  • This study was conducted to examine the changes in the molecular structure and physiological activities of silk fibroin by gamma irradiation. The results of gel permeation chromatography and sodium dodecyl sulfate-polyacrylamide gel electrophoresis showed that the molecular weight of fibroin was increased depending upon the irradiation dose. Secondary structure of fibroin determined by using circular dichroism revealed that the ratio of $\alpha$-helix was increased up to 10 kGy and then decreased depending upon the irradiation dose. Whereas, the ratio of $\beta$-sheet, $\beta$-turn, and random coil were decreased and then increased with an alteration in the $\alpha$-helix secondary conformation. The 2.2-diphenyl-1-picryl-hydrazil (DPPH) radical scavenging activity of fibroin was increased by gamma irradiation at 5 kGy, but was decreased above 10 kGy depending upon the irradiation dose. Also, the inhibition activities of tyrosinase and melanin synthesis of fibroin were increased by gamma irradiation. These results indicated that gamma irradiation could be used as an efficient method to make fibroin more suitable for the development of functional foods and cosmetics.

  • Effects of radiation dose on mechanical properties such as tensile strength, compressive strength, flexural strength, specific gravity and changes of weight and hydrogen content of epoxy resin-type neutron shielding materials to be used for spent fuel shipping casks have been investigated. At radiation dose up to 0.5MGy, the tensile strength, compressive strength and flexural strength of the shielding materials of KNS-115A, KNS-115B and KNS-115C have been increased with increase in the radiation dose. In contract, these mechanical properties have been decreased at radiation dose above 0.5MGy. The amount of radiation dose on the materials of KNS-115A, KNS-115B and KNS-115C has not resulted in a measurable loss of specific gravity and weight of them, whereas the reduction of hydrogen content has been observed.

  • The responses of soybean seeds were evaluated to accelerated aging and gamma irradiation with regard to germination, seed leakage, seed leachate component and dry weight of hypocotyl and primary root of the germinating seed. Accelerated aging significantly reduced the final germination rate while gamma irradiation increased the final germination rate. Furthermore, the interactive effects occurred that the final germination rate of 5-day aged seeds increased considerably in response to 4 Gy of gamma irradiation. The extent to which the electrolyte was leaked from the seeds (conductivity) was significantly affected by accelerated aging and showed a close negative correlation with the germination rate. Gamma irradiation, however, did not significantly affect the electrical conductivity of seed leachate. The accelerated aging significantly increased the concentrations of the particular electrolytes leaked from the seeds while the gamma irradiation did not affect those concentrations. Of the electrolytes leaked from the seeds, Ca and Mg showed relatively lower concentrations while K showed greater concentrations than others. Moreover, N and P showed similar responses to aging treatment. Aging treatment significantly affected dry weight (DW) of hypocotyls and primary root. Also, gamma irradiation decreased DW of hypocotyls and primary root, particularly for 8 Gy associated with 5 days aging treatment. The data were discussed in terms of the relationships of seed vigor with aging treatment and gamma irradiation.

  • Iodine sulfur (IS) process is one of the promising processes for a hydrogen production by using a high temperature heat generated by a very high temperature gas cooled reactor(VHTR) in the nuclear power industries. Even though the IS process is very efficient for a hydrogen production and it is not accompanied by a carbon dioxide evolution, the highly corrosive environment of the process limits its application in the industry. Corrosion tests of selected materials were performed in sulfuric acid to select appropriate materials compatible with the IS process. The materials used in this work were Fe-Cr alloys, Fe-Ni-Cr alloys, Fe-Si alloys, Ni base alloys, Ta, Ti, Zr, SiC, Fe-Si, etc. The test environments were 50 wt% sulfuric acid at $120^{\circ}C$ and 98 wt% at $320^{\circ}C$. Corrosion rates were measured by using a weight change after an immersion. The surface morphologies and cross sectional areas of the corroded materials were examined by using SEM equipped with EDS. Corrosion behaviors of the materials were discussed in terms of the chemical composition of the materials, a weight loss, the corrosion morphology, the precipitates in the microstructure and the surface layer composition.

  • The vinyl-shielded rearing chamber was controlled by automatic electric heater to maintain relatively constant temperature and under this condition, it took 13 to 14 days from egg to adult and 8 days for their preoviposition. The pupal weight was not varied with the advance of generation, whereas was greatly effected by temperature changes. Since only $60\%$ of the pupae was able to classify the sex by pupal weight, a mechanical device will be needed for the further sexing.

  • Two chitinase producing strains CHI2 and CHI4 were isolated from soybean rhizosphere soil. Both the strains belonged to Lysobacter enzymogenes as indicated by 16S rDNA sequence analysis. Though strain CHI2 and CHI4 produced extracellular chitinase, they differ in their chitinolytic activity. CHI4 produced approximately three times the higher amounts of enzyme than that of CHI2 under specified conditions. CHI2 produced $535.67U\;l^{-1}$ of chitinase after 48 h incubation with a specific activity of $3.91U\;mg^{-1}$ of protein while strain CHI4 produced $1584.13U\;l^{-1}$ of chitinase with a specific activity of $10.88U\;mg^{-1}$ protein. SDS-PAGE analysis indicated that the molecular weight of chitinase enzyme was approximately 45 kDa. A faint band with a molecular weight of 55 kDa reveals the possibility for the presence of another kind of chitin binding protein. Mutant library was developed by exposing the isolates to gamma rays at their $LD_{99}$ value (0.23 kGy). Totally, 11 mutants of CHI2 and CHI4 are reported to have enhanced chitinase activity. Several leaky mutant clones with decreased enzyme activity and a defective mutant (CHI2-M16) with complete loss of chitinase activity were also identified. CHI4-M18, CHI4-M8 and CHI4-M29 showed 78.8, 41.5, and 31.9% increased chitinase activity over wild type CHI4.

  • The recovery of Zr from Zircaloy-4 (Zry-4) cladding hulls using a chlorination method was demonstrated for complete conversion of Zr into $ZrCl_4$. A chlorination reaction was performed by reacting Zry-4 hulls for 8 h under a 70 cc/min $Cl_2$ + 70 cc/min Ar flow at $380^{\circ}C$. The initial weight of the reactant (51.7 g) decreased to 0.49 g after 8 h of operation, which is only 0.95wt% of the initial weight. The weight of the total reaction products was 121.7 g with a high Zr purity of 99.80wt%. Fe and Sn were identified as major (0.18wt%) and minor (0.02wt%) impurities of the reaction products, respectively. It was also shown that Zr exhibited a high recovery ratio of 96.95wt% with a relatively small experimental loss of 2.34wt%. Observation of the reaction residues revealed that the chlorination reaction was dominant along the longitudinal direction, and surface oxide layers remained as reaction residues. The high purity and recovery ratio of Zr proposed the feasibility of the chlorination technique as an effective hull waste treatment method.
