Application of Toxicogenomic Technology for the Improvement of Risk Assessment

  • Recently, there has been scientific discussion on the utility of -omics techniques such as genomics, proteomics, and metabolomics within toxicological research and mechanism-based risk assessment. Toxicogenomics is a novel approach integrating the expression analysis of genes (genomic) or proteins (proteomic) with traditional toxicological methods. Since 1999, the toxicogenomic approach has been extensively applied for regulatory purposes in order to understand the potential toxic mechanisms that result from chemical compound exposures. Therefore, this article's purpose was to consider the utility of toxicogenomic profiles for improved risk assessment, explore the current limitations in applying toxicogenomics to regulation, and finally, to rationalize possible avenues to resolve some of the major challenges. Based on many recent works, the significant impact toxicogenomic techniques would have on human health risk assessment is better identification of toxicity pathways or mode-of-actions (MOAs). In addition, the application of toxicogenomics in risk assessment and regulation has proven to be cost effective in terms of screening unknown toxicants prior to more extensive and costly experimental evaluation. However, to maximize the utility of these techniques in regulation, researchers and regulators must resolve many parallel challenges with regard to data collection, integration, and interpretation. Furthermore, standard guidance has to be prepared for researchers and assessors on the scientifically appropriate use of toxicogenomic profiles in risk assessment. The National Institute of Toxicological Research (NITR) looks forward to an ongoing role as leader in addressing the challenges associated with the scientifically sound use of toxicogenomics data in risk assessment.

Ayres Sensory Integration (ASI$^{(R)}$) 중재 교육에 따른 치료사의 치료 수행도 변화 (Delivery of Therapist's Intervention to the Education of Ayres Sensory Integration$^{(R)}$ (ASI$^{(R)}$))

  • 목적: 본 연구의 목적은 ASI$^{(R)}$의 중재 교육을 6명의 작업치료사에게 실시하고, 교육 전과 후에 ASI$^{(R)}$의 중심원리 수행도를 자기-평가, 타인-평가, 전문가-평가로 알아보는 것이다. 연구 방법: 2013년 11월 30일부터 2014년 6월 30일까지 총 6명의 작업치료사를 대상으로 연구가 이루어졌다. 총 8주 동안 6회의 ASI$^{(R)}$ 중재 교육을 실시하였고, 대상자들이 ASI$^{(R)}$ 중재 교육 전에 촬영한 치료 동영상과 교육 후에 촬영한 치료 동영상을 평가하였다. 평가는 교육전과 후에 자기-평가, 타인-평가, 전문가-평가로 실시하였고, 이 자료를 바탕으로 Mann-Whitney, ICC (Intra-Class Correlation)로 분석하였다. 결과: 영역III(평가기록), IV(공간과 도구), V(부모, 교사와의 의사소통)의 구조적 요소 합의 결과는 $79.33{\pm}18.27$점이었다. 평가방법에 따른 ASI$^{(R)}$ 중재 교육 전과 후의 과정요소 차이를 살펴본 결과, 자기-평가에서는 '감각조절 제공', '활동선택에서 협력', '활동의 성공보장', '놀이에 대한 아동의 내적 동기를 지지', '치료적 연합', '과정요소의 합'에서 교육 전보다 교육 후에 점수가 향상되었다. 타인-평가에서는 '물리적 안정성 보장'을 제외한 모든 항목에서 통계학적으로 유의한 차이를 보였다. 전문가-평가에서는 '감각의 기회제공'을 제외한 모든 항목에서 통계학적으로 유의한 차이를 보였다. ASI$^{(R)}$ 중재 교육 전에 실시한 자기-평가와 전문가-평가에서는 '물리적 안정성 보장'항목에서 통계학적으로 유의한 차이를 보였다. 교육 후에 실시한 자기-평가와 전문가-평가에서도 '물리적 안정성 보장' 항목에서 차이를 보였다. 결론: 본 연구의 결과는 정확한 감각통합중재를 위해 ASI$^{(R)}$의 중재 교육이 필요함을 강조하고, 치료사 스스로의 치료적 스타일을 점검하는 것이 중요하다는 것을 제시한다.

