Protein Tertiary Structure Prediction Method based on Fragment Assembly

  • A novel method for ab initio prediction of protein tertiary structures, PROFESY (PROFile Enumerating SYstem), is introduced. This method utilizes secondary structure prediction information and fragment assembly. The secondary structure prediction of proteins is performed with the PREDICT method which uses PSI-BLAST to generate profiles and a distance measure in the pattern space. In order to predict the tertiary structure of a protein sequence, we assemble fragments in the fragment library constructed as a byproduct of PREDICT. The tertiary structure is obtained by minimizing the potential energy using the conformational space annealing method which enables one to sample diverse low lying minima of the energy function. We apply PROFESY for prediction of some proteins with known structures, which shows good performances. We also participated in CASP5 and applied PROFESY to new fold targets for blind predictions. The results were quite promising, despite the fact that PROFESY was in its early stage of development. In particular, the PROFESY result is the best for the hardest target T0161.

  • The present study covers the optimal design for a lens module in a mobile phone camera by using the design of experiments (DOE) and finite element (FE) analysis. FE analyses are performed to investigate the effect of design parameters on the amount of torque required to assemble a barrel and a housing part. The DOE approach is then performed to optimize the design parameters in order to maintain an appropriate torque with less variations.

  • Combination of magnetic recording technology and optical recording technology such as Near Field Optical Recording is watched recently. In order to accomplish this technology, the development of an electromagnetic driven mm-sized mirror shifting laser beam in track direction have to needed. In Near Field Optical Recording System, shifting laser beam in track direction mean as fine tracking and means as coarse tracking. Therefore in Near Field Optical Recording, 2-stage actuator is composed of servo controller in reading or recording information on disc layer. In our research, through design and simulation process of driven mm-sized mirror, we arrange systematically design process of driven mm-sized mirror having good frequency transfer characteristics. Design and simulation processes included modal analysis of spring, calculation of magnetic moment according to the number of turns and geometric configuration of coil and magnetic circuit analysis meaning that calculation of magnetic flux density in air gap of magnetic circuit. After that we design and make parts of driven mm-sized mirror, assemble and evaluate our electriomagnetic driven mm-sized mirror. we compared design values with actual characteristic values and present solution scheme. Through these processes we performed manufacturing of an electromagnetic driven mm-sized mirror having good frequency-domain characteristics and high sensitivity characteristics.

Fate of Donor Centrosome and Microtubule Dynamics of Porcine Somatic Cell Nuclear Transfer Embryos

  • We investigated the microtubule dynamics, including the inheritance of donor centrosomes and the mitotic spindle assembly occurring during the first mitosis of somatic cell nuclear transfer (SCNT) embryos in pigs. SCNT embryos were fixed 15 min and 1 h after fusion in order to assess the inheritance pattern of the donor centrosome. The distribution and dynamic of the centrosome and microtubule during the first mitotic phase of SCNT embryos were also evaluated. The frequency of embryos evidencing $\gamma$-tubulin spots (centrosome) was 93.2% in the SCNT embryos 15 min after fusion. In the majority of the SCNT embryos (61.5%), however, no centrosome was observed 1 h after fusion. The frequency of the embryos with no or abnormal mitotic spindles 20 h after fusion was 19.6%. The $\gamma$-tubulin spots were detected near the nuclei of somatic cells regardless of cell cycle phase, whereas $\gamma$-tubulin spots in the SCNT embryos were observed only during the inter-anaphase transition. These results showed that the donor centrosome is inherited into the SCNT embryos, but failed to assemble the normal mitotic spindles during first mitotic phase in some SCNT embryos.

Designing of a Global Logistics System for the ICGCPS under Considering Overseas Markets

  • This paper proposes a way of designing of a global logistics system for "the international cooperative global complementary production system" (ICGCPS) constructed in ASEAN region. ICGCPS is a global production system with several production bases located in a number of countries. In order to assemble the final products and sell them in the domestic market, each production base produces only special kinds of components and parts with the total demand required all the participating countries, and supplies them to the other production bases each other. In the ICGCPS, there are a number of important decision-making problems such as identifying which countries are suitable to produce specified components and parts, and deciding how to transport components and parts between the production bases. In the initial period of this system, each production base focused on its domestic market so that the final products it produced were sold only in the country where the base was located. Recently, some production bases have expanded sales to overseas markets. Taking this fact into account, we propose a production and transportation planning model in this paper that takes into account the export quantity of the final products, formulating it into a mathematical programming problem. Using this, we propose a way for managing the supply chain processes of the ICGCPS in order to improve performance measurements such as the total lead-time, the inventory quantity at the depot and the average rate of loading. A numerical example is presented to clarify the procedure proposed in this paper.

A New Component Model and Framework for Reuse of Components (컴포넌트 재사용을 지원하는 컴포넌트 모델 및 프레임워크)

  • It is difficult to assemble independently developed software components because of discrepancies between their interfaces. In order to resolve such problem, we propose a new component model, Active Binding Technology, in which each component has its own independently-defined interface for service request that is revealed in its metadata, instead of passively following the interface of a service-providing component. This model Includes the use of the glue component, an interface mediating place, whose template code is automatically generated by reading in the metadata of the components to be combined. We also propose a runtime framework that holds the pool of component instances, completes the assembly of components in the manner of dependency injection, and performs middleware services and real-time system monitoring through glue components. In order to test the practical value of Active Binding Technology, we have made a tool, which supports the development and assembly of Active Binding components.

