Bioreporter bacteria, such as recombinant bioluminescent bacteria, have been used for the detection of specific compounds in complex environmental media. In this study, optimum conditions for the preparation and application of deep-freezed and Iyophilized recombinant bioluminescent strain KG1206 were investigated for the future application on contaminated environmental sites. Genetically engineered microorganism, Pseudomonas putida mt-2 KG1206, contains TOL plasmid and the plasmid inserted $P_{m}$, promoter on the upper part of lux gone in vector pUCD615, and m-toluate and benzoate are considered direct inducers for bioluminescence. Optimum conditions determined for the preparation and application of the deep-freezed and lyophilized strain were followings: cryoprotective agent (24% sucrose), lyophilization time (12 hrs), strain concentration ($OD_{600}=0.6$), reconstitution for freezed strain (quick reconstitution at $35^{\circ}C$), reconstitution for lyophilized strain ($3{\sim}6$ hrs exposure on LB medium), carrying conditions (keep at $20^{\circ}C$ after reconstitution). These results demonstrate the feasibility of deep-freezed or lyophilized state of genetically engineered bioluminescent strain for environmental usage.

Oil leaked from underwound storage tanks and leachate from sanitary landfills have been known as contaminant sources of the high-quality groundwater resources. The mobility of contaminants in the aquifer largely depends on the groundwater flow and the determination of associated hydraulic parameters is essential for a proper remediation of contaminated grnundwater. This study aimed at determining an optimum set of hydraulic parameters for an unconfined sandy aquifer of a laboratory scale through comparison of various methods. Results showed that the specific yield obtained from gravity drainage experiment was an average of 0.20 with minor variations in aquifer depths. and the permeabilities obtained from Dupuit approximation and slug test gave similar values of 5.33 cm/min and 5.85 cm/min but the constant head method gave 0.17 cm/min, which is much ion than the other methods. This experimental evidence reveals that the permeability of the unconfined sandy aquifer could be accurately determined by Dupuit assumption or slug tut rather than by constant head method conducted for a disturbed separate soil column.

In this study, column tests using relatively uniform Jumunjin sand media were conducted to evaluate the feasibility of calcium polysulfide (CaSx, CPS) in removing high concentration of Zn2+ in groundwater. The injected CPS solution reacted rapidly with Zn2+ in artificial groundwater and effectively reduced Zn2+ by more than 99% through metal sulfide precipitation. Since the density (d = 1.27 g/cm3 ) of CPS solution was greater than that of water, CPS solution settled down rapidly while capturing Zn2+ and formed stable CPS layer similar to dense nonaqueous phase liquid. Mass balance analysis on Zn2+ in CPS solution suggested that CPS solution effectively reacted with Zn2+ to form metal sulfide precipitates except for high groundwater seepage velocity of 400 cm/d. With greater groundwater seepage velocity, injected CPS did not completely dissolve at the CPS-water interface, but a partially-misible CPS layer continuously moved and reacted with Zn2++ in the direction of groundwater flow. Since hydraulic conductivity (Kh) decreased slightly due to the generated metal precipitates in the inter-pores of media, injection of CPS solution should be optimized to prevent clogging. As evidenced by both XRF and SEM/EDS results, ZnS precipitates were clearly observed through the reaction between the CPS solution and Zn2+. Further study is warranted to evaluate the feasibility of CPS to remove high-concentration heavy metalcontaminated groundwater in complex and heterogeneous media.

To investigate differences in lead content in soils and paddy rices, affected by air pollutants from the Janghang Smelter, soil samples at the different directions, distances, and depths, surface(0-15cm depth) and subsurface(15-30cm depth) in 1982 and 1990, and rice plants at soil sampling sites in 1990 were separately collected at the Janghang Smelter area affected by combustible waste gas from the smelter chimney. Soil samples were extracted with $4M-HNO_3$ and plant samples were digested with mixture of $HNO_3$ and $HCIO_4$ for analyzing by atomic absorption spectrophotometry. The Pb contents in soils ranged from 10.3 to 644.8mg $kg^{-1}$. The average content in 1990 was higher than that in 1982. The Pb content in soils at sites nearer the center of the smelter was higher than that at sites farther from the center. The highest lead content was found at the east direction, and was low in order of east>north-north east>north east > north. The variation of Pb level in soils at east sites was more considerable than other directions. The Pb level in surface soils was higher than that in subsurface soils. The contaminated radius of Pb was until 3km all at east, north-north east and north east. A significant correlation was found between Pb content in surface soils of 1982 and that in surface soils of 1990, between Pb content in soils and contents of Cd and Zn in soils, and between Pb content in soils and soil properties as organic matter, available silicate, cation exchange capacity, exchangeable $Ca^{++}$, $Mg^{++}$ and $Na^+$. Pb content in brown rice was low in the panicle axis, and brown rice, and Pb content in stem was 3.26 times of that in brown rice. Pb content in brown rice ranged from 2.2 to 9.0 mg $kg^{-1}$.

