye wrinkle was made and analyzed the image. The result was that eye wrinkle was decreased in R1($3.22{\pm}0.85$ to $2.38{\pm}0.32$), R2($3.08{\pm}0.83$ to $2.22{\pm}0.32$), R3($2.29{\pm}0.64$ to $2.29{\pm}0.64$), R4($0.44{\pm}0.14$ to $0.44{\pm}0.14$), and R5($0.98{\pm}0.21$ to $0.98{\pm}0.21$)(P<0.05). Therefore, oriental medicine cosmetics containing traditional herb medicine may play a role anti-wrinkle for eye.

Hypoglycemic Effects of Atractylodis Rhizoma in Rats with Streptozotocin-Induced Hyperglycemia (Streptozotocin에 의해 고혈당을 유발시킨 흰쥐에 미치는 Atractrylodis Rhizoma의 영향에 관한 실험적 연구)

  • A single i.v. dose of streptozotocin (65 mg/kg) given to rats has produced a marked hyper-glycemia (>500 mg serum glucose/dl). Since the Atractylodis Rhizoma is known to have hypo-glycemic action, the water extracts of Atractylodis Rhizoma (ARWE) was given to the streptozotocin-induced (SZ) hyperglycemic rats. To determine whether ARWE has the anti-hyperglycemic effects, two different daily doses of ARWE (i.e.0.2 g/kg and 2.0 g/kg) were given orally to the SZ rats for up to 8 days. Thereupon, serum levels of glucose, insulin, amylase and cholesterol were determined on days 1, 3 and 8, following the initial and repeated daily administrations of ARWE. On day 8, glycogen content and glucose-6-phosphatase activity in the liver were assayed. Results showed that ARWE decreased the serum glucose levels, which had been markedly elevated by the SZ pre-treatment. In support of this, the serum insulin level, which had been quickly lowered by the SZ pre-treatment $(20{\mu}U/ml)$, was quickly elevated in the ARWE dose dependent manner that, at 2.0 g/kg ARWE, the serum insulin level was increased $(20{\mu}U/ml)$ above the normal level $(42{\mu}U/ml)$. Also, the serum amylase level, which was steadily decreasing after the SZ pre-treatment, was restored to the normal level folowing 8 day of ARWE (2.0 g/kg) treatment. Hepatic glycogen content and glucose-6-phosphatase activity, which decreased and increased, respectively in the 52 treatment group, were restored toward the normal level in SZ plus ARWE group.

Income Analysis on the Cultivation of Major Medicinal Herbs (주요 약초류 재배에 대한 소득분석)

  • This study intended to provide basic data required in establishing policies for improving the forestry management and the compensation standard for the loss from non-timber forest products by examining the cost of cultivating key medicinal herbs and the earnings from them to analyze the income. According to the income analysis on the cultivation of medicinal herbs, the average annual income per unit area of Adenophora triphylla var. japonica Hara was the highest as it recorded 14,233,000 won/10a and was followed by Pleuropterus multiflorus TURCZ. which recorded 4,121,000 won/10a, Gastrodia elata Blume 3,766,000 won/10a, Epimedium koreanum Nakai 3,537,000 won/10a, Atractylodes ovata (Thunb.) DC. 2,655,000 won/10a, Aralia continentalis Kitagawat 1,048,000 won/10a, Paeonia lactiflora Pallas 1,025,000 won/10a, and Bupleurum falcatum L. 919,000 won/10a. Compared with the income from major nuts and fruits analyzed in 2014, the average annual income per unit area for medicinal herbs was relatively higher. For Adenophora triphylla var. japonica Hara and Aralia continentalis Kitagawat, soots are used for food and the roots for medicine, it appears that it can become a new income source for the farming and mountain villages. Meanwhile, the price for Paeonia lactiflora Pallas is dropping due to Chinese imports, and also damage to the income from other medicinal herbs due to Chinese imports is expected with the implementation of Korea-China FTA in the future.