Improving Qualification of Safety Manager at Construction Site (건설현장 안전관리자의 자격기준에 관한 연구)

  • As shown in the accident analysis from 2007, it has been found that causes of accidents on large-scale projects are different from those found in general construction projects. A 300-question survey regarding systematic and practical aspects of safety problems at construction sites was distributed to ten (10) different companies. Participants were to respond subjectively, so that the results could be used to assemble the first formal questionnaire survey. They were collected and compiled by an advisory committee for this study. The resulting surveys were then sent to the division chiefs of the top 100 construction companies in Korea, in order to improve the response rate. The Department of Safety & Health Direction, and the :Ministry of Labor, ROK sent the same sheets to medium and small construction companies that placed within 101-200th of all Korea construction companies. The above safety engineers were classified into four (4) levels, from 1st to 4th, followed by the project dollar amount and risk level. Formulae were developed to assign safety engineers to construction sites by engineer level, the project dollar amount, and the project risk level. Conclusions are summarized as follows: 1) Reviewing the assignment system of experienced safety engineers to large scaled projects - The more experienced the engineers assigned to a project, the higher the level of accident prevention. 2) Enforcing the assignment of advanced level safety engineers to large-scaled sites - At least one advanced-level safety engineer should be assigned to construction sites with projects valued at $15million USD (15,000,000,000). 3) For assigning safety engineers by risk level - Twenty models have been developed to calculate the number of safety engineers to be assigned by risk level. In the future, risk level for each job should be established by the government (as is now the practice in Germany).

  • Technological encroachment provides human operators with flood of information that must be analyzed to understand the environment and make judgments that lead to strategic actions. Further, the environment is not static and therefore uncertain, changing its aspect dynamically. Complexity accompanied with its dynamics imposes substantial difficulty to human operators' task. Criticality of having situational understanding becomes more important than ever. Situationalunderstanding requires the human operators possessing tacit knowledge in order for them to make the sense out of the situation while interacting with information from many heterogeneous sources, the notion of sensemaking. Sensemaking refers to the process of developing mental framework to assemble pieces of information representing different aspects of the environment that can be used to develop one's own actionable knowledge to implement their judgments in the uncertain environment. Therefore, judgment process and performance is a key component of sensemaking process. Among many judgment and decision making models, the lens model with its extension can be utilized to partially describe the judgmental aspect of sensemaking. One of the lens model parameters, unmodeled knowledge, can be a corresponding quantitative measure for the tacit knowledge that plays an important role in sensemaking. In this paper, a comprehensive literature for sensemaking is provided to formally define the notion of sensemaking in the military domain. Also, it is proposed that there is a crucial link between the sensemaking and human judgment process and performance from the lens model perspective. Potential implications for experimental framework are also proposed.

Stability Evaluation of Series and Parallel Varistor Combination Using Thermal Image Analysis (열화상 분석을 통한 바리스터의 직렬과 병렬 조합의 안전성 평가)

  • According to the recent revision of KS on the basis of the IEC, equipotential grounding systems has been come into focus and the use of surge protective devices(SPD) has been increased radically in order to operate the power system stably. $Z_nO$ varistor with non-linear resistance, which has an outstanding voltage-current(V-I) characteristic, is mainly used in power system to limit surge voltage and divert surge current. $Z_nO$ varistors are packaged several types based on the circuit assembly to be connected to a.c. power line. When the user assemble the $Z_nO$ varistors into parallel or series circuit package, there are my things to be taken into consideration including functions and thermal stability because they are directly related to the safety. We compare stabilities of each assembly type by measuring residual voltage, discharge current, leakage current and surface temperature concerned to the protection performances between a single device with a 40[kA] of current capacity and parallel or series circuits type of varistor package.

Interaction Between Acid-Labile Subunit and Insulin-like Growth Factor Binding Protein 3 Expressed in Xenopus Oocytes

  • The acid-bible subunit (ALS) associates with the insulinlike growth factor (IGF)-I or II, and the IGF binding protein-3 (IGFBP-3) in order to form a 150-kD complex in the circulation. This complex may regulate the serum IGFs by restricting them in the vascular system and promoting their endocrine actions. Little is known about how ALS binds to IGFBP3, which connects the IGFs to ALS. Xenopus oocyte was utilized to study the function of ALS in assembling IGFs into the ternary complexes. Xenopus oocyte was shown to correctly translate in vitro transcribed mRNAs of ALS and IGFBP3. IGFBP3 and ALS mRNAs were injected in a mixture, and their products were immunoprecipitated by antisera against ALS and IGFBP3. Contrary to traditional reports that ALS interacts only with IGF-bound IGFBP3, this study shows that ALS is capable of forming a binary complex with IGFBP3 in the absence of IGF When cross-linked by disuccinimidyl suberate, the band that represents the ALS-IGFBP3 complex was evident on the PAGE. IGFBP3 movement was monitored according to the distribution between the hemispheres. Following a localized translation in the vegetal hemisphere, IGFBP3 remained in the vegetal half in the presence of ALS. However, the mutant IGFBP3 freely diffused into the animal half, despite the presence of ALS, which is different from the wild type IGFBP3. This study, therefore, suggests that ALS may play an important role in sequestering IGFBP3 polypeptides via the intermolecular aggregation. Studies using this heterologous model will lead to a better understanding of the IGFBP3 and ALS that assemble into the ternary structure and circulate the IGF system.