The objective of this study is to evaluate the effects of changes in soil temperature on biodegradation rate of diesel compounds from a field pilot test using hot air injection process. Total remediation time was estimated from in-situ biodegradation rate and temperature for optimum biodegradation. All tests were conducted by measuring in-situ respiration rates every about 10 days on highly contaminated area where an accidental diesel release occurred. The applied remediation methods were hot air injection/extraction process to volatilize and extract diesel compounds followed by a bioremediation process to degrade residual diesels in soils. Oxygen consumption rate varied from 2.2 to 46.3%/day in the range of 26 to $60^{\circ}C$, and maximum $O_2$ consumption rate was observed at $32.0^{\circ}C$. Zero-order biodegradation rate estimated on the basis of oxygen consumption rates varied from 6.5 to 21.3 mg/kg-day, and the maximum biodegradation rate was observed at $32^{\circ}C$ as well. In other temperature range, the values were in the decreasing trend. The first-order kinetic constants (k) estimated from in-situ respiration rates measured periodically were 0.0027, 0.0013, and $0.0006d^{-1}$ at 32.8, 41.1, and $52.7^{\circ}C$, respectively. The estimated remediation time was from 2 to 9 years, provided that final TPH concentration in soils was set to 870 mg/kg.
The objective of this study was to develop an effective separation and quantification method for kerosene and diesel in a mixed petroleum fuel (gasoline, kerosene, and diesel) contaminated environmental samples. This investigation was directed to prove the hypothesis that if the source of petroleum fuels were identical, the peak-area ratios of a reference n-alkane to other n-alkane peaks should be a constant even at the different concentrations. In addition, experimental recovery rates were determined to select the reference peaks of kerosene and diesel for peak area ratio measurements. The experimental results showed that the peak area ratios were constant among the samples having different concentrations when the ratios were calculated from areas of $C_{l3}$, $C_{l4}$, and $C_{15}$ peaks for kerosene and $C_{l6}$ and $C_{l7}$ peak for diesel as reference n-alkane peaks. The recovery rates were evaluated by comparing the relative peak area ratios of each reference peaks after making pairs of the kerosene and diesel reference peaks in the samples contained a known amount of gasoline, kerosene, and diesel. The recovery rates(%) Were 107.0$_{{\pm}20.6}$/86.6/ sub $\pm$15.9/ for kerosene- $C_{13}$/diesel- $C_{16}$, 99.6$\pm$$_{17.2}$/86.6$_{{\pm}15.9}$ for kerosene- $C_{14}$/diesel- $C_{16}$, 73.9/$\pm$14.4//86.6$_{{\pm}sub 15.9}$ for kerosene- $C_{15}$ /diesel- $C_{16}$, 109.4$_{{pm}0.8}$/75.9$_{{pm}4.7}$ for kerosene- $C_{13}$/diesel- $C_{17}$, 107.4$_{{pm}7.9}$/75.9$_{{pm}4.7}$ for kerosene- $C_{14}$/diesel- $C_{17}$, and 95.7$_{{pm}4.6}$ /75.9/$\pm$14.6//75.9$_{{pm$}4.7}$ for kerosene- $C_{15}$ /diesel- $C_{17.}$ The above experimental results confirm that all of the reference peak pairs of kerosene and diesel are applicable to the quantitative analysis for the mixed fuel contaminated samples, but the kerosene- $C_{15}$ /diesel- $C_{l7}$ peaks are recommended since the pair has a lower standard deviation than the other pairs.s..s.s.s..s..s.s.s.s.s.
Burkholderia sp. D5, a polyaromatic hydrocarbons(PAHs)-degrading bacterium, was isolated from oil-contaminated soil. The bacterium could utilize phenanthrene (Phe) as a sole carbon source but could not use pyrene (Pyr). However, the strain could degrade Pyr when a cosubstrate such as yeast extract (YE) was supplemented. The PAH degradation rate of the bacterium was enhanced by the addition of other organic materials such as YE, peptone and glucose. YE was a particularly effective additive in stimulating cell growth as well as PAH degradation. When 1 g-YE/L was supplemented into the basal salt medium (BSM) with 215 mg-Phe/L, the specific growth rate (0.28 h-1) and Phe-degrading rate (29.30 μmol/L/h) were enhanced approximately ten and two times more than those obtained in the BSM with 215 mg-Phe/L, respectively. Through kinetic analysis, the maximum specific growth rate (μmax) and PAH degrading rate (Vmax) for Phe were obtained as 0.34/h and 289 ${\mu}mol$/L/h, respectively. Also, μmax and Vmax for Pyr were 0.27 h-1 and 50 ${\mu}mol$/L/h, respectively. The degradation rates for each Phe (2.20 μmol/L/h) and Pyr (2.18 μmol/L/h) were lower in mixture substrates than in a single substrate (29.30 ${\mu}mol$/L/h and 9.58 ${\mu}mol$/L/h, respectively). Burkholderia sp. D5 can degrade Phe and Pyr contained in soil, and the PAH degradation rates in soil were 20.03 ${\mu}mol$/L/h for Phe and 1.09 ${\mu}mol$/L/h for Pyr.

