This study examined the possibility on the dietary value and cryopreservation of the marine microalga, Isochrysis galbana. Four cryoprotectants, dimethyl sulfoxide (DMSO), ethylene glycol (EG), glycerol (Gly) and 1.2-propanedial (PD) were tested at the concentrations of 1.0, 1.5, 2.0, 2.5 and 3.0 M respectively. The highest survival rates were obtained with 1.5 and 2.0 M of four cryoprotectants yielding a survival rate of 80%. Cell concentration of $30\;{\times}\;10^4\;cell/ml$ at the initial point of the experiment was increased to $365\;{\times}\;10^4\;cell/ml$ in 1.5 M of dimethyl sulfoxide, $298\;{\times}\;10^4\;cell/ml$ in 1.5 M of ethylene glycol, $512\;{\times}\;10^4\;cell/ml$ in 1.5 M of glycerol, and $385\;{\times}\;10^4\;cell/ml$ in 1.5 M of 1.2-propanedial after five days, respectively. In dietary value experiment, survival rate and growth were not significantly different.
The higher sugar and lower starch in super sweet corn may be due to modified endosperm genes sh, bt series, but its seeds have major limiting factors causing low germination and low seedling vigor, This study was conducted to determine what measurable kernel characteristics during the grain tiling period might be more useful as a guide to optimize harvest date for good seed quality in hybrid super sweet corn production. Artificial crossing in super sweet corn hybrid (Chodangok 1) was made on the same day, and ears were harvested from 18 days to 53 days after pollination at weekly intervals. Kernel weight, moisture content, hardness, endosperm rate, seedling height and storage nutrients such as sugars, protein, starch, and germination rate were measured for the kernels at each harvest. Super sweet corn hybrid, Chodangok 1 presented satisfactory germination rate above 70% when harvested on 39 to 46 days after pollination. Its storage nutrients at that time were 23.7-24.2% in starch content 5.5-5.9% in total sugars, 38.9-46.6% in kernel moisture, and 62.7-64.2% in endosperm rate. Germination rate was extremely high when harvested on 39 days after pollination. The black layer of Chodangok 1 could not be used as an indicator for seed harvest. These results suggested that optimum harvest date seems to be 39 days, and kernel moisture and starch content could be used as indications of kernel maturity in deciding when to harvest fer good seed quality in super sweet corn.
The effects of seed-associated or free linseed oil on fermentation characteristics and long-chain unsaturated fatty acids composition, especially the formation of conjugated linoleic acid (CLA) and octadecenoic acid (trans-11 $C_{18:1}$, $t-C_{18:1}$) by mixed ruminal bacteria were examined in vitro. Concentrate (1% of culture solution, w/v, as-fed basis) with ground linseed (0.6% of culture solution, w/v, DM basis) or linseed oil as absorbed onto ground alfalfa hay was added to 600 ml mixed solution consisting of strained rumen fluid and artificial saliva at the ratio of 1:1 in a glass culture jar. The culture jar was covered with a glass lid with stirrer, and placed into a water-bath ($39^{\circ}C$) and incubated anaerobically up to 24 h. Seed-associated or free linseed oil did not significantly affect the pH and ammonia concentration in the culture solution. Molar percent of acetate tended to increase while that of propionate decreased with the addition of free oil treatment throughout the incubation. Differences in bacterial number were relatively small, regardless of the form of supplements. Decreasing trends in the compositions of linoleic acid ($C_{18:2}$) and linolenic acid ($C_{18:3}$) but increasing trends of stearic acid ($C_{18:0}$), $t-C_{18:1}$ and CLA compositions were found from culture contents up to 12h incubation when incubated with both ground linseed and linseed oil. The compositions of $C_{18:0}$, $C_{18:2}$ and $C_{18:3}$ were greater but those of oleic acid ($C_{18:1}$), $t-C_{18:1}$ and CLA were smaller in a culture solution containing ground linseed than those containing linseed oil. The ratio of $t-C_{18:1}$ to CLA was lower in the culture solutions containing linseed oil up to 12h incubations as compared to those containing ground linseed.
