자운영(Astragalus sinicus)종자의 페놀성 화합물 및 항산화 활성 (Phenolic Compounds from Seeds of Astragalus sinicus and Its Antioxidative Activities)

  • Phytochemical examination of seeds of Astragalus sinicus has led to the isolation and characterization of kaempferol $3-O-{\beta}-D-apiofuranosyl-(1{\rightarrow}2)-{\alpha}-L-rhamnopyranosyl-(1{\rightarrow}6)-{\beta}-D-glucopyranoside$ (1), quercetin $3-O-{\beta}-D-xylopyranosyl-(1{\rightarrow}2)-{\beta}-D-glucopyranoside$ (2), ampelopsin (3), ampelopsin $3'-O-{\beta}-D-xylopyranoside$ (4), ampelopsin $3'-O-{\beta}-Dxylopyranoside$ (5), myricetin (6), myricetin $3'-O-{\beta}-D-glucopyranoside$ (7), myricetin $3'-O-{\beta}-D-xylopyranoside$ (8). Antioxidative activity of these compounds was determined by measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals. Flavanonols(3,4, and 5) showed potent antioxidative activity.

Sclerotium rolfsii에 의한 자운영 흰비단병의 발생 (Occurrence of Stem Rot of Astragalus sinicus Caused by Sclerotium rolfsii in Korea)

  • From 2008 to 2009, the stem rot of Astragalus sinicus L. caused by Sclerotium rolfsii occurred sporadically in Gyeongnam area, Korea. The typical symptom is water-soaking, rotting and wilting on the stem. The infected plants were eventually died. White mycelial mats were spread over lesions, and then sclerotia were formed on stems and near soil line. The sclerotia were globoid in shape, white to brown in color, 1-3 mm in size and the hyphal width was 3-9 μm. The optimum temperature for mycelial growth and sclerotial formation on PDA was 30oC. The typical clamp connections were observed in the hyphae of the fungus grown on PDA. On the basis of mycological characteristics and pathogenicity to host plants, this fungus was identified as Sclerotium rolfsii Saccardo. This is the first report on the stem rot of A. sinicus caused by S. rolfsii in Korea.

자운영(Astragalus sinicus L.) 배축절편으로부터 식물체 재생 (Plant regeneration from hypocotyls explants of Astragalus sinicus L.)

  • 자운영 배양으로부터 신초발생에 대한 최적 배양조건을 조사하기 위하여 배축절편을 0.1, 1.0, 2.0 4.0 mg/L 2,4-D가 첨가된 MS배지에서 6주 동안 배양하였다. MS배지에 2,4-D가 0.1 또는 1.0 mg/L로 첨가된 배지에서 형성된 캘러스로부터 신초발생능 캘러스 형성을 조사하기 위하여 2,4-D와 BA 를 혼합 첨가한 배지에 옮겨 4주 동안 배양하면서 조사하였다. 먼저 2,4-D 농도에 따라 신초와 뿌리를 형성하는 캘러스, 뿌리를 형성하는 캘러스, 캘러스, 진한 갈색으로 변하는 캘러스 등 여러가지 형태발생을 관찰할 수 있었다. 일반적으로 신초와 뿌리를 갖는 캘러스는 2,4-D가 낮은 농도로 첨가 되었을 때 형성되었으며, 반면 신초가 없는 캘러스, 갈변 되는 캘러스는 고농도로 처리되었을 때 형성되었다. 또한 BA와 2,4-D가 조합첨가된 배지에서는 신초발생에 효과적이었으며, 뿌리 발생은 감소하였다. 가장 효과적인 신초발생은 1.0 mg/L 2,4-D 와 0.5 mg/L BA가 조합 첨가되었을 때 얻어졌으며, 이러한 신초들은 1/2 MS기본배지에서 대부분 유식물체로 생육되었으며, 토양에서 정상적으로 적응하여 생장하였다.

  • 본 실험에서 지베레린이 자운영의 생육및 내부 성분에 미치는 영향을 토양재배를 통하여 조사하였다. 잎에 살포한 지베레린의 비교적 적은 농도 차는 자운영의 내부 성분에 현저한 차이를 나타내고 생육의 증가는 지베레린의 농도증가에 관련되었다. 엽녹소 및 Carotene 함량은 지베레린 농도와 상반되고 생장율은 엽록소와 Carotene 함량과 크게 상반되었다. Carotene 함량은 엽록소량과 함께 변하였는데 엽록소와 Carotene의 비는 Beck와 Redman이 얻은 결과처럼 높지 않았다. 엽록소와 Vitamin C의 함량은 발육이 진전함을 따라 차이가 생기는데 이 차이는 엽록소만 유의하다. Anthocyanim 함량은 지베레린의 농도가 증가함에 따라 Sucrose 및 Reducing Sugar의 함량과 함께 감소되었다. Anthocyanim 함량은 Jacob Straus의 옥수수에 대한 실험 결과와 같이 많지 않다.