Symptoms and Prescriptions Based on the Theory for Properties and Tastes of Korean Oriental Herbal Medicines with Regard to the Year When Taeeum Is Affecting the Earth Energy and the dampness Energy Is Partially Over-Abundant (${\urcorner}$태양재천(太陽在泉) 습음소승(濕淫所勝)의 병증(病證)과 기미(氣味)배합 분석${\lrcorner}$)

  • This thesis aims to provide an analytical basis for existing or new prescriptions based on the theory for properties and tastes of Korean oriental herbal medicines. To this end, prescriptions presented in the Various Changes in the Dominations of the Six Energies and their Relations with the Diseases of Plain Questions were analyzed based on the theory for properties and tastes of Korean oriental herbal medicines in Yellow Emperor's Canon of Internal Medicine. This thesis focused on symptoms and prescriptions based on the theory for properties and tastes of Korean oriental herbal medicines, which were described in tire Various Changes in tire Dominations of tire Six Energies and their Relations with the Diseases of Plain Questions with regard to the year when Taeeum is affecting the earth energy and the dampness energy is partially over-abundant. Those symptoms and prescriptions originate from the theory of the five elements and six climates. The symptoms were analyzed from the perspective of physiology. The prescriptions were first analyzed based on the theory for properties and tastes of Korean oriental herbal medicines in Yellow Emperor's Canon of Internal Medicine. Then, a mix formula of oriental medicines pursuant to those prescriptions was studied. Lastly, established prescriptions, which were in conformity with the above prescriptions, were selected. From the physiological point of view, symptoms with regard to the year when Taeeum is affecting the earth energy and the dampness energy is partially over-abundant can be, based on the theory of the five elements and six climates, diagnosed as the symptoms from impaired kidney due to excessive dampness energy. Established prescriptions pursuant to prescriptions based on the theory for properties and tastes of Korean oriental herbal medicines can be broken down into the following: Poria powder with five herbs (Oryeongsan); Divine black bird decoction (Jinmootang); Tetrandra and astragalus decoction (Banggihoanggitang); Licorice, dried ginger, poria and bighead atractylodes decoction (Gamgangryeongchooltang); Spleen-reinforcing powder (Shilbeesan). Therefore, symptoms of six kinds of weather presented in the Various Changes in the Dominations of tire Six Energies and their Relations with tire Diseases of Plain Questions can be analyzed from the physiological point of view. As a result, in addition to the methodology that analyzes existing prescriptions within the boundary of the theory for properties and tastes of Korean oriental herbal medicines in Yellow Emperor's Canon of Internal Medicine, it is expected that a theoretical basis for new prescriptions can be provided by analyzing established prescriptions based on prescriptions from the theory for properties and tastes of Korean oriental herbal medicines.

Interleukin-5 Inhibition Assay of the Oriental Materia Medica Treatment by High Through Screening on the Splenocyte of Mouse (High through screening(HTS)를 이용한 백서(白鼠)의 비장세포(脾臟細胞)에서의 IL-5 분비 조절 한약물(韓藥物) Screening)

  • Backgrounds and Objectives: Asthma is considered to be an inflammatory disease characterized by airway hyperresponsiveness and Pulmonary eosinophilia. And it is known the structure and function of IL-5, its receptor and the mechnism IL-5 triggered eosinophil accumulation and inflammaion of the airways. At this point of view, we assume which oriental materia medics can the splenocyte inhibit from secreting the IL-S in vitro. Material and Methosds: We used the splenocyte of mouse 8 weeks after its birth, and then cnltivated those into the 2 experimental groups and control group for 48 hours. The culture medium of experimental groups were made of $1{\mu}g/ml,\;10{\mu}g/ml$, oriental materia medics, representative. And the culture media of control group was given no oriental materia medica. Then, we assayed the quantity of cytokine-expression by the Sandwich ELISA. The quantifies of cytokine-expression of the experimental groups were compared with that of the control group which was standardized These method were used for the all of oriental materia medica treated. Results: In this study, we demonstrated that 12 oriental materia medica that inhibit the splenocyte from secreting the IL-5 in both $1{\mu}g/ml,\;and\;10{\mu}g/ml$ culture media. Those were Equiseti Herbs, Sophorae Subprostratae Radix, Moutan Radicis Cortex. Trichosanthis Radix, Buddleiae Flos. Cyperi Rhizoma. Benincasae Semen, Armeniacae Semen. Zedoariae Rhizoma, Astragali Semen, Dolichoris Semen. Lilii Bulbus, Asparagi Radix, Atractylodes Rhizoma White, Polygonati Officinallis Rhizoma. Conculusions: These findinga indicate that some oriental materia medica, specially Antipyretics, Herbs for Resolving Phlegm, Relieving Cough and Calming Wheezing and Herbs for Tonifring and Invigorating effects inhibit the splenocyte from secreting the IL-5. And further study experimented in vivo is needed for treating IL-5-driven inflammatory disease including asthma.