A nutritional demand of oyster, Crassostrea gigas larva as part of research for improving of utilization of microalgae being used for the artificial oyster seed production. The change of body growth and biochemical compositions of larvae were investigated during larvae rearing in hatchery. The larvae were cultured in 60 M/T tank and fed mixture 6 different phytoplankton species, Isochrysis galbana (30%), Cheatoceros gracilis (20%), Pavlova lutheri (20%), Phaeodactylum triconutum (10%), Nannochryis oculata (10%) and Tetraselmis tetrathele (10%). The initial feeding amount was $0.3{\times}10^4cells/mL$ at three times a day to D-shaped larva and the feeding amount had been increased 30% gradually every two day since the larvae were raising. The larvae were developed from D shape to pediveliger stage for 12 days. The daily growth of shell length and hight were $5.8{\sim}30.8\;{\mu}m$ and $8.7{\sim}31.4\;{\mu}m$, respectively and weight gains were changed from D shape to pediveliger as follow: wet weight was $0.52{\sim}15.0\;{\mu}g/larva$, dry weight was $0.2{\sim}6.5\;{\mu}g/larva$, and ash free dry weight was $0.1{\sim}8.5\;{\mu}g/larva$. The larvae growth pattern shown a logarithmic phase from D shape to umbone stage but after that stage shown a exponential growth aspect. The crude protein, crude lipid and nitrogen free extract (NFE) of larvae during rearing periods were analyzed as $6.1{\sim}10.6%$, $0.6{\sim}1.1%$ and 1.0-2.7%, respectively. And the total amino acid contents of the larvae during rearing periods were in order as glutamic acid $1.26{\sim}2.24%$, aspartic acid $0.97{\sim}1.70%$, and methionine $0.12{\sim}0.33%$. Of the total fatty acid in the analyzed larvae, the saturated fatty acid (SSAFA) was decreased from 54.3% (D shaped larvae) to 17.1 % (pediveliger) as larvae development but the total mono-unsaturated fatty acid (${\Sigma}MOFA$) and Poly-unsaturated fatty acid (${\Sigma}PUFA$) were increased from 29.9% and 7.8% to 40.6% and 45.6%, respectively. By the way the each fatty acid of the larvae were composed of palmitic acid $9.89{\sim}36.95%$, oleic acid $12.17{\sim}32.29%$, linoleic acid $1.96{\sim}33.55%$, EPA $2.17{\sim}11.58%$ and DHA $1.95{\sim}4.51%$. As a result of this study, the larvae of oyster were demanded a various nutrients for healthy growth and the feeding control, expecially after umbone stage larvae are a rapidly growing time, is very important for success of artificial seed production.
This study was conducted to find the factors affecting somatic embryogenesis in suspension culture of Rehmania glutinosa and investigate the possibility of artificial seed production by encapsulation of somatic embryos. Linsmeier-Skoog medium was appeared as proper for somatic embryogenesis. Sucrose with $3{\sim}5%$ as carbon sources was good for somatic embryogenesis, and both ammonium and nitrate nitrogen were necesary for normal somatic embryo production. BA with NAA or kinetin with NAA were better than the use of cytokinin alone for both somatic embryogenesis and numbers of somatic embryos. $AgNO_3$ as protectant for vitrification of seedlings in vitro culture had no harmful effect on somatic embryos. Sphericity of encapsulated seeds was good at 3% gel of sodium alginate but germination was better at 2.5% sodium alginate level. Artificial seeds were germinated and developed normal shoots and roots under in vitro condition.
This study examines the effects on fertilization rate (FR), hatching rate (HR), and normal individual rate after artificial fertilization using frozen thawed sperm according to the cryoprotectant (DMSO) concentration and the period of cryopreserved sperm of longtooth grouper, Epinephelus bruneus. Performing artificial fertilization using frozen-thawed sperm, after freezing the sperm at different DMSO concentration of 5.0%, 7.5%, 10.0% respectively, FR were (DMSO 5.0%: $99.5{\pm}0.8%$, DMSO 7.5%: $99.5{\pm}0.7%$, and DMSO 10.0%: $99.6{\pm}0.6%$). The results are not significantly different from the control fresh sperm (100%). HR also (DMSO 5.0%: $96.2{\pm}2.3%$, DMSO 7.5%: $95.3{\pm}3.6%$, 10.0%: $96.6{\pm}1.8%$) were not significantly different in each group. The normal individual rate after hatching using with control fresh sperm ($98.4%{\pm}0.5$) and DMSO concentration level of 5.0% ($97.8{\pm}0.1%$) were not significantly different. However, with 7.5% ($97.2{\pm}0.6%$) and 10.0% DMSO concentrations ($95.9{\pm}0.2%$) are lower than the normal individual rate after hatching observed in the control and 5.0% DMSO. Performing artificial fertilization using frozen-thawed sperm at different frozen period (2 days, 2 years, and 3 years), 10% DMSO FR and HR of 3 years (FR; $66.8{\pm}1.8%$, HR: $82.0{\pm}12.9%$) and 2 years (FR; $78.5{\pm}14.8%$, HR: $79.3{\pm}0.6%$) cryopreserved sperm were lower than control (FR; 100%, HR: $91.1{\pm}3.6%$) and 2 days cryopreserved sperm (FR; $99.6{\pm}0.6%$, HR: $96.6{\pm}1.8%$). These results suggest suitable DMSO concentration ranges of cryopreservation sperm for E. bruneus is 5 to 10% and with 2 to 3 years cryopreservation period, cryopreservation sperm can be useful for seed production.