Changes of Seed Quality of Chinese Milk Vetch(Astragalus sinicus L.) During Seed Developmental Stages

  • The objective of this study was to investigate the variation of Chinese milk vetch(Astragalus sinicus L.; CMV) seed quality after flowering. We tagged individual open flowers of CMV at the day of maximum flowering(11 May) in Seoul, Korea. Seed samples were harvested serially at 15, 20, 25 and 30 days after flowering(DAF). To compare with dried seeds, non-dried seeds were tested immediately after harvest and the remaining seeds were placed at room temperature for 4 weeks. Seed length, 1000 seed weight, moisture content, germination rate(GR), mean germination time(MGT), germination speed(GS), germination performance index(GPI) and physical dormancy rate(PDR) were investigated. Seed length increased to 2.6 mm and 1000 seed weight reached up to 2.2 g until 25 DAF. Seed moisture content dramatically decreased from 20 to 25 DAF. Moisture content of non-dried seed(7.5%) was similar to that of dried seed(5.5%) at 25 DAF. The rate of seed viability reached up to 94% at 25 DAF. In case of dried seed, GR increased up to 39% at 25 DAF whereas GR of non-dried seed varied from 5 to 10%. GS and GPI of dried seed were significantly higher than those of non-dried seed since 25 DAF. PDR of dried seed has decreased since 20 DAF, whereas PDR of non-dried seed has increased. GR, GS and GPI increased as PDR decreased. Our results evidenced that PDR might be one of major factor in variation of seed quality, of which development was completed at 25 DAF.

Physical Dormancy in Seeds of Chinese Milk Vetch (Astragalus sinicus L.) from Korea

  • Freshly harvested seed of Chinese milk vetch (Astragalus sinicus L.; CMV) was strongly dormant because of hardseedness. Seeds of freshly harvested germinated only 8% while clipping the seed coat completely overcome the innate dormancy, which indicates inhibition of germination of the seed is mainly due to seed coat (87%). The dormant (intact) hard seeds did not imbibe water whereas the non-dormant (clipped) seeds took up rapidly. In natural environment condition, the hard seed coat dormancy was broken only after 5 months after seed harvest. To break such a strong seed coat dormancy, the chemical and heat treatments were effective. Concentrated sulfuric acid was more effective than dry heat and hot water treatments. Hot water treatment improved germination but the germination percentage was less than 41%. Treatments increased germination due to its effect on the seed coat integrity. A scanning electron microscope reveled that disruption of seed coat layers and subsequent development of numerous crack in the hilum region of the seed and on the seed coat surface of concentrated sulfuric acid treatment and formation of cracks in the dry heat treatments, respectively, were observed in the seed coat surface, which served as water entry points.

중부지방 적합 자운영 (Astragalus sinicus L.) 형질 특성 및 유전적 연관성 분석에 관한 연구 (Study for Morphological and Genetic Characteristics of Chinese Milk Vetch (Astragalus sinicus L.) to Select Suitable Line in Central Area of Korea)

  • 본 연구는 중북부지방 적응성 자운영 선발 및 수집 계통간 형태적, 유전적 근연관계를 분석한 것으로서 총 수집된 16개 자운영 계통 중 파주종은 엽각이 적고, 지하부의 발육이 좋아 형태적 형질이 월동에 적합한 초형이었다. 또한 구례 수집종은 개화기가 타 수집종에 비해 일주일 가량 빨랐다. 유전적 근연관계를 검정하기 위해 8개 primer 조합으로 AFLP를 실시한 결과 총 579개의 밴드를 얻었으며, 이 중 polymorphism을 갖는 밴드는 61.54%인 336개 밴드였다. AFLP 데이터를 이용해 군집분석을 실시한 결과 계통 간 유사도 지수는 0.826~0.939 사이의 값을 가지며, 구례와 파주 수집종은 타 수집종과 유전적 유사도에서 비교적 큰 차이를 보였다.
