Plant Growth-promoting Ability by the Newly Isolated Bacterium Bacillus aerius MH1RS1 from Indigenous Plant in Sand Dune (해안 사구에서 서식하는 토착식물로부터 분리된 근권미생물 Bacillus aerius MH1RS1의 식물성장 촉진 능력 연구)

  • Coastal sand dunes have been seriously damaged caused by the development thoughtless for the environment and coastal erosion and destruction due to artificial structures like coast roads and breakwater. Hereupon, in this study we made a library of rhizobacteria that have the plant growth-promoting ability for plant rhizosphere of indigenous plants inhabiting in a coastal sand dune as well as the strong tolerance to salt, and evaluated the plant growth-promoting ability of these strains. Furthermore, we evaluated the effect of rhizobacteria on the growth rate of saline tolerant plants in sandy soil; selected out the most useful micro-organism for the restoration of a damaged sand dune. The effect of inoculation of strains selected from the first experiment on the growth of Peucedanum japonicum and Arundo donaxes planted in a coastal sand dune was evaluated. As a result, Bacillus aerius MH1RS1 had plant growth promoting activities: indole acetic acid (IAA) production, siderophores and 1-aminocyclopropane-1-carboxylate deaminase (ACC deaminase) activity, and also had a salinity tolerance. Also, in case of Peucedanum japonicum, the length of stems and weights of roots were enhanced by the inoculation of B. aerius MH1RS1. Fresh weights of stems and weights of roots in experimental group were, in particular, increased by 25% comparing with the control group. For an Arundo donax in experimental group, plant length increased by 18%, and weight of roots by 20% which is significant.

Fatty Acid Composition of Concentrated Phytoplanktons by Cold Storage and Their Effects on the Larval Survival of Pacific Oyster, Crassostrea gigas (냉장보관된 농축 먹이생물의 지방산 조성 변화와 농축먹이로 사육한 굴 유생의 생존율)

  • Cold storage of concentrated food phytoplanktons is a useful technique in supplying food organisms for artificial shellfish seed. One month after preservation at $4^{\circ}C$, we have measured survival rate of the concentrated food phytoplanktons, Pavlova lutheli, Isochrysis galbana, Isochrysis aff, galbana and Chaetoceros calcitrans. Thereafter we determined survival rate of oyster lavae fed fresh and concentrated diets and fatty acid compositions of the fresh and concentrated food phytoplanktons. Survival rate of concentrated planktons ranged from $23\%$ to $31\%$ after one month at $4^{\circ}C$. The survival rate of oyster larvae fed cold stored food appeared generally higher than those fed fresh harvested food. Especially, the highest survival rate were found in the larvae fed cold stored concentrated I. aff. galbana. EPA and DHA increased after cold storage and the highest level of DHA was detected in I. aff. galbana. As DHA can role as an important factor in determing nutritional value, it would be better to use concentrated I aff, gaibana kept in cold refrigerator for oyster seed production.

First Report of Bacterial Root Rot Caused by Serratia plymuthica on Panax ginseng (Serratia plymuthica에 의한 인삼 세균뿌리썩음병 발생)

  • In August of 2011, a wilting disease of ginseng was observed at Bongwha, Gyeongbuk province, Korea. Affected plants initially show withering symptoms on leaves of ginseng. As the disease progresses, withering leaves spread downward, eventually encompassing the whole plant. Leaves lose vigor but remain pale green. Symptoms of roots were brown, and soft rots characterized by moist and watery decay of the whole ginseng root, which initiated as small brown, water-soaked lesions of hairy roots and enlarged to the entire roots. The causal organism isolated from the infected roots was identified as Serratia plymuthica based on its physiological and biochemical characteristics, by cellular fatty acid composition (GC-FAME), the utilization of carbon sources (BioLog System), and 16S rRNA sequence of the isolated bacterium were 99% homologous to those of Serratia plymuthica strains. Artificial inoculation of the bacterium produced the same brown or soft rot symptoms on the ginseng roots, from which the same bacterium was isolated. This is the first report of bacterial root rot caused by the Serratia plymuthica in ginseng in Korea. Serratia plymuthica has been used as antagonistic microorganism for biological control on several crop plants. But it was proved pathogen of ginseng at humid condition in this study.

Effects of Light Quality Using LEDs on Expression Patterns in Brassica rapa Seedlings (LED 광원의 다양한 광질이 배추 유묘의 유전자 발현에 미치는 영향)

  • Light with two faces, beneficial and harmful effects is an important signal for every living cell. Optimal adaptation to light environment enhances the fitness of an organism and survival in nature. Understandings of light quality and plant growth provide with the economical guides for artificial light sources like LEDs. Compared with those under white light, the 1 week seedlings of Chinese cabbage (Brassica rapa) under monochromic red and blue light showed normal development and growth. In contrast to extremely long and etiolated hypocotyls of the seedlings under dark, those under far-red etiolated were extremely short. Based on the microarray analysis, blue light induced the vigorous development and growth and two fold changes of transcripts than red light condition. To have insight of gene products under different light qualities conditions, GO term enrichments were calculated and each gene according to their GO terms were categorized. The blue and red lights affected the expressions of genes related to biological process. Especially, the genes related to metabolic process and developmental process and plastid and chloroplast in the cellular component category were induced under blue light. This study provided the molecular biological evidence for various light qualities on the growing process of B. rapa.