One hundred forty four bacterial colonies which were able to degrade crude oil were isolated from soil samples that were contaminated with oil in Daejeon area. Among them, one bacterial strain was selected for this study based on its emulsification activity, growth rate and surface tension activity, and this selected bacterial strain was identified as Pseudomonas sp. HN37 through physiological- biochemical tests and analysis of its 16S rRNA sequence. Pseudomonas sp. HN37 utilize the several aliphatic hydrocarbons, 3,5-dinitrosalicylic acid and 2,4-dichlorophooxyacetic acid as a sole carbon source. And this bacterial strain showed a high resistance to antibiotics such as ampicillin and chloramphenicol, as well as heavy metals such as Ba, Cr, Li and Mn. Also, it was found that the optimal pH and temperature for the cell growth, surface tension, and emulsification activity of Pseudomonas sp. HN37 were pH 6.0-9.0 and $30^{\circ}C$, respectively. The emulsification and surface tension activity was reached the maximum to 1% (V/V) crude oil and 1% (W/V) NaCl concentration. The surface tension of the culture broth was decreased from 62 to 27 dyne/cm after fifteen hours of inoculation in LB media.
To improvement the swelling characteristics of the existing cutoff wall against the moisture, the permeability of the sand, calcium bentonite and solidifier mixture according to the contact with trichloroethylene (TCE) was evaluated. Characteristics analysis and the permeability test of the research materials were performed. The permeability was decreased as the mixing ratio of the calcium bentonite was increased and it was increased as the mixing ratio of the solidifier was increased. In conclusion, when mixing 15% of calcium bentonite and more than 30% of solidifier, the permeability coefficient in the underground water movement was analyzed as more than α × 10-4 cm/sec showing that it does not block the underground water movement. In addition, as the permeability coefficient of mixtures after TCE reaction was analyzed as less than α ×10-7 cm/sec, it satisfied the condition of blocking layer (less than 1.0 × 10-6 cm/sec). Therefore, the calcium bentonite and solidifier can be utilized as barrier that showing the characteristic of percolation ability conversion in soil and underground water contaminated with TCE.

A phenanthrene-degrading bacterium HS362, which is capable of using phenanthrene as a sole carbon and energy source, was isolated from oil contaminated soil. This strain is a gram negative, rod shaped organism that is most closely related to Sphingomonas paucimobilis based on biochemical tests, and belongs to the genus Sphingomonas based on fatty acids analysis. It exhibited more than $99.2{\%}$ nucleotide sequence similarity of 16S rDNA to that of Sphingomonas CF06. Thus, we named this strain as Sphingomonas sp. HS362. It degraded $98{\%}$ of phenanthrene after 10 days of incubation when phenanthrene was added at 500 ppm and $30{\%}$ even when phenanthrene was added at 3000 ppm. Sphingomonas sp. HS362 could also degrade low molecular weight PAHs(Polycyclic aromatic hydrocarbons) such as indole and naphthalene, but was unable to degrade high molecular weight PAHs such as pyrene and fluoranthene. The optimum temperature and pH for phenanthrene degradation were $30^{\circ}C$ and $4{\~}8$, respectively. Sphingomonas sp. HS362 could degrade phenanthrene effectively in the concentration range of NaCl of up to $1{\%}$. Its phenanhrene degrading ability was enhanced by preculture, suggesting the possibility of induction of phenanthrene degrading enzymes. Starch and surfactants such as SDS, Tween 85, and Triton X-100 were also able to enhance phenanthrene degradation by Sphingomonas sp. HS362. It carries five plasmids and one of them, plasmid p4, is considered to be involved in the degradation of phenanthrene according to the plasmid curing experiment by growing at $42^{\circ}C$.