  • Seismic risk management of the built environment is integrated by two main stages, the assessment and the remedial measures to attain its reduction, representing both stages a complex task. The seismic risk of a certain structure located in a seismic zone is determined by the conjunct of the seismic hazard and its structural vulnerability. The hazard level mainly depends on the proximity of the site to a seismic source. On the other hand, the ground shaking depends on the seismic source, geology and topography of the site, but definitely on the inherent earthquake characteristics. Seismic hazard characterization of a site under study is suggested to be estimated by a combination of studies with the history of earthquakes. In this Paper, the most important methods of seismic vulnerability evaluation of buildings and their application are described. The selection of the most suitable method depends on different factors such as number of buildings, importance, available data and aim of the study. These approaches are classified in empirical, analytical, experimental and hybrid. For obtaining more reliable results, it is recommends applying a hybrid approach, which consists of a combination between methods depending on the case. Finally, a recommended approach depending on the building importance and aim of the study is described.

A Dimensionality Assessment for Polytomously Scored Items Using DETECT

  • A versatile dimensionality assessment index DETECT has been developed for binary item response data by Kim (1994). The present paper extends the use of DETECT to the polytomously scored item data. A simulation study shows DETECT performs well in differentiating multidimensional data from unidimensional one by yielding a greater value of DETECT in the case of multidimensionality. An additional investigation is necessary for the dimensionally meaningful clustering methods, such as HAC for binary data, particularly sensitive to the polytomous data.

Fatigue Assessment of Welded Ship Structures

  • Modern ocean-going vessels are highly dynamically loaded all-welded structures. Traditionally, this fact has been taken into account in the design of ship structures by keeping the overall stress level within permissible limits and by limiting notch effects in the design of local structures. With an improving knowledge of loads and the rapid development of calculation means more detailed calculation-based methods are increasingly introduced. A brief review on fatigue assessment methods used in the design of ship structures has been given during the 1993 IIW International Conference in Glasgow. The rapid development during recent years underlines the importace of the subject and justifies its extended discussion in this paper.

  • In this paper, we investigate the Bayesian approach to random effect binomial regression models with improper prior due to the absence of information on parameter. We also propose a method of estimating the posterior moments and prediction and discuss some general methods for studying model assessment. The methodology is illustrated with Crowder's Seeds Data. Markov Chain Monte Carlo techniques are used to overcome the computational difficulties.

  • In this study has investigated and considered product planning methods at the existing product design process and being used the safety evaluation methods at the product planning stage, and has deduced the 16 safety evaluation elements to have to be considered for executing the safety evaluation at the product planning stage. When the safety evaluation is executed from the product planning stage, more safe products will be designed.

  • This paper evaluates the effectiveness of three nonlinear static methods for the prediction of the dynamic response of in-plan irregular buildings. The methods considered are the method suggested in Eurocode 8, a method previously proposed by some of the authors and based on corrective eccentricities and a new method in which two pushover analyses are considered, one with lateral forces applied to the centres of mass of the floors and the other with only translational response. The numerical analyses are carried out on a set of refined models of reinforced concrete framed buildings. The response predicted by the nonlinear static analyses is compared to that provided by nonlinear dynamic analyses. The effectiveness of the nonlinear static methods is evaluated in terms of absolute and interstorey displacements.

  • Before the EIA was established in the Federal Republic of Germany, the impact mitigation principle - a planning instrument, which has its legal foundation in the German nature conservation legislation already had {and still has} the function to valuate environmental impacts. The valuation principles and methods which have been developed in correlation to this instrument are now also used in the EIA. Particularly for the valuation of alternating effects on the different ecological landscape functions and for the valuation of ecological mitigation and compensation measures this valuation methods are used. These methods base on a special kind of modelling nature and environment Following the aim of the nature conservation act, which is to save the capacity of the landscape to perform its essential functions, not the ecological factors (soil, water, air etc.) itselfs are evaluated but the ecological functions of the landscape, which are based on the ecological factors.

지진에 대한 강구조물의 피로손상도 추정법 (Fatigue Damage Assessment for Steel Structures Subjected to Earthquake)

  • Structures subjected to strong seismic excitation may undergo inelastic deformation cycles. The resulting cumulative fatigue damage process reduces the ability of structures and components to withstand seismic loads. Yet, the present earthquake resistance design methods focus mainly on the maximum displacement ductility, ignoring the effect of the cyclic responses. The damage parameters closely related to the cumulative damage need to be properly reflected on the aseismic design methods. In this study, two cumulative damage assessment methods derived from the plastic fatigue theory are investigated. The one is based on the hysteretic ductility amplitude, and the other is based on the dissipated hysteretic energy. Both methods can consider the maximum ductility and the cyclic behavior of structural response. The validity of two damage methods has been examined for single degree of freedom structures with various natural frequencies against two different earthquake excitations.
