Cannabidiol derivatives (1, 2 and 3), 5-fluorouracil (4, 5-FU) and adriamycin (5, AM) were tested for their growth inhibitory effects against human tumor cell lines using two different 3-{4,5-dimeth-ylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay and sulforhodamine B protein (SRB) assay. The light microscopic study showed morphological changes of the treated cells. Disruptions in cell organelles were determined by colorimetric methods; MTT assay and STB assay. These results suggest that cannabidiol (1, CBD) retains the most growth-inhibitory activity against human tumor cell lines.
In developing a useful tool to detect parasitic dynamics in an estuarine ecosystem, a denaturing high-performance liquid chromatography (DHPLC) assay was optimized by cloning plasmid DNA from the grass shrimp Palaemonetes pugio, and its two parasites, the trematode Microphallus turgidus and bopyrid isopod Probopyrus pandalicola. The optimal separation condition was an oven temperature of $57.9^{\circ}C$ and 62-68% of buffer B gradient at a flow rate of 0.45 mL/min. A peptide nucleic acid blocking probe was designed to clamp the amplification of the host gene, which increased the amplification efficiency of genes with low copy numbers. Using the DHPLC assay with wild-type genomic, the assay could detect GC Gram positive bacteria and the bopyrid isopod (P. pandalicola). Therefore, the DHPLC assay is an effective tool for surveying parasitic dynamics in an estuarine ecosystem.

Various kind of whitening agents have been reported in Korea, but standard efficacy protocols are not established yet. So more economical, reproducible standard efficacy assay for whitening agents are needed. As a dermatology specialist, non radio-labeled intracellular melanin assay may be a good candidate for melanogenesis assay and MTT assay with normal human melanocytes may be a good candidate for cell proliferation assay.
We established the model of clonogenic assay with human tumor cell line such as Calu-3 (lung carcinoma), HEC- lB (endometrial adenocarcinoma) , HEp-2 (larnyx carcinoma), Hs-5787 (breast carcinoma), K-562 (chronic myelogenous leukemia), SF-188 (brain carcinoma), SNU-1 (stomach carcinoma) and WiDr (colon carcinoma) . We investigated growth inhibition of solvent (EtOH, MeOH) and water (100$^{\circ}C$, 121$^{\circ}C$) extracts from Korean red ginseng by clonogenic assay. The results of clonogenic assay showed that EtOH extract had growth inhibition against Calu-3, SF-188 and SNU-1, MeOH extract had growth inhibition against Calu-3, Hs-5787, K-562, and WiDr, but water extract at 100$^{\circ}C$ and water extract at 121$^{\circ}C$ had not growth inhibition against used cell lines.
To determine whether ozone is genotoxic at environmentally relevant exposure level, B6C3F1 mice were exposed to 0.5 ppm ozone for 12 weeks, 6 hr/day. Chromosomal aberration, supravital micronucleus and hprt mutation assays were performed. The percentage of abnormal cells was significantly increased at 0.5 ppm ozone when compared to unexposed control in chromosome aberration assay. Significant increase in the frequencies of micro nucleated reticulocytes and 6-thioguanine-resistant ($TG^r$) lymphocytes was also observed in supravital micronucleus assay using peripheral blood and lymphocyte hprt mutation assay, respectively. The results indicate, that under our experimental conditions, 0.5 ppm ozone are genotoxic in exposed B6C3F1 mice.
Objectives : This study was to examine the efficacy of GCT on the hepatoprotective effect in the liver function and immune octivity. Methods : The experiment to investigate the hepatoprotective effect of GCT on the liver damage was conducted with D-galactosamine. The experiments to verify the effects of GCT on the immune activity were conducted by carbon clearance assay, plaque-forming cell SRBC assay of IgM, lymphoproliferation assay of T and B cells, and adherence and phagocytosis of mocrophages. Results: In the damage of liver induced by D-galactosamine, GCT carried hepatoprotective effect on AST. In carbon clearance assay GCT showed significant effect on phagocytosis of Kuffer cells. In the plaque-forming cell assay, GCT improved the formation of IgM. In the lymphoproliferation assay, GCT activated the formation of T and B lymphocytes. In macrophages, GCT activated adherence and phagocytosis. Conclusion : Though further study is needed, our findings suggest that GCT could be recommended as hepatoprotector and immune modulator for liver disease.

Growth inhibitory effect of doenjang(Korean soypaste) methanol extracts in SRB assay using AGS human gastric adenocarcinoma cell, Hep 3B human hepatocellular carcinoma cell and HT 29 human colon cancer cell was studied. The treatment of doenjang methanol extracts(2mg/assay) to the AGS, Hep 3B and HT 29 cancer cells inhibited the growth of the cancer cells by 55%, 60%, and 71%, respectively. Doenjang methanol extracts exhibited the highest inhibitory effect among other soybean fermented foods and original materials in the SRB assay. In addition, to separate active compounds of doenjang methanol extracts, we fractionated the doenjang with hexane, methanol, dichloromethane, ethylacetate and butanol. Growth inhibitory effect on the AGS, Hep 3B, HT 29 and MG 63 cancer cells was the highest in the fractions of dichloromethane and ethylacetate among other solvent fractions of the doenjang. These results showed that some compounds contained in the fractions of dichloromethane and ethylacetate might play a role on the anticanceric effect of doenjang.
80% MeOH extract of black rices fractionated with n-hexane, EtOAc, and n-BuOH. Copper-induced LDL oxidation inhibition assay and ACAT inhibition assay examined with fractions. n-Hexane and EtOAc fractions showed high inhibition activity. We divided n-hexane fraction into three sub-layers(H1 - H3) and EtOAc into eight sub-layers(E1 - E8). H1, H2, E6, and E7 are showed higher inhibition activity than standard in Lp-PLA2 inhibition assay and H1 and E6 are showed higher ACAT inhibition activity than standard.

In the present study, estrogenic or antiestrogenic activity of cypermethrin, a pyrethroid insecticide was investigated. We used immature rat uterotrophic assay, estrogen-responsive calbindin-D9k (CaBP-9k) gene expression assay and luciferase reporter gene assay for measure of estrogenic potential of cypermethrin.(omitted)
The purposes of our study were to optimize the conditions of the screening and testing methods for endocrine disruptors, to characterize these assays using several compounds with well-defined endocrine activity, and to compare the sensitivity between these assays currently undergoing validation. Two in vitro test systems, MCF-7 cells proliferation (E-screen assay) and competitive binding to estrogen receptors (ER) were selected to evaluate the estrogenic effects. 17$\beta$-Estradiol (E2) and diethylstilbestrol (DES) were used as a positive control in vitro test. Also, E2 and ethinyl estradiol (EE) were used as a positive control in vivo uterotrophic assay. In in vitro test, E2 and DES showed a strong estrogenic response at concentration of 1.0 nM. In uterotrophic assay, E2 (0.3 $\mu\textrm{g}$/kg) and EE (0.3 $\mu\textrm{g}$/kg) produced a significant increase in uterus and vagina weight in both immature and ovariectomized rats. Although we did not com-pared the specificity between in vivo and in vitro assays, these assay systems may serve as a good tool for endocrine disruptors screening methods. Our data indicate that these assay systems exhibit some difference in their sensitivity to the same estrogenic compounds. Therefore, as a first rapid screening assay for estrogenic activity qf unknown chemicals, at least two assay systems should probably be carried out with a view of high sensitivity and standardization conditions. Also, a careful validation tests are necessary to obtain a reasonable degree of reproducibility.