Artificial pollination (AP) and gibberellin A4+7 plus benzyladenine (promalin) were applied alone and together. AP was applied at 10% flowering time with 'Senshu' pollen (Malus domestica cv. Senshu). 12mg/L promalin was applied at 0, 10 and 20 days after falling of central flowers, respectively. In promalin treatment with or without AP application methods, fruit length, weight and length/diameter (L/D) were higher than those of control and AP. However, in AP and AP+ promalin application, the number of seeds and seed weight were higher than those of control and promalin. In AP+ promalin treatment, 78.6% fruits showed their uniform fruit shape and so significantly enhanced fruit uniformity compared to other treatments. Also cortex and core thickness of fruits were greater at the apex than that of other treatment. Fruit with L/D ratio over 0.87 were highly produced by applications of AP+promalin and promalin than control and AP only. 'Fuji' apples in good shape can be produced by using of AP + promalin together.

Feeding regimens was investigated for the growth and survival rate of juvenile Maroon clownfish (Premnas biaculeatus) in order to establish a seed production technique for this fish. Three types of feeding regimens were used, and growth and survival were highest under the following regimen: rotifer from day 0, Artemia from day 4, copepod from day 9 and artificial food from day 15, respectively (P<0.05). The average body lengths of the larvae were $6.76{\pm}0.55\;mm$, $7.63{\pm}0.50\;mm$ and $7.33{\pm}0.43\;mm$ in the Nannochloropsis oculata, Isochrysis galbana and control treatments conducted with 3 larvae/L, respectively (P<0.05). There were significant differences in larval growth between the treatments and controls conducted with 6 larvae/L and 9 larvae/L, respectively (P<0.05) but there was no significant difference between the N. oculata and I. galbana treatments (P>0.05). There were no significant differences in larval growth between the treatments at densities of 3-9 larvae/L (P>0.05). At highest density level, the larvae reared in I. galbana had the highest survival rate whereas survival was lowest in the control.
Sargassum fulvellum (Turner) C. Agardh, an edible brown alga is farmed commercially by sexual reproduction and vegetative regeneration. Investigations were made on the phenology, abundance and maturity of reproductive structures in mature fronds, egg release and young germling development under different light conditions (20, 50, 80 and $100{\mu}mol/m^2/s$) and temperatures (5, 10, 15, 20 and $25^{\circ}C$). Monthly sampling was carried out by SCUBA diving at Chungsando on the southwestern coast of Korea from September 2002 to August 2003. The Maximum length of thalli was $104.6{\pm}20.7{\cal}cm$ in March 2003 when the water temperature was $9.0^{\circ}C$ and minimum was $0.8{\pm}0.5{\cal}cm$ in June when the water temperature was $19.5^{\circ}C$. Receptacle formation was observed from February to April. The peak period of egg release for this alga was in April when the water temperature was about $10^{\circ}C$ in nature. In the culture regimes of temperature and irradiance, the egg release of the excised female receptacle was highly affected by temperature. The maximum rate of egg release was $96.7{\pm}5.8{\%}$ under $20^{\circ}C$ and $80{\mu}mol/m^2/s$. The maximum length of young germlings was $3.9{\pm}0.2{\cal}mm$ after 35 days culture under $15^{\circ}C$ and $80{\mu}mol/m^2/s$.

This study aimed to find an optimal diluent and cryoprotective agent (CPA) during cryopreservation of common Korean bitterling Acheilognathus signifier sperm. The bitterling is an endangered species in Korea, and this study will enable conservation and further technical development of artificial seed production. We tested the effects of cryopreservation and toxicity by the type of diluent and CPA. The optimal combination of diluent and CPA for cryopreservation was 300 mM glucose+10% methanol, resulting in a survival rate and sperm activity index (SAI) of $96.3{\pm}1.5%$, and $3.0{\pm}0.0$ respectively, and no significant difference compared to fresh sperm. The survival rate and SAI of post-thawed sperm was $9.7{\pm}1.5%$, $0.8{\pm}0.3$ respectively, which was significantly higher than had been achieved with other diluents and CPAs.
