Diversity of Marine-Derived Aspergillus from Tidal Mudflats and Sea Sand in Korea

  • Aspergillus (Trichocomaceae, Eurotiales, and Ascomycota) is a genus of well-defined asexual spore-forming fungi that produce valuable compounds such as secondary metabolites and enzymes; however, some species are also responsible for diseases in plants and animals, including humans. To date, 26 Aspergillus species have been reported in Korea, with most species located in terrestrial environments. In our study, Aspergillus species were isolated from mudflats and sea sand along the western and southern coasts of Korea. A total of 84 strains were isolated and identified as 17 Aspergillus species in 11 sections on the basis of both morphological characteristics and sequence analysis of the calmodulin gene (CaM) locus. Commonly isolated species were A. fumigatus (26 strains), A. sydowii (14 strains), and A. terreus (10 strains). The diversity of Aspergillus species isolated from mudflats (13 species) was higher than the diversity of those from sea sand (five species). Four identified species-A. caesiellus, A. montenegroi, A. rhizopodus, and A. tabacinus-are in the first records in Korea. Here, we provide detailed descriptions of the morphological characteristics of these four species.

  • Aspergillus는 가장 흔하게 감염을 일으키는 기회 감염균이다. 검사실에서 Aspergillus의 동정은 대부분 형태학적으로 이루어진다. 최근에는 형태학적으로 동정이 안 되거나, 속 수준까지만 동정이 되는 까다로운 균들 동정하기 위하여 분자유전학적인 방법들이 제안되고 있다. 임상검체에서 분리한 475건의 Aspergillus는 Aspergillus fumigatus 257주(54.1%), A. niger 101주(21.3%), A. flavus 43주(9.1%), A. terreus 29주(6.1%), Aspergillus nidulans 2주(0.4%), 그리고 Aspergillus clavatus 1주(0.2%), 그리고 Aspergillus species는 42주(8.8%)이었다. 미동정 균주와 종 구분을 위한 11건의 염기서열분석에서는 Aspergillus fumigatus 5주, Aspergillus falvus 1주, Aspergillus terreus 1주, Aspergillus lentulus 1주 동정되었다. Aspergillus versicolor 2주, 그리고 1주는 Emericella parvathecia로 동정되었다. Aspergillus의 92.2%는 형태학적으로 동정이 가능하였고, 8.8%는 종 수준의 동정이 불가능하였다. 종 수준의 동정을 하기 위해서는 ITS와 D1D2를 이용한 염기서열분석이 유용한 것으로 사료된다.

일균일명 체계에 의한 국내 보고 Aspergillus, Penicillium, Talaromyces 속의 종 목록 정리 (Species List of Aspergillus, Penicillium and Talaromyces in Korea, Based on 'One Fungus One Name' System)

  • 인간의 삶에 큰 영향을 끼치는 Aspergillus, Penicillium 및 그들의 완전세대 속에 속하는 곰팡이의 국내 보고 종 목록을 작성하였다. 기존의 국가 생물종 목록집에는 Aspergillus 14종, Eurotium 4종, Neosartorya 8종, Penicillium 47종, Talaromyces 5종이 보고되었다. 본 논문에서는 국제조류 균류 식물명명규약(ICN)에 따른 일균일명 체계에 따라 Eurotium과 Neosartorya은 Aspergillus 속으로 합치고 Penicillium과 Talaromyces도 새로운 속 개념에 따라 정리하였다. 또한 기존의 국가 생물종 목록에 빠져 있던 77종을 추가하여 Aspergillus, Penicillium 및 그들의 완전세대에 속하는 곰팡이의 국내 보고 종 목록을 Aspergillus 55종, Penicillium 82종, Talaromyces 18종으로 정리하였다.

Aspergillus cumulatus sp. nov., from Rice Straw and Air for Meju Fermentation

  • A new species named Aspergillus cumulatus sp. nov. is described in Aspergillus section Aspergillus (Eurotium state). The type strain (KACC $47316^T$) of this species was isolated from rice straw used in meju fermentations in Korea, and other strains were isolated from the air in a meju fermentation room. The species is characterized by growth at a wide range of water activities and the formation of aerial hyphae on malt extract 60% sucrose agar (ME60S) that resemble a cumulus cloud. Furthermore, A. cumulatus produces yellow ascomata containing small lenticular ascospores (5.1-5.7 ${\mu}m$) with a wide furrow, low equatorial crests, and tuberculate convex surface. The species is phylogenetically distinct from the other reported Aspergillus section Aspergillus species based on multilocus sequence typing using rDNA-ITS, ${\beta}$-tubulin, calmodulin, and RNA polymerase II genes.

Long-Term Investigation of Marine-Derived Aspergillus Diversity in the Republic of Korea

  • Aspergillus species play a crucial role in terrestrial environments as degraders and are well known for producing various secondary metabolites. Recently, Aspergillus species have been discovered in marine environments, exhibiting adaptability to high salinity and producing diverse secondary metabolites with valuable properties. However, limited research has focused on their marine diversity, leading to inaccurate species identification. The current study addresses this gap by investigating diverse marine habitats in the Republic of Korea, including sediment, seawater, seaweed, and marine animals. From three coasts of the Korean Peninsula, 472 Aspergillus strains were isolated from the various marine habitats. A total of 41 species were accurately identified using multigenetic markers: internal transcribed spacer, calmodulin, and b-tubulin. The findings underscore the importance of accurate identification and provide a basis for elucidating the functional role of marine-derived Aspergillus species in marine ecosystems.

  • Two hundred and twenty five strains which belong to the genus Aspergillus were isolated from specimens collected throughout the southern coastal areas of Korea, Fifteen species and one variety throughout the nine species groups including A. itaconicus and A.unguis, which are unrecorded in Korea, were identified according to Raper-Fennell's classification key.

Aspergillus Associated with Meju, a Fermented Soybean Starting Material for Traditional Soy Sauce and Soybean Paste in Korea

  • Aspergillus is an important fungal genus used for the fermentation of Asian foods; this genus is referred to as koji mold in Japan and China. A. oryzae, A. sojae, and A. tamari are used in the production of miso and shoyu in Japan, but a comprehensive taxonomic study of Aspergillus isolated from Meju, a fermented soybean starting material for traditional soy sauce and soybean paste in Korea, has not been conducted. In this study, various Aspergillus species were isolated during a study of the mycobiota of Meju, and the aspergilli were identified based on phenotypic characteristics and sequencing of the ${\beta}$-tubulin gene. Most strains of Aspergillus were found to belong to the following sections: Aspergillus (n = 220), Flavi (n = 213), and Nigri (n = 54). The most commonly identified species were A. oryzae (n = 183), A. pseudoglaucus (Eurotium repens) (n = 81), A. chevalieri (E. chevalieri) (n = 62), A. montevidensis (E. amstelodami) (n = 34), A. niger (n = 21), A. tamari (n = 15), A. ruber (E. rubrum) (n = 15), A. proliferans (n = 14), and A. luchuensis (n = 14); 25 species were identified from 533 Aspergillus strains. Aspergillus strains were mainly found during the high temperature fermentation period in the later steps of Meju fermentation.

Four Unrecorded Aspergillus Species from the Rhizosphere Soil in South Korea

  • The genus Aspergillus is commonly isolated from various marine and terrestrial environments; however, only a few species have been studied in rhizosphere soil. As part of the Korean indigenous fungal excavation project, we investigated fungal diversity from rhizosphere soil, focusing on Aspergillus species. A total of 13 strains were isolated from the rhizosphere soil of three different plants. Based on phylogenetic analysis of β-tubulin and calmodulin and morphological characteristics, we identified five Aspergillus species. A. calidoustus and A. pseudodeflectus were commonly isolated from the rhizosphere soil. Four species were confirmed as unrecorded species in Korea: A. calidoustus, A. dimorphicus, A. germanicus, and A. pseudodeflecuts. The detailed morphological descriptions of these unrecorded species are provided.

Draft Genome Sequences of Three Airborne Aspergilli Series Versicolores

  • The Aspergilli of the section Nidulantes series Versicolores are among the most recurrent molds in indoor environments. These species cause damage to the quality of air. Indeed, they are responsible for allergies, aggravation of asthma and can even cause infections in immunocompromised patients. Molds belonging to the Versicolores series also produce sterigmatocystin, a mycotoxin classified as potential human carcinogen by the International Agency for Research on Cancer (group 2B). Here, we provide for the first time the genome of three species of the series Versicolores: Aspergillus creber, Aspergillus jensenii and Aspergillus protuberus which are the most abundant species of this series in bioaerosols. The genomes of these three species could be assembled with a percentage of completeness of 97.02%, 96.21% and 95.35% for Aspergillus creber, A. jensenii and A. protuberus respectively. These data will allow to study the genes and gene clusters responsible for the expression of virulence factors, the biosynthesis of mycotoxins and the proliferation of these ubiquitous and recurrent molds.

  • 1. Cassava 전분(澱粉)을 이용(利用)하여 곰팡이를 $28^{\circ}C$에서 96시간(時間) 액체정치(液體定置) 배양(培養)한 바 배지중(培地中)의 Cassava 전분함량(澱粉含量)이 6%, 72시간(時間)일 때에 균체량(菌體量)이 가장 많았다. 2. Cassava 전분(澱粉)이 6% 함유(含有)된 배지(培地)에서 곰팡이를 배양(培養)하여 얻은 균체단백질량(菌體蛋白質量)은 Rhizopus는 48시간(時間) 배양(培養)하였을때 가장 많았고, Aspergillus niger 및 Aspergillus fumigatus에서는 72시간(時間) 배양(培養)했을 때 균체단백질량(菌體蛋白質量)이 많았다. 3. Cassava 전분량(澱粉量)을 4%, 6% 및 8%되게 변화(變化)시킨 배지(培地)에서 분리균(分離菌)을 접종(接種)하여 96시간(時間) 배양(培養)하면서 24시간(時間)마다 배양여액(培養濾液) 중(中)의 m-amylase의 활성(活性)을 측정(測定)한 바 8% 농도(濃度)에서 Aspergillus niger 및 Aspergillus fumigatus는 72시간(時間)일 때 높은 D.P 값을 얻었고. Rhizopus 는 144시간(時間)일 때 D.P 값이 높았다.
